Oxytocin as an antidiuretic hormone. I. Concentration dependence of action.

Circulating concentrations of oxytocin increase to 10-40 pM in rats in response to osmotic stimuli, suggesting that oxytocin could play a role in regulation of water balance. The present studies tested whether oxytocin at such concentrations increases osmotic water permeability (Pf) in isolated perfused terminal inner medullary collecting ducts (IMCD). In IMCD segments from Sprague-Dawley rats, 20 pM oxytocin added to the peritubular bath caused a two- to threefold increase in Pf, whereas 200 pM oxytocin increased Pf by five- to sixfold (n = 8, P < 0.01). IMCD from Brattleboro rats, which mani

Short and Mid-Term Predictors of Response to OnabotulinumtoxinA: Real-Life Experience Observational Study.

OBJECTIVE: To identify clinical predictors of excellent response to OnabotulinumtoxinA in patients with chronic migraine (CM) at 6 and 12 months of follow-up. BACKGROUND: Clinical predictors of response to OnabotulinumtoxinA are scarce and have not been clearly reproduced and analyzed in detail. So far, predictors of response to OnabotulinumtoxinA assess response in general or good response, but not an excellent response. METHODS: Cohort study of patients attended in a specialized Headache Clinic in treatment with OnabotulinumtoxinA were classified according to their improvement in frequency

Patterns of immune development in urban preschoolers with recurrent wheeze and/or atopy.

BACKGROUND: Disadvantaged urban children have high rates of allergic diseases and wheezing, which are diseases associated with type 2-biased immunity. OBJECTIVE: We sought to determine whether environmental exposures in early life influence cytokine responses that affect the development of recurrent wheezing illnesses and allergic sensitization. METHODS: A birth cohort of 560 urban families was recruited from neighborhoods with high rates of poverty, and 467 (83%) children were followed until 3 years of age. Cytokine responses were measured in blood cell samples obtained at birth (cord blood

Representation and integration of multiple sensory inputs in primate superior colliculus.

1. The properties of visual-, auditory-, and somatosensory-responsive neurons, as well as of neurons responsive to multiple sensory cues (i.e., multisensory), were examined in the superior colliculus of the rhesus monkey. Although superficial layer neurons responded exclusively to visual stimuli and visual inputs predominated in deeper layers, there was also a rich nonvisual and multisensory representation in the superior colliculus. More than a quarter (27.8%) of the deep layer population responded to stimuli from more than a single sensory modality. In contrast, 37% responded only to visual

Association between type 2 diabetes and short-term clinical response to TNF inhibitors in patients with ankylosing spondylitis: a propensity score-matched retrospective cohort study.

INTRODUCTION/OBJECTIVES: Type 2 diabetes mellitus (T2DM) is a common comorbidity in clinical practice, yet its association with tumor necrosis factor inhibitor (TNFi) response in ankylosing spondylitis (AS) has not been characterized. We hypothesized that comorbid T2DM may be associated with reduced short-term clinical response to TNFi in patients with AS. METHODS: This single-center retrospective cohort study included patients with AS initiating TNFi between January 2020 and December 2025. T2DM was identified from ICD-coded medical records. Propensity score 1:2 nearest-neighbor matching was

Furmonertinib combined with bevacizumab in EGFR-TKI-resistant leptomeningeal metastasis: analysis of the CSF ctDNA molecular response and survival outcomes.

BACKGROUND: Leptomeningeal metastasis (LM) after the development of third-generation epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI) resistance is indicative of a poor prognosis in EGFR-mutant non-small cell lung cancer (NSCLC), and no standardised treatments are currently available. The aims of this study were to evaluate the outcomes of combination therapy with bevacizumab plus high-dose furmonertinib in this setting and to assess the cerebrospinal fluid (CSF) circulating tumour DNA (ctDNA) molecular response as a treatment response biomarker. METHODS: This real-world

Anti-TNF therapy impairs both short- and long-term IgG responses after repeated vaccination.

