Programmable Metal-Organic Framework Biointerfaces Against Pathogens.

Emerging viral diseases continue to pose major challenges to global health, creating demand for materials that can support pathogen control, diagnosis, and therapy. Owing to their tunable structures and versatile biointerfaces, metal-organic frameworks (MOFs) have attracted increasing attention in anti-pathogen applications. While previous studies have often focused on individual functions such as catalysis, biosensing, or drug delivery, a broader perspective on the functional development of MOF-based systems remains limited. In this Review, we summarize recent advances in MOF-enabled pathogen

Overcoming therapeutic resistance in oncology: The prospects of inorganic nanotheranostics.

Cancer remains a leading cause of global mortality, with therapeutic resistance posing a formidable clinical obstacle. Inorganic nanotheranostics-multifunctional platforms integrating diagnostic and therapeutic capabilities-offer a promising strategy to overcome this barrier. This review systematically examines how distinct classes of inorganic nanomaterials (including iron, gadolinium, titanium, gold, manganese, carbon, and silicon-based systems) are engineered to counteract specific resistance mechanisms through unique physicochemical properties and biological interactions. We highlight thei

MASH and the race for liver antifibrotics.

Metabolic dysfunction-associated steatohepatitis (MASH), along with other chronic liver diseases, leads to progressive fibrosis and, ultimately, cirrhosis. Liver fibrosis is a major cause of global morbidity and mortality. Although past efforts to develop antifibrotic drugs have largely failed, recent advances in MASH metabolic therapies offer new hope. These include both indirect-acting agents such as glucagon-like peptide 1 (GLP-1) analogues, which reduce liver fat by promoting weight loss, and therapies with direct-acting mechanisms on the liver, such as thyroid hormone receptor beta (THRβ)

The double-edged role of interferon gamma in cancer immunity.

Interferon-gamma (IFN-γ) is a pivotal cytokine that coordinates the immune response to infections caused by viruses and intracellular pathogens. Knockout (KO) mice lacking IFN-γ exhibited a significant reduction in the capacity of cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells to eliminate tumor cells, resulting in accelerated tumor growth and metastasis. These findings underscore the key role of IFN-γ in regulating the antitumor immune response. However, many advanced tumors exhibit dysregulated IFN-γ signaling, which exerts direct protumoral and immunosuppressive effects, there

Long non-coding RNAs in tuberculosis: Immune regulation and clinical biomarker potential.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains the leading infectious cause of mortality worldwide, with an estimated 1.25 million deaths in 2023. Despite therapeutic advances, the lack of precise early screening tools continues to impede epidemic control. Long non-coding RNAs (lncRNAs) have emerged as essential modulators of host anti-Mtb immunity, and elucidating their functional mechanisms is pivotal for developing effective interventions. This review systematically synthesizes lncRNA-mediated immune regulation during Mtb infection, with emphasis on their roles in mo

[Sleep breathing impairment index: a novel metric for assessing the severity of obstructive sleep apnea].

Obstructive sleep apnea (OSA) is a highly heterogeneous disorder of sleep-related breathing, and intermittent hypoxia during sleep constitutes a key pathophysiological mechanism underlying the increased risk of cardiovascular disease (CVD) in affected individuals. The sleep breathing impairment index (SBII) is a novel, multidimensional index for assessing OSA severity. By integrating the frequency and duration of respiratory events along with the corresponding oxygen desaturation area, SBII quantitatively captures both the occurrence of respiratory disturbances and the associated hypoxemic bur

Age and early life adversity shape heterogeneity of the epigenome across tissues in macaques.

Age and early life adversity (ELA) are key determinants of health, but whether they affect similar physiological mechanisms across tissues is unknown. We generated DNA methylation (DNAm) profiles across 14 tissues in 237 semi-free-ranging rhesus macaques with naturally occurring ELA. Age-associated DNAm was predominantly tissue dependent, yet tissue-specific epigenetic clocks showed that epigenetic aging was relatively consistent within individuals. ELA effects were adversity dependent, but each ELA exerted coordinated effects across tissues. Although ELA targeted many of the same loci as age,

Emerging Oncolytic Viruses: Beyond Adenoviruses and Herpes Simplex Virus.

