Modulation of the cortical processing of novel and target stimuli by drugs affecting glutamate and GABA neurotransmission.

In this double-blind, placebo-controlled study, we examined the effects of subanaesthetic doses of ketamine (an NMDA glutamate receptor antagonist) and thiopental (a GABA-A receptor agonist) on the event-related potential (ERP) correlates of deviant stimulus processing in 24 healthy adults. Participants completed three separate pharmacological challenge sessions (ketamine, thiopental, saline) in a counterbalanced order. EEG data were recorded both before and during each challenge while participants performed a visual 'oddball' task consisting of infrequent 'target' and 'novel' stimuli intermix

Nicotine modulates reorienting of visuospatial attention and neural activity in human parietal cortex.

Prior studies in animals and humans indicate that reorienting of visuospatial attention is modulated by the cholinergic agonist nicotine. We have previously identified neural correlates of alerting and reorienting attention in humans and found that the parietal cortex is specifically involved in reorienting. This study investigates whether the alerting and reorienting systems, especially in the parietal cortex, are modulated by nicotine. We used event-related functional magnetic resonance imaging (fMRI) and studied 15 nonsmoking volunteers under placebo and nicotine (NICORETTE) polacrilex gum

Regional brain activation evoked when approaching a virtual human on a virtual walk.

We investigated the necessity of biological motion for activation of the posterior superior temporal sulcus (pSTS) in circumstances in which the rapid approach of the observer to a virtual human induced the observer to make inferences about the characters intentions. Using a virtual reality environment, subjects experienced themselves walking towards a complex scene composed of animate and/or inanimate objects. During "person" trials, the scene contained a virtual human either making a simple gesture such as scratching his face (Study 1) or standing completely still (Study 2). During "object"

Longitudinal Assessment of Rod Function in Intermediate Age-Related Macular Degeneration With and Without Reticular Pseudodrusen.

PURPOSE: To evaluate rod function longitudinally in intermediate age-related macular degeneration subjects with reticular pseudodrusen (RPD) and without RPD (AMD). METHODS: Retinal sensitivities (505 and 625 nm) during dark adaptation, at 14 locations within the central 12° macula were obtained after photobleaching at baseline and 12-month visits. Pointwise sensitivity differences between both stimuli were used to assess static rod function, while rod intercept time (RIT) and rod recovery rate (RRR) were used to evaluate dynamic function. Changes in function over time were compared between gr

Chromatic pupillography in hemianopia patients with homonymous visual field defects.

PURPOSE: The pupil light reflex is considered to be a simple subcortical reflex. However, many studies have proven that patients with isolated occipital lesions with homonymous hemianopia show pupillary hemihypokinesia. Our hypothesis is that the afferent pupillary system consists of two pathways: one via intrinsically photosensitive retinal ganglion cells (ipRGCs), the other running through the normal RGCs via the visual cortex. The purpose of this study was to test the hypothesis of these two separate pupillomotor pathways. METHODS: 12 patients (59.1 ± 18.8 years) with homonymous hemianopia

Two-color pupillometry in enhanced S-cone syndrome caused by NR2E3 mutations.

PURPOSE: The purpose of this study was to evaluate pupillary light reflexes (PLRs) mediated by rod, cone, and intrinsically photosensitive retinal ganglion cell pathways as indices of outer- and inner-retinal function in patients who have enhanced S-cone syndrome (ESCS) due to NR2E3 mutations. METHODS: Four patients with ESCS (ages 16-23 years) participated in the study. Subjects were tested with long- and short-wavelength single-flash full-field ERG stimuli under light-adapted conditions. They were also tested with an established pupillometry protocol involving 1-s duration, long- and short-

Preservation of episodic memory in semantic dementia: The importance of regions beyond the medial temporal lobes.

Episodic memory impairment represents one of the hallmark clinical features of patients with Alzheimer's disease (AD) attributable to the degeneration of medial temporal and parietal regions of the brain. In contrast, a somewhat paradoxical profile of relatively intact episodic memory, particularly for non-verbal material, is observed in semantic dementia (SD), despite marked atrophy of the hippocampus. This retrospective study investigated the neural substrates of episodic memory retrieval in 20 patients with a diagnosis of SD and 21 disease-matched cases of AD and compared their performance

Temporal and visual source memory deficits among ecstasy/polydrug users.

OBJECTIVES: We wished to investigate whether source memory judgements are adversely affected by recreational illicit drug use. METHOD: Sixty-two ecstasy/polydrug users and 75 non ecstasy users completed a source memory task, in which they tried to determine whether or not a word had been previously presented and if so, attempted to recall the format, location and temporal position in which the word had occurred. RESULTS: While not differing in terms of the number of hits and false positive responses, ecstasy/polydrug users adopted a more liberal decision criterion when judging if a word had

Neural correlates of visual motion processing without awareness in patients with striate cortex and pulvinar lesions.

Patients with striate cortex lesions experience visual perception loss in the contralateral visual field. In few patients, however, stimuli within the blind field can lead to unconscious (blindsight) or even conscious perception when the stimuli are moving (Riddoch syndrome). Using functional magnetic resonance imaging (fMRI), we investigated the neural responses elicited by motion stimulation in the sighted and blind visual fields of eight patients with lesions of the striate cortex. Importantly, repeated testing ensured that none of the patients exhibited blindsight or a Riddoch syndrome. Th

An independent SSVEP-based brain-computer interface in locked-in syndrome.

