Incidence, severity, and time-course of motoneurone dysfunction in myotonic dystrophy: their significance for an understanding of anticipation.

The numbers of functioning motor units and the amplitudes of the maximum evoked muscle responses have been measured in 198 muscles of 102 patients with myotonic dystrophy. Losses of units could be demonstrated in most of the extensor digitorum brevis and thenar muscles but less commonly in the hypothenar groups. A more proximal limb muscle, the soleus, was also shown to be involved frequently. Investigation of two premature infants withmyotonic dystrophy also revealed reductions of functiong units; in one infant clinical improvement was associated with increased muscle innervation. Repeated ex

Anomalous lymphocyte-antigen reaction in relatives of multiple sclerosis patients. A study of a possible genetic factor in the disease.

A combined familial study of multiple sclerosis (MS) in England and in the Rostock area of the GDR using the macrophage electrophoretic mobility (MEM)-LAD test embracing 132 relatives has revealed a closely similar pattern of distribution of "anomalous" LAD (Linoleic Acid Depression) values in relatives (77% type of reaction) to that originally reported in the British study. The anomaly in predominantly associated with females--all mothers of MS patients being affected, whilst daughters and sisters are also represented. In addition unusual full MS type of reaction (90% reduction) has been foun

Limits of using oligonucleotides for allele-selective inhibition at trinucleotide repeat sequences - targeting the CAG repeat within ataxin-1.

Trinucleotide repeats are responsible for many genetic diseases. Previous studies have shown that duplex RNAs (dsRNAs) can be used to target expression of a mutant repeat allele while leaving expression of the wild-type allele untouched, creating opportunities for allele-selective inhibition and better therapeutic outcomes. In contrast to successes with other genes, we report here that we cannot achieve allele-selective inhibition when targeting the expanded CAG repeat within Ataxin-1 (ATXN1), the cause of spinal cerebellar ataxia-1 (SCA1). The most likely explanation for this unfavorable outc

Efficient and cancer-selective gene transfer to hepatocellular carcinoma in a rat using adenovirus vector with iodized oil esters.

Gene therapy for cancer requires efficient, selective gene transfer to cancer cells. In gene therapy for hepatocellular carcinoma (HCC), gene transfer is efficient for small tumors, but not for large tumors. The delivery of anticancer agents and of iodized oil esters as embolic agents through tumor-feeding arteries is known as transarterial embolization. We speculate that genes may be efficiently and selectively transferred for HCC using iodized oil esters because these esters may remain together with a genetic vector within HCC selectively. Hence, we have studied the effect of iodized oil est

The role of genomic oxidative-reductive balance as predictor of complex regional pain syndrome development: a novel theory.

The aftermath of sequencing the human genome has birthed many efforts to utilize an individual's genetic information in order to tailor optimal treatment strategies - so-called personalized medicine. An individual's genetic information may eventually help diagnosis and treatment, as well selecting optimal pharmacologic agents based partly on how well they reach their target, how well they will bind to and produce an effect at their targets, how well they will be metabolized, and the profile of their adverse effects. It also appears that clinicians may be able to utilize an individual's genetic

ERF Transcription Factor PagERF135A Negatively Regulates Drought Tolerance in Poplar by Modulating Sucrose Metabolism and Brassinosteroid Biosynthesis.

Drought is one of the primary environmental constraints limiting poplar growth. Identifying key drought-resistance genes and their regulatory mechanisms is crucial for genetic improvement of drought tolerance in poplar. Time-course transcriptome analysis of 84K poplar (Populus alba × P. glandulosa) under drought stress revealed that the ERF family is among the most responsive transcription factor families, and PagERF135A exhibited the most significant upregulation, suggesting a key regulatory role. Genome-wide identification of the ERF family in 84K poplar identified 276 members, with expansio

Long Non-Coding RNAs: Key Regulators of Economic Traits in Livestock From Mechanisms to Breeding Applications.

