Expressing OsMPK4 Impairs Plant Growth but Enhances the Resistance of Rice to the Striped Stem Borer Chilo suppressalis.

Mitogen-activated protein kinases (MPKs) play a central role not only in plant growth and development, but also in plant responses to abiotic and biotic stresses, including pathogens. Yet, their role in herbivore-induced plant defenses and their underlying mechanisms remain largely unknown. Here, we cloned a rice MPK gene, OsMPK4, whose expression was induced by mechanical wounding, infestation of the striped stem borer (SSB) Chilo suppressalis, and treatment with jasmonic acid (JA), but not by treatment with salicylic acid (SA). The overexpression of OsMPK4 (oe-MPK4) enhanced constitutive and

High resistance of transgenic cabbage plants with a synthetic cry1Ia8 gene from Bacillus thuringiensis against two lepidopteran species under field conditions.

BACKGROUND: Plutella xylostella (Linnaeus) has become the most destructive pest in cabbage throughout the world. Cry1Ia8 cabbage has been developed to reduce pest attacks. To gain a better understanding of the efficacy of Cry1Ia8 cabbage, a homozygous Cry1Ia8 cabbage line A14-5 was produced, and its resistance to P. xylostella, Pieris rapae (Linnaeus) and other lepidopteran pests was evaluated in the field in 2011, 2012 and 2013. RESULTS: Under natural infestation conditions, the homozygous transgenic line was highly resistant against P. xylostella and P. rapae as compared with the untransfor

An invertebrate model to evaluate virulence in Aspergillus fumigatus: the role of azole resistance.

The impact of different mutations in the Aspergillus fumigatus ergosterol biosynthesis pathway on pathogenesis has been evaluated using a simple invertebrate mini host, the caterpillar Galleria mellonella. A set of strains that includes clinical isolates and isogenic mutants with mutations at the cyp51A gene conferring azole resistance were studied. All strains demonstrated a similar in vitro growth pattern and are equally virulent against the insect larvae. These results suggest that in A. fumigatus acquisition of this particular azole-resistance mechanism would not imply any significant chan

Study on the microstructure of African wild silk cocoon shells and fibers.

Silk fibers and cocoon shells from four African wild silkmoths Gonometa postica, Anaphe panda, Argema mimosae and Epiphora bauhiniae-were studied to gain insight into the structure-property-function relations and potential commercial application. The surface and cross-section of cocoon shells and fibers revealed the presence of prominent structural variations. Cocoon shells were multilayered and porous structures constructed from highly cross-linked fibers that are densely packed within the sericin/gum. Fibers had fibrillar sub-structures running along the fiber axis and with greater number an

Caterpillar setae-induced acute anterior uveitis: a case report.

PURPOSE: To report uveitis secondary to ocular penetration of caterpillar hairs (setae). METHODS: Case report. A documented attack of acute anterior uveitis was caused by initially overlooked penetration of caterpillar setae. RESULTS: A 66-year-old man presenting with unilateral hypertensive keratouveitis was treated with antiherpes simplex medication (along with local anti-inflammatory and cycloplegic agents) after anterior chamber paracentesis and serologic testing. Laboratory testing was negative. Resolution occurred after 5 days, and corneal clearing showed a predescemetic caterpillar se

Synthesis of biologically active influenza virus hemagglutinin in insect larvae.

The hemagglutinin of influenza (fowl plague) virus was expressed in larvae of Heliothis virescens by using recombinant Autographa californica nuclear polyhedrosis virus (AcNPV) as a vector. Animals were infected with the recombinant virus either by parenteral injection or by feeding. For oral uptake, recombinant virus occluded in polyhedra obtained from cultured Spodoptera frugiperda cells after coinfection with authentic AcNPV was used. Immunohistological analyses of infected animals revealed that the hemagglutinin was expressed only in those tissues that are also permissive for the replicati

A Wor1-Like Transcription Factor Is Essential for Virulence of Cryptococcus neoformans.

Gti1/Pac2 transcription factors occur exclusively in fungi and their roles vary according to species, including regulating morphological transition and virulence, mating and secondary metabolism. Many of these functions are important for fungal pathogenesis. We therefore hypothesized that one of the two proteins of this family in Cryptococcus neoformans, a major pathogen of humans, would also control virulence-associated cellular processes. Elimination of this protein in C. neoformans results in reduced polysaccharide capsule expression and defective cytokinesis and growth at 37°C. The mutant

Evaluation of the efficacy of antifungal drugs against Paracoccidioides brasiliensis and Paracoccidioides lutzii in a Galleria mellonella model.

