Establishment and structural analysis of human mAb to the E2 component of the 2-oxoglutarate dehydrogenase complex generated from a patient with primary biliary cirrhosis.

We established one Epstein-Barr virus-transformed B cell hybrid clone producing human mAb of the IgG class to the 2-oxoglutarate dehydrogenase complex (OGDC) for the first time from the peripheral B lymphocytes of a patient with primary biliary cirrhosis (PBC). This mAb, designated mAbM37GO37, specifically bound to OGDC and its dissociation constant with OGDC was calculated to be 3.70 x 10(-10) mol/l. mAbM37GO37 stained murine stomach/kidney cryostat sections in a typical immunofluorescence pattern of antimitochondrial antibody (AMA). Western blotting analysis revealed that mAbM37GO37 reacted

Wnt regulates cellular differentiation and retinoid metabolism genes in the mouse stomach.

Although recognized as a key regulator of gastrointestinal tissues, Wnt signaling pathway function in the stomach is poorly understood. This study aimed to define Wnt functions and identify Wnt-regulated genes in the stomach. Reporter mouse analysis localized Wnt signaling to the base and proliferative region in both the corpus and the antrum. Canonical Wnt inhibition in vivo using Sox2-CreERT2; Ctnnb1fl/fl mice reduced epithelial cell proliferation with loss of gastric stem cells. Wnt-regulated genes and potential effector pathways were studied by bulk RNA sequencing (RNA-Seq) analysis of cor

Endogenous oestrogens as neuroprotective modulators in the human retina: Evidence from transcriptomic profiling and aromatase inhibition.

Steroid hormones, particularly estrogens, modulate neuronal survival in the central nervous system and the retina; however, their specific cell-type-specific roles in the human retina remain incompletely characterized. We analyzed the single-cell RNA sequencing dataset E-MTAB-7316 to profile genes from the KEGG steroid hormone biosynthesis and oestrogen signalling pathways. Functional relevance of local oestrogen synthesis was tested in mouse retinal explants treated with the aromatase inhibitor letrozole (20 μM). Over 50% of steroid hormone metabolism genes were expressed in retinal cells, wi

Single-cell and isoform-specific translational profiling of the mouse brain.

The brain displays the richest repertoire of post-transcriptional mechanisms regulating mRNA translation1-11. Among these, alternative splicing has been shown to drive cell-type specificity and, when disrupted, is strongly linked to neurological disorders12-17. However, genome-wide measurements of mRNA translation with isoform sensitivity at single-cell resolution have not been achieved. To address this, we deployed Surveying Ribosomal Targets by APOBEC-Mediated Profiling (Ribo-STAMP) coupled with short-read and long-read single-cell RNA sequencing in the brain18. We generated the first isofor

Candida Tropicalis Biofilm Formation Under Secondary Bile Salt Sodium Deoxycholate.

Candida species are components of the normal intestinal microbiota and are under constant exposure to bacterial metabolites, including secondary bile salts. Secondary bile salts are produced by commensal bacteria in the intestine and not only affect lipid absorption through emulsification but also have great effects on other microorganisms. Here, we examined the effect of a secondary bile salt, sodium deoxycholate (NaDCA), on the formation of biofilms by Candida tropicalis. In contrast to C. albicans, C. tropicalis tended to maintain its absolute biofilm biomass and surface hydrophobicity in t

Transcriptional Reshaping of Bacteriocytes in the Aphid-Serratia Symbiosis.

The facultative endosymbiont Serratia symbiotica significantly influences the ecological fitness of its aphid host. However, the molecular mechanisms by which Serratia affects the host's symbiotic organ, the bacteriocyte, remain poorly understood. Here, we conducted a comparative transcriptomic analysis of bacteriocytes from Serratia- and Serratia+ pea aphid (Acyrthosiphon pisum) strains. Our analysis revealed that Serratia colonization extensively modulates gene expression within bacteriocytes. Key metabolic pathways were significantly altered: genes involved in ribosomal biogenesis and oxida

Exome-based cancer driver gene comprehensive testing can provide a genetic diagnosis for individuals with triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is characterized by aggressive behaviour, high tumor heterogeneity, and an increased likelihood of recurrence and early metastasis. These factors hinder successful treatment. Genetic diagnosis enables personalized clinical recommendations and treatment options. The objective of this study was to validate whole-exome sequencing (WES) and variant prioritization in cancer susceptibility genes (CSG) associated with hereditary cancer (HC) predisposition in TNBC patients (n = 24). We present the development of a reproducible bioinformatic pipeline and its technic

RepID promotes metastatic potential in osteosarcoma through regulation of the PRC1-GATA6 axis.

