Schwann Cell Responses and Plasticity in Different Dental Pulp Scenarios.

Mammalian teeth have evolved as dentin units that enclose a complex system of sensory innervation to protect and preserve their structure and function. In human dental pulp (DP), mechanosensory and nociceptive fibers form a dense meshwork of nerve endings at the coronal dentin-pulp interface, which arise from myelinated and non-myelinated axons of the Raschkow plexus (RP). Schwann cells (SCs) play a crucial role in the support, maintenance and regeneration after injury of these fibers. We have recently characterized two SC phenotypes hierarchically organized within the coronal and radicular DP

An analysis of the measurement principle of smart brackets for 3D force and moment monitoring in orthodontics.

Measuring the three-dimensional (3D) force-moment (F/M) systems applied for correcting tooth malposition is highly desirable for accurate spatial control of tooth movement and for reducing traumatic side effects such as irreversible root resorption. To date, suitable tools for monitoring the applied F/M system during therapy are lacking. We have previously introduced a true-scale orthodontic bracket with an integrated microelectronic stress sensor system for 3D F/M measurements on individual teeth with a perspective for clinical application. The underlying theoretical concept assumes a linear

Evaluation of the safety and efficacy of periodontal applications of a living tissue-engineered human fibroblast-derived dermal substitute. II. Comparison to the subepithelial connective tissue graft: a randomized controlled feasibility study.

BACKGROUND: The subepithelial connective tissue graft, traditionally harvested from the patient's palate, is commonly used for root coverage in periodontal recession defects. This study evaluates the safety and effectiveness of a living human fibroblast-derived dermal substitute (HF-DDS) compared to a connective tissue graft (CTG) for root coverage in these situations. METHODS: Thirteen patients were selected for this study. Each patient had Miller Class I or II bilateral facial recession defects > or =3 mm on two non-adjacent teeth. The test tooth received an HF-DDS graft, while a CTG was pl

Piezo1/PI3K-induced autophagy dysregulation in PDLSCs under mechanical stress during root resorption.

This study aimed to clarify the mechanobiological mechanisms of physiological root resorption in deciduous teeth, focusing specifically on how mechanical stress influences autophagy in periodontal ligament stem cells (PDLSCs) via the Piezo1 ion channel. We established a unilateral occlusal loading model in mice to simulate physiological mechanical stress, while in vitro studies utilized PDLSCs isolated from deciduous teeth at different resorption stages. Techniques included immunofluorescence staining for autophagy markers, Western blot analysis, Piezo1-specific pharmacological modulation, and

Retrieval of a Displaced Mandibular Tooth Fragment From the Floor of the Mouth Using a Combined Navigation System and Endoscope: A Case Report.

BACKGROUND If a tooth fragment is displaced into the floor of the mouth during mandibular third molar extraction, surgical retrieval can be challenging because the fragment cannot be directly visualized in the operative field. Several studies have described the use of navigation systems or endoscopic techniques individually. However, to the best of our knowledge, no reports have described their combined use for this condition. CASE REPORT A 37-year-old man underwent extraction of a left mandibular third molar at a dental clinic in May 2021, during which a root fragment was displaced into the f

The effect of cone beam computed tomography voxel size on age estimation: A three-dimensional surface area analysis study.

Accurate three-dimensional (3D) age estimation relies on cone beam computed tomography (CBCT) image quality. This study investigated the effect of voxel size on age estimation using 3D surface area analysis (3D SAA) of developing tooth apices from different CBCT systems. CBCT scans of 128 developing maxillary second premolars from children aged 7-14 years of Malay and Chinese ethnicities were retrospectively collected using a voxel size of 76 μm. The surface area of the developing apex was measured using the 3D SAA method. A previously developed regression formula, based on 300 μm voxel size d

Development and validation of a machine learning model to predict mandibular third molar impaction and associated complications: A retrospective observational study.

BACKGROUND: Impacted mandibular third molars are common worldwide and may cause complications such as pericoronitis, cysts, and second molar caries. Conventional prediction methods rely on subjective radiographic interpretations and clinical judgment, which can vary among practitioners. Recent advances in machine learning (ML) provide opportunities to develop objective, data-driven models for clinical decision-making in third molar management. AIM: To develop and validate a machine learning-based model for predicting mandibular third molar impaction and associated complications using demograp

Effect of different CAD-CAM post and core materials in endodontically treated teeth without ferrule and with non-circular root canals: 3D finite element analysis.

PURPOSE: Restoring ferrule-lacking, noncircular canal endodontically treated teeth (ETT) poses challenges owing to the increased susceptibility to root fracture and post dislodgement. We aimed to evaluate the influence of different post and core materials on the stress distribution and maximum Von Mises stress in ETT. METHODS: Four three-dimensional models were generated using different customized post and core materials: gold alloy, resin nanoceramic, polyetheretherketone (PEEK), and polyetherketoneketone (PEKK). A static load of 200 N was applied at an angle of 45 ° to the occlusal surface.

Novel standardized method for inducing medication-related osteonecrosis of the jaw in rats and precise quantitative assessment of pathological outcomes.

Medication-related osteonecrosis of the jaw (MRONJ) is a serious complication of antiresorptive therapies, characterized by delayed healing, bone necrosis, and infection following dental procedures. Progress in the understanding of its pathophysiology has been hindered by the lack of standardized animal models. Existing models involving first molar or multiple molar extractions disrupt anatomic landmarks and face technical challenges, such as retained root fragments and inconsistent surgical procedures, which reduce reproducibility. To address these limitations, we developed a novel rat MRONJ

Surface Pre-Reacted Glass-Ionomer Eluate Suppresses Osteoclastogenesis through Downregulation of the MAPK Signaling Pathway.

Surface pre-reacted glass-ionomer (S-PRG) is a new bioactive filler utilized for the restoration of decayed teeth by its ability to release six bioactive ions that prevent the adhesion of dental plaque to the tooth surface. Since ionic liquids are reported to facilitate transepithelial penetration, we reasoned that S-PRG applied to root caries could impact the osteoclasts (OCs) in the proximal alveolar bone. Therefore, this study aimed to investigate the effect of S-PRG eluate solution on RANKL-induced OC-genesis and mineral dissolution in vitro. Using RAW264.7 cells as OC precursor cells (OPC
