What drives adult vaccine hesitancy? Content and face validation of a new measure for U. S. Adults.

BACKGROUND: Vaccine hesitancy is viewed as a complex interaction of behavioral and societal factors that can intervene in the uptake of vaccines or lack thereof. To date, no existing instrument developed for the U. S. adult population measures general vaccine hesitancy in a way that integrates behavioral and cognitive determinants and generalizes across vaccines rather than targeting a specific disease. The development of this tool will be useful for clinicians to counsel patients on vaccination and for vaccine related public health interventions targeted at improving population health. METHO

Differences in Clinical Aspects Between Subacute Thyroiditis Associated with COVID-19 Vaccines and Classical Subacute Thyroiditis.

Subacute thyroiditis (SAT) developed after SARS-CoV-2 vaccines has been less studied. We aimed to compare classical SAT and SAT developed after SARS-CoV-2 vaccines in the context of clinical aspects. Adults with SAT detected in 90 days of COVID-19 vaccination (CoronaVac or Pfizer/BioNTech) were grouped as Vac-SAT. Those with a history of SARS-CoV-2 or upper respiratory tract infection in 6 months before the vaccination, or vaccination with another antiviral vaccine after COVID-19 vaccination were excluded. Those with SAT detected before COVID-19 pandemic were grouped as Classical-SAT. Of total

Vaccines for Prevention of Bluetongue and Epizootic Hemorrhagic Disease in Livestock: A North American Perspective.

Bluetongue (BT) and epizootic hemorrhagic disease (EHD) are noncontagious, insect-transmitted diseases of domestic and wild ruminants caused by related but distinct viruses. There are significant gaps in our scientific knowledge and available countermeasures to control an outbreak of orbivirus-induced disease, whether BT or EHD. Both BT virus (BTV) and EHD virus (EHDV) cause hemorrhagic fevers in susceptible ruminants; however, BT is principally a disease of domestic livestock whereas EHD is principally a disease of certain species of wild, non-African ungulates, notably white-tailed deer. Association of deficiency in antibody response to vaccine and heterogeneity of Ehrlichia risticii strains with Potomac horse fever vaccine failure in horses.

Ehrlichia risticii is the causative agent of Potomac horse fever (PHF), which continues to be an important disease of horses. Commercial inactivated whole-cell vaccines are regularly used for immunization of horses against the disease. However, PHF is occurring in large numbers of horses in spite of vaccination. In a limited study, 43 confirmed cases of PHF occurred between the 1994 and 1996 seasons; of these, 38 (89%) were in horses that had been vaccinated for the respective season, thereby clearly indicating vaccine failure. A field study of horses vaccinated with two PHF vaccines indicated

Recombinant human adenovirus type 5-vectored vaccine expressing avian reovirus σC protein protects against prevalent genotype I avian reovirus in chickens.

Avian reovirus (ARV) is a globally important poultry pathogen causing substantial economic losses due to viral arthritis, enteritis and immunosuppression. Traditional vaccines based on the S1133 strain are less effective against circulating ARV variants due to genetic divergence. The σC protein is the primary antigen inducing neutralizing antibodies against ARV, making it a key target for vaccine development. Here, we constructed a recombinant human adenovirus type 5 (rHAdV5) vector vaccine expressing the σC protein of a prevalent Chinese ARV field strain (SDAU-1) and evaluated its safety, imm

Enhancing memory T cell responses through Soluble Leishmania antigen and natural adjuvants against Leishmania donovani in murine model.

Leishmaniasis, caused by Leishmania spp., is a major public health concern in tropical and subtropical regions and is classified as a neglected tropical disease (NTD). Visceral leishmaniasis (VL), the most severe form, affects the reticuloendothelial system and is often fatal if untreated. Current treatment relies on a limited number of chemotherapeutic agents, and no licensed vaccine is available to prevent infection. This highlights the urgent need for a prophylactic vaccine to protect against the infection and its potentially fatal consequences. Therefore, in the present study, we evaluated

Strategies to enhance DNA vaccine efficacy against emerging arboviruses: lessons from ZIKA and Chikungunya viruses.

