Brain and sense organ anatomy and histology of two species of phyletically basal non-Antarctic thornfishes of the Antarctic suborder Notothenioidei (Perciformes: Bovichtidae).

The predominantly non-Antarctic family Bovichtidae is phyletically basal within the perciform suborder Notothenioidei, the dominant component of the Antarctic fish fauna. In this article we focus on the South Atlantic bovichtids Bovichtus diacanthus, the klipfish from tide pools at Tristan da Cunha, and Cottoperca gobio, the frogmouth from the Patagonian shelf and Falkland Islands. We document the anatomy and histology of the brains, olfactory apparatus, retina, and cephalic lateral line system. We also use the microvascular casting agent Microfil to examine ocular vascular structures. We prov

Seasonal Variations in the Toughness of Leaves: A Case Study Using Griselinia littoralis.

Potential effects of climate change include greater extremes of temperature and increased severity of storms. Many plants have evolved to resist the challenges of winter (freezing, dehydration, and wind) in a process known as cold hardening. Sensing reducing temperatures, they make structural changes at the cellular level to increase their mechanical resistance and prevent damage. Previous work on this topic, though extensive, has been conducted under laboratory conditions rather than in the field, and while many workers have observed changes to cell wall thickness and composition, which imply

Characterization of Retinal Structure in ATF6-Associated Achromatopsia.

PURPOSE: Mutations in six genes have been associated with achromatopsia (ACHM): CNGA3, CNGB3, PDE6H, PDE6C, GNAT2, and ATF6. ATF6 is the most recent gene to be identified, though thorough phenotyping of this genetic subtype is lacking. Here, we sought to test the hypothesis that ATF6-associated ACHM is a structurally distinct form of congenital ACHM. METHODS: Seven genetically confirmed subjects from five nonconsanguineous families were recruited. Foveal hypoplasia and the integrity of the ellipsoid zone (EZ) band (a.k.a., IS/OS) were graded from optical coherence tomography (OCT) images. Ima

Type 2 diabetes disrupts T-tubule and RyR2 organization in male but not in female rat ventricular muscle.

Females with diabetes often experience more severe cardiovascular outcomes than males with diabetes. It is unclear whether these sex-specific outcomes are rooted in distinct cellular alterations of key cardiac excitation-contraction coupling (ECC) proteins, specifically transverse tubules (T-tubules), ryanodine receptors (RyR2), and filamentous actin (F-actin). This study investigated myocardial ECC structures in both sexes under type 2 diabetic conditions and measured cardiac functional outcomes at muscle level. Type 2 diabetes was induced in male and female Wistar rats using a high-fat diet

Structural basis for ITP-selective hydrolysis by a CRISPR-associated Nudix hydrolase.

The nucleoside diphosphates linked to moiety-X (Nudix) hydrolases are a superfamily of proteins defined by the characteristic Nudix box sequence, most of which are involved in clearing the cell of potentially deleterious metabolites and in preventing the accumulation of metabolic intermediates. The Nudix hydrolase associated with CRISPR-Cas-associated adenosine deaminases (CAAD) from Limisphaera ngatamarikiensis (LngNH) mitigates CAAD-induced toxicity by hydrolyzing ITP to IMP and pyrophosphate. Here, we determined crystal structures of LngNH in the apo form and in complex with ITP using catal

First report of novel single-nucleotide polymorphisms and genetic characteristics in the open reading frame of the prion protein gene (PRNP) in bats.

BACKGROUND: Prion diseases are fatal neurodegenerative disorders capable of causing transmissible spongiform encephalopathies (TSEs) and have been reported in humans and several animal species. Genetic polymorphisms in the prion protein gene (PRNP) are known to influence the conformational conversion of the normal cellular prion protein (PrPC) into its pathogenic isoform (PrPSc), thereby affecting disease susceptibility and pathogenesis. However, single-nucleotide polymorphisms (SNPs) in the PRNP gene have not been reported in bats, which remain free from prion disease reports despite their wi

Nanoscale interrogation of α-Synuclein oligomers: Bridging biophysics, interface science, and neurobiology.

