Proteomic Heterogeneity of the Extracellular Matrix Identifies Histologic Subtype-Specific Fibroblast in Gastric Cancer.

Gastric cancer (GC) is a highly heterogeneous disease regarding histologic features, genotypes, and molecular phenotypes. Here, we investigate extracellular matrix (ECM)-centric analysis, examining its association with histologic subtypes and patient prognosis in human GC. We performed quantitative proteomic analysis of decellularized GC tissues that characterizes tumorous ECM, highlighting proteomic heterogeneity in ECM components. We identified 20 tumor-enriched proteins including four glycoproteins, serpin family H member 1 (SERPINH1), annexin family (ANXA3/4/5/13), S100A family (S100A6/8/9

Reduced fibrous capsule elastic fibers from biologic ECM-enveloped CIEDs in minipigs, supported with a novel compression mechanics model.

BACKGROUND: Fibrous capsules (Fb) in response to cardiovascular implantable electronic devices (CIEDs), including a pacemaker (P) system, can produce patient discomfort and difficulties in revision surgery due partially to their increased compressive strength, previously linked to elevated tissue fibers. OBJECTIVE: A preliminary study to quantify structural proteins, determine if biologic extracellular matrix-enveloped CIEDs (PECM) caused differential Fb properties, and to implement a realistic mechanical model. METHODS: Retrieved Fb (-P and -PECM) from minipigs were subjected to biomechanic

Biomimetic cryogel promotes the repair of osteoporotic bone defects through altering the ROS niche via down-regulating the ROMO1.

Osteoporosis is a systemic bone disease that is prone to fractures due to decreased bone density and bone quality, and delayed union or nonunion often occurs in osteoporotic fractures. Therefore, it is particularly important to develop tissue engineering materials to promote osteoporotic fracture healing. In this study, a series of biomimetic cryogels prepared from the decellularized extracellular matrix (dECM), methacrylate gelatin (GelMA), and carboxymethyl chitosan (CMCS) via unidirectional freezing, photo- and genipin crosslinking were applied for the regeneration of osteoporotic fractures

Integrated 3D Printing-Based Framework-A Strategy to Fabricate Tubular Structures with Mechanocompromised Hydrogels.

Several hollow organs perform various crucial functions in the body and must be replaced, repaired, or augmented in many disease conditions. Fabrication of tissue analogues to these hollow organs is incredibly challenging. Still, recent advancements in biofabrication have allowed researchers to pursue the development of several hollow organs such as blood vessels, esophagus, trachea, urethra, and others. Materials like collagen, alginate, elastin, silk, fibrin, etc., have been predominantly used for organ development. However, the focus has been duly shifted toward decellularized extracellular

Bone Fracture-Treatment Method: Fixing 3D-Printed Polycaprolactone Scaffolds with Hydrogel Type Bone-Derived Extracellular Matrix and β-Tricalcium Phosphate as an Osteogenic Promoter.

Bone formation and growth are crucial for treating bone fractures. Improving bone-reconstruction methods using autologous bone and synthetic implants can reduce the recovery time. Here, we investigated three treatments using two different materials, a bone-derived decellularized extracellular matrix (bdECM) and β-tricalcium phosphate (β-TCP), individually and in combination, as osteogenic promoter between bone and 3D-printed polycaprolactone scaffold (6-mm diameter) in rat calvarial defects (8-mm critical diameter). The materials were tested with a human pre-osteoblast cell line (MG63) to dete

Intracoronary Delivery of Porcine Cardiac Progenitor Cells Overexpressing IGF-1 and HGF in a Pig Model of Sub-Acute Myocardial Infarction.

Human cardiac progenitor cells (hCPC) are considered a good candidate in cell therapy for ischemic heart disease, demonstrating capacity to improve functional recovery after myocardial infarction (MI), both in small and large preclinical animal models. However, improvements are required in terms of cell engraftment and efficacy. Based on previously published reports, insulin-growth factor 1 (IGF-1) and hepatocyte growth factor (HGF) have demonstrated substantial cardioprotective, repair and regeneration activities, so they are good candidates to be evaluated in large animal model of MI. We hav

High-Fat-Diet-Induced Hyperglycemia Alters Liver Extracellular Matrix Composition in Mice Model.

