Gamma-aminobutyric acid (GABA) is the major inhibitory neurotransmitter in the brain. However, it is also produced in other sites; notably by pancreatic β cells and immune cells. The function of GABA in the immune system is at an early stage of study, but it exerts inhibitory effects that are relevant to autoimmune diseases. The study of GABAergic interactions in the immune system has centered on three main aspects: 1) the expression of GABA and the relevant GABAergic molecular machinery; 2) the in vitro response of immune cells; and 3) therapeutic applications in autoimmune diseases. T cells

BACKGROUND: Ghrelin is a gastrointestinal peptide hormone (a 28-amino acid peptide) produced primarily by X/A cells in the oxyntic glands of the stomach fundus and cells lining the duodenum cavern. It suppresses insulin secretion and action and commands a significant role in regulating food intake. The aim of the present study was to show that modified laparoscopic sleeve gastrectomy (MLSG), in which a significant part of the gastric fundus and body of the stomach is removed up to 1 inch from the pylorus vein, may contribute to decreasing circulating ghrelin levels.
METHODS: A study populatio

BACKGROUND: A vegetarian diet is considered to promote health and longevity and reduce the risk of cardiovascular diseases and cancer. However, a vegetarian diet may be deficient in some nutrients. Exclusion of animal products in vegetarian diets may affect the status of certain B-vitamins, and further cause the rise of plasma homocysteine concentration.
OBJECTIVE: The nutritional status of various B-vitamins (B(1), B(2), B(6), B(12), folic acid) and the concentration of homocysteine in blood plasma of omnivores (n = 40), vegetarians (n = 36) and vegans (n = 42) in Austria was evaluated.

An experiment was conducted to study the variation in the precaecal amino acid (AA) and energy digestibility of 4 cultivars of white-flowering peas (Pisum sativum) for broiler chickens. The 4 cultivars were grown and harvested under the same agronomic and environmental conditions. One basal diet and 8 diets including each pea cultivar at inclusion rates of either 150 or 300 g/kg were used. Peas were included at the expense of starch. Hence, the differences in dietary crude protein and AA concentrations resulted only from the inclusion of peas. Titanium dioxide was included as an indigestible m

The pineal gland serves the function of a neuroendocrine transducer converting information about day length into the nocturnal release of melatonin. Melatonin acts on the brain, particularly on the hypothalamus, to affect several biological rhythms. By employing autoradiography and 2-[(125)I]melatonin as a radioligand, the hypothalamic suprachiasmatic nucleus (SCN) and the pars tuberalis of the adenohypophysis have been identified as sites for melatonin binding exhibiting dissociation constants (K(d)s) in the 10(?10) M range. These sites were also revealed in test-tube binding assays employing

BACKGROUND: The pathophysiology of pain in patients with symptomatic thoracic disc herniation (TDH) remains poorly understood. Mere mechanical compression of the spinal cord and/or the exiting nerve root by a prolapsed disc cannot explain the pathogenesis of pain in all cases. Previous studies report a direct correlation between the levels of proinflammatory cytokines in disc biopsies and the severity of leg pain in patients with lumbar disc herniation. A similar correlation in patients with TDH has not been investigated.
PURPOSE: To correlate the cerebrospinal fluid (CSF) expression of cytok

Alzheimer's disease (AD) remains an unmet medical challenge, as there are no effective therapies that alter the disease's progression. While approaches have targeted molecules like acetylcholine (ACh) and glutamate, these strategies have provided only limited benefits and do not address the complex molecular mechanisms underlying AD development. This review suggests that β-alanine (3-aminopropanoic acid) is an underexplored neurotransmitter that could serve as a potential AD drug target. Existing evidence indicates that β-alanine modulates GABAergic and glutamatergic neurotransmission, thereby

Fluorinated liquid crystal monomers (FLCMs), emerging contaminants from electronic waste, exhibit environmental persistence and bioaccumulative potential. Despite their growing detection in humans and aquatic systems, the subcellular mechanisms of their metabolic toxicity remain poorly understood. Here, zebrafish (Danio rerio) embryos and larvae were exposed to five representative FLCMs (0.05-50 µg/L) to investigate mitochondrial and metabolic disruption. Exposure to environmentally relevant concentrations significantly reduced basal respiration by 25.0-49.8 %, ATP-linked oxygen consumption by

BACKGROUND: Deceased-donor kidneys experience cellular stress before undergoing transplantation. To alleviate this, preservation techniques were developed, including normothermic machine perfusion (NMP).
METHODS: Here, we performed kidney NMP on discarded human kidneys for up to 24 h. Volume management was regulated either by urine recirculation (UR) or urine replacement (NUR) with Ringer's lactate. Notably, UR led to longer perfusion times compared with NUR. To investigate kidney NMP metabolic traits with or without UR over time, we performed longitudinal metabolomics analyses of perfusates

