Efferent projections of the suprachiasmatic nucleus: II. Studies using retrograde transport of fluorescent dyes and simultaneous peptide immunohistochemistry in the rat.

In a previous study (Watts et al., '87) we reexamined the projections of the suprachiasmatic nucleus (SCh) with the PHA-L method and found that they could be divided conveniently into six groups of fibers. By far the densest projection ends just dorsal to the SCh in a comma-shaped region designated the "subparaventricular zone," although some fibers continue on through the paraventricular nucleus of the hypothalamus to end in the overlying midline thalamus, and others continue on to end in the dorsomedial nucleus, the region around the ventromedial nucleus, and the posterior hypothalamic area.

Automated MRI-based parcellation of a putative core auditory region in individual human Heschl's gyrus.

The human auditory cortex exhibits substantial anatomical variability, making precise individual-level delineation challenging. Although recent advances in imaging and machine learning techniques have enabled the delineation of approximate boundaries of the human primary auditory cortex, existing approaches often fail to adequately capture individual anatomical variability. Here, we propose an openly accessible, fully automated MRI-based pipeline for identifying a putative core auditory region within Heschl's gyrus by integrating intracortical myelin estimates with cortical curvature. A data-d

Impaired prepulse inhibition in APP/PS1 mice is accompanied by substantial morphological changes in neurons of the central auditory system and hippocampus.

Auditory dysfunction is increasingly recognized as a non-cognitive feature of Alzheimer's disease (AD). We examined auditory processing and neuronal morphology in APPswe/PSEN1dE9 (APP/PS1) transgenic mice, a model of AD. Ten-month-old male APP/PS1 and wild-type (WT) littermates were tested for auditory thresholds using auditory brainstem responses (ABR) and for sensorimotor gating using prepulse inhibition (PPI) of the acoustic startle reflex. ABR thresholds did not differ between groups across tested frequencies (2-16 kHz), indicating preserved peripheral hearing. In contrast, APP/PS1 mice sh

Parallel Encoding of Speech in Human Frontal and Temporal Lobes.

Models of speech perception are centered around a hierarchy in which auditory representations in the thalamus propagate to primary auditory cortex, then to the lateral temporal cortex, and finally through dorsal and ventral pathways to sites in the frontal lobe. However, evidence for short latency speech responses and low-level spectrotemporal representations in frontal cortex raises the question of whether speech-evoked activity in frontal cortex strictly reflects downstream processing from lateral temporal cortex or whether there are direct parallel pathways from the thalamus or primary audi

Hearing loss is an early biomarker in APP/PS1 Alzheimer's disease mice.

Alzheimer's disease (AD) is a neurodegenerative disease characterized by a progressive loss of memory and cognitive decline. Over the last decade, it has been found that defects in sensory systems could be highly associated with AD. Hearing is an important neural sense. However, little is known about hearing functional changes in AD. In this study, APP/PS1 AD mice (Jackson Lab: Stack No. 004462) were used. Hearing function was assessed by auditory brainstem response (ABR), distortion product otoacoustic emission (DPOAE), and cochlear microphonics (CM) recordings. Wild-type (WT) littermates ser

White matter differences between essential tremor and Parkinson disease.

OBJECTIVE: To assess white matter integrity in patients with essential tremor (ET) and Parkinson disease (PD) with moderate to severe motor impairment. METHODS: Sedated participants with ET (n = 57) or PD (n = 99) underwent diffusion tensor imaging (DTI) and fractional anisotropy, mean diffusivity, axial diffusivity, and radial diffusivity values were computed. White matter tracts were defined using 3 well-described atlases. To determine candidate white matter regions that differ between ET and PD groups, a bootstrapping analysis was applied using the least absolute shrinkage and selection op

The influence of aging on the number of neurons and levels of non-phosporylated neurofilament proteins in the central auditory system of rats.

In the present study, an unbiased stereological method was used to determine the number of all neurons in Nissl stained sections of the inferior colliculus (IC), medial geniculate body (MGB), and auditory cortex (AC) in rats (strains Long Evans and Fischer 344) and their changes with aging. In addition, using the optical fractionator and western blot technique, we also evaluated the number of SMI-32-immunoreactive (-ir) neurons and levels of non-phosphorylated neurofilament proteins in the IC, MGB, AC, and visual cortex of young and old rats of the two strains. The SMI-32 positive neuronal pop

Exercise increases blood flow to locomotor, vestibular, cardiorespiratory and visual regions of the brain in miniature swine.

1. The purpose of these experiments was to use radiolabelled microspheres to measure blood flow distribution within the brain, and in particular to areas associated with motor function, maintenance of equilibrium, cardiorespiratory control, vision, hearing and smell, at rest and during exercise in miniature swine. Exercise consisted of steady-state treadmill running at intensities eliciting 70 and 100 % maximal oxygen consumption (V(O(2),max)). 2. Mean arterial pressure was elevated by 17 and 26 % above that at rest during exercise at 70 and 100 % V(O(2),max), respectively. 3. Mean brain blood

Functional impairment due to white matter ischemia after middle cerebral artery occlusion in cats.

We recorded regional cerebral blood flow, somatosensory evoked potentials, and auditory evoked potentials in the thalamic relay nuclei (ventral posterior lateral nucleus and medial geniculate body) and in the somatosensory and auditory cortices during and after 1 hour of transient left middle cerebral artery occlusion in nine cats. Regional cerebral blood flow was also measured in the thalamocortical tracts of five of these cats. Additionally, the integrity of thalamocortical connections was tested by retrograde labeling of the thalamic nuclei with horseradish peroxidase in eight cats (three o

The human brain at stages 21-23, with particular reference to the cerebral cortical plate and to the development of the cerebellum.

