A new analog of calcitriol, 19-nor-1,25-(OH)2D2, suppresses parathyroid hormone secretion in uremic rats in the absence of hypercalcemia.

The active metabolite of vitamin D, calcitriol (1 alpha,25-(OH)2D3), suppresses parathyroid hormone (PTH) gene transcription. Although 1 alpha,25-(OH)2D3 is effective in suppressing secondary hyperparathyroidism (SH) in uremic patients, the mandatory use of large amounts of calcium salts to control serum phosphorus may preclude, in some patients, the use of ideal therapeutic doses of 1 alpha,25-(OH)2D3 because of hypercalcemia. We have studied a new analog of calcitriol, 19-nor-1 alpha,25-(OH)2D2, that possesses low calcemic and phosphatemic activity. Uremic rats received vehicle, 1 alpha,25-(

Differential diagnosis and psychopharmacology of dual disorders.

There are multiple potential interactions of substances of abuse with psychiatric illnesses. The individual agents, the stage of abuse, withdrawal, or recovery, and the patient's psychiatric illness must be considered as an integrated whole to ascertain the appropriate interventions. Polysubstance abuse can cause still more complicated interactions. Assessment of differential treatment needs for both the psychiatric illness component and the substance use disorder component permits both elements to be approached, often simultaneously, and can facilitate treatment of both. Ignoring either compo

False nonresponders to anti-calcitonin gene-related peptide monoclonal antibodies: A real-world analysis beyond migraine frequency reduction.

OBJECTIVES/BACKGROUND: Reimbursement criteria for anti-calcitonin gene-related peptide (CGRP) monoclonal antibodies (MAbs) typically rely on predefined reductions in monthly migraine days (MMD). However, this approach may fail to capture the full spectrum of treatment benefits, potentially leading to premature discontinuation in patients experiencing meaningful improvements in other parameters. METHODS: This was a retrospective observational cohort study of prospectively collected data from a multicenter headache unit (Hospital Universitari de Bellvitge and Hospital de Viladecans, Barcelona,

Effects of C/N ratio and carbon-source composition on nutrient removal in a dual-sludge membrane bioreactor system (A2N-MBR).

Microbial competition that is driven by substrates plays an essential role in the stable denitrifying phosphorus removal (DPR) of low carbon wastewater treatment. This paper used a dual-sludge membrane bioreactor (A2N-MBR) to evaluate C/N ratios of 3-6 using acetate, followed by a comparison of acetate, propionate and their mixture at C/N = 5. Stoichiometric analysis estimated substrate allocations among denitrifying polyphosphate-accumulating organisms (DPAOs), denitrifying glycogen-accumulating organisms (DGAOs) and denitrifying ordinary heterotrophic organisms (DOHOs), while 16S rRNA gene s

2'-Fucosyllactose enhances indole-3-lactic acid production by Bifidobacterium bifidum DNG-08 and alleviates colitis-associated bone loss: Associations with microbiota remodeling and intestinal AHR-related responses.

Inflammatory bowel disease is frequently associated with an imbalance in bone remodeling and a decline in bone mass, however, there is a lack of systematic research on microbiome-based intervention strategies targeting colitis-associated bone loss. In this study, Bifidobacterium bifidum DNG-08 (DNG-08) was selected from four candidate strains based on its favorable 2'-fucosyllactose (2'-FL) utilization and indole-3-lactic acid (ILA) production. Genomic annotation revealed that DNG-08 harbors functional genes related to fucosylated oligosaccharide utilization and the conversion of tryptophan to

Bisphenol A induces hepatotoxicity via a LAMP3-dependent impairment of mitophagic processing and reduced PI3K-AKT-mTOR signaling.

Bisphenol A (BPA) is a ubiquitous environmental toxicant, but the mechanisms underlying BPA-associated hepatic injury remain incompletely understood. This study investigated the involvement of lysosome-associated membrane protein 3 (LAMP3) in mitophagy-related alterations and downstream survival signaling. Human hepatocellular carcinoma-derived Huh7 and PLC5 cells and a mouse model of 4-week BPA exposure were used. Cell proliferation and apoptosis were assessed using CCK-8, EdU, TUNEL, and flow cytometry; mitochondrial function and morphology were evaluated using JC-1, MitoTracker, reactive ox

Screening and functional identification of CtMYB2 as a positive regulator of flavonoid biosynthesis in safflower (Carthamus tinctorius L.).

