Arthritis and sepsis caused by Staphylococcus aureus: can the tissue injury be reduced by modulating the host's immune system?

In recent years, a number of virulence factors that are either expressed, secreted or sequestered by staphylococci have been shown to affect the outcome of the infective process. Secretion of superantigens and exposure of the immune system to sequestered bacterial DNA aggravates the inflammatory response of the infected host to a point that can be life threatening. This exaggerated inflammatory response is mediated by cytokines originating from activated T cells and macrophages. Recent studies show that downregulation of lymphocyte and macrophage responses (e.g. by treatment with corticosteroi

Clinical experience and outcomes of community-acquired and nosocomial methicillin-resistant Staphylococcus aureus in a northern Australian hospital.

Methicillin-resistant Staphylococcus aureus (MRSA) is a well-recognized cause of hospital-acquired sepsis. We reviewed the clinical features of a new variant of community-acquired MRSA originally described from the Kimberley region of northern Western Australia (WA MRSA). This strain has become an increasing cause of community- and hospital-acquired sepsis at Royal Darwin Hospital (RDH) in the Northern Territory, especially in Aboriginal Australians from remote communities. Fifty percent of WA MRSA was community-acquired, with 76% in Aboriginals. Like the MRSA from eastern Australia (EA MRSA),

Outcome of Staphylococcus aureus bacteremia according to compliance with recommendations of infectious diseases specialists: experience with 244 patients.

To determine whether recommendations of infectious diseases specialists affect outcome for patients, we evaluated 244 hospitalized patients with Staphylococcus aureus bacteremia. We offered our management recommendations to each patient's physicians and then assessed the clinical outcome for both patients for whom our consultative advice was followed and those for whom our advice was not heeded. All patients were followed up for 12 weeks after their first positive blood culture. Our management advice was followed for 112 patients (45.9%) and partially or completely ignored for 132 patients (54

Correlation of histopathologic and bacteriologic changes with cytokine expression in an experimental murine model of bacteremic Staphylococcus aureus infection.

Staphylococcus aureus infections are often life threatening. Relatively little is known about the host response to these infections, in particular, the role played by cytokines. We established a mouse model of bacteremic S. aureus infection to correlate bacteriologic findings and pathologic changes with cytokine gene expression. Bacterial density in blood and tissue was highest at 1 h and minimal by 48 h. Despite the rapid clearance of bacteria, pathologic abnormalities and inflammatory cytokines were detected after clearance of the bacteria. The number of infiltrating inflammatory cells, as w

Effect of Merocel vaginal sponge on growth of Staphylococcus aureus and production of toxic shock syndrome-associated toxins.

BACKGROUND: Staphylococcus aureus toxic shock syndrome toxin-1 and enterotoxin B are the major causes of toxic shock syndrome. These toxins are produced in sufficient concentrations to produce illness in the presence of certain tampons. This necessitates evaluating tampons, as well as wound dressings for their effects on S. aureus growth and toxin production. STUDY DESIGN: In this study, the Merocel vaginal sponge was evaluated both in vitro and in vivo in a rabbit model for effect on S. aureus. The Merocel sponge was tested in Erlenmeyer shake flasks containing growth media and in dialysis t

Reducing surgical site infection in implant-based breast reconstruction: Impact of preoperative Staphylococcus aureus screening and decolonisation.

BACKGROUND: Implant-based breast reconstruction (BR) is widely performed after curative or prophylactic mastectomy. Surgical site infection (SSI) is a frequent and dreaded complication that is known to compromise the outcomes of BR. Nasal carriage of Staphylococcus aureus is a known risk factor for SSI. Preoperative nasal screening followed by decolonisation in case of positive result has been offered at the Oscar Lambret Centre since 2018. In this study, we evaluated the association between the realisation of an S. aureus screening and decolonisation programme on the risk of SSI after breast

Recurrent Invasive Staphylococcus aureus Infections in a Non-Neutropenic Hemodialysis Patient With Myelodysplastic Syndrome: A Case Report.

INTRODUCTION: Myelodysplastic syndromes (MDS) may be overlooked in patients undergoing hemodialysis (HD) because anemia is commonly attributed to renal anemia and treated with erythropoiesis-stimulating agents. MDS is also associated with an increased risk of bacterial infections due to neutropenia and qualitative immune dysfunction. We report recurrent invasive methicillin-susceptible Staphylococcus aureus (MSSA) infections in a patient undergoing HD with concomitant MDS. CASE PRESENTATION: The patient was a 76-year-old man undergoing HD for end-stage kidney disease due to nephrosclerosis wh

Co-infection models of Staphylococcus aureus and respiratory viruses reveal synergistic interactions counteracted by a bithiazole derivative.

Viral-bacterial co-infections represent a major contributor to global morbidity and mortality. In influenza virus infections, secondary bacterial pneumonia occurs in approximately 20-30% of cases and is associated with an increased risk of shock, mechanical ventilation, and even mortality. Bacterial superinfections are frequent causes of mortality in influenza severe infections and have been a major cause of morbidity and mortality also in COVID-19 patients. Staphylococcus aureus has been identified as one of the most prevalent bacterial pathogens among hospitalized and non-hospitalized patien

Deep Herpes in an Adolescent With Severe Atopic Dermatitis and Concurrent Methicillin-Resistant Staphylococcus aureus (MRSA) Bacteremia.

