CO2-modified atmosphere improves the flavor quality of low-salt Xuecai by regulating microbial communities and metabolic functions.

Low-salt pickled vegetables are often limited by their poor flavor and the accumulation of biogenic amines (BAs). In the present study, the effects of CO2-modified atmosphere (CMA) technology on the dynamics of flavor compounds, microbial communities, and metabolic functions in low-salt Xuecai during pickling were investigated. In comparison with low-salt pickling under natural air conditions, a CMA effectively prevented excessive acidification, enriched volatile metabolites (e.g., isothiocyanates, alcohols, and esters), and minimized the accumulation of bitter-tasting amino acids, resulting i

Nanotechnology-Driven Strategies to Combat Antimicrobial Resistance in the One Health Era.

Antimicrobial resistance (AMR) has emerged as one of the greatest global public health threats, compromising the effectiveness of antibiotics and jeopardizing advances in modern medicine. The extensive use and misuse of antimicrobial agents in human medicine, veterinary practice, agriculture, and aquaculture have accelerated the emergence and spread of multidrug-resistant microorganisms. Because resistant pathogens, antibiotic residues, and resistance genes continuously circulate among humans, animals, and the environment, AMR is now recognized as a complex One Health challenge requiring coord

Different DNA input amounts and library preparation kits yield comparable resistome characterization using target-enriched metagenomic sequencing.

INTRODUCTION: Target-enriched metagenomic sequencing improves detection of antimicrobial resistance genes (ARGs) relative to standard shotgun metagenomics, but whether DNA input amount influences resistome characterization has not been systematically evaluated. METHODS: We evaluated target-enriched sequencing using fecal microbial communities from multiple host species and compared results across library preparation kits, DNA input amounts, and sequencing approaches. Composite fecal samples from livestock and humans and individual canine samples were processed using three library preparation

Enhanced cellulosic ethanol production by increasing p-benzoquinone tolerance of Saccharomyces cerevisiae in adaptive evolution.

p-Benzoquinone, a lignin-derived inhibitor generated during lignocellulosic biomass pretreatment, severely restricts microbial fermentation and remains a major obstacle to efficient cellulosic ethanol production. In this study, p-benzoquinone concentrations in different lignocellulosic hydrolysates ranged from 130 to 210 mg/L under high-solids conditions. Growth analysis showed that p-benzoquinone strongly inhibited Saccharomyces cerevisiae DQ1 in a concentration-dependent manner, with 100 mg/L p-benzoquinone causing 68.5% growth inhibition. Adaptive evolution under gradually increasing p-benz

Multi-omics analysis reveals associations among endophytic microbiome shifts, host transcriptional responses, and metabolic variation across variegated leaf sectors of Aspidistra elatior.

Leaf variegation in Aspidistra elatior provides a useful system for exploring localized plant-microbiome associations, yet the biological factors linked to sector-specific chlorosis remain unclear. To investigate potential relationships among endophytic microbial communities, host transcriptomic responses, and metabolic variation, we integrated 16S/ITS amplicon sequencing, transcriptomics, and widely targeted LC-MS/MS-based metabolomics across normal green (GG), adjacent green (SG), and chlorotic spot (SS) sectors. SS sectors showed sector-associated bacterial community patterns characterized

Propionate attenuates osteoarthritis progression by regulating the gut-joint axis.

INTRODUCTION: Osteoarthritis (OA) is a degenerative joint disorder characterized by cartilage degradation, inflammation, and pain. Growing evidence indicates that dysregulation of the gut-joint axis contributes to OA progression. This study investigated the therapeutic potential of propionate, a gut-derived short-chain fatty acid, in OA. METHODS: A monosodium iodoacetate (MIA)-induced OA rat model was used to evaluate the effects of propionate on pain and inflammation through behavioral assessments, histological analysis, and gut microbiota profiling. The intestinal environment was further as

A cloaked glutamate decarboxylase sustains the GABA shunt in Mycobacterium tuberculosis.

Despite the exponential growth in genome sequencing, the functional annotation of genes, particularly the discovery of novel enzymatic activities, remains a formidable challenge. This gap is exacerbated by annotation biases that propagate assumptions about enzyme function across homologous sequences. Here, we report the identification of an unannotated enzymatic activity within a presumed well-characterized enzyme family. The gene rv2531c, under strong purifying selection across Mycobacterium tuberculosis strains, has been annotated as a member of the lysine-ornithine-arginine (KOR) decarboxyl

Integrated analysis of the diabetic foot ulcer microbiome and host transcriptome supports a microenvironment-microbiota-host repair framework.

BACKGROUND: Diabetic foot ulcers (DFUs) arise from interacting clinical, microbial, and host processes, yet public datasets differ substantially in design, scale, and evidentiary strength. METHODS: We curated eight public DFU cohorts and defined an evidence hierarchy. PRJNA287759 was reprocessed from raw 16 S reads; outcome tests used one baseline sample per patient (74 healed and 15 non-healed/adverse). GSE134431 compared 13 DFU with 8 diabetic foot skin samples using limma. Host discrimination underwent fold-confined leave-one-sample-out validation, 100 repeated nested five-fold analyses, 2

Genetic diversity of spotted fever group Rickettsia and Dabieshan tick virus detected in ticks collected from suburban forest areas of Gifu, Central Japan.