The development of the human brain during the eighth embryonic week was studied in serial sections of 22 embryos, and graphic reconstructions were prepared. The cortical plate appears in stage 21 in the area of the future insula and is an excellent feature for staging. The internal capsule contains neocortical fibres. Its three main outlets begin to be present in stage 22 and lead to epithalamus, to dorsal thalamus, and to mesencephalon. At this time a well developed lateral olfactory tract can be seen. The anterior commissure appears in stage 23. A clear developmental relationship between cla

Accumulation of amyloid precursor protein-like immunoreactivity in rat brain in response to thiamine deficiency.

Thiamine deficiency (TD) is a classical model of impaired cerebral oxidation. As in Alzheimer's disease (AD), TD is characterized by selective neuronal loss, decreased activities of thiamine pyrophosphate-dependent enzymes, cholinergic deficits and memory loss. Amyloid beta-protein (A beta), a approximately 4 kDa fragment of the beta-amyloid precursor protein (APP), accumulates in the brains of patients with AD or Down's syndrome. In the current study, we examined APP and A beta immunoreactivity in the brains of thiamine-deficient rats. Animals received thiamine-deficient diet ad libitum and d

Macrostructural Changes of the Acoustic Radiation in Humans with Hearing Loss and Tinnitus Revealed with Fixel-Based Analysis.

Age-related hearing loss is the most prevalent sensory impairment in the older adult population and is related to noise-induced damage or age-related deterioration of the peripheral auditory system. Hearing loss may affect the central auditory pathway in the brain, which is a continuation of the peripheral auditory system located in the ear. A debilitating symptom that frequently co-occurs with hearing loss is tinnitus. Strikingly, investigations into the impact of acquired hearing loss, with and without tinnitus, on the human central auditory pathway are sparse. This study used diffusion-weig

Noninvasive topical loading for manganese-enhanced MRI of the mouse visual system.

PURPOSE: To evaluate topical loading as an alternative to intravitreal injection for Mn(2+)-enhanced magnetic resonance imaging (MEMRI) of the visual system. METHODS: Topical administration of 0.5 to 1.5 M MnCl(2) and intravitreal injections with 0.5 μL 100 mM and 2 μL 1 M MnCl(2) for mouse MEMRI were conducted, followed by immunohistochemistry. In another mouse group, two topical administrations of 1 M Mn(2+) were applied to the same animals 7 days apart, to evaluate the use of MEMRI in a time course study. Dynamic imaging was also conducted to reveal how Mn(2+) travels to the retina. MEMRI

Inhibitory effect of state independent ponto-geniculo-occipital waves on seizure occurrence induced by local application of penicillin into the temporal lobe amygdala.

PURPOSE: In order to explore the possible inhibitory role of the phasic phenomena of REM sleep ponto-geniculo-occipital (PGO) waves over epilepsy, seizure activity produced by topic administration of Na-penicillin (PCN) has been analyzed during sustained PGO waves irrespective of current state. PGO waves were induced by the injection of carbachol in the peribrachial area. METHODS: The development of acute experimental epilepsy was compared among nine chronically implanted, adult, male cats, by means of polygraphic 23 h recordings. Our protocol consisted of sets of 4 trials: carbachol; PCN; ca

Amblyopia decreases activation of the corticogeniculate pathway and visual thalamic reticularis in attentive rats: a 'focal attention' hypothesis.

In rats which were rendered monocular amblyopic by lid suturing one eye during a critical period, the intensity of neuronal activation in parts of the monocular segments of the striate cortex (layers 4 and 6) and lateral geniculate nucleus, and in the visual segment of the thalamic reticular nucleus, was determined after exploration of a novel-complex environment. Quantitative analysis of the number of Fos-labelled neurons per unit area showed that, in comparison to the structures contralateral to the normal eye, in the side contralateral to the deprived amblyopic eye there is a gradient of di

Highly variable distribution of HSV-1-specific DNA in human geniculate, vestibular and spiral ganglia.

Viral reactivation in temporal ganglia is the suspected cause of Bell's palsy, vestibular neuritis and sudden hearing loss. Since the distribution of latent herpes simplex type 1 (HSV-1) in geniculate, vestibular and spiral ganglia of individual human temporal bones could have implications for the explanation of isolated as well as combined disorders of these three cranial nerves, we examined these ganglia in 18 human temporal bones of adults by nested polymerase chain reaction. In all of the temporal bones HSV-1 specific DNA was detected: 10/18 (56%) of the geniculate, 11/18 (61%) of the vest

Elicited PGO waves in rats: lack of 5-HT1A inhibition in putative pontine generator region.

Ponto-geniculo-occipital (PGO) waves and an elicited analogue (PGOE) may be recorded in the pons of rats. Cholinergic cells in the pedunculopontine tegmental (PPT) and laterodorsal tegmental (LDT) nuclei are implicated in the generation of PGO waves. Serotonin (5-HT) may inhibit the generation of PGO waves, and possibly PGOE. We examined the role of 5-HT1A receptor mechanisms in the generation of auditory-elicited PGOE in rats. Administration of 8-OH-DPAT [8-hydroxy-2-(n-dipropylamino)tetralin] into PPT did not significantly affect PGOE amplitude or response frequency. Binding of [3H]CN-IMI ([

Complex visual hallucinations in macular degeneration.

A previously healthy elderly patient with a recent onset of macular degeneration presented for evaluation of elaborate complex visual hallucinations. The patient's psychiatric evaluation and level of cognitive functioning were normal. A diagnosis of organic hallucinosis secondary to macular degeneration was made, and the hallucinations ceased with increased sensory stimulation in the hospital. Numerous diagnoses were considered and are discussed, including Charles Bonnet hallucinations and peduncular hallucinosis.