BACKGROUND: Safflower (Carthamus tinctorius L.), an annual herb of the Asteraceae family, possesses dried tubular florets that are commonly used in traditional Chinese medicine to promote blood circulation and remove blood stasis. Flavonoids are the main active constituents of safflower, yet their regulatory mechanisms remain largely unclear. While MYB transcription factors have been widely reported to participate in regulating flavonoid biosynthesis, few transcription factors that regulate this pathway have been functionally characterized in safflower. RESULTS: Based on safflower transcripto

A multifunctional ZIF-8@CuO/collagen-binding peptide hydrogel for photothermal-enhanced antibacterial and infected skin wound management.

Owing to continuous bacterial proliferation, a persistent inflammatory response, and impaired vascularization, infected skin wounds remain a major clinical challenge. Herein, we fabricated a multifunctional skin-repair material by incorporating ZIF-8@CuO nanoparticles and a collagen-binding vascular endothelial growth factor mimetic peptide (C-VEGFMP) into a gelatin methacryloyl (GelMA) hydrogel. The ZIF-8@CuO nanoparticles were prepared by controllably depositing copper oxide (CuO) onto the surface of zeolitic imidazolate framework-8 (ZIF-8). ZIF-8@CuO can endowed the hydrogel with potent ant

Xinfeng capsule regulates pyroptosis and ameliorates cardiac fibrosis in rheumatoid arthritis through lncRNA GAS5 and TLR4/ROS/miR-21 pathways.

Xinfeng capsule (XFC) is a traditional Chinese medicinal compound that has been used clinically for several years to treat rheumatoid arthritis (RA)-related cardiac dysfunction. Nevertheless, the mechanism through which XFC treats RA-induced cardiac impairment remains unclear. This study investigates the mechanism by which XFC-containing serum regulates pyroptosis and ameliorates cardiac fibrosis in RA through the lncRNA GAS5 and TLR4/ROS/miR-21 pathways. A cardiac fibrosis model using human myocardial cells was established to study lncRNA GAS5, miR-21, TLR4, and NLRP3 expressions. Immunofluor

The Neural Symphony in the Periodontium: Health, Inflammation, and Regeneration.

Periodontal nerves, comprising sensory, sympathetic, and parasympathetic fibers, are essential components of the periodontium. Beyond their classic sensory and proprioceptive functions, these neural networks actively modulate periodontal immune responses, bone remodeling, and tissue regeneration. They orchestrate tissue homeostasis by releasing specific neuropeptides, neurotransmitters, and neurotrophic factors. The progression of periodontitis is intimately linked to neural dysregulation, where proinflammatory mediators (e.g., excessive substance P and sympathetic catecholamines) overpower pr

GOLPH3 promotes alveolar epithelial glycocalyx shedding and barrier disruption in lipopolysaccharide-induced acute lung injury via the ERK/MMP2/9 axis.

BACKGROUND: Acute lung injury (ALI) is a life-threatening syndrome with high mortality, characterized by alveolar epithelial barrier disruption and excessive inflammatory injury. The epithelial glycocalyx, a layer critical for barrier integrity, undergoes shedding during ALI. However, the underlying regulatory mechanisms remain poorly understood. This study investigates the role of Golgi phosphoprotein 3 (GOLPH3) in glycocalyx-mediated barrier dysfunction in ALI. METHODS: A Lipopolysaccharide (LPS)-induced ALI model was established in MLE-12 cells and C57BL/6 mice. Bioinformatics analysis of

Emergent symbiont strains provide thermally robust protection against co-evolved and novel parasitoids of introduced pea aphids.

Climate change and biological invasions pose synergistic threats; however, organisms may rapidly adapt through microbial symbiosis. We investigated how defensive symbionts in invasive pea aphids, Acyrthosiphon pisum, respond to emerging threats. Previously rare strains of the protective symbiont Hamiltonella defensa increased from <0.5% to 58% in aphid populations over just a few years. Bioassays revealed that these strains confer reciprocal, enemy-specific defences. One strain (C11) protected against Praon pequodorum, a native parasitoid that only began attacking pea aphids post-introduction,

Brown midrib mutants in sorghum and their applications in renewable energy production.

