Hypoxia-induced transcriptional reprogramming in sugar beet seedlings: Linking energy conservation and metabolic maintenance.

Sugar beet (Beta vulgaris L.), as a major sugar crop, faces a significant threat to its yield and industrial development due to hypoxia stress, a critical environmental factor limiting its growth. Here, we investigate the response mechanisms of sugar beet leaves and roots to hypoxia stress at different developmental stages (first and third true leaf stages). Hypoxia significantly reduced plant height, leaf area, and root area. Key photosynthetic parameters, including net photosynthetic rate, stomatal conductance, and intercellular CO2 concentration, decreased, leading to impaired photosyntheti

Cinnamyl alcohol acyltransferase PmCAAT1 specifically catalyzes the formation of the characteristic volatile cinnamyl acetate of Prunus mume.

Three cinnamyl alcohol acyltransferases (PmCAAT1-3) were identified in Prunus mume, with PmCAAT1 showing high substrate preference for cinnamyl alcohol and PmCAAT2 exhibiting broader substrate specificity including linalool. Prunus mume is renowned for its distinctive fragrance, with cinnamyl acetate serving as one of its key aroma components. However, the genes responsible for cinnamyl acetate biosynthesis have remained unidentified. In this study, a total of 116 acyltransferase family members were identified from the whole-genome data of P. mume var. tortuosa. Among these, three cinnamyl alc

Nitrogen Status Rewires Transcriptional Regulation of Dhurrin, a Dual-Purpose Defense Metabolite in Sorghum bicolor.

Dhurrin, a cyanogenic glucoside, plays an important role in Sorghum bicolor physiology and defense. The concentration of dhurrin in sorghum is influenced by both nitrogen status and stage of plant organ development. While nitrogen resupply activates the expression of genes for dhurrin biosynthesis, the molecular mechanisms underlying this regulation remain unclear. In this study, we investigated the transcriptional response of sorghum to nitrogen resupply following growth under nitrogen-limiting conditions. Using a time-course design, we measured hydrogen cyanide potential (HCNp), growth, and

Circadian Biology and Gynecological Diseases: Genetic Mechanisms and Potential Therapeutic Targets Underlying the Causal Link Between Insomnia and Ovarian Cysts.

BACKGROUND: Although circadian disruption has been implicated in gynecologic malignancies, genetic evidence for a causal relationship between insomnia and ovarian cysts remains lacking. The biological pathways linking sleep disturbance to ovarian pathology are therefore poorly understood, limiting mechanistic insights and therapeutic development. METHODS: We employed a two-sample Mendelian Randomization (MR) framework using large-scale Genome-Wide Association Study (GWAS) datasets of European ancestry to examine the effects of seven sleep-related traits on five gynecologic outcome types. We f

Unveiling the hidden threat: flupyradifurone soluble liquid's toxic impact on silkworms.

Bombyx mori Linnaeus (Lepidoptera: Bombycidae) is an economically significant insect that consistently contributes to the enhancement of the national economy in all countries with predominant sericulture industries. Flupyradifurone, a butenolide insecticide, is recognized for its minimal toxic effects on bees when applied at standard dosages; however, its potential toxicity or threat to B. mori has not been fully elucidated. This study aims to investigate and evaluate the toxicity of flupyradifurone soluble liquid (SL) on B. mori by administering various concentrations to third instar silkworm

SegJointGene: joint cell segmentation and spatial gene prioritization by information entropy guided convolutional neural networks.

Spatial sequencing technologies enable the single-cell-level study of molecular organization in tissues. Revealing such spatial patterns relies on accurate cell segmentation. In complex tissues with dense cell packing, segmentation based solely on nuclear staining is insufficient for accurate cell boundary detection. This limitation arises because accurate segmentation necessitates the delineation of cell morphology, which is driven by molecular activities such as cytoskeletal dynamics, cell-cell adhesion, and intercellular signaling. Thus, integrating molecular information, including gene or

Update on Congenital Cranial Dysinnervation Disorders (CCDDs).

