Diabetes in pregnancy: islet cell proliferation in the fetal rat pancreas.

Since it has not been possible to reproduce, in the rat, the hyperplasia of the islets of Langerhans observed in the fetus in human diabetic pregnancy, the rate of proliferation of the endocrine pancreas of fetuses of manifest diabetic rats has been studied. Rats were rendered diabetic by streptozotocin injections before mating. At days 20 or 22 of gestation the pregnant rats were injected with colchicine and sacrificed at 1-h intervals. The mitotic indices of the fetal endocrine pancreas were determined and plotted against the time after colchicine injection. The production of new cells (i.e.

Retardative effects of a corticosteroid hormone upon chondrocyte growth in the mandibular condyle of neonatal mice.

This study examined the influence of triamcinolone hexacetonide, a long-acting synthetic analog of cortisol on the proliferative activity and subsequent development of chondroprogenitor cells in condylar cartilage of neonatal mice. It became evident that a single injection of relatively low doses of the hormone significantly reduced the uptake and incorporation of [3H]thymidine followed by a significant decrease in the number of young cartilage cells. A unique feature was the fact that at the same time condyles of triamcinolone-treated mice revealed an increase in the number of mesenchyme-like

Light and electron microscopic study of cellular proliferation in carcinoid heart disease.

An ultrastructural and histochemical study of the subendocardial lesion in carcinoid heart disease showed six different cell types within a myxoid matrix. The matrix, composed of a mucopolysaccharidic ground substance, collagen, and reticluin fibers, contained stem cells, four types of fusiform cells (fibrocytes, fibroblasts, myofibroblasts, and smooth muscle cells), and intermediary cell type. Our observations suggest that the humoral mediators of the carcinoid syndrome may induce the differentiation of a subendocardial stem cell into contractile elements.

The Virtual Biotech: A multi-agent AI framework for therapeutic discovery and development.

Drug development requires evidence integration across biological scales and modalities, but relevant tools are fragmented. We introduce the Virtual Biotech, an organization of artificial intelligence (AI) agents modeled on a drug-development company, with agentic divisions spanning target discovery, safety assessment, modality selection, and clinical development. We demonstrate its utility at three drug-development decision points. First, over 37,000 agents annotated outcomes from 55,984 trials and found that drugs targeting cell-type-specific genes were 48% more likely to reach market with 32

Zonation, Zonation, Zonation: The Real Estate of the Liver.

The liver serves as a central hub for a diverse set of functions including metabolic homeostasis, detoxification, and protein synthesis. While appearing homogeneous, hepatocytes, the major workhorse in the liver, demonstrate spatial identity within the lobule, which in turn dictates gene and protein expression and, eventually, function. Presenting as an axis from the portal triad to the central vein, this organization has been conventionally referred to as metabolic zonation. In recent years, the heterogeneity in expression and function is now understood to extend well beyond hepatocytes and m

Intraoperative hypertensive crisis in a dog with functional paraganglioma of the gall bladder.

A 15-year-old spayed female mongrel presented with anorexia and an abdominal mass. The mass originated from the gall bladder and was surgically resected along with divisionectomy of the central hepatic division. Paroxysmal hypertension and tachycardia were noted during manipulation of the mass. Following resection, arterial blood pressure decreased significantly. Histopathological analysis confirmed a diagnosis of neuroendocrine neoplasm. Immunohistochemical staining for synaptophysin and chromogranin A yielded diffuse and strong positive results, while gastrin was positive in only 10% of the

Post-mortem lung tissue: the fossil record of the pathophysiology and immunopathology of severe COVID-19.

The lungs are the main site that is affected in severe COVID-19, and post-mortem lung tissue provides crucial insights into the pathophysiology of severe disease. From basic histology to state-of-the-art multiparameter digital pathology technologies, post-mortem lung tissue provides snapshots of tissue architecture, and resident and inflammatory cell phenotypes and composition at the time of death. Contrary to early assumptions that COVID-19 in the lungs is a uniform disease, post-mortem findings have established a high degree of disease heterogeneity. Classic diffuse alveolar damage represent

Comprehensive analysis of circRNA expression profiles and circRNA-associated competing endogenous RNA networks in the development of mouse thymus.

The thymus plays an irreplaceable role as a primary lymphoid organ. However, the complicate processes of its development and involution are incompletely understood. Accumulating evidence indicates that non-coding RNAs play key roles in the regulation of biological development. At present, the studies of the circRNA profiles and of circRNA-associated competing endogenous RNAs (ceRNAs) in the thymus are still scarce. Here, deep-RNA sequencing was used to study the biological mechanisms underlying the development process (from 2-week-old to 6-week-old) and the recession process (from 6-week-old t

Human SPG11 cerebral organoids reveal cortical neurogenesis impairment.

Spastic paraplegia gene 11(SPG11)-linked hereditary spastic paraplegia is a complex monogenic neurodegenerative disease that in addition to spastic paraplegia is characterized by childhood onset cognitive impairment, thin corpus callosum and enlarged ventricles. We have previously shown impaired proliferation of SPG11 neural progenitor cells (NPCs). For the delineation of potential defect in SPG11 brain development we employ 2D culture systems and 3D human brain organoids derived from SPG11 patients' iPSC and controls. We reveal that an increased rate of asymmetric divisions of NPCs leads to p

Olfactory Nerve Schwannoma: A Case Report and Review of the Literature.

