Potential use of mesenchymal stem cells in human meniscal repair: current insights.

The menisci of the human knee play an important role in maintaining normal functions to provide stability and nutrition to the articular cartilage, and to absorb shock. Once injured, these important structures have very limited natural healing potential. Unfortunately, the traditional arthroscopic meniscectomy performed on these damaged menisci may predispose the joint toward early development of osteoarthritis. Although a very limited number of studies are available, mesenchymal stem cells (MSCs) have been investigated as an alternative therapeutic modality to repair human knee meniscal tears

The role of Plasmodium knowlesi in the history of malaria research.

In recent years, a malaria infection of humans in South East Asia, originally diagnosed as a known human-infecting species, Plasmodium malariae, has been identified as a simian parasite, Plasmodium knowlesi. This species had been subject to considerable investigation in monkeys since the 1930s. With the development of continuous culture of the erythrocytic stages of the human malarial parasite, Plasmodium falciparum in 1976, the emphasis in research shifted away from knowlesi. However, its importance as a human pathogen has provoked a renewed interest in P. knowlesi, not least because it too c

The Microbiome: The Trillions of Microorganisms That Maintain Health and Cause Disease in Humans and Companion Animals.

The microbiome is the complex collection of microorganisms, their genes, and their metabolites, colonizing the human and animal mucosal surfaces, digestive tract, and skin. It is now well known that the microbiome interacts with its host, assisting in digestion and detoxification, supporting immunity, protecting against pathogens, and maintaining health. Studies published to date have demonstrated that healthy individuals are often colonized with different microbiomes than those with disease involving various organ systems. This review covers a brief history of the development of the microbiom

The mesenchyme in malignancy: a partner in the initiation, progression and dissemination of cancer.

The tumor microenvironment presents an exciting opportunity for innovative prognostic and therapeutic approaches to human cancer. The diverse cellular and extracellular contribution to tumor growth argues that prevention and cure of human cancers will result only from a multifaceted approach to cancer therapy. In this review we provide a foundation for considering the mesenchymal contribution to the tumor microenvironment. We address normal mesenchymal development, physiological interactions between the epithelium and stroma and the cellular hierarchy within these compartments. We focus on can

Novel inhibitors to Taenia solium Cu/Zn superoxide dismutase identified by virtual screening.

We describe in this work a successful virtual screening and experimental testing aimed to the identification of novel inhibitors of superoxide dismutase of the worm Taenia solium (TsCu/Zn-SOD), a human parasite. Conformers from LeadQuest(®) database of drug-like compounds were selected and then docked on the surface of TsCu/Zn-SOD. Results were screened looking for ligand contacts with receptor side-chains not conserved in the human homologue, with a subsequent development of a score optimization by a set of energy minimization steps, aimed to identify lead compounds for in vitro experiments.

Human papillomavirus: molecular and cytologic/histologic aspects related to cervical intraepithelial neoplasia and carcinoma.

Cervical cancer is unique among human cancers because it was the first cancer discovered to be virtually solely attributable to the effects of an infectious agent. Numerous epidemiologic and laboratory studies have confirmed a strong causal association between human papillomavirus infection and the development of premalignant and malignant lesions of the uterine cervix, and human papillomavirus-mediated malignant transformation is an ideal model system for the study of virally mediated carcinogenesis. Neoplastic transformation of affected cervical epithelium appears to be a direct consequence

Epidemiological studies on Fasciola hepatica in Gafsa Oases (south west of Tunisia).

Epidemiological investigations on Fasciola hepatica fasciolasis were carried out from July 2004 to June 2005 in the Gafsa oases (Tunisia) after the detection of a human case. Three habitats were studied: one in El Gsar and two in Ain Soltan. The prevalence of human infection was 6.6%. The presence of the parasite was detected through serology in 14.3% of cattle, 35% of sheep and 68.4% of goats. The plants Apium nodiflorum, Oxalis cernua and Sonchus maritimus were suspected to be at the origin of animal contamination and Apium nodiflorum was incriminated in human infection. The prevalence of th

[Association of human histocompatibility antigens with ophthalmological disorders].

