Lectin-Micropatterned Biointerfaces Regulate Cytoskeletal Organization and Viscoelastic Phenotypes of Bladder Cancer Cells.

Altered glycosylation is a hallmark of cancer progression and provides opportunities to modulate cell adhesion through glycan-lectin interactions. In this study, we investigate how lectin-mediated adhesion and substrate micropatterns influence cytoskeletal organization and viscoelastic properties of bladder cancer cells. Bladder cancer cell linesHCV29, 5637, and T24 cellsrepresenting different malignancy stages were cultured on homogeneous and micropatterned lectin-functionalized substrates. Lectins recognizing N-glycans (Dolichos biflorus agglutinin, DBA; Wheat germ agglutinin, WGA) and fuc

Fiber Diameter Differentially Regulates Function of Retinal Pigment and Corneal Epithelial Cells on Nanofibrous Tissue Scaffolds.

Biomaterials have significant functions as tissue scaffolds to support cells for regeneration. Nanofibrous scaffolds which mimic the architecture of the extracellular matrix are well suited to support epithelial cells for ocular tissue engineering. This study aimed at investigating the role of scaffold architecture, if any, on the response of ocular epithelial cells. Thus, we have cultured two different types of ocular epithelial cells on nanofibrous scaffolds of two different diameters to evaluate their generic and cell-specific properties. Human adult retinal pigment epithelial (ARPE-19) and

Alpha fetoprotein (AFP) participates in the build up of hematopoietic cells in the early embryonic stage: an abortion case observation.

BACKGROUND: At the 3rd week of human embryo, some cell clumps are formed by the hyperplasia of mesenchymal cells at the germ layer of the yolk sac wall. These cell clumps are known as blood islands. The cells in the center of the blood islands further develop into primitive blood cells, such as hematopoietic stem cells. The blood island in the yolk sac further develops into the extramedullary hematopoietic tissue in 1 week at the 3rd to 4th week. CASE PRESENTATION: A 32-year-old pregnant woman who missed menstruation for 42 days discovered that her pregnancy required an abortion. The tissue c

Projections to the pontine nuclei from choline acetyltransferase-like immunoreactive neurons in the brainstem of the cat.

By use of retrograde transport of horseradish peroxidase-wheat germ agglutinin (HRP-WGA) in combination with monoclonal antibodies against choline acetyltransferase (ChAT), we show that putative cholinergic inputs to the feline pontine nuclei originate from cells in the dorsolateral pontine tegmentum. These cells form a loosely arranged continuum that nevertheless may be subdivided into two groups on the basis of differences in cell morphology. One group consists of double-labeled cells in the periventricular gray substance medial to, and partly merging with, the nucleus locus coeruleus. The o

Spatiotemporal mapping reveals microbiota-dependent maintenance of functional competence during postnatal immune maturation.

Postnatal immune maturation is commonly viewed as a developmental milestone associated with weaning. However, how postnatal microbiota exposure shapes immune cell heterogeneity across tissues, and whether this maturation is self-sustaining or requires continuous microbial input, remain unclear. Here, we conducted spatiotemporal single-cell transcriptomic profiling of 120,649 immune cells isolated from 10 tissues in both germ-free (GF) and specific pathogen-free (SPF) mice from weaning to adulthood. We demonstrate that microbial colonization drives the expansion and functional maturation of dis

Directed conversion of porcine expanded pluripotent stem cells into trophoblast stem cells.

Trophoblast stem cells (TSCs) serve as a valuable system for investigating the signaling pathways that regulate extraembryonic lineage commitment. Although trophoblast specification has been extensively characterized in humans and rodents, comparable tools and molecular frameworks remain underdeveloped in pigs, a species of significant agricultural and biomedical relevance. This study identifies defined biochemical conditions that enable efficient conversion of porcine (p) expanded pluripotent stem cells (EPSCs) into induced TSCs (iTSCs). The results demonstrate that combined inhibition of TGF

Functional Analysis of the Halastavi árva Virus (HalV) Internal Ribosome Entry Site.