Sleep deprivation induces profound metabolic disturbances in the brain, while regular physical exercise is recognized for promoting neuroprotection and energy balance. This study investigated the effects of chronic treadmill exercise on the cortical and hippocampal metabolic profiles of sleep-deprived Swiss mice using the entire 1H-NMR spectrum. Male mice (n=48) were assigned to four groups: Control, Exercise (EX), Sleep Deprivation (SD), and Exercise before Sleep Deprivation (EX+SD). The EX group underwent 8 weeks of aerobic training, and SD was induced by 72-h total sleep deprivation. Brain

BACKGROUND: The health of elderly individuals is closely linked to their protein intake and the abundance of intestinal microbiota. To investigate the impact of a compound powder made from mulberry leaf and konjac (hereinafter referred to as 'compound powder') on regulating the structure of intestinal microbiota in 15-month-old BALB/c mice that were fed a high-beef-protein diet, 16S rRNA high-throughput sequencing, reverse transcription quantitative polymerase reaction, western blot, and other biochemical methods were used to analyze the differences in intestinal microbiota, protein metabolism

Glutamate carboxypeptidase-II (GCPII) is a zinc-dependent metalloenzyme implicated in numerous neurological disorders. The pharmacophoric requirements of active-site GCPII inhibitors makes them highly charged, manifesting poor pharmacokinetic (PK) properties. Herein, we describe the discovery and characterization of catechol-based inhibitors including L-DOPA, D-DOPA, and caffeic acid, with sub-micromolar potencies. Of these, D-DOPA emerged as the most promising compound, with good metabolic stability, and excellent PK properties. Orally administered D-DOPA yielded high plasma exposures (AUCpla

Poisoning with pesticides containing convulsive substances is the cause of death of thousand people in the world. Convulsive syndrome can also develop with infection of the central nervous system, intracranial hemorrhages and strokes, alcohol withdrawal syndrome, eclampsia. Up to 10% of all of the people on the Earth had experienced seizures. The most common cause of seizures is epilepsy. Existing drugs are not always effective and have side effects. Alternative therapies are being developed. Valproates (derivatives of valproic acid) are one of the most studied drugs of choice for epilepsy and

BACKGROUND: Frontotemporal lobar degeneration is a heterogeneous disorder entailing a semantic variant of primary progressive aphasia (svPPA). A subtype of frontotemporal dementia associated with glutamate receptor subunit 3 (GluA3) antibody of the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor (AMPAR) was recently identified. Here, we describe the novelty of a svPPA associated with anti-glial fibrillary acid protein (GFAP) antibodies.
METHODS: To diagnose this 68-year-old woman we conducted a clinical examination, neuropsychological testing, CSF analysis, MRI and 18F-fluorodeo

Cyanide (CN-) pollution in agricultural systems impairs amino acid metabolism in rice plants, hence decreasing their quality and yield. Meanwhile, little is known about the effects of CN- assimilation on the innate pool of proline (Pro) and its synthesis-related amino acids (Pro-AAs) in rice plants. In this study, a hydroponic experiment was carried out to investigate the effect of exogenous KCN on indigenous levels of Pro-AAs, i.e., Pro, glutamate (Glu), arginine (Arg), and ornithine (Orn) in rice seedlings fertilized with either nitrate (NO3-) or ammonium (NH4+) through the biochemical and R

BACKGROUND: Celiac disease (CeD) is an autoimmune intestinal disorder caused by gluten protein consumption in genetically predisposed individuals. As biopsy sampling is an invasive procedure, finding novel noninvasive serological markers for screening of at-risk CeD population is a priority. Metabolomics is helpful in monitoring metabolite changes in body fluids and tissues. In the present study, we evaluated serum metabolite levels of CeD patients relative to healthy controls with the aim of introducing new biomarkers for population screening.
METHOD: We compared the serum metabolic profile

UNLABELLED: The principal intent of the investigation was to examine the influence of joint application of methyl jasmonate (MeJA, 10 μM) and a nitric oxide-donor sodium nitroprusside (SNP, 100 μM) to wheat plants grown under cadmium (Cd as CdCl2, 100 μM) stress. Cd stress suppressed plant growth, chlorophylls (Chl), and PSII maximum efficiency (F v /F m ), but it elevated leaf and root Cd, and contents of leaf proline, phytochelatins, malondialdehyde, and hydrogen peroxide, as well as the activity of lipoxygenase. MeJA and SNP applied jointly or singly improved the concentrations of key antio