This study investigated the prevalence and genetic diversity of tick-borne pathogens in the forested suburbs of Gifu in central Japan, where human cases of Japanese spotted fever and severe fever with thrombocytopenia syndrome (SFTS) have recently been reported. A total of 130 ticks representing eight species were analyzed for spotted fever group Rickettsia (SFGR) and viruses in the family Phenuiviridae using PCR and reverse transcription-PCR, respectively. While the causative agents of Japanese spotted fever (Rickettsia japonica) and SFTS were not detected in this study, SFGR-specific genes w

Engineering a versatile Aspergillus niger platform for efficient and scalable biosynthesis of polyketides.

Polyketides are a diverse class of natural products with broad pharmacological activities and substantial industrial relevance; however, their scalable biosynthesis is often constrained by inefficient heterologous expression platforms. In this study, a systematic evaluation of multiple microbial hosts identified Aspergillus niger HL-1 as an efficient fungal chassis for polyketide production. Subcellular compartmentalization analysis revealed that cytosolic biosynthesis significantly outperformed peroxisomal localization, leading to a 5.88-fold increase in triacetic acid lactone (TAL) productio

Distinct Host and Bacterial Profiles in Murine Single Versus Co-Infection with Foal-Isolated Streptococcus equi and Streptococcus dysgalactiae.

This study investigated the interactive pathogenic characteristics of co-infection with Streptococcus equi (S. equi) and Streptococcus dysgalactiae (S. dysgalactiae) isolated from diseased foals. Integrating genomic analysis, in vitro phenotypic assays, and a murine intranasal infection model, we comparatively analyzed the biological traits and pathogenic differences between single and dual infections. Comparative genomic analysis revealed divergent virulence profiles between the two isolates, and both species harbored open pan-genomes. In vitro, the biofilm biomass of strain F1 was higher tha

Integrated multi-omics analysis uncovers Zearalenone-induced ovarian and intestinal inflammation is mediated by the tumor necrosis factor-α.

As a widely encountered foodborne mycotoxin, Zearalenone (ZEN) has the potential to accumulate along the food chain, posing threats to public health and safety. However, the precise mechanisms by which ZEN causes tissue damage remain incompletely understood. Here, we established in vivo (40 μg/kg) and in vitro (30 μM) models to elucidate the underlying processes of ZEN-induced damage to the ovary and intestine. Our results demonstrated that, following ZEN exposure, pro-inflammatory signaling factors, including NLR family pyrin domain containing 3 (NLRP3) and Interleukin 6 (IL6), were significa

Multi-omics reveals different signatures of obesity-prone and obesity-resistant mice.

Obesity-prone (OP) and obesity-resistant (OR) individuals demonstrate significant metabolic differences, potentially influenced by variations in the gut microbiome. However, the influence of host-microbiota interactions on obesity susceptibility remains unknown. We performed an integrative multi-omics approach to explore microbial, metabolic, and genetic differences in high-fat diet (HFD)-fed OP and OR mice, with additional analyses of gut microbiota variations in humans. In OP mice, the dynamic gut microbiota was characterized by a stable presence of Longibaculum, while Kineothrix predominate

10058-F4 Mediated inhibition of the biofilm formation in multidrug-resistant Staphylococcus aureus.

Antimicrobial resistance (AMR) is a global concern that undermines microbial disease treatment and prevention. WHO and World Bank's EcoAMR report predicts that AMR could cause 39 million deaths and $3.4 trillion in annual GDP losses by the year 2050. This is particularly critical with S. aureus, a cause of diverse infections like skin abscesses and pneumonia, where antibiotic resistance increases mortality and hinders treatment. Biofilms are one of the major causes of multi-drug resistance in S. aureus, and their inhibition can restore antibiotic sensitivity. In this study, through screening o

PD-L1 deficiency exacerbates colitis severity by remodeling gut microbiota in inflammatory bowel disease.

BACKGROUND: Inflammatory bowel disease (IBD) is a chronic autoimmune disorder driven by gut microbiota dysbiosis. As an essential immune checkpoint, Programmed death-ligand 1 (PD-L1) has been implicated in modulating gut microbiota composition. However, the precise role of PD-L1 in shaping metagenomic profiles during IBD-associated colitis remains unexplored. METHODS: DSS-induced colitis was established in both PD-L1 knockout (Pdcd1lg1-/-) mice and wild-type (wt) control mice. Clinical parameters, including disease activity index (DAI), body weight changes, colon length, and histopathological

Targeting the cGAS-STING Pathway to Modulate Immune Inflammation in Diabetes and Cardiovascular Complications: Mechanisms and Therapeutic Insights.

Type 2 diabetes mellitus (T2DM), characterized by insulin resistance and chronic hyperglycemia, markedly increases the incidence and mortality of cardiovascular disease (CVD). Emerging preclinical evidence identifies the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway as a critical mediator of diabetic cardiovascular inflammation. Metabolic stressors in T2DM-hyperglycemia, lipotoxicity, and mitochondrial dysfunction-induce leakage of mitochondrial and microbial double-stranded DNA into the cytosol, where it engages cGAS and activates STING. Subsequent TBK1/IRF3 and
