Acute myeloid leukemia (AML) is driven by a combination of genetic alterations and non-mutational mechanisms that disrupt normal hematopoiesis and support leukemic cell survival. While the mutational landscape of AML is well characterized, the non-genetic processes that sustain leukemic maintenance remain comparatively less understood. Using human AML cell lines and murine models of AML, we identify BCL2-associated transcription factor 1 (BCLAF1) as a key regulator of leukemic progression through control of mRNA processing. BCLAF1 physically associates with core spliceosome components and regu

The proteolytic processing of the amyloid precursor protein (APP) is a core pathological event in Alzheimer's disease (AD) pathogenesis, yet the global genetic regulatory networks modulating this process have not been fully characterized. To systematically identify novel regulators of APP cleavage, we performed a genome-wide CRISPR/Cas9 knockout screen utilizing an optimized UAS-GAL4-based cellular reporter, and identified genetic modulators governing amyloidogenic and non-amyloidogenic processing. The screen uncovered distinct functional gene clusters regulating the APP, prominently involving

Pioglitazone is thought to increase the risk of heart failure exacerbation in vulnerable individuals, primarily through fluid retention and plasma volume expansion. However, the precise molecular and cellular mechanisms remain incompletely understood. This narrative review aims to collect, organise, and evaluate current evidence on the pathophysiological mechanisms driving pioglitazone-related heart failure. We categorised preclinical and clinical data into key pathophysiological domains and graded evidence strength as high, moderate, or limited. The key domains implicated in this process incl

Breast cancer (BC) is a complex, clinically and morphologically heterogeneous disease for which integral morphological and molecular genetic parameters are of key importance. This paper describes the evolution of semiquantitative tumor grading systems, beginning with the classical Greenough, Patey-Scarff, and Bloom-Richardson systems and ending with modern mathematical methods based on the grade of BC malignancy. Integral prognostic indices are presented in detail, including the Nottingham Prognostic Index (NPI), its modifications (NPI+, METASTASIS), the Total malignancy score (TMS), as well a

Endometriosis is a common gynecological disorder characterized by the growth of endometrial-like tissue outside the uterine cavity, leading to chronic inflammation, pelvic pain, and infertility. Reliable mouse models are essential for investigating the molecular, hormonal, immune, and environmental mechanisms underlying disease development and progression. Conventional methods for identifying endometriotic lesions often rely on genetically engineered fluorescent reporter mice, which are time-consuming and costly to generate. Here, we present a simple, non-genetic approach to inducing and visua

Alzheimer's disease (AD) is increasingly recognized as a disorder of innate immune dysregulation within the central nervous system. The triggering receptor expressed on myeloid cells 2 (TREM2), a microglial immunoreceptor, has emerged as a pivotal genetic risk factor for late-onset AD, underscoring the critical role of neuroimmune interactions in disease pathogenesis. This review synthesizes recent advances concerning TREM2's modulation of core microglial functions, including phagocytosis, inflammatory signaling, cellular metabolism, and survival, processes that are essential for responding to

INTRODUCTION: Aspirin is not recommended for dementia prevention due to limited benefit. We investigated whether genetic subgroups may benefit.
METHODS: In the ASPirin in Reducing Events in the Elderly (ASPREE) trial (n = 13,541), we screened 5182 polygenic risk scores (PGSs) for aspirin interactions on dementia and major bleeding. After quality control, 1848 PGSs were analyzed using Cox models for aspirin×PGS interaction, adjusted for age, sex, apolipoprotein Estatus, and ancestry. Interactions were Bonferroni-corrected and examined by quintiles.
RESULTS: Platelet-related PGS accounted for

Dengue virus (DENV) is a rapidly evolving arbovirus responsible for significant morbidity and mortality worldwide. Understanding its evolutionary trajectory is essential for tracking viral emergence, transmission dynamics, and the factors driving its geographic expansion. This chapter provides a comprehensive overview of the genetic diversification and phylogenetic pathways of DENV, focusing on serotype evolution and the classification of genetic lineages. We discuss molecular phylogenetics as a key tool for elucidating the evolutionary relationships among DENV strains and highlight the applic

Follicular-cell-derived thyroid cancers that progress from differentiated tumors to poorly differentiated or anaplastic states commonly lose thyroid lineage identity, radioiodine avidity, and favorable clinical behavior. Genetic and signaling alterations within tumor cells explain part of this transition, but they do not fully account for the coexistence of different differentiation states within the same molecular subtype or even within the same lesion. Increasing functional evidence indicates that dedifferentiation is maintained by reciprocal interactions between malignant cells and the immu

Scaffold integrity is essential for the activity of proteins that function through protein-protein interactions rather than catalytic output. RAF1 exemplifies this duality: although it is a bona fide kinase and a core component of the MAPK cascade, its tumor-promoting role is largely kinase-independent, relying instead on scaffold-mediated suppression of apoptosis. Genetic Raf1 ablation in KRAS-driven lung adenocarcinoma mouse models induces tumor regression without systemic toxicity, making it an attractive candidate for targeted protein degradation. Chemogenetic systems like the dTAG platfor

