The role of pigs and slaughterhouses in the transmission of Listeria monocytogenes into the meat processing chain.

Listeria (L.) monocytogenes is one of the most dangerous foodborne pathogens. Many different food products are known to be the source of listeriosis including pork. However, the information on the role of pigs in L. monocytogenes epidemiology is very limited. In this study, we analysed the occurrence of Listeria spp. in 800 faecal samples from Austrian pigs, in swab samples from the farm environment (n = 280) and from two corresponding slaughterhouses (n = 305). L. monocytogenes was rarely present in pigs (1.5%), more often in their farm environment (7.86%) and commonly in the two slaughterhou

Allele-resolved monosome and polysome sequencing identifies cis-variants affecting mRNA translation.

To prioritize germline genetic variants affecting mRNA fate at the translational level, we used sucrose gradient-based isolation of 80S monosomes and polysomes, followed by RNA sequencing in RPE-1 cells, under mock and nutlin treatment conditions. Differential gene expression analysis confirmed a canonical p53 response. Heterozygous SNPs and SNVs were identified from the RNA-sequencing data, and allelic fractions were calculated for total, monosomal, and polysomal mRNAs. Variants showing reproducible allelic fraction differences across fractions beyond experimental variability were defined as

Genome-Wide Association Study of Vertical Jump Performance Among Elite Badminton Players.

Vertical jump performance is known to be a moderately heritable trait. However, previous studies on sport genetics have largely relied on candidate-gene approaches, which do not adequately reflect the polygenic nature of explosive performance, particularly among elite badminton players. Therefore, the aim of the present study was to identify genetic variants associated with lower-limb explosive performance, assessed via vertical jump measures, among elite Turkish badminton players using a genome-wide association study (GWAS) approach. The present study included 90 elite male (n = 47) and femal

The devil is in the enhancer: decoding regulatory variation at the ZNF827 locus in SCAD.

Genome-wide association studies have identified numerous susceptibility loci for spontaneous coronary artery dissection (SCAD), yet translating non-coding genetic associations into molecular mechanisms remains a major challenge. In the present issue, Liu et al. provide functional evidence linking the SCAD-associated variant rs13128814 at the ZNF827 locus to the regulation of a vascular smooth muscle cell (SMC)-specific enhancer. The authors show that the risk allele creates a de novo binding site for nuclear factor I (NFI) transcription factors within a developmentally regulated putative enhan

Multi-omics analysis of perfluorooctanoic acid and glioblastoma: insights from Mendelian randomization, network toxicology, and molecular docking.

Glioblastoma (GBM) is the most lethal primary brain tumor with limited treatment options. Perfluorooctanoic acid (PFOA), a ubiquitous "forever chemical" classified as a group 1 carcinogen by the IARC, can cross the blood-brain barrier and accumulate in neural tissue. This study aimed to determine the causal relationship between genetically predicted higher circulating PFOA levels and GBM risk, elucidate underlying molecular mechanisms, and identify potential therapeutic targets using an integrated multi-omics framework. Two-sample Mendelian randomization (MR) analysis was conducted using 23 si

Large-Scale Genotype-Based Trait Imputation With Multi-Ancestry GWAS Data.

Genome-wide association studies (GWAS) have been instrumental in identifying genetic variants associated with complex traits and diseases, including Alzheimer's disease (AD). However, traditional GWAS approaches often focus on European populations, which may lead to loss of power and limit the generalizability of findings across diverse ancestries. On the other hand, LS-Imputation, a nonparametric trait imputation method, leverages GWAS summary statistics and genotype data to impute missing traits, which can then be used for GWAS and other downstream analyses. Although LS-Imputation has been a

ACTN3 rs1815739 and BDNF rs6265 Polymorphisms May Not Be Associated with Handgrip Strength in Elite Wrestlers.

BACKGROUND/OBJECTIVES: Handgrip strength (HGS) is a widely used indicator of upper-limb muscular strength and a practical proxy for neuromuscular performance across both clinical and athletic contexts. Although HGS is heritable, evidence supporting specific genetic contributors in elite athletes remains limited. Thus, the present study investigated the associations of two functional polymorphisms, BDNF rs6265 (p.Val66Met) and ACTN3 rs1815739 (p.R577X), with HGS performance in elite wrestlers, integrating neuromuscular and muscle fiber-related biological pathways. Genome-wide association studies of infant and toddler temperament in European and multi-ancestry populations.

Early temperament, such as socio-emotional development and activity level, varies widely, yet its underlying biological associations are not understood. We identified genetic variation associated with infant and toddler temperament using genome-wide association meta-analyses. We studied parent-rated emotionality, activity, shyness and sociability (n = 43,963-72,663) in the second and third postnatal years and a cross-age average. Cross-age single nucleotide polymorphism heritabilities for emotionality, activity, shyness and sociability were 6.79% (95% confidence interval (CI), (4.71%, 8.87%)),

Genetic correlation between inflammatory bowel disease and educational attainment: unveiling shared genetic mechanisms.

