Post-Traumatic Stress Disorder Symptoms Are Related to Cognition after Complicated Mild and Moderate Traumatic Brain Injury but Not Severe and Penetrating Traumatic Brain Injury.

Although post-traumatic stress disorder (PTSD) has been associated with worse cognitive outcomes after mild traumatic brain injury (TBI), its impact has not been evaluated after more severe TBI. This study aimed to determine whether PTSD symptoms are related to cognition after complicated mild, moderate, severe, and penetrating TBI. Service members (n = 137) with a history of complicated mild/moderate TBI (n = 64) or severe/penetrating TBI (n = 73) were prospectively enrolled from United States Military Treatment Facilities. Participants completed a neuropsychological assessment one year or mo

Plasma membrane ordering agent pluronic F-68 (PF-68) reduces neurotransmitter uptake and release and produces learning and memory deficits in rats.

A substantial body of evidence indicates that aged-related changes in the fluidity and lipid composition of the plasma membrane contribute to cellular dysfunction in humans and other mammalian species. In the CNS, reductions in neuronal plasma membrane order (PMO) (i.e., increased plasma membrane fluidity) have been attributed to age as well as the presence of the beta-amyloid peptide-25-35, known to play an important role in the neuropathology of Alzheimer's disease (AD). These PMO increases may influence neurotransmitter synthesis, receptor binding, and second messenger systems as well as si

Screening Techniques for Drug Discovery in Alzheimer's Disease.

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by progressive and irreversible impairment of memory and other cognitive functions of the aging brain. Pathways such as amyloid beta neurotoxicity, tau pathogenesis and neuroinflammatory have been used to understand AD, despite not knowing the definite molecular mechanism which causes this progressive disease. This review attempts to summarize the small molecules that target these pathways using various techniques involving high-throughput screening, molecular modeling, custom bioassays, and spectroscopic detection tools. Creutzfeldt-Jakob disease: A case report.

Creutzfeldt-Jakob Disease (CJD) is a rare, fatal neurodegenerative disorder that is caused by prion proteins. Patients often present with rapidly progressive dementia, ataxia, myoclonus, memory impairment, visual problems, and changes in personality. In this case report, we aimed to address the course of a 62 year old female who presented with progressive decline in cognitive function and died within 6 months of presentation. The patient underwent cerebrospinal fluid testing, MRI brain, and electroencephalography during her stay in the hospital. Ultimately, an autopsy was performed, which demo

Functional brain alterations during self-reference processing in adolescents with sexual abuse-related post-traumatic stress disorder: A preliminary report.

We proposed to explore the functional brain changes during a self-reference processing (SRP) task in adolescents with sexual abuse-related post-traumatic stress disorder (N = 10), compared with healthy adolescents (N = 10). While patients showed no behavioral disturbances in (SRP), they exhibited changes in activity and connectivity in regions involved in emotional regulation (amygdala and dorsal prefrontal cortex) and semantic memory (temporal and ventrolateral prefrontal regions). These preliminary results suggest that these alterations may have an effect on self-esteem which may contribute

Late-onset Leber hereditary optic neuropathy mimicking Susac's syndrome.

Leber hereditary optic neuropathy (LHON) is a mitochondrial disorder characterized by bilateral painless optic atrophy and blindness. It usually occurs in young men in association with three major mutations in the mitochondrial genome (mtDNA). We report a patient with a history of alcohol abuse who developed at age 63 years visual impairment, sensorineural hearing loss, and memory dysfunction, suggestive of Susac's syndrome. The patient carried the heteroplasmic mt. 11778G>A mutation on the T2e mtDNA haplogroup. It remains unclear if chronic alcohol abuse combined with the mitochondrial geneti

Domains of childhood teasing and psychopathological symptoms in Dutch adolescents.

This study examined relations between memories for childhood teasing and symptoms of social anxiety, depression, and eating disorders in 130 Dutch adolescents. Participants completed the Teasing Questionnaire-Revised which measures how much people recall having been teased during childhood, as well as the Social Anxiety Scale for Children, the Child Depression Inventory, and the Children's Eating Attitude Test. Analysis yielded significant positive correlations between self-reported recollections of childhood teasing and various symptoms. Furthermore, some evidence indicated specific domains o

A novel mutation (L250V) in the presenilin 1 gene in a Japanese familial Alzheimer's disease with myoclonus and generalized convulsion.

This study reports a novel presenilin 1 (PS1) gene mutation in a Japanese family with Alzheimer's disease (AD). Two patients developed progressive memory disorder with disorientation around 50 years of age and showed myoclonus with frequent tonic-clonic seizures several years later. Direct sequencing of the proband's PS1 gene revealed a novel mis-sense mutation (leucine-to-valine at residue 250 (L250V)). This mutation was found in both patients, but not in a normal family member or normal Japanese control subjects. Thus, L250V is a novel PS1 gene mutation responsible for familial AD (FAD) in J

Food and hypersensitivity in functional dyspepsia.

It has long been known that stress affects both the stomach and colon, as shown by the very high prevalence of gastrointestinal symptoms among patients with psychiatric illness. The source may be limbic or peripheral, involving encoded memories or physiological changes. It is important to realise that physical symptoms such as those of functional dyspepsia do not only mean that the patient has a stomach disorder which needs to be identified and treated with specific pharmacological remedies, they often represent, in metaphorical and symbolic form, a state of disharmony brought about a specific

[Conduction aphasia and phonemic disorder].