Viral internal ribosome entry sites (IRESs) are specialized RNA structures that facilitate cap-independent translation as a strategy to usurp the host translational machinery. The Type 6 IRESs are the most streamlined mechanism to date, as they adopt a three pseudoknot RNA structure to initiate factorless translation initiation by directly recruiting the ribosome and drive translation. The Halastavi árva virus (HalV) IRES represents the most minimalistic subclass identified to date, whereby the IRES lacks specific pseudoknot domains that bind to the 40S subunit but instead recruits pre-assembl

The nuclear receptor NR6A1 plays an oncogenic role through reprogramming glycolysis in tumors.

NR6A1 is a member of the nuclear receptor superfamily of ligand-activated transcription factors and can bind to conserved DNA sequences, acting as a transcriptional repressor. NR6A1 has been reported to promote prostate cancer, gastric cancer, and testicular germ cell tumor progression. In the present study, we confirmed the oncogenic role of NR6A1 in several types of cancer cells, including HeLa, TFK1, and A549 cells, and revealed that NR6A1 knockdown led to a decrease in lung adenocarcinoma cell proliferation, a reduction in glucose consumption, a reduction in lactic acid production, and dec

NQO1 antagonizes PM2.5-induced apoptosis of Sertoli cells by activating the UPRmt pathway.

The mechanism by which NQO1 antagonizes PM2.5-induced apoptosis in Sertoli cells through activation of the mitochondrial unfolded protein response (UPRmt) was investigated. A reproductive toxicity model was established using TM4 cells exposed to PM2.5 (50 μg/mL, 24 h). Transcriptome sequencing and bioinformatics analysis identified NQO1 as the target gene, playing a key role in redox regulation and apoptosis. Phenotypic analysis showed that PM2.5 exposure significantly increased intracellular ROS levels (P < 0.01), induced structural disintegration of mitochondrial cristae (as observed by TEM)

Research progress on the mechanism underlying the application of mesenchymal stem cells in the treatment of male infertility.

Male infertility has become an increasingly prominent health issue worldwide. This review systematically evaluated the therapeutic application of different types of MSCs in various male infertility models. The therapeutic effects of MSCs are attributed to mechanisms such as in vivo and in vitro differentiation into germ cells, improved antioxidant capacity of testicular tissue, inhibited secretion of inflammatory factors and elevated anti-inflammatory level of testicular tissue, prevention of excessive apoptosis of testicular tissue cells, restoration of the normal secretion of sexual hormone

Clonal Parabacteroides from Gut Microfistulous Tracts as Transmissible Cytotoxic Succinate-Commensal Model of Crohn's Disease Complications.

Crohn's disease (CD) has been traditionally viewed as a chronic inflammatory disease that cause gut wall thickening and complications, including fistulas, by mechanisms not understood. By focusing on Parabacteroides distasonis (presumed modern succinate-producing commensal probiotic), recovered from intestinal microfistulous tracts (cavernous fistulous micropathologies CavFT proposed as intermediate between 'mucosal fissures' and 'fistulas') in two patients that required surgery to remove CD-damaged ilea, we demonstrate that such isolates exert pathogenic/pathobiont roles in mouse models of CD

Embryonic spatiotemporal expression pattern of Folded gastrulation suggests roles in multiple morphogenetic events and regulation by AbdA.

In Drosophila, the signaling pathway activated by the ligand Folded gastrulation (Fog) is among the few known G protein-coupled receptor (GPCR) pathways to regulate cell shape change with a well-characterized role in gastrulation. However, an understanding of the spectrum of morphogenetic events regulated by Fog signaling is still lacking. Here, we present an analysis of the expression pattern and regulation of fog using a genome-engineered Fog::sfGFP line. We show that Fog expression is widespread and in tissues previously not associated with the signaling pathway including germ cells, trache

Progress Toward Genetic Rescue of the Northern White Rhinoceros (Ceratotherium simum cottoni).

The northern white rhinoceros (NWR) is functionally extinct, with only two nonreproductive females remaining. However, because of the foresight of scientists, cryopreserved cells and reproductive tissues may aid in the recovery of this species. An ambitious program of natural and artificial gametes and in vitro embryo generation was first outlined in 2015, and many of the proposed steps have been achieved. Multiple induced pluripotent stem cell lines have been established, primordial germ cell-like cells have been generated, oocytes have been collected from the remaining females, blastocysts h

The cancer testis antigen TDRD1 regulates prostate cancer proliferation by associating with snRNP biogenesis machinery.