Regenerative medicine integrates interdisciplinary approaches towards restoring the function of diseased organs. This study examined alterations that occurred in the liver under a high-fat diet (HFD) with the development of obesity and fatty liver, and changes in metabolic homeostasis and glucose levels, in mice. HFD nutrition causes hyperglycemia, leading to the formation and accumulation of advanced glycation end-products (AGEs) promoting protein post-translational modifications (PTMs) and introducing crosslinking in the extracellular matrix (ECM). Using histological and gene expression anal

Small intestine submucosa decorated 3D printed scaffold accelerated diabetic bone regeneration by ameliorating the microenvironment.

The 3D printed scaffolds constructed from polymers have shown significant potential in the field of bone defect regeneration. However, the efficacy of these scaffolds can be markedly reduced in certain pathological conditions like diabetes, where an altered inflammatory microenvironment and diminished small blood vessels complicate the integration of these polymers with the host tissue. In this study, the bioactivity of a 3D-printed poly(lactide-co-glycolide) (PLGA) scaffold is enhanced through the integration of hydroxyapatite (HA), icariin (ICA), and small intestine submucosa (SIS), a form o

Conserved and tissue-specific immune responses to biologic scaffold implantation.

Upon implantation into a patient, any biomaterial induces a cascade of immune responses that influences the outcome of that device. This cascade depends upon several factors, including the composition of the material itself and the location in which the material is implanted. There is still significant uncertainty around the role of different tissue microenvironments in the immune response to biomaterials and how that may alter downstream scaffold remodeling and integration. In this study, we present a study evaluating the immune response to decellularized extracellular matrix materials within

Strategies for whole lung tissue engineering.

Recent work has demonstrated the feasibility of using decellularized lung extracellular matrix scaffolds to support the engineering of functional lung tissue in vitro. Rendered acellular through the use of detergents and other reagents, the scaffolds are mounted in organ-specific bioreactors where cells in the scaffold are provided with nutrients and appropriate mechanical stimuli such as ventilation and perfusion. Though initial studies are encouraging, a great deal remains to be done to advance the field and transition from rodent lungs to whole human tissue engineered lungs. To do so, a var

Enhanced decellularization efficiency in combination of low-concentration chemical and physical methods on the human umbilical arteries.

BackgroundCardiovascular diseases are the leading cause of mortality worldwide, with coronary artery bypass grafting being the most effective treatment for severe cases. While autografts are preferred, donor veins are often limited. Human umbilical arteries (hUAs) show promise as an alternative. However, to make vascular graft by decellularization, a traditional chemical method can damage tissue structure and function.ObjectiveThis study aims to evaluate the shortening of treatment time and the hUA decellularization efficiency of the perfusion bioreactor systems.MethodshUAs were perfused with

Reconstruction of the orbital floor using supercritical CO2 decellularized porcine bone graft.

Orbital floor fractures subsequently lead to consequences such as diplopia and enophthalmos. The graft materials used in orbital floor fractures varied from autografts to alloplastic grafts, which possess certain limitations. In the present study, a novel porcine bone matrix decellularized by supercritical CO2 (scCO2), ABCcolla® Collagen Bone Graft, was used for the reconstruction of the orbital framework. The study was approved by the institutional review board (IRB) of Kaohsiung Medical University Chung-Ho Memorial Hospital (KMUH). Ten cases underwent orbital floor reconstruction in KMUH in

In vivo remodeling of human cell-assembled extracellular matrix yarns.

Cell-assembled extracellular matrix (CAM) has been used to produce vascular grafts. While these completely biological vascular grafts performed well in clinical trials, the in vivo remodeling and inflammatory response of this truly "bio" material has not yet been investigated. In this study, human CAM yarns were implanted subcutaneously in nude rats to investigate the innate immune response to this matrix. The impact of processing steps relevant to yarn manufacturing was evaluated (devitalization, decellularization, gamma sterilization, and twisting). We observed that yarns were still present

Thermosensitive Hydrogel Derived from a Human Amniotic Membrane Promotes Diabetic Wound Healing.

