Effects of neuromuscular electrical stimulation on voluntary muscle activation and peripheral muscle contractility following short-term bed rest.

Disuse induces a disproportionate loss of muscle force compared with muscle mass, with unclear effects on voluntary muscle activation (VA) and peripheral contractility. Furthermore, the effect of neuromuscular electrical stimulation (NMES) as a disuse countermeasure remains uncertain. We investigated the effects of NMES during bed rest on neuromechanical function to improve our understanding of the mechanisms underlying disuse-induced reductions in muscular force. Young (n = 16, 25 years old) and old (n = 16, 71 years old) adults underwent 5 days of bed rest. One leg received NMES (3 × 30 min/

Impact of peripheral conditioning on reperfusion injury following primary percutaneous coronary intervention in diabetic and non-diabetic STEMI patients.

BACKGROUND: Peripheral conditioning induces transient ischemia, promoting antioxidant production in ischemia-affected tissues, which helps reduce heart reperfusion injury in ST-elevation myocardial infarction (STEMI) patients. This study compares troponin and creatine kinase-MB (CK-MB) levels among STEMI patients with and without remote conditioning. METHODS: This study included 160 patients treated for STEMI at a tertiary care centre. The study protocol involved cyclic inflation and deflation of a blood pressure cuff on the brachial region in four cycles of 5 min each. Markers of myocardial

D- and L-Amino Acid Blood Concentrations Are Affected in Children With Duchenne Muscular Dystrophy.

Duchenne muscular dystrophy (DMD) is an X-linked disease caused by the absence of functional dystrophin in the muscle cells. Recent untargeted metabolomics studies identified amino acid metabolism alterations as biochemical pathways potentially involved in DMD pathogenesis. Here, in a well-characterised cohort of DMD children and paediatric controls, we investigated by high-performance liquid chromatography (HPLC) the serum profile of a selected pool of amino acids in D- and L-configuration, including L-glutamate, L-glutamine, glycine, L-aspartate, D-aspartate, L-asparagine, L-serine, and D-se

Carvacrol alleviates diabetic cardiomyopathy in rats: Targeting Cuproptosis pathway.

Diabetic cardiomyopathy (DCM) is a myocardial injury induced by chronic hyperglycemia. Recent literature has reported that hyperglycemia causes cardiac copper overload, triggering copper-induced cell death (cuproptosis). Copper leads to the dysregulation of ferredoxin-1 (FDX-1) and lipoic acid synthase (LIAS), two enzymes essential for the lipoylation process in the Krebs cycle, and are considered key markers of cuproptosis. We aim to explore the protective effects of carvacrol (CAR) against DCM by targeting cuproptosis. Diabetes was induced by a high-fat diet/streptozotocin (35 mg/kg) model.

Development and validation of an interpretable clinical scoring model to monitor the progression of preclinical Alzheimer's disease.

BACKGROUND: Monitoring the progression of preclinical Alzheimer's disease (AD) is challenging due to the absence of obvious cognitive impairment, yet it's crucial for determining the optimal timing for disease-modifying treatments. METHODS: This prognostic study used data from the Anti-Amyloid Treatment in Asymptomatic Alzheimer’s Disease (A4) study. The development cohort included 412 participants from the placebo arm, and external validation was performed using 342 participants from the treatment arm. All participants had baseline brain amyloid-beta (Aβ) positron emission tomography (PET) st

Magnesium-assisted hydrogen improves isoproterenol-induced heart failure.

Heart failure (HF) is a leading cause of mortality among patients with cardiovascular disease and is often associated with myocardial apoptosis and endoplasmic reticulum stress (ERS). While hydrogen has demonstrated potential in reducing oxidative stress and ERS, recent evidence suggests that magnesium may aid in hydrogen release within the body, further enhancing these protective effects. This study aimed to investigate the cardioprotective effects of magnesium in reducing apoptosis and ERS through hydrogen release in a rat model of isoproterenol (ISO)-induced HF. Magnesium was administered o

EMD missense variant causes X-linked isolated dilated cardiomyopathy with myocardial emerin deficiency.

Pathogenic variants in the EMD gene cause X-linked Emery-Dreifuss muscular dystrophy type 1 (EDMD1), typically presenting with joint contractures and skeletal muscle atrophy, followed by atrial arrhythmias, cardiac conduction defects, and atrial dilatation. Although an association with isolated dilated cardiomyopathy (DCM) has been suggested, evidence is currently insufficient to verify the gene-disease association. We investigated the causality of a missense variant, c.23C>G, p.Ser8Trp, in EMD in a large family with a history of DCM and suspected sudden cardiac death (SCD) in males. DCM was d