Long non-coding RNAs (lncRNAs) have emerged as critical regulators of economically important traits in livestock, including growth, reproduction, disease resistance and product quality. Unlike protein-coding genes, lncRNAs orchestrate gene expression at epigenetic, transcriptional and post-transcriptional levels, acting as molecular switches, scaffolds, decoys, and competing endogenous RNAs (ceRNAs). This review synthesises current knowledge on key lncRNA mechanisms such as ceRNA networks, liquid-liquid phase separation (LLPS) and RNA-targeting clustered regularly interspaced short palindromic

Microbes as Key Players During Evolution: I From the Beginnings to the Appearance of Eukaryotes.

Life on Earth originated in the Precambrian, around 4 billion years ago. This review focuses on microbes (viruses, bacteria, archaea) which developed in these prehistoric times and their roles during the later evolution of protists (eukaryotes). Among the different theories of life evolution, the author favors the theory of the RNA world as having been first. Over the past few decades, new molecular biological technologies and findings have changed the paradigm of mutation and selection as being the main driving force of biological evolution. Instead, microbes and microbe-derived genetic eleme

Genome-wide CRISPR screens identify shared and context-specific regulators of podocyte resistance to oxidative injury.

Podocyte injury is a convergent driver of glomerular disease progression, yet the genetic determinants that govern podocyte vulnerability to oxidative stress remain incompletely defined. Here, we performed genome-wide CRISPR-Cas9 knockout screens in differentiated human podocytes exposed to two complementary injury models: Adriamycin (ADR), a clinically relevant inducer of podocyte damage, and hydrogen peroxide (H2O2), a direct oxidative stress challenge. Human podocytes were transduced with a genome-wide sgRNA library, differentiated under non-permissive conditions and subjected to optimized

Natural variation in GmRVE4d facilitates soybean latitudinal adaptation by regulating GmPRR5a-dependent flowering and maturity.

Timely flowering and maturity are crucial for plant reproduction and environmental adaptation. Light-dark cycle-associated regulatory networks integrate photoperiodic cues with intrinsic developmental programs and play pivotal roles in flowering, maturity and environmental adaptation. Soybean is a short-day crop with strong photoperiod responsiveness; however, the molecular mechanisms by which these networks mediate soybean adaptation to diverse environments remain largely elusive. Here, we combine genome-wide association analysis with deep learning-based assessment of soybean maturity variati

Pharmacogenetics in pediatric cardiology.

Children with congenital and acquired heart disease frequently require complex and prolonged pharmacotherapy, often involving off-label cardiovascular medications with narrow therapeutic ranges. Consequently, variability in drug response and medication-related adverse events remains a significant clinical challenge in pediatric cardiology. Pharmacogenetics provides a framework to individualize drug selection and dosing by incorporating genetic variability alongside established clinical and developmental factors. This review outlines the current clinical utility and emerging role of pharmacogen

An Intronic Structural Variant in GhEBS1 Drives Cotton Fruit Size Divergence by Modulating Chromatin Accessibility and Gene Expression.

Fruit size is not only a crucial fitness trait in plant evolution but also an important agronomic target in crop domestication and improvement, yet its regulatory mechanisms remain poorly characterized. Here, we identified an F-box E3 ubiquitin ligase gene, GhEBS1 (ENLARGED BOLL SIZE 1), as a positive regulator of cotton boll size via map-based cloning. An 845-bp structural variant in the promoter-proximal intronic region of GhEBS1 alters local chromatin accessibility and histone modifications, specifically H3K27 and H3K9 methylation levels and acetylation, thereby modulating GhEBS1 expression

Diagnostic and Therapeutic Challenges Related to HER2 Heterogeneity in Gastric Cancer: Bridging Molecular Pathology and Clinical Decision-Making.

HER2 testing represents a cornerstone of the treatment algorithm in advanced gastric and gastroesophageal junction adenocarcinoma (GC), yet its evaluation remains complex due to tumor heterogeneity and methodological variability. Unlike breast cancer, HER2 expression in GC is often incomplete and heterogeneous, resulting in discordant results between biopsies, resections, and metastatic sites. Both spatial and temporal HER2 heterogeneity are key determinants of testing reproducibility, diagnostic accuracy, and treatment selection and response in GC. Optimizing sampling through multiple, well-t

Evolutionary trajectories of myelodysplastic syndromes/neoplasms.