BACKGROUND: Recently, it has been questioned whether vaccination of patients with inflammatory (auto)immune diseases under anti-tumor necrosis factor (TNF) treatment leads to impaired vaccine-induced immune responses and protection against breakthrough infections. However, the effects of TNF blockade on short- and long-term immune responses after repeated vaccination remain unclear. Vaccination studies have shown that initial short-term IgG antibodies (Abs) carry highly galactosylated and sialylated Fc glycans, whilst long-term IgG Abs have low levels of galactosylation and sialylation and are

Investigation of the utility of the 1.1B4 cell as a model human beta cell line for study of persistent enteroviral infection.

The generation of a human pancreatic beta cell line which reproduces the responses seen in primary beta cells, but is amenable to propagation in culture, has long been an important goal in diabetes research. This is particularly true for studies focussing on the role of enteroviral infection as a potential cause of beta-cell autoimmunity in type 1 diabetes. In the present work we made use of a clonal beta cell line (1.1B4) available from the European Collection of Authenticated Cell Cultures, which had been generated by the fusion of primary human beta-cells with a pancreatic ductal carcinoma

Murine host response to Neisseria gonorrhoeae upper genital tract infection reveals a common transcriptional signature, plus distinct inflammatory responses that vary between reproductive cycle phases.

BACKGROUND: The emergence of fully antimicrobial resistant Neisseria gonorrhoeae has led global public health agencies to identify a critical need for next generation anti-gonococcal pharmaceuticals. The development and success of these compounds will rely upon valid pre-clinical models of gonorrhoeae infection. We recently developed and reported the first model of upper genital tract gonococcal infection. During initial characterization, we observed significant reproductive cycle-based variation in infection outcome. When uterine infection occurred in the diestrus phase, there was significant

Activation of Liver FGF21 in hepatocarcinogenesis and during hepatic stress.

BACKGROUND: FGF21 is a promising intervention therapy for metabolic diseases as fatty liver, obesity and diabetes. Recent results suggest that FGF21 is highly expressed in hepatocytes under metabolic stress caused by starvation, hepatosteatosis, obesity and diabetes. Hepatic FGF21 elicits metabolic benefits by targeting adipocytes of the peripheral adipose tissue through the transmembrane FGFR1-KLB complex. Ablation of adipose FGFR1 resulted in increased hepatosteatosis under starvation conditions and abrogation of the anti-obesogenic action of FGF21. These results indicate that FGF21 may be a

Differential expression of interferon responsive genes in rodent models of transmissible spongiform encephalopathy disease.

BACKGROUND: The pathological hallmarks of transmissible spongiform encephalopathy (TSE) diseases are the deposition of a misfolded form of a host-encoded protein (PrPres), marked astrocytosis, microglial activation and spongiosis. The development of powerful gene based technologies has permitted increased levels of pro-inflammatory cytokines to be demonstrated. However, due to the use of assays of differing sensitivities and typically the analysis of a single model system it remained unclear whether this was a general feature of these diseases or to what extent different model systems and rout

What Is the Relationship Between Clinician-reported and Patient-reported Outcomes in Orthopaedic Surgery?

BACKGROUND: Patient-reported outcome measures (PROMs) and clinician-reported outcome measures (CROMs) are designed to capture different dimensions of treatment response: the patient's experience and the clinician's assessment, respectively. Because of that, some degree of discordance between them is expected. However, the nature and magnitude of PROM-CROM correlations across orthopaedic subspecialties have not been systematically characterized. Understanding where they converge and where they diverge may help clinicians and researchers select instrument pairings that capture complementary dime

Causes of Death and Physicians' Knowledge-Producing Responsibility-How Expectations Shape Interest in 'Finding Out What Happened'.

