The Shaggy Dog Story of Enteric Signaling: Serotonin, a Molecular Megillah.

Historically and quantitatively, the enteric site of serotonin (5-HT) storage has primacy over those of any other organ. 5-HT, by the name of "enteramine", was first discovered in the bowel, and the gut produces most of the body's 5-HT. Not only does the bowel secrete 5-HT prodigiously but it also expresses a kaleidoscopic abundance of 5-HT receptors. The larger of two enteric 5-HT stores is mucosal, biosynthetically dependent upon tryptophan hydroxylase1 (TPH1), and located in EC cells. Mechanical stimuli, nutrients, luminal bacteria, and neurotransmitters such as acetylcholine and norepineph

Store-operated Ca2+ entry and Ca2+ responses to hypothalamic releasing hormones in anterior pituitary cells from Orai1-/- and heptaTRPC knockout mice.

Store operated Ca2+ entry (SOCE) is the most important Ca2+ entry pathway in non-excitable cells. However, SOCE can also play a pivotal role in excitable cells such as anterior pituitary (AP) cells. The AP gland contains five different cell types that release six major AP hormones controlling most of the entire endocrine system. AP hormone release is modulated by Ca2+ signals induced by different hypothalamic releasing hormones (HRHs) acting on specific receptors in AP cells. TRH and LHRH both induce Ca2+ release and Ca2+ entry in responsive cells while GHRH and CRH only induce Ca2+ entry. SOC

Chief complaints, diagnoses, and medications prescribed seven weeks post-Katrina in New Orleans.

BACKGROUND: In the aftermath of Hurricane Katrina, widespread flooding devastated the New Orleans healthcare system. Prior studies of post-hurricane healthcare do not consistently offer evidence-based recommendations for re-establishing patient care post-disaster. The primary objective of this study is to examine associations between patient characteristics, chief complaints, final diagnoses, and medications prescribed at a post-Katrina clinic to better inform strategic planning for post-disaster healthcare delivery (e.g., charitable donations of medications and medical supplies). METHODS: Th

Endogenous prostaglandins are physiological regulators of endocrine cells in the gastroduodenal mucosa of the rat.

BACKGROUND/AIM: To investigate whether endogenous prostaglandins participate in the regulation of the gastrointestinal endocrine cell system. METHODS: Sprague-Dawley rats were treated with 1 mg/kg indomethacin subcutaneously or indomethacin subcutaneously and 500 microg/kg oral prostaglandin E2 or solvents for 2 months. Endocrine cells were visualized by using immunohistochemistry and by the Sevier-Munger silver stain on specimens from the gastroduodenal mucosa, and their total volume was estimated, using standard stereological methods. Plasma and gastrointestinal tissue concentrations of reg

Interaction of environmental chemicals with the estrogen and progesterone receptors from the oviduct of the American alligator.

Reports of reproductive abnormalities in the American alligator from Lake Apopka, Florida, have been linked to a spill of DDT and other pesticides suspected of having hormonelike activity. To determine whether environmental chemicals had the potential to function as exogenous hormones in the American alligator, we examined the ability of chemicals to bind the estrogen receptor (aER) and progesterone receptor (aPR) in a protein extract prepared from the oviduct of the alligator. In competition binding assays with [3H]17 beta-estradiol, some DDT metabolites showed inhibition of [3H]17 beta-estra

The pathobiology of the human enterochromaffin-like cell.

The significance of the enterochromaffin-like (ECL) cell as a critical endocrine regulator of gastric fundic mucosal function has only recently been recognized. Although the percentage of these cells present in the human fundic mucosa is less than that in rodents, the observation that they secrete histamine and are probably important modulators of parietal cell function has resulted in their attaining some considerable biological significance. The further identification of gastrin and somatostatin receptors on the surface of the ECL cells has suggested that other neurohormonal influences may b

Pubertal development: normal, precocious and delayed.

The present concepts on the neuroendocrine mechanisms which trigger pubertal development and modulate the progression towards sexual maturity have been reviewed. Essentially, puberty is presented as a continuum, the programming of which is initiated prenatally and which ends in adult life when all hormonal secretions become autoregulated. This continuum is dependent on a delicate equilibrium between CNS neurohormones (GnRH), neurotransmitters (biogenic amines), pituitary gonadotrophin (FSH, LH) secretion and the end-organ response (testis or ovary) through the activation of specific membrane r

Interactions of bile acids and gut microbiota modulate neurological health: a comprehensive review on mechanisms and therapeutic potential of dietary phytochemicals.

Bile acids (BAs), classically regarded as detergents for dietary lipid absorption, have emerged as pivotal signaling molecules with systemic endocrine functions. The discovery of the Farnesoid X Receptor (FXR) and Takeda G protein-coupled receptor 5 (TGR5) as BAs-activated receptors unveiled their profound influences on glucose, lipid, and energy metabolism. BAs are first synthesized in hepatocytes and further metabolized by gut microbes, can either circulate in enterohepatic system or be found in circulations to exert various effects. More recently, the gut-brain axis has been identified as a

Maternal microbiota and fetal brain development: a multidisciplinary review of cognitive, neurological, and metabolic outcomes.