Congenital cranial dysinnervation disorders (CCDDs) are a group of rare, nonprogressive conditions characterized by abnormal development of the cranial motor nerves and variable ocular motility deficits, ptosis, incomitant strabismus, and facial palsy. Advances in genetics and neuroimaging have revealed that these disorders result from defects in neuronal differentiation or axon guidance of the cranial motor neurons. Duane retraction syndrome, the most common CCDD, results from the absence of the abducens nerve and innervation of the lateral rectus by oculomotor nerve axons; causative genes in

Tracing the Evolutionary Dynamics and the Transcriptional Partitioning of Terpene Synthase Gene Family in Coffea arabica and Its Diploid Progenitors.

Terpenes represent a diverse class of plant secondary metabolites that play important roles in development, ecological interactions, and aroma, and are synthesized by the terpene synthase (TPS) gene family. To investigate the evolutionary history and regulatory diversification of this family in coffee, we performed a genome-wide analysis of TPS genes in Coffea arabica and its diploid progenitors, C. canephora and C. eugenioides. Using domain-based searches targeting the PF01397 module, we identified 69 TPS genes in C. arabica, 40 in C. canephora, and 55 in C. eugenioides. Phylogenetic classifi

Neuroimaging-genetic integration reveals shared structural and functional brain alterations in major depressive disorder.

BACKGROUND: Major depressive disorder (MDD) is linked to widespread disruptions in brain structure and function. Nevertheless, the extent to which structural and functional abnormalities overlap in MDD, and whether they share a common genetic basis, remains largely unclear. METHODS: We performed voxel-wise meta-analyses to identify structural alterations in gray matter volume (GMV) and functional changes using regional homogeneity (ReHo) and amplitude of low-frequency fluctuations (ALFF). MDD-associated genes were prioritized using the H-MAGMA framework with large-scale genome-wide associatio

Osteochondrosis in horses: An overview of genetic and other factors.

Osteochondrosis (OC) is a frequent manifestation of developmental orthopaedic disease, and its severe clinical presentation is known as OC dissecans (OCD). OC is defined as a disruption of the endochondral ossification process in the epiphyseal cartilage, and this disease has been reported in different mammalian species, including humans, dogs, pigs, and horses. OCD is an important cause of lameness in sport horses and is a common cause of impaired orthopaedic potential, whose clinical signs may be of minimal magnitude or manifest as severe joint effusion or clinically noticeable lameness. The

Multi-ancestry genome-wide association meta-analysis identifies novel associations and informs genetic risk prediction for Hirschsprung disease.

BACKGROUND: Hirschsprung disease (HSCR) is a rare, congenital disease characterized by the absence of enteric ganglia in the hindgut. Common genetic variation contributes substantially to the heritability of the disease yet only three HSCR-associated loci were identified from genome-wide association studies (GWAS) thus far. METHODS: We performed the largest multi-ancestry meta-analysis of GWAS to date, totalling 1250 HSCR cases and 7140 controls. Prioritized candidate genes were further characterized using single-cell transcriptomic data of developing human and mouse gut for their roles in de

Unveiling role of HSP70 genes for development and survival of Indian malaria vector Anopheles culicifacies.

Heat shock proteins (HSPs) play a pivotal role in maintaining cellular homeostasis and mediating stress responses across diverse organisms. Among them, the HSP70 family is crucial for protein folding and stress regulation. However, its functions remain underexplored in mosquito species, particularly in major Indian malaria vectors such as Anopheles culicifacies (Ac). This study aims to contribute to mosquito control by investigating the role of HSP70 in An. culicifacies. Given the persistent global challenge posed by malaria, understanding the regulatory mechanisms of HSP70 is essential for de

An ApiAP2 Family Transcriptional Factor PfAP2-06B Regulates Erythrocyte Invasion Indirectly in Plasmodium falciparum.