Clinical knowledge production is fundamentally centred on patient problems, improving the lives of patients. If patients die, physicians also need to administer death. What is their responsibility to generate knowledge about causes of death? Based on semi-structured interviews with 35 physicians, we show what sources physicians draw on, what they consider sufficient for making statements about causes of death and when and on what grounds they consider autopsies needed. We broaden and generalise the question of how physicians report causes of death by viewing it as a case of knowledge-producing

Love-Centered Therapy and the First Story Cycle: A Dyadic Process Model of Implicit Relational Activation, Autonomic Co-Regulation, and Sexual Field Disruption.

Couples often recognize the patterns that keep them stuck long before they become capable of interrupting them. Love-Centered Therapy (LCT) is a clinically derived process model that synthesizes attachment theory, somatic psychology, interpersonal neurobiology, and decolonial scholarship. LCT organizes intervention through three interconnected constructs: learned templates for intimate relationships (the First Story), the repetitive unsatisfying interactions those templates produce (the First Story Cycle), and the emotions generated by that ongoing interaction (the Relational Field). Intervent

Ordered Electromagnetic Dissipation via Admittance Scaling.

A long-standing challenge in electromagnetic dissipative systems is that dissipation and phase evolution are fundamentally entangled through the underlying admittance dynamics, leading to oscillatory and path-dependent response trajectories under tuning. In this Letter, we identify an admittance-scaling mechanism that restructures the evolution pathway of the effective input admittance, enabling broadband dissipation control through magnitude scaling while maintaining an approximately invariant phase manifold. The resulting response evolution avoids the oscillatory behavior associated with con

MAVS undergoes aggregation under hypoxia to enhance the hypoxia signaling pathway by promoting the generation of mitochondrial ROS.

The mitochondrial antiviral signaling protein (MAVS) is a key component of the innate immune response against RNA viruses. Its involvement in other cellular stresses is largely unknown. Here, we found that hypoxia induces MAVS aggregation, leading to the stabilization of HIF1/2α proteins and the enhancement of hypoxia signaling. Mechanistic studies revealed that MAVS enhances TRAF6 binding to ECSIT, which catalyzes ECSIT polyubiquitination. This promotes mitochondrial ROS generation and likely inhibits PHD2 enzymatic activity, thereby reducing HIF1/2α proteasomal degradation. Disrupting MAVS i

Neoadjuvant immunotherapy in a solitary, isolated peritoneal mesothelioma involving the abdominal wall: a case report.

Mesothelioma is a rare neoplasm with a minority of cases originating from the peritoneum. Patients are generally treated with a combination of cytoreductive surgery, hyperthermic intraperitoneal chemotherapy, and various systemic therapies. In recent years, expression of programmed death ligand 1 (PD-L1) has been investigated in mesothelioma with implications for targeted therapy. In this case report, we present a patient with solitary isolated peritoneal mesothelioma of the right flank with abdominal and chest wall involvement and high PD-L1 positivity. The patient demonstrated a remarkable m

Luteinizing hormone-induced changes in the structure of mammalian preovulatory follicles.

Ovulation of a mammalian oocyte from its follicle, which occurs in response to luteinizing hormone (LH), requires complex restructuring of the ∼20 layers of surrounding somatic cells. This chapter describes the cellular architecture of preovulatory follicles, the localization of the receptors for LH, and the LH-induced changes in follicular structure, focusing on mice and other small mammals. The multiple interrelated processes that result in ovulation include breakdown of existing extracellular matrix, generation of new extracellular matrix, thinning of the follicular apex where the oocyte wi

Multiple faces of anxiety: a frontal lobe perspective.

Marked dysregulation of the human prefrontal cortex (PFC) and anterior cingulate cortex (ACC) characterises a variety of anxiety disorders, and its amelioration is a key feature of treatment success. Overall treatment response, however, is highly variable, and about a third of patients are resistant to treatment. In this review we hypothesise that a major contributor to this variation in treatment response are the multiple faces of anxiety induced by distinct forms of frontal cortex dysregulation. Comparison of findings from humans and non-human primates reveals marked similarity in the functi