Oncolytic virotherapy represents an innovative therapeutic modality in oncology, exploiting the selective tropism of viruses to target and eradicate malignant cells. Unlike conventional cancer treatments such as chemotherapy and radiotherapy, oncolytic viruses (OVs) exhibit a dual mechanism of action: direct tumor cell lysis and potent immune activation. This approach transforms tumors into in situ vaccines, generating durable anti-tumor immune memory. The field has garnered substantial attention following the regulatory approval of talimogene laherparepvec (T-VEC), a genetically engineered he

Machine learning for discovering anticancer compounds from plants and elucidating their mechanisms.

Plants are important sources of bioactive compounds with demonstrated anticancer and cancer-preventive properties. However, traditional methods for discovering and deciphering the mechanisms of these compounds are often slow and labor-intensive. The integration of machine learning (ML) provides a computational approach for bioactivity prediction, candidate prioritization, and mechanistic hypothesis generation. Algorithms including support vector machines (SVM) and graph neural networks (GNN) are increasingly employed to support multiple stages from compound screening to experimentally guided m

Tracking Tool for Fiddler Crabs in Natural Settings to Promote a Model Organism for Synchrony.

Synchronization has been studied across vastly different spatiotemporal scales in physical, biological, and social systems. Research into the mechanisms that give rise to temporal order, however, has been, by and large, mostly theoretical. Here, we seek to advance the fiddler crab as a model organism for collective synchronization. To attract mates, males of many fiddler crab species wave their claws in sync. These crabs are found in many places around the world, so they are generally accessible and observable. Translating observation (e.g., from recorded video) into actionable data, however,

Topological defects mediate collective transport of confluent cells.

Collective cell migration governs a range of physiological and pathological processes, from tissue morphogenesis to cancer invasion, in which topological defects arise as an inevitable consequence of frequent cellular rearrangement and migration. Here, we employ the active vertex model to investigate structural defects generated in the wake of transported cells. We find that while the drag coefficient of a cell in a perfect lattice is anisotropic, the threshold drag force required to mobilize the cell is isotropic. Remarkably, we find that dragging two neighboring cells along the direction of

Signaling lipids: An overview of emerging physiological functions.

Lipids are now recognized as central regulators of cellular signaling, extending well beyond their traditional roles in membrane structure and energy storage. As spatially confined and rapidly inducible messengers, signaling lipids integrate membrane dynamics, metabolism, and signal transduction to control processes like inflammation, immune responses, vesicular trafficking, cytoskeletal organization, and cell fate decisions. This review highlights key mechanistic principles underlying lipid signaling specificity, including localized biosynthesis, enzymatic turnover, and receptor engagement. W

Neuroimmunology of Epilepsy: Mechanisms, Treatments, and Clinical Insights.

Immune-mediated epilepsies represent an etiologically unique category defined by immune activation within the central nervous system that initiates a pathophysiological cascade resulting in seizures, encephalopathy, and sometimes epileptogenesis with long-term epilepsy. Once considered rare and largely confined to paraneoplastic or postinfectious syndromes, immune-mediated and inflammatory mechanisms are now recognized across a broad spectrum ranging from acute autoimmune encephalitis with reversible seizures, new-onset refractory status epilepticus, or febrile illness-related refractory statu

Bridging Nano-Interface Interactions and Organ-Specific Toxicity: A Review of Machine Learning for Nanomaterials Risk Assessment.

Risk assessment of engineered nanomaterials (ENMs) is essential for protecting human health and the environment. Traditional hazard assessments rely primarily on in vivo testing, which faces technical challenges in extrapolation validity, ethical dilemmas, and high costs. Machine learning (ML) models offer alternative approaches that are aligned with the 3R principles (Replacement, Reduction, and Refinement) for reducing animal use. ML methods help address the economic, ethical, and temporal limitations of traditional nanotoxicology while advancing mechanistic understanding. This review presen