Many studies have been trying to identify genetic markers for ophthalmological diseases, including, among others, the HLA (Human Leukocyte Antigens). Localized on the short arm of chromosome 6, the human leukocyte antigen system is well known for its capacity to confer susceptibility or resistance to different diseases. In view of its accentuated polymorphism, the strength and type of association differs with the disease and sometimes, with the studied ethnic-racial group. The development of molecular methods to typify HLA alleles and recent updates of their nomenclature has contributed to a b

Embryology of myelomeningocele and anencephaly.

The authors review current views on of the embryogenesis of the neural tube defects (NTDs) myelomeningocele and anencephaly. In this context, the following four approaches to the study of NTDs are discussed: normal morphogenesis and timing of early human neural development from conception to the ascent of the conus medullaris; mechanical and molecular biology of neural tube closure derived from experimental and animal models; morphological and biomechanical features of the NTDs myelomeningocele and anencephaly; and the experimental evidence for the importance of both genetic and environmental

Human caliciviruses and pediatric gastroenteritis.

Surveillance using molecular diagnostic methods developed in the past decade shows that human caliciviruses, particularly the Noroviruses, are the most important cause of pediatric acute gastroenteritis, next to rotavirus. Noroviruses also cause severe gastroenteritis in children, requiring hospitalization. The Sapovirus, a morphologically and genetically distinct genus of calicivirus, cause mainly milder endemic pediatric gastroenteritis. Noroviruses and Sapoviruses are genetically and antigenetically diverse, and multiple genetic types cocirculate in the same community. Human caliciviruses s

Pityriasis versicolor.

UNLABELLED: Pityriasis versicolor is a common superficial fungal infection of the skin. It is caused by Malassezia spp., which are normal human saprophytes. Under certain conditions, both exogenous and endogenous, the fungus can convert from a yeast to a pathogenic mycelial form. This alteration results in mild inflammation of the skin, and in characteristic clinical and histological changes. The taxonomy of Malassezia spp. has recently been modified to include six obligatorily lipophilic species, all of which can be found on human skin, plus one non-obligatorily lipophilic species, which only

Use of the C3H/He Lyme disease mouse model for the recovery of a Spanish isolate of Borrelia garinii from erythema migrans lesions.

A skin biopsy from a patient with erythema migrans was inoculated into C3H/He mice and into culture medium. A Borrelia garinii strain named Rio1 was isolated from both a direct BSK medium culture and a mouse ear-punch biopsy culture. Inoculating human tissue into mice produced a disease resulting in severe inflammation of the left tibio-tarsal joint, development of perivascular infiltrates as seen in ear-punch biopsies and the spread of spirochaetes along the skin, far from the inoculation site. The isolation of this strain confirms the circulation of this Borrelia species in Spain as a human

Cyanobacterial toxins: occurrence, modes of action, health effects and exposure routes.

Cyanobacterial toxins are produced by terrestrial- fresh-, brackish- and sea-water cyanobacteria of cosmopolitan occurrence. These toxins present acute and chronic hazards to human and animal health and are responsible for isolated, sporadic animal fatalities (mammals, fish, birds) each year. Human health problems are associated with the ingestion of, and contact with cyanobacterial blooms and their toxins. Modes of action of cyanobacterial neurotoxins, hepatotoxins and skin irritants are considered. Recent indications of the accumulation of cyanobacterial toxins in fish, their effect on crop

[Insulin-like growth factors and ovarian function].

Insulin-like growth factors (IGF) are important in physiology and function of the human ovary. Intraovarian peptides may have a potential for local modulation of follicular development serving as intercellular mediator between the ovarian cells. IGFs constitute a family of homologous, low molecular weight, single chain polypeptide growth factors named for their remarkable structural and functional similarity to insulin. Studies have suggested its synthesis in several extrahepatic tissues wherein it may play autocrine or paracrine regulatory roles. IGF-1 stimulates DNA synthesis in human granul

Expression of human immunodeficiency virus type 1 in the nervous system of transgenic mice leads to neurological disease.

