Inflammatory and degenerative phases resulting from anterior cruciate rupture in a non-invasive murine model of post-traumatic osteoarthritis.

Joint injury is the predominant risk factor for post-traumatic osteoarthritis development (PTOA). Several non-invasive mouse models mimicking human PTOA investigate molecular mechanisms of disease development; none have characterized the inflammatory response to this acute traumatic injury. Our aim was to characterize the early inflammatory phase and later degenerative component in our in vivo non-invasive murine model of PTOA induced by anterior cruciate ligament (ACL) rupture. Right knees of 12-week-old C57Bl6 mice were placed in flexion at a 30° offset position and subjected to a single com

Smash landing in elite male badminton players: Effects of fatigue and task predictability via explainable machine-learning analysis.

Smash landings in badminton carry high injury risk, worsened by fatigue. Assess how fatigue and task predictability shape lower-limb biomechanics in elite male players and identify fatigue markers via explainable ML. Fourteen elites performed anticipated and unanticipated smash-landings before and after fatigue. Synchronous 3-D kinematics/kinetics were analyzed with a 2 × 2 repeated-measures ANOVA. XGBoost models based on anticipated-task trials were used to classify fatigue status; SHAP ranked feature importance. Fatigue and unpredictability independently affected peak coronal-plane knee mome

Disturbed multi-planar muscular support of the knee after anterior cruciate ligament reconstruction.

This study compared lower-limb muscle contributions to multi-planar knee loading between individuals with anterior cruciate ligament reconstruction (ACLR) and healthy controls during sidestep cutting. Eleven ACLR participants with quadrupled hamstring autografts and eleven matched healthy controls performed pre-planned sidestep cutting. Whole-body kinematics, ground reaction forces, and electromyograms (EMG) were recorded. An EMG-informed neuromusculoskeletal modelling approach was used to estimate muscle contributions to anteroposterior force (in bodyweights, BW), varus/valgus moment (BW*Heig

Histological effects of intra-articular allogeneic leukocyte-rich and leukocyte-poor platelet-rich plasma in healthy rabbit knees: an exploratory study.

OBJECTIVE: Platelet-rich plasma (PRP) is used to treat knee osteoarthritis and cartilage injuries, but its effects on healthy joint tissues are unclear. This exploratory study compared histological responses to allogeneic leukocyte-rich (LR) and leukocyte-poor (LP) PRP versus saline in healthy rabbit knees. RESULTS: Eighteen knees from nine healthy male Japanese white rabbits received a single intra-articular injection of saline, LR-PRP (Vets), or LP-PRP (Cellaid) (n = 6 knees/group). At 4 weeks, trochlear cartilage and suprapatellar pouch synovium were evaluated by hematoxylin-eosin histolog

Influences of footstrike patterns and overground conditions on lower extremity kinematics and kinetics during running: Statistical parametric mapping analysis.

This study investigated lower extremity biomechanics when running on different surfaces among runners with different footstrike patterns. Thirty rearfoot strikers (RFSs) and non-rearfoot strikers (nRFSs) ran at 3.3 m/s on a specially designed indoor track covered with three surfaces: artificial grass, synthetic rubber, and concrete. A motion capture system with ten cameras combined a force plate was used to collect marker trajectory and ground reaction force (GRF) during the running stance phase. A two-way analysis of variance with statistical parametric mapping was employed to evaluate differ

Biomechanics of horizontal meniscus tear and healing during knee flexion: Finite element analysis.

Meniscus horizontal tear is a common injury that mostly occurs in middle-aged and elderly people, and the effect of repair surgery directly affects the functional recovery of the knee joint and prevention of degenerative joint diseases. However, the stress concentration in a horizontal tear is not well understood. The primary objective of this study was to examine the reparative mechanisms involved in addressing horizontal tears of the meniscus and to elucidate the alterations in mechanical behavior throughout the subsequent postoperative healing stages. Based on clinical MRI scan data of norm

Genicular Artery Embolisation in Patients with Osteoarthritis of the Knee (GENESIS 2): Protocol for a Double-Blind Randomised Sham-Controlled Trial.

Knee osteoarthritis is a leading cause of chronic disability and economic burden. In many patients who are not surgical candidates, existing treatment options are insufficient. Clinical evidence for a new treatment approach, genicular artery embolisation (GAE), is currently limited to single arm cohort, or small population randomised studies. This trial will investigate the use of a permanent embolic agent for embolisation of abnormal genicular arterial vasculature to reduce pain in patients with mild to moderate knee osteoarthritis. Up to 110 participants, 45 years or older, with knee pain fo

Polymicrobial infection presenting as non-clostridial gas gangrene in a non-diabetic trauma patient. A case report and review of literature.

BACKGROUND: Clostridium species are known to be the primary causative organism of gas gangrene. Non-clostridial gas gangrene (NCGG) is another rare necrotizing entity often associated with an underlying disease, particularly diabetes mellitus, and has a high mortality rate. CASE REPORT: A 16-year-old, immunocompetent male was referred to us after four days, following a roadside accident, with a degloving injury over the thigh and knee along with fractures around the knee. Although clinico-radiologically suspicious of gas gangrene, the initial smear report was negative for any Gram-positive ba

Effect of knee joint angle on individual hamstrings morphology quantified using free-hand 3D ultrasonography.

Exercise responses and injury rates differ between individual hamstrings and this may be linked with their morphology. The aim of this study was to compare muscle length and tendon dimensions between the individual hamstrings at two knee joint angles using free hand three-dimensional ultrasound (3D US). Muscle-tendon length and distal tendon cross-sectional area (CSA), volume, length and echogenicity of biceps femoris long (BFlh) and short (BFsh) head, semimembranosus (SM) and semitendinosus (ST) of 16 individuals were measured using free-hand 3D US at 0° (full extension) and 45° of knee flexi

The Value of Phenotypes in Knee Osteoarthritis Research.

BACKGROUND: Over the past decade, phenotypes have been used to help categorize knee osteoarthritis patients relative to being subject to disease, disease progression, and treatment response. A review of potential phenotype selection is now appropriate. The appeal of using phenotypes is that they most rely on simple physical examination, clinically routine imaging, and demographics. The purpose of this review is to describe the panoply of phenotypes that can be potentially used in osteoarthritis research. METHODS: A search of PubMed was used singularly to review the literature on knee osteoart
