Activating transcription factor 2 (ATF2), a ubiquitously expressed member of the basic region leucine zipper (bZIP) family of transcription factors activated by mitogen activated protein kinase (MAPK) pathways, is important in the mediation of cellular stress responses, development and transformation. We have previously reported the differential expression of active ATF2 in the human myometrium throughout pregnancy and labour, and identified and partially characterized a novel splice variant ATF2-small (ATF2-sm). To further understand the role of these factors in the myometrium, we have used g

BACKGROUND: Cryptorchidism is one of the most common reproductive abnormalities in rams, which seriously harms the reproductive capacity of rams and causes significant economic losses to the sheep industry. However, there are few studies elucidating its hereditary predisposition in sheep.
RESULTS: In the present study, the transcriptome and proteome of the cryptic (CT) and contralateral (CLT) testis from four unilaterally cryptorchid rams, and the normal testis (NT) from four healthy rams were analyzed using RNA-seq and TMT-based proteomics, respectively. A total of 10,357, 10,175, and 132 di

Branching morphogenesis in murine submandibular glands (SMG) is regulated by growth factors, extracellular matrix (ECM) and many biological processes through interactions between the epithelium and the mesenchyme. MicroRNAs (miRNAs) are a set of small, non-protein-coding RNAs that regulate gene expression at the post-transcriptional level. We hypothesized that branching morphogenesis is partly regulated by miRNAs. Forty-four miRNAs and novel miRNA candidates were detected in SMG at embryonic day 13 by a cloning method combined with Argonaute-2 immunoprecipitation. MicroRNA21 (miR-21) expressio

The paired box transcription factor PAX6 is a master regulator of eye development. Pathogenic PAX6 variants predominantly cause aniridia, resulting in a wide range of ocular anomalies affecting both the anterior and posterior segments of the eye. However, a detailed examination of the transcriptomic alterations that occur with these variants during early eye development has not yet been undertaken. To investigate this, human-induced pluripotent stem cells (hiPSC)-derived 3D optic vesicle-like organoids were generated up to day 35 from an aniridia patient with a heterozygous nonsense PAX6R261X

Ovarian cancer stem-like cells (OCSCs) are major drivers of tumor recurrence and chemoresistance. Although PIWI-interacting RNAs (piRNAs) have emerged as important regulators of cancer stem cell (CSC) biology, their roles in OCSCs remain poorly understood. We performed a systematic analysis of public datasets to identify ovarian cancer-associated piRNAs and selected piR-823 and piR-20326 for functional validation. Their expression was evaluated by quantitative reverse transcription polymerase chain reaction (qRT-PCR)following piRNA mimic and inhibitor transfections in three-dimensional (3D) ov

Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive malignancy with a five-year survival rate of approximately 13%. Patients with PDAC also have an elevated incidence of venous thromboembolism (VTE), despite prophylactic anticoagulation. Thus, there is an urgent need for therapeutic strategies that target tumor progression and hypercoagulability. Protein S (PS), a physiological anticoagulant encoded by the PROS1 gene, has recently been shown to inhibit PDAC growth in preclinical models. To further examine the physiological relevance of intratumoral PS in PDAC, we performed a meta-an

The Hedgehog (Hh) signaling pathway plays an essential role in embryonic development, and its aberrant activation is associated with pancreatic cancer progression. However, the function of Suppressor of Fused (SUFU), a critical negative regulator of the Hh pathway, in pancreatic cancer has been poorly studied. In this study, we found that SUFU knockdown significantly inhibits pancreatic cancer cell growth, migration, and proliferation while inducing apoptosis. We identified a direct interaction between JOSD1 and SUFU, whereby JOSD1 stabilizes SUFU protein by inhibiting its ubiquitination and p

Apricot (Prunus armeniaca L.) is an economically significant fruit crop worldwide with remarkable germplasm diversity, and its fruit quality is primarily determined by key metabolites (e.g., sugars, organic acids, and flavonoids). However, current apricot genome assemblies remain incomplete, with substantial unanchored regions that hamper analyses of genomic evolution and the regulatory mechanisms underlying fruit quality. Here, we generated the first gap-free telomere-to-telomere (T2T) de novo assembly of the P. armeniaca genome (248.3 Mb) and annotated 22 786 protein-coding genes across eigh

Our fi ndings identify NtWHY1 as a potential genetic target for improving cold tolerance in crops. WHIRLY1 is a plant-specific transcription factor localized to both the nucleus and plastids, playing important roles in plant growth and stress responses. However, its function in tobacco under cold stress remains unclear. In this study, we found that NtWHY1 expression was strongly induced by cold (-4 °C). Surprisingly, overexpression of NtWHY1 enhanced cold sensitivity, causing more severe leaf wilting. Physiological analysis revealed higher accumulation of reactive oxygen species (ROS) and malo