OBJECTIVE: Steady-state visually evoked potential (SSVEP)-based brain-computer interfaces (BCIs) allow healthy subjects to communicate. However, their dependence on gaze control prevents their use with severely disabled patients. Gaze-independent SSVEP-BCIs have been designed but have shown a drop in accuracy and have not been tested in brain-injured patients. In the present paper, we propose a novel independent SSVEP-BCI based on covert attention with an improved classification rate. We study the influence of feature extraction algorithms and the number of harmonics. Finally, we test online c

Impaired texture segregation but spared contour integration following damage to right posterior parietal cortex.

We examined the relations between texture segregation and contour integration in patients with deficits in spatial attention leading to left or right hemisphere extinction. Patients and control participants were presented with texture and contour stimuli consisting of oriented elements. We induced regularity in the stimuli by manipulating the element orientations resulting in an implicit texture border or explicit contour. Participants had to discriminate curved from straight shapes without making eye movements, while the stimulus presentation time was varied using a QUEST procedure. The resul

Clutter modulates the representation of target objects in the human occipitotemporal cortex.

Target objects required for goal-directed behavior are typically embedded within multiple irrelevant objects that may interfere with their encoding. Most neuroimaging studies of high-level visual cortex have examined the representation of isolated objects, and therefore, little is known about how surrounding objects influence the neural representation of target objects. To investigate the effect of different types of clutter on the distributed responses to target objects in high-level visual areas, we used fMRI and manipulated the type of clutter. Specifically, target objects (i.e., a face and

Effects of citalopram and escitalopram on fMRI response to affective stimuli in healthy volunteers selected by serotonin transporter genotype.

This study was designed to assess whether functional magnetic resonance imaging (fMRI) following antidepressant administration (pharmaco-fMRI) is sufficiently sensitive to detect differences in patterns of activation between enantiomers of the same compound. Healthy adult males (n=11) participated in a randomized, double-blind, cross-over trial with three medication periods during which they received citalopram (racemic mixture), escitalopram (S-citalopram alone), or placebo for 2 weeks. All participants had high expression serotonin transporter genotypes. An fMRI scan that included passive vi

Neural correlates of fear of movement in high and low fear-avoidant chronic low back pain patients: an event-related fMRI study.

The fear-avoidance model postulates that in chronic low back pain (CLBP) a fear of movement is acquired in the acute phase, which leads to subsequent avoidance of physical activity and contributes to the pain syndrome's becoming chronic. In the present event-related functional magnetic resonance imaging (fMRI) study of the neural correlates of the fear of movement, 60 women (30 CLBP patients, 15 healthy controls, and 15 women with spider phobia; mean age 46.8±9.8 years) participated. The CLBP patients were divided into a high and low fear-avoidant group on the basis of the Tampa Scale of Kines

Volitional reduction of anterior cingulate cortex activity produces decreased cue craving in smoking cessation: a preliminary real-time fMRI study.

Numerous research groups are now using analysis of blood oxygen level-dependent (BOLD) functional magnetic resonance imaging (fMRI) results and relaying back information about regional activity in their brains to participants in the scanner in 'real time'. In this study, we explored the feasibility of self-regulation of frontal cortical activation using real-time fMRI (rtfMRI) neurofeedback in nicotine-dependent cigarette smokers during exposure to smoking cues. Ten cigarette smokers were shown smoking-related visual cues in a 3 Tesla MRI scanner to induce their nicotine craving. Participants

Inactivation of the parietal reach region causes optic ataxia, impairing reaches but not saccades.

Lesions in human posterior parietal cortex can cause optic ataxia (OA), in which reaches but not saccades to visual objects are impaired, suggesting separate visuomotor pathways for the two effectors. In monkeys, one potentially crucial area for reach control is the parietal reach region (PRR), in which neurons respond preferentially during reach planning as compared to saccade planning. However, direct causal evidence linking the monkey PRR to the deficits observed in OA is missing. We thus inactivated part of the macaque PRR, in the medial wall of the intraparietal sulcus, and produced the h

fMRI reactivity to high-calorie food pictures predicts short- and long-term outcome in a weight-loss program.

Behavioral studies have suggested that food cues have stronger motivating effects in obese than in normal-weight individuals, which may be a risk factor underlying obesity. Previous cross-sectional neuroimaging studies have suggested that this difference is mediated by increased reactivity to food cues in parts of the reward system in obese individuals. To date, however, only a few prospective neuroimaging studies have been conducted to examine whether individual differences in brain activation elicited by food cues can predict differences in weight change. We used functional magnetic resonanc

Implicit and explicit reward learning in chronic nicotine use.

BACKGROUND: Chronic tobacco use is related to specific neurobiological alterations in the dopaminergic brain reward system that can be termed "reward deficiency syndrome" in dependent nicotine consumers. The close linkage of dopaminergic activity and reward learning led us to expect implicit and explicit reward learning deficits in dependent compared to non-smokers. Smokers who maintain a less regular, occasional use may also, to a lesser extent, show implicit reward learning deficits. The purpose of our study was to examine the behavioral effects of the neurobiological alterations on reward r

Contralesional cross-over in chronic neglect: visual search patterns reveal neglect of the ipsilesional hemispace.

INTRODUCTION: With neuropsychological rehabilitation approaches neglect patients can learn to compensate for the reduced awareness of the contralesional hemispace that is often observed after right brain damage. Here, we report contralesional "cross-over" deviations in line bisection that are hypothesized to be a result of focusing on the contralesional hemispace while the intact hemispace is "neglected". We investigate whether this unexpected pattern of deviation is related to defects in the visual field, motor intention/hypokinesia deficits or deficits in working memory. METHODS: Neglect pa