Extracellular matrix (ECM) hydrogels are recognized as promising biomaterials for regenerative medicine owing to their ability to recapitulate the native tissue microenvironment. The human amniotic membrane (AM), readily available and posing little to no ethical concerns, is rich in ECM components with inherent wound-healing potential. This study aimed to develop and characterize thermosensitive hydrogels derived from a decellularized AM and assess their therapeutic potential for diabetic wound healing. The native AM was subjected to detergent-enzymatic decellularization to remove the cellular

Bioengineered MSC-derived extracellular vesicles in intervertebral disc therapeutics: A systematic review.

Intervertebral disc degeneration (IVDD) is a major contributor to chronic low back pain, representing a significant global health burden. Mesenchymal stem/stromal cell-derived extracellular vesicles (MSC-EVs) have emerged as a promising cell-free therapeutic strategy, with preclinical evidence demonstrating their ability to modulate inflammation, oxidative stress, apoptosis, senescence, and extracellular matrix (ECM) degradation. However, several challenges limit translation, including inconsistent EV isolation methods, incomplete mechanistic characterization, lack of standardized dosing, and

Production and assessment of decellularized pig and human lung scaffolds.

The authors have previously shown that acellular (AC) trachea-lung scaffolds can (1) be produced from natural rat lungs, (2) retain critical components of the extracellular matrix (ECM) such as collagen-1 and elastin, and (3) be used to produce lung tissue after recellularization with murine embryonic stem cells. The aim of this study was to produce large (porcine or human) AC lung scaffolds to determine the feasibility of producing scaffolds with potential clinical applicability. We report here the first attempt to produce AC pig or human trachea-lung scaffold. Using a combination of freezing

ROS-responsive 3D biological scaffold delivers hypoxia-primed extracellular vesicles for targeted modulation of neuroinflammation in intracerebral hemorrhage.

BACKGROUND: Emerging evidence suggests that paracrine mechanisms may underlie the therapeutic effects of human umbilical cord mesenchymal stem cell-derived extracellular vesicles (EVs) (hUCMSC-exos) in mitigating neuroinflammation following intracerebral hemorrhage (ICH). Hypoxic preconditioning enhances the paracrine efficacy of hUCMSC-exos. Building on prior studies [1, 2], we developed a ROS-responsive three-dimensional (3D) biological scaffold encapsulating hypoxia-primed EVs (Hypo-Exos) for sustained release under reactive oxygen species (ROS)-rich conditions. METHODS: The 3D biological

Bioprinted dECM particles-laden microgels enhance the mesenchymal stromal cell paracrine effect for myocardial infarction treatment.

Despite guideline-directed reperfusion and pharmacological therapies, myocardial infarction (MI) continues to pose substantial clinical challenges, with high incidence and mortality rates. Mesenchymal stromal cell (MSC) transplantation represents a promising therapeutic strategy, yet its efficacy remains limited by poor survival and unstable paracrine function of transplanted cells within the hostile infarct microenvironment. Here, we developed an integrated microcarrier system via digital light processing, which encapsulates MSCs within microgels functionalized with cardiac-derived decellular

A xenogeneic developmental matrix hydrogel with MnO2-gallic acid enables spatiotemporal immunometabolic reprogramming and endochondral bone repair in steroid-associated osteonecrosis.

Steroid-associated osteonecrosis (SON) is characterized by a refractory bone defect microenvironment involving oxidative stress, persistent inflammation, and impaired vascularization, which limits the efficacy of conventional graft materials. Here, we developed a temporally programmable composite hydrogel, GelMA/dECM@MnO2-GA, by integrating human induced pluripotent stem cell (iPSC)-derived decellularized extracellular matrix (iPSC-dECM) with an MnO2-gallic acid (GA) reactive oxygen species (ROS)-responsive module. Following implantation, elevated ROS may induce the release of MnO2 and GA, whi