Obligate intracellular parasites must efficiently invade host cells to complete their life cycle and facilitate transmission. For the malaria-causing parasite Plasmodium falciparum, the invasion of an erythrocyte is a critical process, and thereby a key target for intervention strategies. In this study, we investigate the role of the ApiAP2 family transcription factor PfAP2-06B (PF3D7_0613800) in the intraerythrocytic developmental cycle of P. falciparum and focus on its regulation of genes involved in erythrocyte invasion. Conditional knockdown of PfAP2-06B resulted in a defect in asexual gro

Maternal preconception stress produces sex-specific effects at the maternal:fetal interface to impact offspring development and phenotypic outcomes†.

Entering pregnancy with a history of adversity, including adverse childhood experiences and racial discrimination stress, is a predictor of negative maternal and fetal health outcomes. Little is known about the biological mechanisms by which preconception adverse experiences are stored and impact future offspring health outcomes. In our maternal preconception stress (MPS) model, female mice underwent chronic stress from postnatal days 28-70 and were mated 2 weeks post-stress. Maternal preconception stress dams blunted the pregnancy-induced shift in the circulating extracellular vesicle proteom

Genetic basis of variation in thermal developmental plasticity for Drosophila melanogaster body pigmentation.

Seasonal differences in insect pigmentation are attributed to the influence of ambient temperature on pigmentation development. This thermal plasticity is adaptive and heritable, and thereby capable of evolving. However, the specific genes contributing to the variation in plasticity that can drive its evolution remain largely unknown. To address this, we analysed pigmentation and pigmentation plasticity in Drosophila melanogaster. We measured two components of pigmentation in the thorax and abdomen: overall darkness and the proportion of length covered by darker pattern elements (a trident in

Leveraging multiple approaches for the detection of pathogenic deep intronic variants in developmental and epileptic encephalopathies: A case report.

About 50% of individuals with developmental and epileptic encephalopathies (DEEs) are unsolved following genetic testing. Deep intronic variants, defined as >100 bp from exon-intron junctions, contribute to disease by affecting the splicing of mRNAs in clinically relevant genes. Identifying deep intronic pathogenic variants is challenging and resource intensive, and interpretation is difficult due to limited functional annotations. We aimed to identify deep intronic variants in individuals suspected to have unsolved single gene DEEs. In a research cohort of unsolved cases of DEEs, we searched

Identification of a circRNA-miRNA-mRNA network to explore the effects of circRNAs on Holstein bull testis after sexual maturity.

Spermatogenesis is an extremely sophisticated and complex process and is regulated not only by a large number of genes, but also by a large number of epigenetic factors. Although existing studies have demonstrated that circRNAs plays an important regulatory role in spermatogenesis, there is still insufficient information to properly understand the regulatory role and mechanism of circRNA action. We addressed this issue by examining the testes of two Holstein bull developmental stages; three 8-week-olds (young bull, YB) and three 80-week-olds (adult bull, AB), randomly selected from the same br

Toward novel treatment against filariasis: Insight into genome-wide co-evolutionary analysis of filarial nematodes and Wolbachia.

Infectious diseases caused by filarial nematodes are major health problems for humans and animals globally. Current treatment using anti-helminthic drugs requires a long treatment period and is only effective against the microfilarial stage. Most species of filarial nematodes harbor a specific strain of Wolbachia bacteria, which are essential for the survival, development, and reproduction of the nematodes. This parasite-bacteria obligate symbiosis offers a new angle for the cure of filariasis. In this study, we utilized publicly available genome data and putative protein sequences from seven

Sex-Specific Whole-Transcriptome Analysis in the Cerebral Cortex of FAE Offspring.

Fetal alcohol spectrum disorders (FASDs) are associated with systemic inflammation and neurodevelopmental abnormalities. Several candidate genes were found to be associated with fetal alcohol exposure (FAE)-associated behaviors, but a sex-specific complete transcriptomic analysis was not performed at the adult stage. Recent studies have shown that they are regulated at the developmental stage. However, the sex-specific role of RNA in FAE offspring brain development and function has not been studied yet. Here, we carried out the first systematic RNA profiling by utilizing a high-throughput tran