Paracoccidioides brasiliensis and P. lutzii belong to a group of thermodimorphic fungi and cause paracoccidioidomycosis (PCM), which is a human systemic mycosis endemic in South and Central America. Patients with this mycosis are commonly treated with amphotericin B (AmB) and azoles. The study of fungal virulence and the efficacy and toxicity of antifungal drugs has been successfully performed in a Galleria mellonella infection model. In this work, G. mellonella larvae were infected with two Paracoccidioides spp. and the efficacy and toxicity of AmB and itraconazole were evaluated in this mode

In vivo efficacy of telavancin/colistin combination therapy in a Galleria mellonella model of Acinetobacter baumannii infection.

Treatment of Acinetobacter baumannii infections is challenging owing to widespread multidrug resistance and the lack of novel agents. There is now considerable interest in the potential of unorthodox combination therapies such as colistin and glycopeptides (e.g. vancomycin and teicoplanin), since potent synergy can be demonstrated in vitro. A simple invertebrate model (Galleria mellonella) has been developed to assess the in vivo activity of antimicrobial therapies and was used to investigate the efficacy of colistin combined with the lipoglycopeptide telavancin in the treatment of A. baumanni

Immunochemical and proteomic technologies as tools for unravelling toxins involved in envenoming by accidental contact with Lonomia obliqua caterpillars.

The accidental contact with Lonomia obliqua caterpillar causes local and systemic symptoms (such as fibrinogen depletion), leading, in some cases, to serious clinical complications (acute renal failure and intracranial haemorrhage). Fortunately, a successful therapeutical approach using anti-Lonomic serum, produced in horses against L. obliqua's bristle extract, has already been put in place. However, a global view of immunogenic toxins involved in the coagulation disorders could help to elucidate the envenoming process. In the present study, our aim was to identify bristle extract's immunogen

Comparative ultrastructure and blood-brain barrier in diapause and nondiapause larvae of the European corn borer Ostrinia nubilalis (Hübner).

Ultrastructural examination of diapause and nondiapause larval brains of the European corn borer disclosed anatomical differences that may be related to the insect's "blood-barrier". The perineurial type I cells are quite closely appressed in the diapause brain, but thrown into extensive folds with large intercellular spaces in the nondiapause brain. The perineurial type II cells of diapause and nondiapause larvae are basically similar in general ultrastructure, and most likely form the basis for the "blood-brain barrier." Horseradish peroxidase penetration studies indicated that the outer mar

High-Throughput DNA Barcoding Reveals Multi-Trophic Networks of Plants, Leafminers and Parasitoids in a Tropical Dry Forest.

Tropical dry forests host highly diverse insect communities and complex trophic interactions, yet these networks remain difficult to resolve using conventional molecular or rearing-based approaches. Here, we integrate long-read DNA barcoding, non-target sequence co-amplification, and host plant taxonomy to reconstruct tri-trophic interactions among leaf-mining insects, their host plants, and associated parasitoids in a Mexican tropical dry forest. Using Single Molecule, Real-Time (SMRT) sequencing on the PacBio Sequel IIe platform, we generated high-fidelity barcodes from individual leaf-miner

Functional Characterization of Peripheral Neurons and a Receptor Recognizing Sex Pheromones in Hyphantria cunea (Erebidae).

Hyphantria cunea (Lepidoptera: Erebidae) is difficult and costly to control as a quarantine pest found globally. Sex pheromone trapping is an effective measure for its population monitoring and control; however, the peripheral neural mechanism of sex pheromone recognition in H. cunea remains unclear. An electrophysiological analysis showed that both male and female moths of H. cunea responded to four components of sex pheromones and the responses of male moths were stronger than those of the female moths. We identified three types of trichoid sensilla (ST) responsive to sex pheromones using th

Semiochemicals from Trichosanthes anguina (Cucurbitaceae) plants influence behavior in Diaphania indica.

