Research progress on the mechanism underlying the application of mesenchymal stem cells in the treatment of male infertility.

Male infertility has become an increasingly prominent health issue worldwide. This review systematically evaluated the therapeutic application of different types of MSCs in various male infertility models. The therapeutic effects of MSCs are attributed to mechanisms such as in vivo and in vitro differentiation into germ cells, improved antioxidant capacity of testicular tissue, inhibited secretion of inflammatory factors and elevated anti-inflammatory level of testicular tissue, prevention of excessive apoptosis of testicular tissue cells, restoration of the normal secretion of sexual hormone

Morphometric, Biomechanical and Macromolecular Performances of β-TCP Macro/Micro-Porous Lattice Scaffolds Fabricated via Lithography-Based Ceramic Manufacturing for Jawbone Engineering.

Effective bone tissue regeneration remains pivotal in implant dentistry, particularly for edentulous patients with compromised alveolar bone due to atrophy and sinus pneumatization. Biomaterials are essential for promoting regenerative processes by supporting cellular recruitment, vascularization, and osteogenesis. This study presents the development and characterization of a novel lithography-printed ceramic β-TCP scaffold, with a macro/micro-porous lattice, engineered to optimize osteoconduction and mechanical stability. Morphological, structural, and biomechanical assessments confirmed a re

Engineered Animal Models Designed for Investigating Ethanol Metabolism, Toxicity and Cancer.

Excessive consumption of alcohol is a leading cause of lifestyle-induced morbidity and mortality worldwide. Although long-term alcohol abuse has been shown to be detrimental to the liver, brain and many other organs, our understanding of the exact molecular mechanisms by which this occurs is still limited. In tissues, ethanol is metabolized to acetaldehyde (mainly by alcohol dehydrogenase and cytochrome p450 2E1) and subsequently to acetic acid by aldehyde dehydrogenases. Intracellular generation of free radicals and depletion of the antioxidant glutathione (GSH) are believed to be key steps i

Transplantation of periodontal ligament cell sheets expressing human β‑defensin‑3 promotes anti‑inflammation in a canine model of periodontitis.

Periodontitis is a chronic oral inflammatory disease caused by microorganisms. Human β‑defensin‑3 (HBD‑3) is an endogenous antimicrobial peptide that inhibits a broad spectrum of microorganisms. Cell sheet technology has been widely applied in tissue and organ reconstructions. In the current study, it was aimed to investigate the anti‑inflammatory effect of periodontal tissue engineered by HBD‑3 gene‑modified periodontal ligament cell (PDLC) sheets, and to identify a suitable method of promoting the regeneration of periodontal tissues. Western blot analysis and antimicrobial tests were used to

Flow interactions with cells and tissues: cardiovascular flows and fluid-structure interactions. Sixth International Bio-Fluid Mechanics Symposium and Workshop, March 28-30, 2008, Pasadena, California.

Interactions between flow and biological cells and tissues are intrinsic to the circulatory, respiratory, digestive and genitourinary systems. In the circulatory system, an understanding of the complex interaction between the arterial wall (a living multi-component organ with anisotropic, nonlinear material properties) and blood (a shear-thinning fluid with 45% by volume consisting of red blood cells, platelets, and white blood cells) is vital to our understanding of the physiology of the human circulation and the etiology and development of arterial diseases, and to the design and development

On the track of mesothelial progenitor cells from the peritoneal cavity transplanted to immunodeficient mice.

Mesothelial progenitor cells have been reported to reside in either the monolayer of mesothelium, submesothelium or within the peritoneal cavity as free floating cells. As a putative plasticity has been reported for the mesothelial progenitor cells and considering the potential implications of the establishment of a novel resource of stem/progenitor cells in gene and cell therapeutics and tissue engineering, we conducted an in vivo tracking of transplanted mesothelial cells. In order to induce immunodeficiency, the recipient mice were treated with 32 mg kg(-1) of daily Cyclosporine. On days 14

Ectopic expression of the gastric inhibitory polypeptide receptor gene is a sufficient genetic event to induce benign adrenocortical tumor in a xenotransplantation model.