Schwannomas are benign tumors, which arise from the Schwann cells of the central or peripheral nerves. They form 8% of all intracranial tumors and most of the cases arise from vestibular division of the 8 th cranial nerve. Rare cases are shown to arise from the olfactory or optic nerve, being devoid of myelin sheath. Up to date and according to our best of knowledge, 66 cases have been reported till now. Here we present a review of the literature and a case report of a 56-year-old male with an accidently discovered anterior cranial fossa schwannoma, following a road traffic accident. Tumor was

A novel nonsense GPSM2 mutation in a Yemeni family underlying Chudley-McCullough syndrome.

Mutations in the G Protein Signaling Modulator 2 (GPSM2) cause the autosomal recessive disorder Chudley-McCullough syndrome (CMS), which is characterized by profound congenital sensorineural hearing loss with various abnormalities in the brain. This phenotypic combination is attributed to the role played by GPSM2 in the establishment of planar polarity and spindle orientation during asymmetric cell divisions. Here we present two brothers from a Yemeni family who were diagnosed clinically with CMS then tested for GPSM2 mutations using Sanger sequencing. Consequent to sequencing, in silico tools

Cenpj/CPAP regulates progenitor divisions and neuronal migration in the cerebral cortex downstream of Ascl1.

The proneural factor Ascl1 controls multiple steps of neurogenesis in the embryonic brain, including progenitor division and neuronal migration. Here we show that Cenpj, also known as CPAP, a microcephaly gene, is a transcriptional target of Ascl1 in the embryonic cerebral cortex. We have characterized the role of Cenpj during cortical development by in utero electroporation knockdown and found that silencing Cenpj in the ventricular zone disrupts centrosome biogenesis and randomizes the cleavage plane orientation of radial glia progenitors. Moreover, we show that downregulation of Cenpj in po

Microcephaly-associated protein WDR62 regulates neurogenesis through JNK1 in the developing neocortex.

Mutations of WD40-repeat protein 62 (WDR62) have been identified recently to cause human MCPH (autosomal-recessive primary microcephaly), a neurodevelopmental disorder characterized by decreased brain size. However, the underlying mechanism is unclear. Here, we investigate the function of WDR62 in brain development and the pathological role of WDR62 mutations. We find that WDR62 knockdown leads to premature differentiation of neural progenitor cells (NPCs). The defect can be rescued by wild-type human WDR62, but not by the five MCPH-associated WDR62 mutants. We demonstrate that WDR62 acts upst

Optimizing combination therapies with existing and future CML drugs.

Small-molecule inhibitors imatinib, dasatinib and nilotinib have been developed to treat Chromic Myeloid Leukemia (CML). The existence of a triple-cross-resistant mutation, T315I, has been a challenging problem, which can be overcome by finding new inhibitors. Many new compounds active against T315I mutants are now at different stages of development. In this paper we develop an algorithm which can weigh different combination treatment protocols according to their cross-resistance properties, and find the protocols with the highest probability of treatment success. This algorithm also takes int

Induction of meiotic micronuclei in spermatocytes in vivo by aflatoxin B1: light and transmission electron microscopic study in Swiss mouse.

Dietary aflatoxins produce a disease state known as aflatoxicosis, and disruption of spermatogenesis is one of its serious consequences. Towards finding the cellular targets in spermatogenic compartment for aflatoxin toxicity, aflatoxin B(1) (AFB(1)) was administered to 90-day-old Swiss mouse through i.p. route at a daily dose of 20mug per kg body weight for 7, 15, 35 and 45 days. The testis and epididymis were subjected to light- as well as transmission electron microscopic analysis. One of the newer observations was occurrence of meiotic micronucleate giant spermatocytes in seminiferous epit

Cadherins as regulators for the emergence of neural nets from embryonic divisions.

Cadherins are a large family of cell adhesion molecules that are expressed in a spatially restricted fashion during vertebrate CNS development. Each cadherin shows a characteristic expression pattern that differs from that of other cadherins. Early in development, the cadherin expression domains relate to the neuromeric organization of the vertebrate CNS. Later, as functional structures (brain nuclei, cortical regions, fiber tracts and synapses) emerge, the expression patterns of each cadherin become restricted to subsets of these structures that form parts of specific neural nets. Cadherins t

New concepts regarding the pathogenesis of amebiasis.

Our understanding of the pathogenesis of amebiasis has progressed significantly since Dr. Braude's description of the major clinical syndromes of amebic liver abscess. His hypothesis that invasive amebic strains must be resistant to complement-mediated lysis has been confirmed. We have also shown that Entamoeba histolytica activates complement by a unique mechanism, i.e., cleavage of C3 by an extracellular cysteine proteinase. Cysteine proteinases are important virulence factors encoded by at least three genes; one gene, acp1, is unique to invasive strains. The amebic cysteine proteinases are

[Histological observations of the capsules induced by Contortylenchus diplogaster v. Lins (Nematoda: Allantonematidae) in the adipose tissue of Ips sexdentatus Boern. (Coleoptera: Scolytidae)].

The evolution of the capsules induced by C. diplogaster in the fat body of I. sexdentatus is followed by histological methods from the pupal stage to the oviposition of the host. Early in the pupal stage of the insect, the parasitic female of C. diplogaster has not oviposited yet and the nuclei of the capsule that surrounds it are gathered in tetrads; some of these nuclei are in a well forwarded degenerescence. These observations suggest that the syncytial capsule should result both from an amitotic division of the nuclei followed by a degenerescence of some of them and from a fusion of cells.
