Impact of CP12 deletion on inorganic carbon acquisition and Rubisco partitioning in Chlamydomonas reinhardtii.

The small chloroplastic protein CP12 has multiple functions, including the regulation of enzymes in the Calvin-Benson-Bassham cycle. Here, we investigated its role in the acclimation of Chlamydomonas reinhardtii to varying CO2 availability. We show that phosphoribulokinase can interact with CP12 in conditions where the Calvin-Benson-Bassham cycle is active. Compared to the wild type, at high CO2  C. reinhardtii ΔCP12 mutants have less phosphoribulokinase and ribulose-1,5-bisphosphate (RuBP), indicating that the regeneration of RuBP is regulated, in part, by CP12. Chlamydomonas reinhardtii has

Elevated methionine in plants: the effect on development, stress responses, epigenetics, and nutritional biofortification.

Methionine (Met) is an essential amino acid that limits the nutritional value of many crop plants, yet its steady-state level in plant tissues is remarkably low. At the same time, Met is a central metabolic hub supporting protein synthesis and, through S-adenosylmethionine (SAM), drives the production of hormones, vitamins, polyamines, and epigenetic marks. The primary objective of this review is to understand why and how plants maintain low steady-state Met levels, and what occurs when Met content is elevated, a question of growing importance for Met biofortification. We first summarize evide

Functional characterization of bicarbonate transporters from the cyanobacterial SbtA2 family and subsequent expression in tobacco.

Cyanobacteria rely on bicarbonate (HCO3-) as the primary inorganic carbon (Ci) source for photosynthesis in aquatic environments. To use this Ci source, cyanobacteria employ CO2-concentrating mechanisms that elevate cytoplasmic HCO3- via plasma membrane transporters, enhancing carboxylation by carboxysomal Rubisco. The sodium-dependent SbtA1 transporter family is well characterized in freshwater cyanobacteria, but the related SbtA2 family, prevalent in marine α-cyanobacteria, remains uncharacterized. Here, we report functional characterization of SbtA2 homologues from marine Synechococcus spp.

Assimilation dynamics of xylem-transported CO2 by woody tissue photosynthesis revealed with 11C- and 13C-labelling.

A considerable quantity of locally respired CO2 in stems can be assimilated via woody tissue photosynthesis (Pwt). Hence, Pwt locally provides carbon fuelling radial stem growth and therefore plays a major role in the stem carbon balance. It is, however, not straightforward to quantify or visualize its dynamic behaviour, especially on small spatial scales. To assess Pwt we traced xylem-transported 11CO2 in detached branches of Populus tremula L. and used positron autoradiography to visualize and quantify its dynamics on a submillimetre scale, while manipulating light exposure to the branch. Wheat leaf dark respiration acclimates more strongly at night than in the day when responding to nocturnal warming.

Rising night temperatures pose a significant threat to wheat productivity, yet the physiological basis of wheat adaptation to nocturnal warming remains poorly understood. We evaluated leaf photosynthetic and respiratory traits in response to warm nights under temperature-controlled environments in 10 Australian wheat cultivars released between 1901 and 2012. When exposed to warmer nights, rates of leaf net CO2 assimilation measured at 25 °C (Anet25) remained stable across cultivar release date despite declines in photosynthetic capacity (Vcmax and J1500) in newer cultivars. In most cultivars l

Exploring the diversity of the CO2-concentrating mechanism in different C4 subtypes.

C4 plants have traditionally been classified into NADP-malic enzyme (NADP-ME), NAD-malic enzyme (NAD-ME), and phosphoenolpyruvate carboxykinase (PEPCK) subtypes based on the predominant C4 acid decarboxylating enzyme. To investigate the relative contributions of malate and aspartate to C4 pathway fluxes in each subtype, we performed 13CO2 pulse-chase labelling experiments on four C4 grass species: Zea mays and Setaria viridis (NADP-ME), Panicum miliaceum (NAD-ME), and Megathyrsus maximus (PEPCK). Only a proportion (8-50%) of the total malate pool in the leaves is photosynthetically active, whe

Temporal profiling of floret developmental asynchrony for wheat-fertility studies.