Aberrant expression of ectopic G protein-coupled receptors (GPCRs) in adrenal cortex tissue has been observed in several cases of ACTH-independent macronodular adrenal hyperplasias and adenomas associated with Cushing's syndrome. Although there is clear clinical evidence for the implication of these ectopic receptors in abnormal regulation of cortisol production, whether this aberrant GPCR expression is the cause or the consequence of the development of an adrenal hyperplasia is still an open question. To answer it, we genetically engineered primary bovine adrenocortical cells to have them exp

An engineered probiotic Escherichia coli Nissle 1917 strain for Amuc_1100 delivery in a CT26 colorectal tumor model.

Engineered probiotics provide a microbial platform for the localized delivery of bioactive proteins. Escherichia coli Nissle 1917 (EcN) is an attractive bacterial chassis for this purpose because of its ability to colonize tumors and its genetic tractability. Amuc_1100, an outer membrane protein of Akkermansia muciniphila, has shown antitumor and immunomodulatory effects after oral administration. However, whether EcN-based delivery can improve the antitumor performance of microbiome-derived proteins such as Amuc_1100 remains unclear. Here, EcN was engineered with a pelB-His-Amuc_1100 expressi

One organ, two engines: Divergent mitochondrial phenotypes in anterior and posterior crab gills.

Gills are multifunctional organs that integrate respiration with homeostasis, including energy demanding processes such as osmoregulation and excretion. In osmoregulating decapod crustaceans, two spatially segregated gill types differ in function, ultrastructure and membrane composition, as well as in their responses to environmental change. Yet mitochondrial function in crab gills remains poorly characterized. Here, for the first time, we used high-resolution respirometry with a substrate-uncoupler-inhibitor titration protocol to characterize the mitochondrial phenotypes in anterior (respirat

Engineered Tandem Thymosin Peptide Promotes Corneal Wound Healing.

PURPOSE: Thymosin beta-4 (TB4) is a small peptide upregulated in injured tissues, playing roles in cell migration, angiogenesis, inflammation, and oxidative stress. Studies show TB4 significantly promotes corneal wound healing after injury, leading to a clinical trial (RGN-259), with full US Food and Drug Administration approval still pending. Current limitations to TB4-based therapies are a short half-life and high peptide synthesis costs, limiting large-scale applications. Here, we engineered a tandem thymosin beta-4 (tTB4) peptide with improved therapeutic potential and scalability for corn

RNA interference-mediated hookworm control by gut-dwelling lactic acid bacteria.

BACKGROUND: The complex life cycle of geohelminth parasites make it difficult to manage repeated infections that occur in endemic areas, that requires the development of new therapeutic strategies. Human hookworm Ancylostoma sp. and Necator sp. are a serious problem in many areas of the world where the old age anti-helminthics are ineffective. To address this, we have tried a non-traditional therapeutic approach for a sustainable solution to manage parasite infections. AIMS AND OBJECTIVES: In this study, we attempted to develop a new type of therapy using value-added probiotics-producing anti

Engineering diabetic human skin equivalent for in vitro and in vivo applications.

The prevalence of diabetes is increasing worldwide. Diabetes contributes to 70% of all non-traumatic lower-limb amputations, which are directly caused by diabetic foot ulcers (DFU) that are difficult to heal. Non-healing diabetic ulcers represent one of modern society's most difficult medical challenges. One of the promising initiatives to treat DFU is the grafting of autologous skin or stimulating the skin cells at the edge of the wound to proliferate and close the wound. The present study was to engineer a diabetic human skin equivalent (DHSE) that contains fibroblasts and keratinocytes extr

Mesenchymal stem cells engineered to express selectin ligands and IL-10 exert enhanced therapeutic efficacy in murine experimental autoimmune encephalomyelitis.