BACKGROUND: A potential genetic link between inflammatory bowel disease (IBD) and educational attainment has been suggested. Understanding the underlying mechanisms of this genetic relationship is crucial for advancing the knowledge of their co-occurrence patterns. METHODS: Using genome-wide association study data for both IBD and educational attainment, a multiphase analytical approach was applied to examine their genetic correlation. The study involved three phases: first, linkage disequilibrium score regression and high-definition likelihood models were used to assess genome-wide genetic c

WDR12 and HIVEP3 are contributors to cognitive preservation in Amish SuperAgers.

INTRODUCTION: Cognitive SuperAgers (SAs) are individuals aged 80+ with exceptional episodic memory performance for their age, exceeding middle-aged adult norms. This study integrates family- and association-based methods to identify genetic variants associated with SAs in the Midwestern Amish population. METHODS: Eighty-three Amish SAs were grouped into 16 pedigrees for parametric and non-parametric linkage analysis. Variants in linked regions (heterogeneity logarithm of the odds [HLOD] or Kong and Cox logarithm of the odds [LOD*] ≥ 3) were tested for association with SAs using two contrasts:

Predominantly Independent Genetic Control Between Growth and Visceral White Nodules Disease Resistance Revealed by High-Density Linkage Map and QTL Mapping in Larimichthys crocea.

The large yellow croaker (Larimichthys crocea) is a key mariculture species in China, however, its industry is threatened by visceral white nodules disease (VWND) caused by the bacterium Pseudomonas plecoglossicida. A significant challenge in breeding is the potential genetic trade-off between growth and disease resistance. To investigate their genetic relationship, we constructed a high-density SNP-based genetic linkage map for L. crocea using a F1 full-sib family (n = 150). The map comprised 24 linkage groups with 32,429 bin markers and an average interval of 0.051 cM. Based on this map, we

Exploratory Mendelian randomization analysis: The genetic susceptibility of CT examination preference and the risk of hypertrophic cardiomyopathy.

Hypertrophic cardiomyopathy (HCM) is a common hereditary heart disease, characterized by myocardial hypertrophy, reduced ventricular cavity, and abnormal myocardial fiber structure. Computed tomography (CT) is a commonly used imaging tool in the diagnosis and risk assessment of cardiovascular diseases. This study aims to conduct an exploratory Mendelian randomization (MR) analysis to investigate the genetic association between the propensity to undergo CT (inferred from genetic variations) and the risk of hypertrophic cardiomyopathy (HCM). In this study, a two-sample MR method was employed. Us

Assessing the causal association between obstructive sleep apnea and the risk of atrial fibrillation.

Atrial fibrillation (AF) and obstructive sleep apnea (OSA) are associated, however the majority of the research to date has been observational in nature and is unable to definitively prove a causative relationship. Using Mendelian randomization (MR) analysis and meta-analysis, this study attempts to assess the causal link between AF and OSA in more detail. Six genome-wide association studies (GWAS) datasets were subjected to de novo 2-sample MR analysis, and a series of analyses were carried out to guarantee the validity of the MR findings. In order to better elucidate the relationship between

Mosaic Accumulation of Somatic Genetic Variation and Estimates of Age in the Long-Lived Reef-Building Coral Acropora palmata.

Somatic genetic variation (SoGV) accumulates during an organism's lifetime and is traditionally viewed as detrimental rather than adaptive due to links with cancer and senescence. However, in modular organisms like corals, deleterious mutations can be purged at the cellular or polyp level, while adaptive mutations may rise in frequency as polyps create genetically distinct modules. Quantifying the somatic genetic landscape in corals is necessary to understand the role these mutations may have in coral and clonal animal development and evolution. Here, we catalogue somatic genetic variation in

Mutations in ERG11, TAC1B, and CDR1 reduce fluconazole accumulation in drug-resistant Candidozyma auris isolates.

Fluconazole (FLC)-resistant Candidozyma auris isolates have reduced drug accumulation compared to azole-susceptible isolates. Of 119 C. auris isolates, 83 out of 87 resistant isolates (~95%) had extremely low fluconazole uptake, whereas 30 out of 32 susceptible isolates (~93%) had high fluconazole uptake. In search of a genetic explanation for this phenomenon, we compared metadata for TAC1B and CDR1 single-nucleotide polymorphisms (SNPs) and found overlap with many but not all isolates that are FLC resistant. We found that CDR1 mutations are common in resistant isolates from Clade 1, and TAC1B

Genetic landscape of adult executive function reveals a cell-type-specific developmental origin.

Executive function is an essential cognitive domain for typical human behavior which is disrupted in neurodevelopmental and neurodegenerative disorders, but little is known about its underlying molecular basis. To address this, we perform genome-wide association studies (GWAS) using three different measures of executive function in UK Biobank (N = 84,238) and NIHR BioResource's Genes and Cognition (N = 9932) study participants, followed by a meta-analysis. The trail-making alphanumeric (TMA) measure is the most heritable phenotype (h²=7-26%), associated with 18 independent loci that exhibit a