Wheat grain number integrates fate of individual florets, strongly affected by environmental stress during sensitive stages like meiosis. Because development is asynchronous across tillers, spikelets, and florets, it is hard to distinguish stress tolerance from stress escape. Based on 2400 destructive measurements of spike and anther length together with non-destructive morphological measurements from 158 plants grown in four experiments in controlled-environment, we developed a framework to track individual floret developmental stages at plant level. We applied it in two case studies for conn

Sensitization rather than acclimation to chronic atmospheric aridity in invasive and native dryland C4 grasses.

Despite extreme heat and limited water availability, invasive plant species are rapidly expanding in arid ecosystems, threatening native biodiversity. To understand the physiological mechanisms underlying invasive success in water-limited ecosystems, we analyzed photosynthetic, hydraulic and morphological traits of the invasive African grass Cenchrus ciliaris and three native North American dryland grasses (Bouteloua gracilis, Schizachyrium scoparium, and Sporobolus cryptandrus) in a full factorial growth chamber experiment combining two vapor pressure deficit (VPD) levels and three soil moist

Nitric oxide interacts with brassinosteroid signalling to regulate plant growth and development in an organ-specific manner.

Nitric oxide (NO) is a plant gasotransmitter that regulates plant growth and development by interacting with different regulatory pathways. Among these, brassinosteroids (BRs) have become candidate phytohormones that interact antagonistically and closely with NO to regulate plant growth. Seedling growth in NO-deficient mutants is enhanced and hyper-responds to BR treatments, while NO overaccumulator mutants show some developmental defects. Interestingly, this hyper-response was also observed in mutants that had both constitutively activated BR signalling and reduced NO levels. BR signalling mu

Single-nucleus RNA sequencing reveals the developmental trajectory and transcriptional regulatory networks of trichomes in Artemisia annua.

Sweet wormwood (Artemisia annua) produces and deposits artemisinin in its glandular trichomes but the molecular mechanisms of trichome development remain poorly understood. Here, we conduct single-nucleus RNA sequencing of 10-day-old A. annua seedlings, revealing the specific RNA expression of the leaf trichome cells. The reconstruction of the developmental trajectory of trichome cells not only identifies novel expressed genes, but also elucidates trichome development involved in photosynthesis, auxin biosynthesis, and cutin biosynthesis. By integrating the developmental trajectory of trichome

Developmental roles of LSD1/KDM1A-like (LDL) proteins in plants.

LYSINE-SPECIFIC DEMETHYLASE 1-like (LDL) proteins are conserved FAD-dependent amine oxidases that serve as pivotal regulators in plants. While animal systems typically rely on a single LSD1/KDM1A enzyme, the Arabidopsis thaliana genome encodes an expanded family of LDL homologues (FLD, LDL1, LDL2, and LDL3), resulting in substantial subfunctionalization and specialized recruitment mechanisms. This review explores the diverse developmental roles of plant LDLs, ranging from flowering time and circadian clock regulation to heterochromatin maintenance and epigenetic regulation. We discuss the redu

Using ΦPSII and leaf temperature as indicators of non-steady-state photosynthesis and stomatal conductance during stepwise changes in light intensity.

Quantifying the kinetics of net CO2 assimilation (A) and stomatal conductance (gs) under fluctuating light typically relies on gas exchange measurements, which are slow and thus unsuited for high-throughput phenotyping. As a result, faster, non-invasive phenotyping methods are needed to further evaluate these traits at a larger scale. However, first the relationship between non-steady-state parameters must be examined in greater detail. In this study, we aimed to determine whether variations in non-steady-state values of chlorophyll fluorescence and leaf temperature reflect differences in key

The co-chaperone AtDJB3 regulates HSC70-1-mediated expression of heat shock genes and thermotolerance in Arabidopsis.

Heat stress disrupts protein homeostasis, triggering the heat shock response (HSR) to maintain cellular proteostasis. A key aspect of this response is the release of heat shock factors (HSFs) from HSP70-mediated attenuation under heat shock conditions, making HSP70 a central regulator of HSR. However, the role of HSP70 co-chaperones in this process remains largely unexplored in plants. Our study identifies AtDJB3, a heat-inducible class II J-domain protein (JDP), as a critical modulator of HSR. Microscopy and cell fractionation show that loss of AtDJB3 impairs HSC70-1 recruitment to heat-induc

Humidity and temperature interact in breaking physical dormancy in Fabaceae seeds from the Brazilian semiarid region.