Myelodysplastic syndromes/neoplasms (MDS) are a heterogeneous group of myeloid malignancies characterized by clonality, cytopenia, bone marrow dysplasia, and a variable risk of progression to acute myeloid leukemia (AML). With few therapeutic advances beyond the introduction of hypomethylating agents twenty years ago, MDS remains associated with poor overall survival, limited curative options, and frequent relapse or resistance to available treatments. Breakthroughs in genomic and single-cell technologies have transformed our understanding of MDS pathogenesis, revealing that disease initiation

Mapping the peptide interaction fingerprint of the Behçet's disease-associated HLA-B∗51.

The strongest genetic risk factor for Behçet's disease, a relapsing inflammatory disorder marked by recurrent mucocutaneous ulcers and uveitis, is HLA-B∗51:01, a class I major histocompatibility complex (MHC class I) allele that presents intracellular peptides to CD8+ T cells. The molecular mechanisms linking the peptide preferences of this allele to dysregulated immunity remain unclear, limiting efforts to design peptide-based modulators of antigen presentation. Here, we define HLA-B∗51:01's peptide selection rules by mapping the "interaction fingerprint" of 36 self-peptides using an extensiv

Determinants of Epitope Immunodominance Hierarchies in Protein Antigens at the Molecular, Structural, and Cellular Levels, Including MHC-Dependent Mechanisms.

BACKGROUND: Epitope immunodominance describes the phenomenon whereby some antigenic epitopes preferentially elicit immune responses, whereas other epitopes remain subdominant or immunologically silent. Epitope immunodominance hierarchies are the result of coordinated molecular, structural, and cellular mechanisms. These hierarchies emerge from selective immune recognition processes that balance effective immunity and self-tolerance mechanisms. OBJECTIVE: The aim of this study is to describe the molecular, structural, and cellular parameters that mediate epitope immunodominance in antigens. In

Metabolite-Pool Networks and Biological-Non-Enzymatic Synergy in Volatile Flavor Formation by Lactic Acid Bacteria in Fermented Foods: A Review.

Volatile flavor compounds (VFCs) are major determinants of sensory quality in fermented foods. In lactic acid bacteria (LAB)-containing systems, their formation reflects precursor metabolism, strain-specific metabolic capacity, environmental and community regulation, and non-enzymatic chemistry. This review develops an integrated precursor-pool-flux framework for LAB-associated VFC formation. Carbohydrate/citrate-, protein/amino-acid-, and lipid-related routes provide the principal precursor inputs, which intersect to different extents at three functional metabolite pools centered on α-keto ac

Redox Homeostasis and Oxidative Stress in Schizophrenia: Glutathione-NMDA-Neuroimmune Convergence and a Hypothesis-Generating Iron-Lipid Redox Extension.

Schizophrenia is a heterogeneous neurodevelopmental disorder in which genetic liability, developmental exposures, illness stage, treatment, and metabolic or inflammatory comorbidity may shape redox biology. This narrative review evaluates the human and mechanistic evidence for impaired redox adaptation as a convergence mechanism linking glutathione (GSH) regulation, N-methyl-D-aspartate receptor hypofunction, parvalbumin-interneuron and perineuronal-net vulnerability, mitochondrial-glial dysfunction, and neuroimmune signalling. The findings do not support a uniform oxidative abnormality across

CD117 epitope-shielded hematopoietic stem cell transplantation with toxin-free conditioning and in vivo selection ameliorates a β-thalassemia model.

Clinical evidence demonstrates that ex vivo gene therapy and genome engineering of hematopoietic stem and progenitor cells (HSPCs) could represent one-time cures. However, while genome editing itself has become increasingly efficient and precise, the toxic conditioning required for hematopoietic stem cell transplantation remains a major barrier to broad clinical implementation of these otherwise curative therapies. In particular, the use of busulfan for myeloablative conditioning constitutes a major safety concern. While preclinical studies established CD117 as a promising target for antigen-s