Systemic administration of mesenchymal stem cells (MSCs) affords the potential to ameliorate the symptoms of Multiple Sclerosis (MS) in both preclinical and clinical studies. However, the efficacy of MSC-based therapy for MS likely depends on the number of cells that home to inflamed tissues and on the controlled production of paracrine and immunomodulatory factors. Previously, we reported that engineered MSCs expressing P-selectin glycoprotein ligand-1 (PSGL-1) and Sialyl-Lewis(x) (SLeX) via mRNA transfection facilitated the targeted delivery of anti-inflammatory cytokine interleukin-10 (IL-1

BaTiO3-based Piezoelectric Wireless Neuromodulation: From Molecular Mechanisms to Clinical Translation Challenges.

Neurological injuries remain difficult to treat due to the complex microenvironment and the limited intrinsic regenerative capacity of neural tissues. Recently, wireless neuromodulation based on piezoelectric nanomaterials, particularly barium titanate (BaTiO3), has emerged as a promising alternative to conventional implantable electrical stimulation. Polar BaTiO3 ceramics generally have higher piezoelectric charge coefficients than conventional Polyvinylidene Fluoride (PVDF) and Poly(vinylidene fluoride-co-trifluoroethylene) (P(VDF-TrFE)) films, are more resistant to degradation than ZnO, and

Hydroxyapatite-decorated Fmoc-hydrogel as a bone-mimicking substrate for osteoclast differentiation and culture.

Hydrogels are water-swollen networks with great potential for tissue engineering applications. However, their use in bone regeneration is often hampered due to a lack of materials' mineralization and poor mechanical properties. Moreover, most studies are focused on osteoblasts (OBs) for bone formation, while osteoclasts (OCs), cells involved in bone resorption, are often overlooked. Yet, the role of OCs is pivotal for bone homeostasis and aberrant OC activity has been reported in several pathological diseases, such as osteoporosis and bone cancer. For these reasons, the aim of this work is to

Three-Dimensional Scaffolds for Bone, Soft Tissue, and Periodontal Regeneration.

The oral cavity presents three distinct regenerative challenges: restoration of alveolar bone volume and stability, augmentation of soft mucogingival tissues, and regeneration of the tooth-supporting periodontium. In each of these clinical contexts, conventional approaches remain limited by the morbidity associated with autologous tissue harvesting and by the difficulty in predictably reconstructing the complex architecture and functional integration required for the long-term stability of these distinct tissues. Although currently available biomaterials have demonstrated clinical effectivenes

Bioenergetic Materials for Tissue Regeneration: Modulating Metabolism to Promote Cellular Anabolism.

Tissue regeneration is an energy-demanding process that requires adequate ATP production to support cellular proliferation, biosynthesis, and tissue remodeling. Under pathological and age-related conditions, bioenergetic deficits impair intrinsic regenerative capacity by disrupting mitochondrial function, redox homeostasis, and anabolic signaling. Although conventional scaffold- and cell-based bioengineering strategies have advanced regenerative medicine, they generally do not directly address the metabolic dysfunction that limits tissue repair. Recently, bioenergetic materials (BEMs) have eme

Digital light processing of reactive ceramic-polymer bone scaffolds enables In situ hydroxyapatite formation.

The incorporation of calcium phosphates to additive manufacturing, particularly digital light processing (DLP), has gained increasing attention for the fabrication of bone tissue engineering scaffolds. DLP enables the production of patient-specific constructs with complex architectures and high spatial resolution. In this study, we develop photo-crosslinkable resin formulations based on poly(ethylene glycol) diacrylate (PEGDA) loaded with reactive α-tricalcium phosphate (α-TCP) particles for DLP printing. Using a liquid crystal display-based DLP (LCD-DLP) system, composite scaffolds were direc