Conduction aphasia is usually described as a repetition impairment. Semiology or pathophysiology cannot be explained with this definition. We report a single case particularly demonstrative. The patient showed spontaneous speech, denomination, repetition and reading impairments. Main errors were phonemic paraphasia. No arthric disorder nor comprehension impairment was observed. Damage of supramarginalis gyrus and Wernicke's area was found. A cognitive analysis suggested that the phonological buffer and the working memory were impaired. Implication for rehabilitation, which included segmentatio

Linking epigenetic age acceleration to self-reported daily memory lapses: evidence from the National Study of Daily Experiences.

DNA methylation algorithms, such as DunedinPACE, are increasingly used to study the mechanisms of aging and to identify associations with risk factors in adult development and aging. Growing evidence suggests that an increased pace of epigenetic aging is associated with cognitive impairment and dementia, but the correlation between epigenetic aging and subjective cognitive complaints (eg, forgetting a name or forgetting to take your medication), an early marker of dementia risk, remains unexplored. Using data from The National Study of Daily Experiences (NSDE) and Midlife in the United States

Medial entorhinal-hippocampal desynchronization parallels the emergence of memory impairment in a mouse model of Alzheimer's disease pathology.

Alzheimer's disease (AD) is a neurodegenerative disease characterized by progressive impairments in episodic and spatial memory, as well as circuit and network-level dysfunction. While functional impairments in medial entorhinal cortex (MEC) and hippocampus (HPC) have been observed in patients and rodent models of AD, it remains unclear how communication between these regions breaks down in disease, and what specific physiological changes are associated with the onset of memory impairment. Here, we use silicon probes to simultaneously record neural activity in MEC and HPC before or after the o

From painful memories to present relief: Enhancing CBT efficacy for depression through imagery rescripting in a randomized controlled trial.

INTRODUCTION: Imagery Rescripting (ImRs) is an evidence-based technique for modifying the emotional meaning of distressing autobiographical memories, which are common in depression. This randomized controlled trial examined whether adding three ImRs sessions to ongoing Cognitive Behavioral Therapy (CBT) improves outcomes compared with an active imagery-based control condition. METHOD: Sixty-six patients with major depressive disorder (MDD) receiving outpatient CBT were randomized to three sessions of either (1) ImRs or (2) guided imagery (GI), both delivered as adjunctive interventions to rou

Hypothalamic PVH Tyrosine Hydroxylase-Positive (Predominantly Dopaminergic) Neurons Mediate the Bidirectional Control of Neuropathic Pain and Comorbid Memory Impairments.

BACKGROUND: Neuropathic pain often co-occurs with memory impairment, yet the underlying neural mechanisms remain unclear. Here, we identify tyrosine hydroxylase-positive (TH+) neurons in the paraventricular hypothalamic nucleus (PVH), the majority of which are dopaminergic, as a key node in the circuit mediating this comorbidity. METHODS: A neuropathic pain model was established in C57BL/6J mice through sciatic nerve chronic constriction injury (CCI). Mechanical and thermal hypersensitivity were assessed using Von Frey and cold plate tests, respectively. Memory function was evaluated using no

SirT2 Inhibition is Associated with Improvements in Depression-like Behavior and Memory Impairment in Olfactory Bulbectomized Mice.

Increased silent information regulator protein 2 (SirT2) expression in the prefrontal cortex (PFC) has been reported to be associated with the development of depression; however, its functional contribution to depression-like behavior and memory impairment remains incompletely understood. In this study, we examined alterations in SirT2 expression in the PFC of olfactory bulbectomized (OBX) mice, a well-established model of depression, and evaluated the effects of AK-7, a selective SirT2 inhibitor, on OBX-induced depression-like behavior and memory impairment. On day 21 after surgery, OBX mice

Mild Traumatic Brain Injury Alters Hippocampal Sharp-Wave Ripple Properties: Implications in Memory Impairments.

A large body of evidence has shown that sharp wave ripples (SWRs), highly synchronized oscillations of the local field potential (LFP) in the CA1 pyramidal layer of the hippocampus, play a prominent role in memory consolidation. SWRs typically occur in periods of non-REM sleep, but have also been observed during awake rest periods of animals engaged in cognitive tasks. These events have been observed in rodents, non-human primates, and humans, suggesting they are evolutionarily conserved and contribute to memory formation. Numerous neurological diseases and insults, including traumatic brain i

Effects of Osteopontin Combined with Milk Fat Globule Membrane Proteins on Scopolamine-Induced Learning and Memory Impairment in Mice.

This study investigated the effects of osteopontin (OPN) combined with milk fat globule membrane (MFGM) proteins on scopolamine-induced learning and memory impairment in mice. A dementia model was established through intraperitoneal injection of scopolamine, followed by random allocation into seven experimental groups: blank control, model control, OPN alone, MFGM alone, and three combination groups (low-, intermediate-, and high-dosage OPN + MFGM). Cognitive performance was evaluated using the Morris water maze test, accompanied by quantification of hippocampal acetylcholinesterase (AChE) act
