Antimicrobial resistance patterns and characterization of integrons of Shigella sonnei isolates in Seoul, 1999-2008.

A total of 66 Shigella sonnei isolates from 1999 to 2008 in Seoul was analyzed for their antimicrobial resistance, carriage of integron, and the patterns of Pulsed-field gel electrophoresis (PFGE). A high level of antimicrobial resistance to streptomycin (100%), trimethoprim/sulfamethoxazole (95%), tetracycline (94%), nalidixic acid (65%), and ampicillin (41%) was observed among S. sonnei isolates. Fourteen profiles of antimicrobial resistance were identified with the most common resistance profile being nalidixic acid, streptomycin, tetracycline, and trimethoprim/sulfamethoxazole (35%). PCR a

NMR-based metabolic profiling and metabonomic approaches to problems in molecular toxicology.

We have reviewed the main contributions to the development of NMR-based metabonomic and metabolic profiling approaches for toxicological assessment, biomarker discovery, and studies on toxic mechanisms. The metabonomic approach, (defined as the quantitative measurement of the multiparametric metabolic response of living systems to pathophysiological stimuli or genetic modification) was originally developed to assist interpretation in NMR-based toxicological studies. However, in recent years there has been extensive fusion with metabolomic and other metabolic profiling approaches developed in p

Diversity among clinical isolates of penicillin-resistant Streptococcus mitis: indication for a PBP1-dependent way to reach high levels of penicillin resistance.

A total of 12 non-epidemiologically related clinical isolates of Streptococcus mitis that showed different levels of resistance to penicillin were studied. Membrane-protein profiles and penicillin-binding protein (PBP) patterns showed a great polymorphism; and patterns of 4-7 PBPs, with sizes that ranged from approximately 101 kDa to approximately 40 kDa, were detected in each strain. No association could be found between PBP pattern and resistance level to penicillin among these isolates. Arbitrarily primed PCR confirmed the genetic diversity among this group of streptococci. One of the isola

Facial dysmorphology, roentgenographic measurements, and clinical genetics.

Roentgenographic measurements are used in the facial diagnosis of a male patient (proband) and his mother with unusual craniofacies. Twenty-four measurements (from both the PA and LA x-ray headplates) defining the major anatomic areas of the head and face were chosen as the overall descriptors for the characterization and recognition of craniofacial pattern profile (CFPP). The measurements were transformed into Z-scores. By using the sigma z (i.e., standard deviation of the z-scores) value, the CFPP deviations were estimated. The proband and mother have aberrant, i.e., dysmorphic, craniofacial

Multilayered Insights into Poorly Differentiated, BRAFV600E-Positive, Thyroid Carcinoma in a Rapidly Developing Goiter with Retrosternal Extension: From En "Y" Cervicotomy to SPECT/CT-Positive Lung Metastases.

Poorly differentiated thyroid malignancy, a rare histological type of aggressive thyroid malignancy with associated difficulties and gaps in its histological and molecular characterization, might lead to challenging clinical presentations that require a prompt multimodal approach. This case study involved a 56-year-old, non-smoking male with a rapidly developing goiter (within 2-3 months) in association with mild, non-specific neck compressive symptoms. His medical history was irrelevant. A voluminous goiter with substernal and posterior extension up to the vertebral bodies was detected using

Clinical, biochemical, and genetic spectrum of MADD in a South African cohort: an ICGNMD study.

BACKGROUND: Multiple acyl-CoA dehydrogenase deficiency (MADD) is an autosomal recessive disorder resulting from pathogenic variants in three distinct genes, with most of the variants occurring in the electron transfer flavoprotein-ubiquinone oxidoreductase gene (ETFDH). Recent evidence of potential founder variants for MADD in the South African (SA) population, initiated this extensive investigation. As part of the International Centre for Genomic Medicine in Neuromuscular Diseases study, we recruited a cohort of patients diagnosed with MADD from academic medical centres across SA over a three

Memory-Based Learning Disabilities: A Syndrome without a Pigeonhole-Towards a Mechanism-First Assessment Framework.

Memory-based learning disabilities represent a distinct and underrecognized clinical entity that current diagnostic classification systems fail to capture. This theoretical synthesis argues that these difficulties constitute an identifiable syndrome defined by neuropsychological vulnerabilities in declarative memory systems, particularly those dependent on hippocampal function. Evidence from cognitive neuroscience, developmental neuropsychology, and educational psychology converges to identify a distinctive phenotypic profile that differentiates memory-based learning disabilities from other de

Spatiotemporal dynamics of ecto-5'-nucleotidase (CD73) in mouse retina under physiological conditions.

Adenosine is essential for energy metabolism and acts as a neuromodulator in the central nervous system. The retina is a highly energy-demanding neural tissue, and dysregulation of adenosine metabolism contributes to retinal diseases. Although adenosine pathways form extensive compensatory networks, the dynamics of ecto-5'-nucleotidase (CD73), a key enzyme for extracellular adenosine generation, remain elusive. Here, we map its spatiotemporal profile from development to adulthood using two transgenic mouse lines. We found that CD73 became expressed in the rod-photoreceptor lineage by postnatal

Regulatory T cell therapies: biological foundations, engineering strategies, and clinical translation.