The mosquito-borne arboviruses Zika and Chikungunya pose serious health threats worldwide, often co-circulating and causing co-infections in Aedes endemic regions. While no licensed vaccine is currently available for Zika virus (ZIKV), recent progress has led to the approval of vaccines for Chikungunya virus (CHIKV) in few countries, highlighting the need for improved vaccine strategies. Zika virus (ZIKV) presents unique challenges, including its neurotropism, association with congenital abnormalities, and potential for sexual and vertical transmission, whereas Chikungunya virus (CHIKV) is cha

Genotype-matched recombiant inactivated Newcastle disease virus vaccine confer protection against genotype Ⅻ challenge in geese with maternal antibodies.

The majority of Newcastle disease virus (NDV) strains circulating in China, which exhibit a tendency toward increased virulence, are from genotype Ⅻ infections obtained from waterfowl. Based on our previous findings, the genotype Ⅻ strain E115 was non-pathogenic in geese but induced notable clinical symptoms in chickens without mortality. However, in recent years, we isolated the genotype Ⅻ strain E117 from dead geese and confirmed its lethality in geese through animal regression tests. Genotype-matched vaccination has been proposed as a potential solution. In this study, two NDV vaccination s

Identification of a protective antigen reveals the trade-off between iron acquisition and antigen exposure in a global fungal pathogen.

Systemic infections caused by Cryptococcus claim over 161,000 lives annually, with global mortality rate close to 70% despite antifungal therapies. Currently, no vaccine is available. To develop an effective multivalent vaccine against this free-living opportunistic eukaryotic pathogen, it is critical to identify protective antigens. We previously discovered ZNF2oe strains elicit protective host immune responses and increase the abundance of antigens present in the capsule, which is required for its immunoprotection. Capsule is a defining feature of Cryptococcus species and composed of polysac

Novel Oral papaya-based preventive and therapeutic vaccine for controlling different zoonotic Taeniid parasites: Current advances.

Zoonotic infections caused by Taenia and Echinococcus species are the most significant Taeniid parasitic diseases affecting human and animal health, particularly in regions with poor sanitation. Vaccination of intermediate hosts has proven effective in controlling these parasitic infections. In the case of Echinococcosis, targeted immunization programs have led to marked reductions in transmission, although global eradication remains unachieved. Similarly, sustained vaccination studies for cysticercosis have demonstrated the feasibility of interrupting transmission cycles. This review summariz

Evaluation of a Quadrivalent Shigella flexneri Serotype 2a, 3a, 6, and Shigella sonnei O-Specific Polysaccharide and IpaB MAPS Vaccine.

BACKGROUND: Shigellosis is the leading cause of diarrheal deaths worldwide and is particularly dangerous in children under 5 years of age in low- and middle-income countries. Additionally, the rise in antibiotic resistance has highlighted the need for an effective Shigella vaccine. Previously, we have used the Multiple Antigen-Presenting System (MAPS) technology to generate monovalent and quadrivalent Salmonella MAPS vaccines that induce functional antibodies against Salmonella components. METHODS: In this work, we detail the development of several monovalent vaccines using O-specific polysac

The Current State and Progress of Mpox Vaccine Research.

On July 23, 2022, the World Health Organization (WHO) declared the monkeypox (mpox) outbreak a "Public Health Emergency of International Concern." Since 2022, outbreaks of mpox in many countries around the world have primarily resulted in fatalities among immunocompromised individuals, such as untreated HIV/AIDS patients. Since the eradication of smallpox was declared by the WHO in 1980, the global vaccination against smallpox has been gradually discontinued. China also stopped routine smallpox vaccination in 1981. The protective effect of the smallpox vaccine has decreased over time due to ag

Heterologous DNA Prime- Subunit Protein Boost with Chikungunya Virus E2 Induces Neutralizing Antibodies and Cellular-Mediated Immunity.

Chikungunya virus (CHIKV) has become a significant public health concern due to the increasing number of outbreaks worldwide and the associated comorbidities. Despite substantial efforts, there is no specific treatment or licensed vaccine against CHIKV to date. The E2 glycoprotein of CHIKV is a promising vaccine candidate as it is a major target of neutralizing antibodies during infection. In this study, we evaluated the immunogenicity of two DNA vaccines (a non-targeted and a dendritic cell-targeted vaccine) encoding a consensus sequence of E2CHIKV and a recombinant protein (E2*CHIKV). Mice w

Manifestations of adverse events post Sinovac vaccine immunization at Wirasakti Hospital, Kupang.