Herpes simplex virus (HSV) infection is a well-recognized complication of atopic dermatitis, most commonly presenting as eczema herpeticum. Deep herpes is a recently described clinicopathologic manifestation of HSV infection, characterized by viral involvement of cutaneous stromal cells with limited or absent epidermal infection, which can make diagnosis challenging when classic histopathologic findings are absent. We report a 17-year-old adolescent with severe atopic dermatitis who presented with fever and a rapidly progressive painful erosive eruption. Evaluation revealed methicillin-resista

Skin Infection Pathogenicity Associated with a Canine Microbiome Resident: Polygenic Architecture of Virulence Factors in Staphylococcus pseudintermedius.

Staphylococcus pseudintermedius is a common opportunistic pathogen in companion animals and a leading cause of skin and soft tissue infections (SSTIs). Despite its clinical relevance, the genomic determinants underlying pathogenicity and the transition from commensal carriage to invasive infection remain poorly understood. This study aimed to identify the genomic determinants of SSTI pathogenic potential in S. pseudintermedius and to determine whether pathogenicity is driven by single, major-effect virulence genes or by polygenic genome-wide genetic architecture. Using accessory gene-based and

The transsulfuration pathway metabolite α-ketobutyrate drives RIPK1-lactate axis-dependent autophagy to alleviate Staphylococcus aureus infection.

Breastfeeding anchors infant immunity and long-term health, but its benefits are threatened by Staphylococcus aureus (S. aureus) mastitis, an increasingly prevalent condition driven by antimicrobial resistance and therapeutic limitations. Beyond compromising maternal wellness, mastitis threatens the safety and continuity of breast milk, highlighting a critical need for innovative intervention strategies. Herein, we demonstrated that α‑ketobutyrate (α-KB), a metabolite of the transsulfuration pathway, mitigated S. aureus‑induced inflammation, oxidative stress, and blood-milk barrier (BMB) disru

Retrospective Study on Antimicrobial Susceptibility of Staphylococcus hyicus Strains Isolated in Northern Italy Between 2022 and 2025.

Sudden outbreaks of exudative epidermitis (EE) are managed with antibiotics. This retrospective observational study evaluated the antimicrobial susceptibility of Staphylococcus (S.) hyicus strains isolated in Northern Italy between January 2022 and April 2025. Isolates were recovered from clinical cases submitted to IZSLER diagnostic laboratories. To ensure data independence, only one isolate per pig farm or strains from the same farm isolated at least 6 months apart were included in the final analysis. Strains resulted from bacteriological examinations on skin swabs (65) and synovial fluid (1

Training a phage to expand its host range: directed evolution against Staphylococcus aureus from diabetic foot infections.

Diabetic foot infections (DFIs) are a major complication of diabetes frequently involving multidrug-resistant Staphylococcus aureus. Their persistence and therapeutic complexity underscore the urgent need for alternatives to conventional antibiotics. Phage therapy offers a promising solution, though its clinical application is often limited by the narrow host range of individual phages. Here, SAVM02, a recently characterized staphylococcal Kayvirus, was trained using directed evolution against DFI isolates. After 20 iterative passages on a mixed panel of susceptible and non-susceptible S. aure

Antibiotic Resistance and Genomic Diversity of Methicillin-Resistant Staphylococcus aureus Clonal Complex 45 Isolates in Kuwait Hospitals.

Background/Objectives: Methicillin-resistant Staphylococcus aureus (MRSA) causes hospital- and community-acquired infections. MRSA is a highly diverse strain that includes several epidemic clones, including CC45. A previous study conducted among MRSA isolates in Kuwait identified CC45 in two isolates in the early 2000s. This study provides an update on the prevalence and molecular characteristics of CC45 among MRSA isolates in Kuwait hospitals, during 2016-2022. Methods: A total of 13,276 MRSA isolates were collected during 2016-2022 and typed using antibiogram, DNA microarray, Staphylococcal

Secreted factors of Staphylococcus aureus promote co-invasion with Candida albicans by inducing hypha formation and invasion.

Interkingdom interactions between Candida albicans and Staphylococcus aureus promote lethal dissemination of the bacterium. During this process, C. albicans hypha invasion aids S. aureus dissemination through Als1p/Als3p-facilitated co-invasion. The effects of S. aureus on C. albicans hypha formation and invasion are, however, unknown. In this study, we used both liquid mDMEM-DMP as well as a previously constructed semi-solid adaptation of the medium (mDMEM-DMPA) to study the effects of C. albicans/S. aureus co-culturing on hypha formation and invasion. Semi-solid-based co-culturing significan

Antiseptic Susceptibility and Sub-Inhibitory Concentration-Induced Biofilm Response in Staphylococcus epidermidis from Abdominal Surgical Site Skin.

BACKGROUND: Staphylococcus epidermidis, a commensal skin pathogen, is a key pathogen in surgical site infections (SSIs). This study characterized clinical isolates obtained from an abdominal operation to guide decolonization therapy. METHODS: In total, 146 S. epidermidis isolates obtained from pre-operative skin swabs of patients undergoing gynecological procedures were analyzed. PCR was performed to detect mecA, efflux pump genes (qacA/B, smr, etc.), and biofilm-associated genes (icaA-D, aap). Methicillin-resistant S. epidermidis (MRSE) isolates were typed by multi-locus sequence typing. Bro

Molecular detection of antibiotic resistance genes in Staphylococcus aureus isolated from poultry farms of Kathmandu Valley, Nepal.

BACKGROUND: Antimicrobial resistance (AMR) has emerged as a critical global health challenge and is currently addressed through a "One Health" approach that investigates its occurrence in humans, animals, and the environment, as well as the transmission pathways linking these reservoirs. In commercial poultry farms, antibiotics are routinely used to treat bacterial infections that are economically significant as well as to promote weight gain. However, data on AMR in Staphylococcus aureus (S. aureus) of poultry origin remain scarce in Nepal. Antimicrobial resistance in S aureus, particularly t