Osteosarcoma is the most common primary malignant bone tumor and is characterized by a high propensity for metastasis, most frequently to the lungs and lymph nodes. Despite extensive research, the key modulators and their functional roles driving osteosarcoma metastasis remain poorly defined. Here, we identify RepID (Replication initiation determinant protein), a substrate receptor which is crucial for the chromatin recruitment of the Cullin-RING ubiquitin E3 ligase 4 (CRL4) complex, as a upstream regulator of epithelial-mesenchymal transition (EMT)-associated gene expression that promotes ost

Genomic Signatures of MASLD: How Genomics Is Redefining Our Understanding of Metabolic Liver Disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver condition globally, driven by strong genetic and environmental components. This review summarizes recent advances in understanding the genetic architecture of MASLD. Genome-wide association studies (GWAS) have identified several key risk variants, primarily in genes such as PNPLA3, TM6SF2, GCKR, and MBOAT7, which influence hepatic lipid metabolism and disease progression. By utilizing surrogate markers of MASLD, researchers have also identified numerous putative MASLD-associated genes, warranti

Integrated analysis of transcriptome and metabolome reveal the complex molecular regulatory network of chicken house PM2.5 induced lung injury.

Fine particulate matter (PM2.5) in poultry houses is the main component of air pollution. Long-term exposure to high concentrations of PM2.5 could easily damage the respiratory system of animals. In this study, we first investigated the changes in lung tissue morphology and serum cytokines in SPF chickens after exposure to poultry house PM2.5 and further conduct a combined analysis of its lung transcriptome and metabolome. The results showed that chicken exposed to PM2.5 led to decrease in body weight and cause inflammatory damage such as pulmonary fibrosis in chickens. It also significantly u

In vitro and in vivo study on the effects of nutritive media compared with water on the resuscitation of a live attenuated Salmonella Typhimurium vaccine.

In this study, a commercially available Salmonella Typhimurium vaccine (Vaxsafe ST) reconstituted in either water or nutrient broth, was tested to understand its effects on the in vitro invasion of Caco-2 cells and its transcriptional regulation post-reconstitution. Over time, the invasiveness of Vaxsafe ST was significantly higher following reconstitution in nutrient broth compared with water. Incubation temperature post-reconstitution did not significantly affect the invasion rate. Transcriptome data showed that Vaxsafe ST reconstituted in nutrient broth upregulated genes involved in the two

Transcriptome identification of differential mammary genes of Kazakh horses during early pregnancy.

Kazakh mares have attracted widespread attention with their outstanding lactation traits. Lactation is a complex dynamic process regulated by multiple factors. The extensive application of transcriptome sequencing technology enables researchers to further explore this biological issue. This study selected three pregnant and three non-pregnant Kazakh mares as the research subject. Their mammary glands were taken for transcriptome sequencing. The results show that there are 9 lncRNAs and 122 mRNAs differentially expressed between the two groups. GO enrichment analysis shows that there are 175 mo

The roles of TAF1 in neuroscience and beyond.

The transcriptional machinery is essential for gene expression and regulation; dysregulation of transcription can result in a range of pathologies, including neurodegeneration, cancer, developmental disorders and cardiovascular disease. A key component of RNA polymerase II-mediated transcription is the basal transcription factor IID, which is formed of the TATA box-binding protein (TBP) and 14 TBP-associated factors (TAFs), the largest of which is the TAF1 protein, encoded on the X chromosome (Xq13.1). TAF1 is dysregulated in X-linked dystonia-parkinsonism and congenital mutations in the gene

Genome scan reveals several loci associated with torus palatinus.

OBJECTIVE: Torus Palatinus (TP) is a common trait with an unclear aetiology. Although prior studies suggest a hereditary component, the genetic factors that influence TP risk remain unknown. The purpose of this study is to identify genetic variants associated with TP. MATERIALS AND METHODS: We assessed the TP status of 829 individuals from various ancestral backgrounds using 3D palate scans. We then carried out a genome-wide association study (GWAS) to identify common variants associated with TP. We also performed gene-based tests across the exome to investigate the role of low-frequency codi

Transcriptome profiling of triploid Crassostrea gigas gills indicates the host immune mechanism against bacterial infection.

As an important member of global aquaculture, oysters (Crassostrea gigas) have significant economic value. With the development of commercial aquaculture, the frequent occurrence of diseases caused by Vibrio alginolyticus has become a hindrance to high-density aquaculture. Gill tissue, as an important component of immune system of the oysters, plays the key point in the face of invasion by foreign substances. Compared to the diploid oyster, the triploid oyster presents a higher growth rate and lower growth investment, making it a more ideal model for studying oyster immune defense. In this stu
