NEK2 promotes oral squamous cell carcinoma progression and serves as a diagnostic and therapeutic target.

NIMA - related kinase 2 (NEK2), belonging to the NIMA family, is intricately linked to tumorigenesis and tumor progression. Nevertheless, its biological functions and underlying mechanisms in oral squamous cell carcinoma (OSCC) remain elusive. The protein expression level of NEK2 in clinical OSCC samples was evaluated using immunohistochemical staining, and its prognostic impact on OSCC patients was analyzed via Kaplan-Meier survival analysis. Additionally, RT-qPCR and Western blot were performed to examine NEK2 mRNA and protein expression levels in OSCC cell lines (SCC4 and Cal27). Scratch wo

Increased expression of mitochondrial enzyme MTHFD2 associated with clinicopathological features and prognosis of oral squamous cell carcinoma.

BACKGROUND: This study investigated the expression profile of MTHFD2 at both transcript and protein levels in oral squamous cell carcinoma (OSCC). In addition, its association with clinicopathological parameters, patient survival, and immune-related features was systematically evaluated. METHODS: Publicly available transcriptomic data from The Cancer Genome Atlas were analyzed to assess MTHFD2 expression in OSCC, followed by experimental validation in an independent cohort of 42 OSCC patients. Quantitative real-time PCR was employed to validate MTHFD2 transcript expression in OSCC tissues. Pr

MAGEA Family Gene Expression Influences Survival in Intestinal-Type Gastric Cancer.

Gastric cancer (GC) is a heterogeneous disease characterized by diverse molecular profiles and distinct histological subtypes, each with differing prognoses and therapeutic responses. Melanoma-associated antigen-A (MAGEA) genes encode cancer testis antigens frequently expressed in tumor tissue and in limited amounts in normal tissues, making them a promising target for cancer immunotherapy. While therapies targeting MAGEA family members have demonstrated promise across various cancers, clinical trials have been limited by substantial toxicities, underscoring the need to precisely identify pati

Supervising the recurrence of pancreatic ductal adenocarcinoma using CtDNA-based MIR129-2 methylation detection.

BACKGROUND: Surgery remains the primary treatment for pancreatic ductal adenocarcinoma (PDAC), which has remarkably high morbidity and mortality rates. However, recurrence is prevalent after resection. The effectiveness of circulating tumor DNA (ctDNA) in recurrence prediction has been demonstrated, and methylation has advantages over traditional biomarkers in terms of early-stage specificity. These advances indicate the need for a ctDNA-based methylation marker for PDAC prognosis, which would be valuable in clinical practice. METHODS: MIR129-2 promoter methylation markers were identified via

SOX13 is a novel prognostic biomarker and associates with immune infiltration in breast cancer.

BACKGROUND: The transcription factor, SOX13 is part of the SOX family. SOX proteins are crucial in the progression of many cancers, and some correlate with carcinogenesis. Nonetheless, the biological and clinical implications of SOX13 in human breast cancer (BC) remain rarely known. METHODS: We evaluated the survival and expression data of SOX13 in BC patients via the UNLCAL, GEPIA, TIMER, and Kaplan-Meier plotter databases. Immunohistochemistry (IHC) was used to verify clinical specimens. The gene alteration rates of SOX13 were acquired on the online web cBioportal. With the aid of the TCGA

Construction and multiple validations of a robust ferroptosis-related prognostic model in bladder cancer: A comprehensive study.

Ferroptosis is iron-dependent programmed cell death that inhibits tumor growth, particularly in traditional treatment-resistant tumors. Prognostic models constructed from ferroptosis-related genes are lacking; prognostic biomarkers remain insufficient. We acquired gene expression data and corresponding clinical information for bladder cancer (BC) samples from public databases. Ferroptosis-related genes from the ferroptosis database were screened for clinical predictive value. We validated gene expression differences between tumors and normal tissues through polymerase chain reaction and wester

Comprehensive analysis of gene expression profiles to identify differential prognostic factors of primary and metastatic breast cancer.

Breast cancer accounts for nearly half of all cancer-related deaths in women worldwide. However, the molecular mechanisms that lead to tumour development and progression remain poorly understood and there is a need to identify candidate genes associated with primary and metastatic breast cancer progression and prognosis. In this study, candidate genes associated with prognosis of primary and metastatic breast cancer were explored through a novel bioinformatics approach. Primary and metastatic breast cancer tissues and adjacent normal breast tissues were evaluated to identify biomarkers charact

Spatial and Transcriptomic Analysis of Perineural Invasion in Oral Cancer.