Regulatory T cell (Treg) therapy has emerged as a promising strategy to control pathological immune responses in autoimmunity, graft-versus-host disease, and solid organ transplantation. Most clinical studies to date have relied on autologous Tregs expanded ex vivo, an approach that has demonstrated safety and feasibility but remains limited by variable cell quality, restricted scalability, and complex manufacturing requirements. To address these constraints, multiple alternative strategies are being developed, including the induction of regulatory phenotypes in conventional T cells, the engin

Senescent-Like Myofibers Contribute to Anti-Regenerative Cytokine Signaling in Duchenne Muscular Dystrophy.

Duchenne muscular dystrophy (DMD) is a genetic muscular disease characterized by progressive muscle degeneration. p16 is expressed in skeletal muscles and induces cellular senescence in a rat model of DMD, whereas its ablation enhances muscle regeneration. However, the mechanism underlying this phenomenon remains unclear. This study aimed to elucidate the mechanism for p16-induced DMD exacerbation. RNA-seq analysis revealed p16-dependent upregulation of cytokine gene expression in DMD rat skeletal muscles, which also altered the systemic blood cytokine profile. Furthermore, the effect of an al

Development of a HiBiT-tagged reporter Akabane virus and its application in antiviral drug screening.

Akabane virus (AKAV), an arthropod-borne pathogen causing severe reproductive failure in ruminants, poses a major threat to livestock industry. Given the lack of vaccines and antiviral drugs against AKAV, the development of rapid antiviral screening tools is urgently needed. To establish a quantifiable platform for AKAV research, we constructed a reporter virus (rAKAV-L-HiBiT) by inserting the 11-amino-acid HiBiT subunit of NanoLuc luciferase into the C-terminus of the L protein. The recombinant virus was characterized for growth kinetics, genetic stability, and luciferase activity. rAKAV-L-Hi

Immune Toxicities in AAV Gene Therapy: Overview for Clinicians.

Gene therapy using recombinant adeno-associated virus (rAAV) vectors has emerged as a transformative therapeutic modality for genetic disorders, demonstrating high transduction efficiency and a generally favorable safety profile during pre-clinical development. However, serious adverse events, including thrombotic microangiopathy, acute respiratory distress syndrome, hepatotoxicity, myocarditis, cytokine storm, and hemophagocytic lymphohistiocytosis, have been observed across multiple gene therapy clinical trials. Significant efforts have been made to understand the toxicities that cause these

The polymerized protein response (PPR) is activated by genetic variants that polymerize in the ER and is mediated by NFκBp50.

In previous studies we have shown that accumulation of the polymerogenic variant ATZ that causes α1-antitrypsin deficiency (ATD) activates NFκB DNA binding. Here we discovered that this response, which we are naming the polymerized protein response (PPR), is characterized by NFκB p50 homodimers, distinct from the p50/p65 heterodimers in the canonical NFκB inflammatory response and the unfolded protein response (UPR). The PPR is also activated by other disease associated variants that polymerize in the ER whereas disease-associated variants that do not form polymers activate p50/p65 heterodimer

Evaluating mitapivat for the treatment of alpha or beta thalassemia.

INTRODUCTION: Thalassemia is a group of diverse genetic disorders with worldwide distribution that affects hemoglobin synthesis. Until recently, the therapeutic approach to thalassemia was symptomatic, relying on red blood cell transfusions, treatment of comorbidities and of disease-related complications. However, novel therapeutic agents have recently been developed and are gradually being integrated into routine clinical practice. AREAS COVERED: One of the most promising agents is mitapivat (AG348), an oral pyruvate kinase activator that enhances the erythrocytic adenosine triphosphate (ATP

Beyond CFTR: Ivacaftor's role in restoring cellular redox balance and preventing ferroptosis.

Cystic fibrosis (CF), is a genetic disorder caused by dysfunctional Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) protein, characterized by progressive pulmonary decline and elevated oxidative stress and inflammation. CFTR modulators, including potentiators such as ivacaftor, represent the cornerstone of current therapeutic strategies. We present compelling evidence that ivacaftor, in contrast to other CFTR modulators, exhibits a CFTR-independent ability to mitigate oxidative injury by inhibiting lipid peroxidation, a cascade of oxidative damage, and by blocking ferroptosis, a spe

Pathogenic cytokines in thrombotic microangiopathies: molecular insights and therapeutic targets.

Thrombotic microangiopathies (TMAs) are a heterogeneous group of disorders characterized by endothelial damage, microvascular thrombosis, thrombocytopenia, and microangiopathic hemolytic anemia. While the initiating triggers may differ-ranging from infections and autoimmune diseases to genetic complement dysregulation-a unifying pathophysiological feature is injury to the vascular endothelium. Recent advances have highlighted the critical role of pro-inflammatory cytokines in mediating endothelial dysfunction, contributing to both the initiation and propagation of thrombotic events in TMAs.