α-Synuclein (α-syn) is an intrinsically disordered presynaptic protein. In synucleinopathies, it undergoes a structural transition into β-sheet-rich conformers that promote the formation of amyloid fibrils and pathological inclusions. Although fibrillar aggregates have been studied extensively, soluble oligomers, which may have the greatest neurotoxic potential, remain poorly understood because of their transient nature and structural heterogeneity. This review critically examines α-syn oligomeric species, with particular emphasis on advanced biophysical and surface characterization techniques

Structure-Guided Immunoinformatics for the Rational Design of a Multi-Epitope Vaccine Against Batai Orthobunyavirus.

Background/Objectives: Batai orthobunyavirus (BATV) is an emerging mosquito-borne zoonotic pathogen for which no licensed vaccine is currently available. The viral envelope glycoprotein plays an important role in viral attachment and host immune recognition, making it a potential target for rational vaccine design. Methods: In this study, an immunoinformatics-based framework was used to design and evaluate a multi-epitope vaccine candidate targeting the BATV envelope glycoprotein. Selected B-cell, cytotoxic T-lymphocyte (CTL), and helper T-lymphocyte (HTL) epitopes were assembled using appropr

Nuclear molecular imaging of venous thromboembolism: targeting clot-specific molecular components for improved diagnosis and management.

Venous thromboembolism (VTE), manifesting as deep vein thrombosis and pulmonary embolism, is a common, recurrent and potentially fatal condition. Anticoagulation remains the cornerstone of therapy, yet determining optimal treatment duration (eg, decision to shorten or extend anticoagulant treatment) is difficult due to the uncertain balance between long-term bleeding risk and recurrence. Future advances in vascular nuclear molecular imaging could critically help support this decision-making process. Current VTE diagnostic imaging relies on structural or perfusion-based techniques to infer thro

Structural and functional insights into a mesophilic cold shock protein CspA with enhanced precision.

Cold-shock proteins (CSPs) are highly conserved nucleic acid-binding proteins that act as chaperones during cellular adaptation to low temperatures. Here, we present a comprehensive structural and dynamic characterization of Escherichia coli CspA using high-resolution NMR spectroscopy. The solution structure of CspA (PDB ID: 30IB) is supported by extensive NMR experimental restraints, minimal violations, and favorable stereochemistry, establishing it as a well-converged NMR structure. Crucially, we investigated backbone dynamics across multiple timescales, with a particular focus on the micros

Conformational insights into the unfolding pathway of RNA recognition motif (RRM) domain of TAF15 associated with amyotrophic lateral sclerosis.

The RNA recognition motif (RRM) domain of TAF15 protein plays an essential role in RNA binding and regulation of diverse cellular processes. However, the molecular mechanism underlying its structural stability and folding behaviour associated with disease state remains poorly understood. Herein, we performed an extensive molecular dynamics (MD) simulations study to investigate the conformational dynamics and unfolding pathway of the TAF15-RRM domain in 8 M urea. The elevated temperatures (300-500 K) were employed to accelerate the unfolding and capture different conformational states. The resu

Will I see you again? The promise, power, and problems of retinal regeneration in fish.

The vertebrate neural retina, the image forming component of the eye, offers a useful, clinically-relevant system for understanding nervous system development and regeneration. Within vertebrates, the teleost fish retina specifically has been the subject of intense study for its regenerative response to neuronal damage - a response that does not take place in mammals. While the field has recently focused upon molecular and cellular processes that initiate this response and generate new neurons, it is also important to evaluate the ultimate "success" of this process. True neural regeneration re

Transcriptional signatures of cortical morphometric variability in temporal lobe epilepsy.

OBJECTIVE: Widespread cognitive deficits in temporal lobe epilepsy (TLE) are associated with macroscale brain structural changes. However, the correlation between the changes in the cortical morphological similarity of the TLE and gene expression remains unclear. METHODS: Structural magnetic resonance imaging (MRI) was collected from 31 TLE patients and 35 healthy controls to construct an individual-level Morphometric INverse Divergence (MIND) network. Changes in cortical morphological similarity attributable to TLE and their cognitive correlates were assessed. Using a public transcriptional

Construction of biomimetic hydroxyapatite bilayer scaffolds via DLP 3D printing for bio-root regeneration.