PURPOSE: Perineural invasion (PNI), a common occurrence in oral squamous cell carcinomas, is associated with poor survival. Consequently, these tumors are treated aggressively. However, diagnostic criteria of PNI vary and its role as an independent predictor of prognosis has not been established. To address these knowledge gaps, we investigated spatial and transcriptomic profiles of PNI-positive and PNI-negative nerves. EXPERIMENTAL DESIGN: Tissue sections from 142 patients were stained with S100 and cytokeratin antibodies. Nerves were identified in two distinct areas: tumor bulk and margin.

The Transcoelomic Ecosystem and Epithelial Ovarian Cancer Dissemination.

Epithelial ovarian cancer (EOC) is considered the deadliest gynecological disease and is normally diagnosed at late stages, at which point metastasis has already occurred. Throughout disease progression, EOC will encounter various ecosystems and the communication between cancer cells and these microenvironments will promote the survival and dissemination of EOC. The primary tumor is thought to develop within the ovaries or the fallopian tubes, both of which provide a microenvironment with high risk of causing DNA damage and enhanced proliferation. EOC disseminates by direct extension from the

Human induced pluripotent stem cell-derived three-dimensional cardiomyocyte tissues ameliorate the rat ischemic myocardium by remodeling the extracellular matrix and cardiac protein phenotype.

The extracellular matrix (ECM) plays a key role in the viability and survival of implanted human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs). We hypothesized that coating of three-dimensional (3D) cardiac tissue-derived hiPSC-CMs with the ECM protein fibronectin (FN) would improve the survival of transplanted cells in the heart and improve heart function in a rat model of ischemic heart failure. To test this hypothesis, we first explored the tolerance of FN-coated hiPSC-CMs to hypoxia in an in vitro study. For in vivo assessments, we constructed 3D-hiPSC cardiac tissues (3

Distally based peroneal artery perforator-plus fasciocutaneous flap in the reconstruction of soft tissue defects over the distal forefoot: a retrospectively analyzed clinical trial.

BACKGROUND: Distally based peroneal artery perforator-plus fasciocutaneous (DPAPF) flaps are widely used for reconstructing soft-tissue defects of the lower extremity. However, reports on the reconstruction of the defects over the distal forefoot using the DPAPF flaps are scarce. Herein, we describe our experience on the reconstruction of these defects using DPAPF flaps in a considerable sample size. METHODS: Between February 2005 and August 2019, a total of 56 DPAPF flaps in 56 patients were used to reconstruct soft-tissue defects in the forefoot. In order to reduce the length of fascial ped

Homing of chloromethylbenzoyl ammonia-labeled bone marrow mesenchymal stem cells in an immune-mediated bone marrow failure mouse model in vivo.

Aplastic anemia is an abnormal immune reaction disease in which T lymphocytes destroy hematopoietic stem and progenitor cells because of immune hyperactivity. Bone marrow mesenchymal stem cells (BMSCs) have hematopoietic supporting and immune regulation functions. This study investigated BMSCs homing in mice transplantation models after bone marrow failure. BALB/c mice were randomly divided into three groups: normal control, bone marrow failure model, and BMSC transplantation group. Chloromethyl benzamido-labeled BMSCs of BALB/c mice were transplanted through tail vein injection in mouse model

Down-regulation of miR-221 inhibits proliferation of pancreatic cancer cells through up-regulation of PTEN, p27(kip1), p57(kip2), and PUMA.

Pancreatic cancer is the fourth leading cause of cancer related death in the US and exhibits aggressive features with short survival rate and high mortality. Therefore, it is important to understand the molecular mechanism(s) involved in the aggressive growth of pancreatic cancers, and further design novel targeted therapies for its treatment with better treatment outcome. In the present study, we found that the expression of miR-221 was significantly up-regulated in pancreatic cancer cell lines and tumor tissues compared to normal pancreatic duct epithelial cells and normal pancreas tissues.

The use of field-based mesocosm systems to assess the effects of uranium milling effluent on fathead minnow (Pimephales promelas) reproduction.

Northern Saskatchewan, Canada is home to a uranium milling operation that discharges a complex milling effluent containing nutrients, cations and anions, and many metals including selenium (Se). Se has the potential to accumulate in a system even when water concentrations are low. This study evaluated the effects of treated uranium milling effluent and contaminated sediment in combination and in isolation to determine the contribution and importance of each source to fathead minnow (Pimephales promelas) reproduction and survival. Trios of fathead minnows were allocated to one of four treatment