Ataxin-1 (ATXN1) was originally identified as a gene in which abnormal expansion of glutamine encoding CAG repeats causes inherited neurodegenerative disease spinocerebellar ataxia type 1. Spinocerebellar ataxia type 1 is characterized by alterations in movement, cognition, and mood, with severe pathology in the cerebellum and brain stem. Studies in spinocerebellar ataxia type 1 mouse models indicated that CAG expansion predominantly causes pathogenic ATXN1 gain-of-function in the cerebellum. Conversely, mice lacking ATXN1 are severely impaired in cognitive tests and exhibit abnormalities in c

Metabolic dependencies are emerging as promising therapeutic targets in cancer, including acute myeloid leukemia (AML). Several metabolic vulnerabilities have been identified in AML cells, including a requirement for balanced sphingolipid metabolism to maintain survival and proliferation. Here we describe a novel function of the RAS superfamily small GTPase ARF6 in maintaining sphingolipid homeostasis in AML. Genetic depletion of ARF6 inhibited the proliferation of AML cell lines and reduced colony formation of primary AML CD34+ cells. Mechanistically, ARF6 promotes conversion of ceramide to s

Acute myeloid leukemia (AML) is a highly heterogeneous malignant hematological neoplasm. Although standard diagnostic procedures have been established, traditional methods still face limitations with regard to efficiency, accuracy, and standardization. In recent years, artificial intelligence (AI) has demonstrated notable advantages in medical image analysis, flow cytometry interpretation, and genetic data modeling, offering new approaches for adjunctive diagnosis of AML. This review systematically summarizes recent research advances in adjunctive diagnosis of AML, categorizing current AI-base

Age-related macular degeneration (AMD) is a complex, multifactorial retinal degenerative disease that is influenced by both genetic and environmental factors. However, the strongest risk factor for AMD is advanced age. Several physiological processes are observed in aging tissues including a low level of chronic inflammation (inflammaging), changed lipid and energy metabolism, and senescence. Nevertheless, whereas everyone ages, only a subset of the population develops AMD. The purpose of this review is to delineate the differences on a cellular and molecular level between natural aging change

Implantation is a complex and tightly regulated process essential for the establishment of pregnancy. It involves dynamic interactions between a receptive uterus and a competent embryo, orchestrated by ovarian hormones such as estrogen and progesterone. These hormones regulate proliferation, differentiation, and gene expression within the three primary uterine tissue types: myometrium, stroma, and epithelium. Advances in genetic manipulation, particularly the Cre/loxP system, have enabled the in vivo investigation of the role of genes in a uterine compartmental and cell type-specific manner, p

The inherited arrhythmia syndromes long QT syndrome, Brugada syndrome and catecholaminergic polymorphic ventricular tachycardia are rare cardiac channelopathies associated with a higher risk for malignant arrhythmias and sudden cardiac death. Adequate and timely clinical management of patients remains a challenge and is often based on expert opinions and (small) cohort studies. Particularly, identification of high-risk patients continues to be difficult due to limitations in diagnostic tools and risk prediction models. Importantly, underlying causes determining the large variability in disease

Nephronophthisis (NPHP) is a recessive tubulointerstitial nephropathy and a leading genetic cause of kidney failure in children and young adults. The most common genetic cause is a homozygous deletion of NPHP1, which encodes nephrocystin-1, a protein essential for primary cilium structure and cell junctions. Using personalized medicine and deep phenotyping, we investigated a family with three siblings carrying a homozygous NPHP1 deletion. We compared kidney biopsy tissue and human urine-derived renal epithelial cells (hURECs) from these individuals. Bulk RNA-seq on patient hURECs revealed alte

Early-life abilities involved in perceiving, producing and engaging with music (musicality) may shape later (social) communication and language abilities. Here, we investigate phenotypic and genetic relationships linking musicality and communication abilities by studying information from preschool and school-aged children of the Avon Longitudinal Study of Parents and Children (N = 4169-6737 per measure, age 0.5-17 years). Using structural models, we identified relationships between latent musicality and speech- and cognition-related variables (r > 0.30). Consistently, polygenic scores for rhyt

Alzheimer's disease (AD) is an incurable neurodegenerative disorder that leads to dementia. This study employs explainable machine learning models to detect dementia cases using blood gene expression, single nucleotide polymorphisms (SNPs), and clinical data from Alzheimer's Disease Neuroimaging Initiative (ADNI). Analyzing 623 ADNI participants, we found that the Support Vector Machine classifier with Mutual Information (MI) feature selection, trained on all three data modalities, achieved exceptional performance (accuracy = 0.95, AUC = 0.94). When using gene expression and SNP data separatel