PREMISE: In semiarid ecosystems such as the Brazilian Caatinga, physical seed dormancy (PY) is critical for plant persistence when water is unpredictable, but how soil moisture and temperature regulate PY release and seed longevity in species of native Fabaceae that have different growth forms is unknown. METHODS: Seeds of the herbaceous species Centrosema pascuorum and Crotalaria retusa and tree species Mimosa ophthalmocentra, M. tenuiflora, and Piptadenia retusa were collected in the Caatinga and tested in the field and laboratory to assess dormancy, viability, and germination. Branching Varies with Light Limitation Scenarios in relation with Changes in Carbon Source-Sink Dynamics.

Bud outgrowth is a major component of plant architectural plasticity and is influenced by light conditions. While the inhibitory effect of low light intensity on branching is well documented, the underlying regulators remain debated and, especially, the role of sugar availability has never been thoroughly evaluated. Here, we combined experiments with a computational approach quantifying carbon source-sink balance in single-axis rose plants to investigate how continuous and transient light limitation regulate bud outgrowth. Continuous low light reduced photosynthesis, leading to decreased sugar

Lineage-specific response to melatonin in stomatal regulation across vascular plants.

Melatonin (N-acetyl-5-methoxytryptamine) is a conserved indoleamine that regulates diverse physiological processes, including stomatal movement in angiosperms. However, whether this regulation is conserved across different plant lineages remains uncertain. Here, we examined the stomatal responses to exogenous melatonin in 17 representative plant species: seven angiosperms, two gymnosperms, seven ferns, and one lycophyte. In seed plants, melatonin induced dose-dependent stomatal closure, accompanied by the accumulation of reactive oxygen species (ROS), nitric oxide (NO), and cytosolic Ca2+; thi

Intercellular Concentration Gradients of 3-Phosphoglycerate and Triose-Phosphate Demonstrate Operation of an Energy Shuttle in NAD-Malic Enzyme and Phosphoenolypyruvate Carboxykinase C4 Subtypes.

In C4 photosynthesis, incoming CO2 is incorporated in mesophyll cells (MC) into 4-carbon acids that diffuse to bundle sheath cells (BSC) and are decarboxylated to generate a high CO2 concentration that suppresses the oxygenation reaction of Rubisco. Decarboxylation can occur by NADP-malic enzyme, (NADP-ME), NAD-malic enzyme (NAD-ME) or phosphoenolpyruvate carboxykinase (PEPCK). NADP-ME generates NADPH in the BSC chloroplast and species that use it as the major route for decarboxylation typically have dimorphic BSC chloroplasts with little or no photosystem II. They operate an energy shuttle: m

Structural and regulatory features associated with elevated accumulation of the rice ERFVII transcription factor SUB1A-1.

Flooding-induced hypoxia severely limits crop productivity, and Group VII Ethylene Response Factors (ERFVIIs) function as central transcriptional regulators of physiological and transcriptional acclimation to hypoxia. In rice, the ERFVII transcription factor SUB1A-1 confers strong submergence tolerance. It displays unusually high steady-state protein levels and reduced sensitivity to oxygen-dependent turnover despite retaining a canonical N-terminal degron motif that promotes rapid turnover of other ERFVIIs under normoxia. The basis of this property remains incompletely resolved. Here, we inve

Protein persulfidation: a ubiquitous modification regulating a broad spectrum of biological processes.

Hydrogen sulfide signaling occurs mainly through protein persulfidation, an important post-translational modification and a highly dynamic process in plants. Beyond enzyme activity, persulfidation affects protein localization, structure, and interactions. This review provides an overview of the mechanisms governing this modification and the pivotal role of the thioredoxin system mediating depersulfidation, which is essential for maintaining redox homeostasis. Proteomic studies in Arabidopsis thaliana and crop species reveal that protein persulfidation is a widespread post-translational modific

Pollinator-mediated fitness differences do not fully explain the maintenance of gynodioecy in Sidalcea campestris.

PREMISE: The maintenance of gynodioecy, female and hermaphroditic coexistence in plants, requires females to have a reproductive advantage over hermaphrodites in offspring quality, quantity, or both. Pollinators can influence this by mediating the reproductive success of females and hermaphrodites. We measured differences in pollinator-mediated fitness components between female and hermaphroditic plants in Sidalcea campestris to evaluate the role of pollinator-mediated selection in maintaining gynodioecy. METHODS: We studied sex-biased pollinator dynamics across 28 gynodioecious populations o
