Stress granules and tau pathology in Alzheimer's disease: A scoping review of bidirectional interactions, liquid-liquid phase separation, and proteostatic consequences.

BACKGROUND: Alzheimer's disease (AD) is characterized by progressive neurodegeneration driven by amyloid-β (Aβ) plaques and tau neurofibrillary tangles. Stress granules (SGs), dynamic ribonucleoprotein condensates formed under cellular stress have been implicated in several neurodegenerative disorders, but their role in AD pathogenesis remains incompletely understood. METHODS: Following PRISMA-ScR guidelines, we systematically searched PubMed, Embase, Scopus, Web of Science, and Cochrane Library (through February 2026) for peer-reviewed studies investigating SGs in AD models or human tissue.

Peli1 promotes sepsis-induced inflammation and coagulopathy by maintaining lipg stability in macrophages.

Sepsis is marked by dysregulated inflammation and coagulopathy, with macrophages playing a central role. However, the molecular mechanisms linking macrophage dysfunction to sepsis-associated coagulopathy remain unclear. This study investigated the role of the E3 ubiquitin ligase Peli1 in sepsis progression and its downstream mechanism. Public scRNA-seq data from sepsis patients were analyzed to identify Peli1-associated immune signatures. Peli1 expression was validated in monocytes from sepsis patients and in CLP-induced septic mice. Peli1-deficient iBMDMs and myeloid cell-specific Peli1-defic

Mutant neutrophils and NETosis in clonal hematopoiesis: an emerging contributor of thromboinflammation and organ dysfunction.

UNLABELLED: Age-related proliferation of indeterminate capacity hematopoietic stem and progenitor cells bearing somatic mutations, mainly in DNMT3A, TET2, and ASXL1, is referred to as clonal hematopoiesis of indeterminate potential (CHIP). In addition to the well-known premalignant effects, CHIP causes systemic effects and predisposes to cardiovascular disease, thromboembolism, and dysfunction of multiple organs. Mutant neutrophils have been increasingly recognized as contributors to this pathology. These cells exhibit aberrant phenotypes and epigenetic alterations associated with pro-inflamma

Proteomic comparison of hippocampal neurofibrillary tangles in PART, intermediate Alzheimer's disease and advanced Alzheimer's disease.

Alzheimer's disease (AD) is characterised by the intraneuronal aggregation of phosphorylated Tau (pTau) into neurofibrillary tangles and by the extracellular deposition of β-amyloid (Aβ). Tau pathology restricted to the hippocampal formation is frequently observed in the elderly brain in the absence of any Aβ deposition and considered as "primary age-related tauopathy" (PART). Here, we applied an unbiased proteomic approach to determine how concomitant Aβ pathology modifies the neurofibrillary tangle proteome. Neurofibrillary tangles were isolated by dissecting Tau pSer202/pThr205 "AT8" immuno

Presymptomatic pharmacological inhibition of mGluR5 improves survival in a mouse model of prion diseases.

Toxic signaling of oligomeric protein species via binding to the cellular prion protein (PrPC) is implicated in various neurodegenerative diseases, including Alzheimer’s (AD) and Parkinson’s disease (PD). Metabotropic glutamate receptor 5 (mGluR5) has been identified as a PrPC signaling partner, and pharmacological inhibition of mGluR5 was shown to improve cognitive performance and rescue long-term-potentiation (LTP) impairment in both in vitro and in vivo models of AD and PD. Prion diseases are another group of fatal neurodegenerative disorders that are characterized by templated misfolding o

Therapeutic Plasma Exchange in Immune-Mediated Thrombotic Thrombocytopenic Purpura: From Cornerstone to Contextualized Therapy.

Immune-mediated thrombotic thrombocytopenic purpura (iTTP) is a rare, life-threatening thrombotic microangiopathy caused by severe ADAMTS13 deficiency due to anti-ADAMTS13 autoantibodies. The resulting persistence of ultra-large von Willebrand factor (VWF) multimers promotes uncontrolled platelet adhesion and aggregation in the microcirculation, leading to thrombocytopenia, microangiopathic hemolytic anemia, and ischemic organ injury. Therapeutic plasma exchange (TPE) has transformed the prognosis of iTTP by removing circulating autoantibodies and replenishing functional ADAMTS13, and it remai

Nanoemulsion Technology in Oral Drug Delivery: A Path to Enhanced Solubility and Bioavailability.

Nanoemulsions (NEs) are submicron-sized colloidal dispersions (20-500 nm) consisting of oil and aqueous phases stabilized by surfactants and cosurfactants. Despite their thermodynamic instability, NEs maintain kinetic stability, preventing separation and aggregation. This stability distinguishes them from microemulsions, which are thermodynamically stable and formed spontaneously. The emulsification process involves a reduction in Gibbs surface free energy facilitated by emulsifiers that lower interfacial tension, crucial for compensating for the high surface area associated with small droplet

Brain Transcriptome Changes Associated With an Acute Increase of Protein O-GlcNAcylation and Implications for Neurodegenerative Disease.

Enhancing protein O-GlcNAcylation by pharmacological inhibition of the enzyme O-GlcNAcase (OGA) has been considered as a strategy to decrease tau and amyloid-beta phosphorylation, aggregation, and pathology in Alzheimer's disease (AD). There is still more to be learned about the impact of enhancing global protein O-GlcNAcylation, which is important for understanding the potential of using OGA inhibition to treat neurodegenerative diseases. In this study, we investigated the acute effect of pharmacologically increasing O-GlcNAc levels, using the OGA inhibitor Thiamet G (TG), in normal mouse bra