OBJECTIVE: The prenatal period represents a critical window for the programming of the central nervous system. This multidisciplinary narrative review aims to synthesize current evidence regarding the maternal microbiota-gut-fetal brain axis, examining how maternal gut composition may influence fetal neurogenesis, synaptogenesis, and long-term neurobehavioural outcomes. MECHANISM: We analyse three primary pathways of influence: the systemic translocation of neuroactive bacterial metabolites (specifically short-chain fatty acids), the modulation of the maternal immune system leading to materna

Brain-heart interaction: direct and indirect circuits in health and diseases.

BACKGROUND: Epidemiological data indicate a significant global increase in both comorbidity and mutually induced rates between cardiovascular and neurological diseases. Among these, cardiovascular and cerebrovascular diseases share eleven co-localized loci. The critical role of the brain-heart axis in these processes has been increasingly recognized. This axis mediates dynamic bidirectional crosstalk via neuroendocrine, neuroimmune and related pathways, with key mediators including the hypothalamic-pituitary-adrenal (HPA) axis, gut-derived metabolites, extracellular vesicles (EVs), and autonom

Extracellular Matrix-Mediated Neuroendocrine Control of the Thymus.

BACKGROUND: The thymus is the primary lymphoid organ responsible for the generation of a self-tolerant and immunocompetent T-cell repertoire. Its function is not autonomous but is finely tuned by a complex neuroendocrine network, integrating signals from the central nervous system and the endocrine system to modulate thymopoiesis. While the roles of classical hormones and neurotransmitters in regulating thymic epithelial cell (TEC) function and T-cell development are increasingly recognized, the mechanistic pathways by which these signals are integrated within the thymic microenvironment still

Arginine Vasopressin Deficiency and Oxytocin Deficiency in the Endocrine Clinic.

The hypothalamo-neurohypophysial system consists of specialized neurons in the supraoptic and paraventricular nuclei that project to the central nervous system and the posterior pituitary, where they secrete arginine vasopressin (AVP) and oxytocin (OXT) into the systemic circulation. AVP is the key endocrine regulator of water balance via V2 receptor-mediated aquaporin-2 insertion in renal collecting ducts and modulates vascular tone via V1 receptors. OXT plays a central role in labor and lactation, but also influences metabolism, social behavior, emotional processing, and stress regulation. A

Chronic exposure to F-53B disrupts sperm quality and steroidogenic regulation in male rats.

Chlorinated polyfluoroalkyl ether sulfonic acid (Cl-PFESAs, trade name F-53B), an emerging substitute for perfluorooctane sulfonates (PFOS) used as mist suppressor in electroplating in China, has raised increasing attention due to its environmental persistence, bioaccumulation, and potential health risks. F-53B exposure has been associated with multi-system toxicities, including hepatotoxicity, immunotoxicity, and developmental toxicity, etc. However, its chronic effects on mammalian reproductive system remain poorly understood. In this study, male Sprague-Dawley (SD) rats were exposed to F-53

Chronic Stress and the Brain-Kidney Axis in Renal Injury: From Neuroendocrine Dysregulation to Kidney Damage.

Chronic stress is a pervasive psychophysiological burden and an independent risk factor for chronic kidney disease (CKD). Large-scale cohort studies confirm that stress exposure significantly increases CKD risk and accelerates renal decline, even after adjusting for traditional confounders. However, the neuroendocrine mechanisms translating stress into direct kidney damage remain unclear. This review focuses on the brain-kidney axis, a bidirectional framework integrating neural, endocrine, and immune signals. Under chronic stress, sustained overactivation of the hypothalamic-pituitary-adrenal

Developmental neurotoxicity of bisphenol F and bisphenol S in animal model systems: A literature review.

Neurodevelopmental disorders have complex etiologies, stemming both from genetic and environmental risk factors, including gestational exposure to bisphenol A (BPA). BPA is an endocrine-disrupting chemical widely used in the synthesis of plastics and epoxy-resins. In 2012, the Food and Drug Administration issued a ban on the use of BPA in certain baby and childhood products, which contributed to the proliferation of BPA-free products. To make products without BPA, plastic and epoxy manufacturers often use chemical analogs, including bisphenol F (BPF) and bisphenol S (BPS). However, the structu

Impact of the environmental endocrine disruptor 4-octylphenol on reproductive function in pubertal male mice.

4-Octylphenol, a major environmental degradation product of alkylphenols (Aps) used in industry, is an endocrine-disrupting chemical with significant estrogenic activity. It is one of the most toxic Aps, poorly biodegradable in the environment, and can accumulate in organisms through the food chain, thereby affecting the male reproductive system. In this study, the effect of 4-octylphenol on male reproductive health was analyzed. Male pubertal mice were exposed to 4-octylphenol at doses of 0, 1, 10, and 100 mg/kg once daily for 28 days. Our findings indicated that pubertal exposure to 4-octylp

Profiles of Circulating Exosomal miRNA in Patients with SAPHO Patients by High-Throughput Sequencing.

BACKGROUND: Synovitis, acne, pustulosis, hyperostosis, and osteitis (SAPHO) syndrome is a rare disease that is characterized by autoinflammatory lesions on both bones and skin. The diverse manifestations and limited understanding of its etiology have hindered the diagnosis and treatment of this condition. SAPHO syndrome is also classified as a primary inflammatory osteitis. The onset of osteoarticular involvement in this disease is typically gradual, and the identification of associated biomarkers may be crucial for accurate diagnosis, effective treatment, and a better understanding of its und