BACKGROUND: Since it was first discovered, COVID-19 has spread worldwide and has been declared a pandemic by the World Health Organization. One of the various efforts made to prevent the spread of COVID-19 is the development of the COVID-19 vaccine. One of the vaccines developed and used by the Indonesian government is the Sinovac vaccine. However, like vaccines in general, there are always vaccine safety problems which are better known as adverse events following immunization (AEFI). OBJECTIVE: The purpose of this study is to determine the AEFI manifestations that occurred in Sinovac vaccina

Comparative Biodistribution Study of Baculoviral and Adenoviral Vector Vaccines against SARS-CoV-2.

Various types of vaccines have been developed against COVID-19, including vector vaccines. Among the COVID-19 vaccines, AstraZeneca's chimpanzee adenoviral vaccine was the first to be commercialized. For viral vector vaccines, biodistribution studies are critical to vaccine safety, gene delivery, and efficacy. This study compared the biodistribution of the baculoviral vector vaccine (AcHERV-COVID19) and the adenoviral vector vaccine (Ad-COVID19). Both vaccines were administered intramuscularly to mice, and the distribution of the SARS-CoV-2 S gene in each tissue was evaluated for up to 30 days

Adenovirus-Based Vaccines and Thrombosis in Pregnancy: A Systematic Review and Meta-analysis.

BACKGROUND: Rare cases of thrombosis and thrombocytopenia (thrombosis with thrombocytopenia syndrome [TTS]) have been associated with 2 coronavirus disease 2019 adenovirus vector vaccines: the ChAdOx1 nCoV-19 Vaxzevria vaccine (Oxford/AstraZeneca) and the JNJ-7836735 Johnson & Johnson vaccine (Janssen). It is unknown if TTS is a class-mediated effect of adenovirus-based vaccines or if it could worsen known hypercoagulable states. Since most cases of TTS happen in women of childbearing age, pregnancy is a crucial risk factor to assess. Understanding these risks is important for advising vaccine

Control of SARS-CoV-2 infection after Spike DNA or Spike DNA+Protein co-immunization in rhesus macaques.

The speed of development, versatility and efficacy of mRNA-based vaccines have been amply demonstrated in the case of SARS-CoV-2. DNA vaccines represent an important alternative since they induce both humoral and cellular immune responses in animal models and in human trials. We tested the immunogenicity and protective efficacy of DNA-based vaccine regimens expressing different prefusion-stabilized Wuhan-Hu-1 SARS-CoV-2 Spike antigens upon intramuscular injection followed by electroporation in rhesus macaques. Different Spike DNA vaccine regimens induced antibodies that potently neutralized SA

Addressing Vaccine-Induced Immune Thrombotic Thrombocytopenia (VITT) Following COVID-19 Vaccination: A Mini-Review of Practical Strategies.

In response to the COVID-19 pandemic, several vaccines were developed and rolled out at unprecedented speed, and notwithstanding this rapid pace of development, the results from initial clinical trials involving tens of thousands of adult subjects generally indicated that most vaccines were remarkably effective and safe, with no major safety warnings noted. However, with more than 2 billion vaccination doses administered to date, reports of rare adverse events following immunization (AEFI) are beginning to emerge. In late February 2021, atypical thrombotic events following immunization with th

Therapeutic ISCOMATRIX™ adjuvant vaccine elicits effective anti-tumor immunity in the TRAMP-C1 mouse model of prostate cancer.

Cancer vaccine development has proven challenging with the exception of some virally induced cancers for which prophylactic vaccines exist. Currently, there is only one FDA approved vaccine for the treatment of prostate cancer and as such prostate cancer continues to present a significant unmet medical need. In this study, we examine the effectiveness of a therapeutic cancer vaccine that combines the ISCOMATRIX™ adjuvant (ISCOMATRIX) with the Toll-like receptor 3 agonist, polyinosinic-polycytidylic acid (Poly I:C), and Flt3L, FMS-like tyrosine kinase 3 ligand. We employed the TRAMP-C1 (transge