Patients infected with the human immunodeficiency virus type 1 (HIV-1) frequently develop central and peripheral nervous system complications, some of which may reflect the effect of the virus itself. In order to elucidate the pathogenic mechanisms of HIV in neurological disease in a small animal model, we generated transgenic mice expressing the entire HIV genome under control of the promoter for the human neurofilament NF-L gene. The transgene was predominantly expressed in anterior thalamic and spinal motor neurons. Animals developed a neurological syndrome characterized by hypoactivity and

Inhibitory effect of a new androstenedione derivative, 14 alpha-hydroxy-4-androstene-3,6,17-trione (14 alpha-OHAT) on aromatase activity of human uterine tumors.

The development of human uterine estrogen-dependent tumors is considered to be closely related to estrogen biosynthesis. This study examined whether or not 14 alpha-hydroxy-4-androstene-3,6,17-trione (14 alpha-OHAT), a new 4-androstene-3, 17-dione derivative synthesized microbiologically, inhibits estrogen biosynthetase (aromatase) activities of human uterine tumors (i.e. uterine endometrial cancer, uterine leiomyoma and uterine adenomyosis tissues). 14 alpha-OHAT inhibited aromatase activity in all uterine tumors, dose-dependently (0.1-10 microM). Moreover, 14 alpha-OHAT did not show the bind

Biological characterization of 10-(2-propynyl) estr-4-ene-3, 17-dione (MDL 18,962), an enzyme-activated inhibitor of aromatase.

MDL 18,962 was shown to be a highly specific, potent (Ki = 3-4 nM), enzyme-activated inhibitor of aromatase with minimal intrinsic endocrine properties. The affinity of MDL 18,962 was higher for human and baboon placental aromatase than for rhesus placental or rodent ovarian aromatase. These species differences necessitated the development of a novel model of peripheral aromatase utilizing human enzyme. Human choriocarcinoma trophoblast xenografts in athymic nude mice were used for pharmacologic and pharmacokinetic evaluation of MDL 18,962. The ED50 for inhibition of aromatase activity in thes

Rabies immune globulin (human) in the prevention of rabies.

The development of rabies immune globulin of human origin is discussed. Antirabies serum of equine origin when used in conjunction with rabies vaccine, enhances significantly the chances of survival after a severe bite by a rabid animal. However, those who receive the equine serum may develop serum sickness or even anaphylaxis. To circumvent this risk, while preserving the benefit, many workers have attempted to develop rabies immune globulin of human origin. Success was impeded by the difficulty in obtaining rabies immune plasma with the needed antibody level. Despite this and other obstacles

Age estimation of individuals aged 5-23 years based on dental development of the Indonesian population.

Dental development can be used to estimate age for forensic purposes. However, most of the currently available methods are less reliable for the Indonesian population due to population variability. This study presents a new method and evaluates other methods that utilize dental development to estimate the age of Indonesian people. Panoramic radiographs of 304 young Indonesian people aged 5-23 years old were analysed for deciduous tooth root resorption, permanent tooth calcification, and eruption. The extent of tooth root resorption was determined based on AlQahtani's modified Moorrees et al. m

Utilization of Model-Informed Drug Development in Early Clinical Development: Case Study of a Hb Modulator for Treatment of Sickle Cell Disease.

Model-informed drug development (MIDD) has an important role to play at various stages of drug development. In this case study, the application of MIDD in the early clinical development of a small-molecule inhibitor of sickle hemoglobin (Hb) polymerization is highlighted. Starting from the setting of quantitative criteria, to using a combination of PKPD and semi-mechanistic RBC modeling, and key biomarker analysis, based on data from a healthy participant first-in-human (FIH) study and historical data, we chart the strategic approaches planned and taken to inform decisions for the program, and