Digital light processing (DLP) 3D printing of hydroxyapatite (HA) bioceramics offers a promising strategy for fabricating complex, patient-specific bio-root scaffolds with high resolution and bioactivity. However, achieving adequate mechanical integrity while guiding the formation of a periodontal ligament (PDL)-like enthesis through structural optimisation remains a major challenge. Inspired by interfacial micromorphological cues that regulate both intrinsic mechanical properties and cellular responses, this study proposed a novel bone-root integrated design for a DLP 3D-printed bio-root scaf

Conditionally folded disordered regions contribute to adaptation and regulation in Schistosomamansoni.

Schistosoma mansoni, a causative agent of schistosomiasis, has a complex life cycle that requires extensive molecular adaptation to diverse host environments. Intrinsically disordered proteins (IDPs) and regions (IDRs) are highly flexible protein segments that play key roles in cellular regulation, signaling, and host-pathogen interactions. In this study, we conducted a proteome-wide analysis of IDRs and conditionally folded intrinsically disordered regions (CF-IDRs) in the S. mansoni proteome. 10,749 protein sequences were retrieved from UniProt and reduced to 9638 non-redundant proteins. Single-cell analyses of intestinal epithelium reveal the dysregulation of gut immune microenvironment in systemic lupus erythematosus.

BACKGROUND: The small intestine harbors a rich array of intestinal intraepithelial lymphocytes (IELs) that interact with structural cells to collectively sustain gut immune homeostasis. Dysregulation of gut immune homeostasis was implicated in the pathogenesis of multiple autoimmune diseases, however, whether this homeostasis is disrupted in a lupus autoimmune background remains unclear. METHODS: We performed single-cell RNA sequencing (scRNA-seq) analyses to elucidate immune and structural milieu in the intestinal epithelium of MRL/Lpr lupus mice (Lpr mice) and MRL/Mpj control mice (Mpj mice

Detailed Assessment of Acrel Ganglion Through Cadaveric Dissection, Histology, and Ultrasonography.

Background Acrel's "pseudo-ganglion" is a ganglioform structure found on the floor of the fourth dorsal extensor compartment. The function of Acrel's "pseudo-ganglion" is undetermined within published literature; however, recent histological studies suggest it to be devoid of nerve cell bodies and is, therefore, a "pseudo-ganglion." Advances in ultrasound technology have made it possible to better visualize nerves in vivo. This study aims to investigate any ganglioform structure at the termination of the posterior interosseous nerve, using ultrasound, cadaveric dissection, and histological stu

rJararacin, a recombinant disintegrin from Bothrops jararaca venom: Exploring its effects on hemostasis and thrombosis.

Integrins are a family of heterodimeric transmembrane receptors which link the extracellular matrix to the cell cytoskeleton. These receptors play a role in many cellular processes: adhesion, proliferation, migration, apoptosis, and platelet aggregation, thus modulating a wide range of scenarios in health and disease. Therefore, integrins have been the target of new antithrombotic drugs. Disintegrins from snake venoms are recognized by the ability to modulate the activity of integrins, such as integrin αIIbβ3, a fundamental platelet glycoprotein, and αvβ3 expressed on tumor cells. For this rea

Icariin-loaded 3D-printed porous Ti6Al4V reconstruction rods for the treatment of necrotic femoral heads.

Avascular necrosis of the femoral head is a prevalent hip joint disease. Due to the damage and destruction of the blood supply of the femoral head, the ischemic necrosis of bone cells and bone marrow leads to the structural changes and the collapse of the femoral head. In this study, an icariin-loaded 3D-printed porous Ti6Al4V reconstruction rod (referred to as reconstruction rod) was prepared by 3D printing technology. The mechanical validity of the reconstruction rod was verified by finite element analysis. Through infilling of mercapto hyaluronic acid hydrogel containing icariin into the po