Prostate cancer is the most commonly diagnosed noncutaneous cancer in American men. TDRD1, a germ cell-specific gene, is erroneously expressed in more than half of prostate tumors, but its role in prostate cancer development remains elusive. In this study, we identified a PRMT5-TDRD1 signaling axis that regulates the proliferation of prostate cancer cells. PRMT5 is a protein arginine methyltransferase essential for small nuclear ribonucleoprotein (snRNP) biogenesis. Methylation of Sm proteins by PRMT5 is a critical initiation step for assembling snRNPs in the cytoplasm, and the final snRNP ass

NECL2 regulates blood-testis barrier dynamics in mouse testes.

The adhesion protein nectin-like molecule 2 (NECL2) is involved in spermatogenesis and participates in the connections between Sertoli cells and germ cells. Necl2 deficiency leads to infertility in male mice. We found that NECL2 is relatively highly expressed on the cell membranes of preleptotene spermatocytes. It is known that preleptotene spermatocytes pass through the blood-testis barrier (BTB) from the base of the seminiferous tubules to the lumen to complete meiosis. We hypothesized that the NECL2 protein on the surfaces of preleptotene spermatocytes has an effect on the BTB when crossing

High doses of clethodim-based herbicide GrassOut Max poses reproductive hazard by affecting male reproductive function and early embryogenesis in Swiss albino mice.

Clethodim is a widely used and approved class II herbicide, with little information about its impact on the reproductive system. Herein, we investigated the male reproductive toxicity of clethodim using a mouse model. GrassOut Max (26% clethodim-equivalent) or analytical grade clethodim (≥90%) were given orally to male mice for 10 d in varying doses. All parameters were assessed at 35 d post-treatment. Significant decrease in testicular weight, decreased germ cell population, elevated DNA damage in testicular cells and lower serum testosterone level was observed post clethodim based herbicide

VCP promotes tTAF-target gene expression and spermatocyte differentiation by downregulating mono-ubiquitylated H2A.

Valosin-containing protein (VCP) binds and extracts ubiquitylated cargo to regulate protein homeostasis. VCP has been studied primarily in aging and disease contexts, but it also affects germline development. However, the precise molecular functions of VCP in the germline, particularly in males, are poorly understood. Using the Drosophila male germline as a model system, we find that VCP translocates from the cytosol to the nucleus as germ cells transition into the meiotic spermatocyte stage. Importantly, nuclear translocation of VCP appears to be one crucial event stimulated by testis-specifi

A human induced pluripotent stem cell model from a patient with hereditary cerebral small vessel disease carrying a heterozygous R302Q mutation in HTRA1.

Cerebral autosomal recessive arteriopathy with subcortical infarcts and leukoencephalopathy (CARASIL) is an inherited cerebral small vessel disease (CSVD) caused by biallelic mutations in the high-temperature requirement serine peptidase A1 (HTRA1) gene. Even heterozygous mutations in HTRA1 are recently revealed to cause cardinal clinical features of CSVD. Here, we report the first establishment of a human induced pluripotent stem cell (hiPSC) line from a patient with heterozygous HTRA1-related CSVD. Peripheral blood mononuclear cells (PBMCs) were reprogrammed by the transfection of episomal v

MMPs, ADAMs and ADAMTSs are associated with mammalian sperm fate.

Metalloproteinases include matrix metalloproteinases and disintegrin metalloproteinases. They are important members of the ECM degradation and reconstruction process and are associated with tissue development and disease. The ECM is a three-dimensional network of large molecules consisting of a variety of proteins. It is a physical scaffold for organs, and all types of cells can be found within the ECM. The testicle, where sperm are produced, is an organ that is constantly in dynamic flux. Metalloproteinases can regulate testicular tissue development and the maturation of sperm by affecting th

Lectin-Functionalized Polyethylene Glycol for Relief of Mucosal Dryness.

The importance of lubrication between oral surfaces provided by the salivary film is most acutely apparent when it is disrupted, a prevalent consequence of salivary gland hypofunction experienced with aging, a symptom of certain diseases, or a side effect of some medical interventions. Sufferers report difficulty with speech and oral food processing and collectively is detrimental to quality of life. Polyethylene glycol (PEG) is widely employed as a successful biocompatible boundary lubricant in engineering and biomedical applications. It is hypothesized that the immobilization of PEG to biolo