Phosphate (Pi) homeostasis is critical for rice growth and development. Previous studies have shown that OsHMGB1, a chromatin-associated protein, positively regulates Pi accumulation by modulating the chromatin accessibility of phosphate starvation-responsive (PSR) genes. OsHMGB1 is induced under Pi-deficient conditions, and its protein level declines more rapidly under Pi-sufficient conditions, suggesting the existence of post-translational regulation. However, the mechanism underlying OsHMGB1 protein degradation remains unclear. Here, we demonstrate that the degradation of OsHMGB1 under Pi-s

Amelogenesis imperfecta is a hereditary enamel defect arising from dental epithelium dysfunction. Although keratinocyte differentiation factor 1 (KDF1) acts as an intracellular regulator in epithelial cells, the underlying disease mechanism of the patient-derived KDF1 missense mutation in amelogenesis remains unclear. Here, we show that a patient-derived KDF1 mutation (c.908 G > C, p.R303P) causes enamel defects by disrupting cell adhesion and Hippo-YAP signaling. Immunohistochemistry revealed strong KDF1 expression throughout dental epithelium development, particularly at the cell membrane. K

Global warming increasingly threatens crop productivity, necessitating new varieties that combine high yield potential with climate resilience. Optimizing plant architecture, particularly by reducing leaf angle (LA), enables dense planting and enhanced light capture, yet the mechanisms coordinating architecture improvement with heat tolerance remain largely unknown. Here, we identify the microRNA ZmMIR319A as a key regulator that simultaneously regulates LA and thermotolerance in maize. ZmMIR319A promotes LA by post-transcriptionally repressing Teosinte branched1/Cycloidea/PCF (TCP) transcript

Prostate cancer (PC) progression and mortality are largely driven by therapeutic resistance, increasingly linked to dynamic tumor-stroma interactions. However, the molecular basis of cancer-fibroblast crosstalk remains incompletely understood, particularly the contribution of Interleukin (IL)-30, an emerging regulator of PC aggressiveness. Here, we demonstrate that IL30 produced by PC cells reprograms stromal fibroblasts via IL6Rα/gp130 signaling, activating AKT and TGF-β/BMP pathways to drive their proliferation and differentiation into pro-angiogenic cancer-associated fibroblasts (CAFs). INTRODUCTION: Faba bean (Vicia faba L.) is a high-yielding, protein-rich grain legume with considerable potential for sustainable cropping systems. However, its genetic improvement has been constrained by a large, complex genome and historically limited genomic resources. We aimed to identify agronomically relevant QTLs in a diverse faba bean panel using genome-wide associations studies (GWAS) with rigorous control of false positives due to model violations.
METHODS: The NORFAB faba bean diversity panel was evaluated for agronomic traits over four years (2018-2021) at two locations in Denmark

Enterohemorrhagic Escherichia coli (EHEC) comprises a prominent group of extracellular human pathogens that specifically colonize the colonic epithelium, leading to severe gastrointestinal diseases and representing a significant global public health threat. During infection, EHEC O157:H7-the most prevalent serotype - tightly adheres to intestinal epithelial cells and induces the formation of characteristic attaching and effacing (A/E) lesions. However, the molecular mechanisms governing this close interaction remain incompletely understood. In this study, we employed transposon-directed insert

Myogenesis is a stepwise process encompassing myogenic progenitor proliferation, lineage commitment, differentiation, myocyte fusion, and myotube maturation, and it is orchestrated by myogenic regulatory factors (MRFs) together with signaling pathways that coordinate these transitions. Long noncoding RNAs (lncRNAs) have emerged as important regulators of muscle development and regeneration, yet how lncRNAs integrate with canonical signaling networks to shape myogenic progression remains incompletely defined. Here, we identify a novel myocyte-enriched, Notch-repressed myogenic lncRNA (NRMLncR,

Leptin is a central regulator of appetite and energy metabolism in vertebrates, and its deficiency in mammals typically results in hyperphagia and severe obesity. In fish, however, the physiological roles of leptin remain less clearly defined, partly due to the whole genome duplication (WGD) and divergent expression patterns. In this study, we generated a leptin A (lepa) loss-of-function mutant in medaka (Oryzias latipes) using CRISPR/Cas9 to investigate the function of leptin signaling. Phenotypic analysis revealed that female homozygous mutants exhibited significantly reduced body length and

Chronic noise exposure is increasingly recognized as an environmental risk factor for metabolic disorders; however, whether and how circadian dysregulation contributes to noise-induced hepatic lipid abnormalities remains unclear. Here, we investigated the role of circadian regulation in chronic noise-induced hepatic lipid metabolic alterations using transcriptomic, metabolomic, and genetic approaches. Thirty-day noise exposure in C57BL/6 J mice induced hepatic lipid accumulation and mild histopathological changes without affecting body weight or food intake. Noise exposure altered the temporal

Adolescence represents a sensitive window of neural plasticity, when social experiences exert enduring influences on emotional and cognitive development. Adverse social conditions, such as social instability stress (SIS), disrupt peer relationships and lead to long-term changes in social and affective behaviour. Here, we used RNA sequencing to characterize the immediate (P46) and enduring (P70) molecular consequences of adolescent SIS in male and female rats in two stress-sensitive regions: the dorsal hippocampus (dHPC) and basolateral amygdala (BLA). SIS produced region- and sex-specific tran