Lignin is a complicated phenolic polymer that is found in the secondary cell walls of plants. It is necessary for plant mechanical strength, vascular integrity, and stress tolerance. In sorghum (Sorghum bicolor L. Moench), the amount and composition of lignin greatly affect forage digestibility, biomass recalcitrance to bioenergy production and biochemical conversion process efficiency. Brown midrib (bmr) mutants, which exhibit reddish-brown pigmentation of the vasculature, have been a classic and potent tool for identifying lignin biosynthesis and engineering cell wall traits. Over the last 6

Suppression of Glucosylceramide Synthase Reverses Drug Resistance in Cancer Cells Harboring Homozygous p53 Mutants.

Glucosylceramide synthase (GCS) catalyzes ceramide glycosylation in response to cell stress that produces glucosylceramide and other glycosphingolipids. GCS overexpression is a cause of drug resistance and enriches cancer stem cells (CSCs) during cancer chemotherapy. Previous studies showed that GCS modulates the expression of p53 mutants and oncogenic gain-of-function (GOF) in heterozygous knock-in cell models (TP53 R273H-/+). However, it is unclear whether GCS can modulate the effects of homozygous p53 mutations, which are common in many cancer cases. We report herewith that inhibition of GC

Clinical value of miR-329-3p in thalassemia and its regulation of TNRC6B expression.

Background β-thalassemia is a common monogenic genetic disorder, characterized by reduced or absent synthesis of β-globin chains. High fetal hemoglobin (HbF) levels can alleviate the severity of anemia in β-thalassemia, and miRNAs can regulate the expression of globins. MiR-329-3p is a miRNA that is differentially expressed in β-thalassemia. Aim this study mainly investigated the expression of miR-329-3p in the peripheral blood of children with β-thalassemia, analyzed its clinical diagnostic value in β-thalassemia, and further studied the regulatory effects of miR-329-3p on its target genes TN

Immunoregulatory effect of metformin in monocytes exposed to SARS-CoV-2 spike protein subunit 1.

BACKGROUND: Severe coronavirus disease 2019 (COVID-19) is characterized by a hyperinflammatory state associated with an exacerbated inflammatory activation of monocytes and macrophages in the respiratory tract. Metformin has been identified as a potent monocyte inflammatory suppressor, and it has been demonstrated to attenuate inflammation in COVID-19. The mechanisms underlying metformin's anti-inflammatory effects are, however, unclear. We thus sought to investigate metformin's main interactions and their respective isolated effects in modulating monocyte inflammatory response to severe acute

Aging and Western Diet Synergistically Impair Hepatic Thyroid Hormone Signaling to Promote Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) in Mice.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is primarily driven by a Western-style diet and exacerbated with aging, yet underlying mechanisms remain unclear. Given the essential role of thyroid hormone (TH) in MASLD progression, we hypothesized that impaired intrahepatic TH action during aging promotes MASLD progression and severity of MASH with fibrosis. We evaluated hepatic TH metabolism in young (18-24 weeks) and old (108-120 weeks) C57BL/6J mice fed either a normal chow diet (NCD) or a Western diet with fructose (WDF) for 8 weeks. Liver histology, metabolic parameters,

A p53-responsive microRNA-100 released from cardiomyoblasts serves as an early biomarker for acute coronary syndrome.

Coronary artery disease, a leading cause of mortality, underscores the need for biomarkers that can predict acute coronary syndrome (ACS) occurrence and outcomes. MicroRNAs (miRNAs) are increasingly recognized as potential markers. This study aimed to identify a plasma miRNA signature in ACS patients undergoing elective cardiac catheterization and to investigate miR-100's potential as an ACS prognostic biomarker. Plasma samples from 100 patients with suspected ACS were analyzed. qPCR revealed significantly elevated plasma miR-29a-3p, miR-100, miR-192, and miR-194-5p in ACS patients, and multip