BACKGROUND: First to third instars of Diaphania indica (Saunders) (Lepidoptera: Crambidae) feed on the lower surface of leaves, while fourth and fifth instars gregariously consume leaves of Trichosanthes anguina L. After defoliating, the caterpillar also attacks flowers and fruits of the plant and finally, results in loss of crop yield. Therefore, behavioral responses of D. indica adults were investigated to volatiles from undamaged (UD), insect-damaged (ID, plants after feeding by D. indica larvae) and jasmonic acid (JA) treated T. anguina plants. RESULTS: Females showed attraction to volati

Infectivity of entomopathogenic nematodes against the legume pod-borer, Maruca vitrata Fabricius, infesting pigeon pea.

The legume pod-borer, Maruca vitrata Fabricius (Lepidoptera: Crambidae) (LPB), is an important insect pest of pigeon pea. Chemical pesticides are generally employed to manage this pest, but because of the soil residue issues and other environmental hazards associated with their use, biopesticides are also in demand. Another benign alternative is to use entomopathogenic nematodes (EPNs) to manage this vital pest. In the present study, the infectivity of ten native EPNs was evaluated against LPB by assessing their penetration and production in the LPB. The effectiveness of the promising EPNs aga

WSR-88D doppler radar detection of corn earworm moth migration.

Corn earworm (Lepidoptera: Noctuidae) (CEW) populations infesting one crop production area may rapidly migrate and infest distant crop production areas. Although entomological radars have detected corn earworm moth migrations, the spatial extent of the radar coverage has been limited to a small horizontal view above crop production areas. The Weather Service Radar (version 88D) (WSR-88D) continuously monitors the radar-transmitted energy reflected by, and radial speed of, biota as well as by precipitation over areas that may encompass crop production areas. We analyzed data from the WSR-88D ra

Multifunctional Properties of the Gut Symbiont Enterococcus gallinarum Associated With Eudocima materna Caterpillars in Host Plant Detoxification.

Gut symbiotic bacteria are highly dynamic and contribute to digestion, development, immunity, detoxification, and environmental adaptation in insect hosts. In fruit-sucking moths (FSM), particularly Eudocima materna (Lepidoptera: Noctuidae), adults are destructive frugivorous pests, whereas larvae feed exclusively on toxic plants such as Tinospora cordifolia. This study investigates the gut bacterial diversity of FSM larvae, with an emphasis on their role in host plant adaptation. Nine bacterial strains were isolated, including Enterobacter hormaechei, Klebsiella aerogenes, Mammaliicoccus sciu

Differential pathogenicity of heterologous nucleopolyhedroviruses on Spodoptera frugiperda corresponds to modulated expression of immune-response-related genes.

The fall armyworm Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) is a notorious invasive pest wreaking havoc on various crops globally. Nucleopolyhedroviruses (NPVs) are viral pathogens that specially target lepidopteran pests. However, the homologous virus, Spodoptera frugiperda multiple nucleopolyhedrovirus (SfMNPV), has not been commercialized in China. Therefore, understanding the molecular mechanisms underlying heterologous virus-host interactions can inform the design of virus-based insecticides for controlling S. frugiperda. The pathogenicity of the four heterologous NPVs o

Sugar Diet Affects Odor Reception but Variation in Sugar Concentration Plays Minimal Role in the Response of the Parasitoid, Microplitis croceipes (Hymenoptera: Braconidae), to Host-Related Plant Volatiles.

Parasitoids utilize various sugar resources in nature, and rely on odor cues from plants to locate their food and hosts. However, lack of sugar in the diet may negatively impact odor reception in parasitoids, thus affecting foraging efficiency. We used Microplitis croceipes (Cresson) (Hymenoptera: Braconidae), a larval endoparasitoid of Heliothis virescens (F.) (Lepidoptera: Noctuidae), as a model species to test the hypothesis that variation in sugar diet of parasitoids affects their olfactory response to host-related odors. Heliothis virescens is a major pest of cotton and other important cr

Courtship behavior of the castniid palm borer, Paysandisia archon: potential roles of male scents and visual cues in a day-flying moth.

The castniid palm borer, Paysandisia archon (Burmeister) (Lepidoptera: Castniidae), is a South American moth that in the last ten years has become a major pest of palm trees in the Mediterranean region. Current knowledge on the reproductive behavior of this diurnal moth suggests the importance of both visual and chemical cues, in particular the production of a male pheromone emitted during a specific scratching behavior. Male-produced scents have diverse functions in lepidopteran sexual communication but generally act during courtship behavior, leading to complex, stereotyped courtship sequenc
