Enzyme-linked immunosorbent assay for diagnosis of Amphimerus spp. liver fluke infection in Humans.

BACKGROUND: Amphimerus spp. is a liver fluke that infects humans and domestic animals. It is highly prevalent in some Ecuadorian communities. Currently, diagnosis is based on the microscopic observation of eggs in faeces, but this has variable sensitivity. More sensitive methods are needed for diagnostic testing. OBJECTIVE: The main objective of this work was to develop an enzyme-linked immunosorbent assay (ELISA) using crude antigens from Amphimerus spp. adult worms to detect anti-Amphimerus IgG in human sera. METHODS: Crude somatic antigens were obtained from adult Amphimerus spp. worms. H

Immunoglobulin G (IgG) subclass and IgE responses in human paragonimiases caused by three different species.

In 40 cases of human paragonimiases caused by Paragonimus westermani (20 cases), P. miyazakii (10 cases), and P. skrjabini (10 cases), responses of serum immunoglobulin G (IgG), IgG subclasses, and IgE were analyzed by immunoblotting with crude antigens prepared from egg, 4-week-old juvenile, and adult forms of P. westermani. The 32- and 35-kDa proteins in the adult extracts showed specific reactions regardless of the causative species (39 of 40 cases; 98%). Sera of patients infected with P. westermani and P. miyazakii reacted strongly with the 28-, 46-, and 94-kDa proteins of egg extracts, wh

Evolutionary dynamics of endogenous densoviruses NS1 proteins reveal ancient codivergence with their Platyhelminth hosts.

Endogenous viral elements serve as molecular fossils of ancient viral infections, providing valuable evidence for reconstructing the evolutionary history of modern viruses and their hosts. These genomic remnants also provide crucial benchmarks for calibrating viral evolutionary timelines. Here, our flanking sequence analysis reveals that densovirus NS1 sequences are chromosomally integrated in Platyhelminth genomes. Furthermore, their evolutionary dynamics reflect an ancient host-virus codivergence event dating to the Ediacaran-Cambrian transition (approximately 692 million years ago, MYA). Ph

Clinical, Laboratory and Radiological Features of Paragonimiasis Misdiagnosed as Pulmonary Tuberculosis.

BACKGROUND: Paragonimiasis presents with nonspecific symptoms and radiologic findings, allowing for the possibility of misdiagnosis. Diagnosis is generally delayed due to lack of suspicion and presentation similar to pulmonary tuberculosis. METHODS: A prospective observational study was carried out on 20 subjects at Civil Service Hospital of Nepal from March 2015 to June 2019 who presented with eosinophilia and pulmonary symptoms, and were treated empirically with Anti-tubercular therapy for suspicion of pulmonary tuberculosis. RESULTS: The median age of the patient was 34 years. Mean blood

Pleural lump after paragonimiasis treated by thoracoscopy: A case report.

BACKGROUND: Paragonimiasis is a parasitic disease that has multiple symptoms, with pulmonary types being common. According to our clinical practices, the pleural effusion of our patients is full of fibrous contents. Drainage, praziquantel, and triclabendazole are recommended for the treatment, but when fibrous contents are contained in pleural effusion, surgical interventions are necessary. However, no related reports have been noted. Herein, we present a case of pulmonary paragonimiasis treated by thoracoscopy. CASE SUMMARY: A 12-year-old girl presented to our outpatient clinic complaining o

Sequences and gene organization of the mitochondrial genomes of the liver flukes Opisthorchis viverrini and Clonorchis sinensis (Trematoda).

Opisthorchis viverrini and Clonorchis sinensis are important trematodes infecting humans and animals, belonging to the family Opisthorchiidae. In the present study, we sequenced the nearly complete mitochondrial (mt) DNA (mtDNA) sequences of O. viverrini from Laos, obtained the complete mtDNA sequences of C. sinensis from China and Korea, and revealed their gene annotations and genome organizations. The mtDNA sequences of O. viverrini, C. sinensis (China isolate), C. sinensis (Korea isolate) were 13,510, 13,879, and 13,877 bp in size, respectively. Each of the three mt genomes comprises 36 gen

Development of enzyme-linked immunosorbent assay for serodiagnosis of human paragonimiasis.

BACKGROUND & OBJECTIVE: Although human paragonimiasis is known to occur in Manipur and Arunachal Pradesh, there is no indigenous immunodiagnostic test available in India. Sputum examination for detection of eggs is less sensitive method for diagnosis of paragonimiasis and unfortunately, the eggs are not always present in the sputum of infected individuals. Due to overlapping clinical manifestations and similarities between X-ray picture in pulmonary paragonimiasis and pulmonary tuberculosis, chances of diagnostic confusion can increase. Therefore, the objective of this study was to develop an

Paragonimiasis: A continuing public health problem in northeast India.

PURPOSE: Trematode species belonging to the genus Paragonimus are the source of the food-borne parasitic zoonosis known as paragonimiasis. The disease has been identified as an outbreak in northeastern India since the early 1980s and is frequently diagnosed as tuberculosis because of similar clinical and radiological findings. This study is an attempt to update the clinic-radiological features and geographical distribution of the disease in recent times. METHODS: We retrospectively collected and analysed the data of 37 paragonimiasis suspected cases attending three healthcare facilities in Ma

Molecular Identification of the Human Pathogen Amphimerus sp. in the Freshwater Snail Aroapyrgus sp. in Ecuador.

Here, we report for the first time the snail intermediate host for the Amphimerus liver fluke, a foodborne trematodiasis. In Ecuador, Amphimerus of the Opisthorchiidae family, infects humans, cats, and dogs, in the tropical Pacific-coast region. Opisthorchiidae comprising also Clonorchis sinensis, Opisthorchis sp., and Metorchis sp., have complex life cycles involving a definitive and two intermediate hosts. We identified morphologically and investigated the presence and prevalence of Amphimerus cercaria and DNA in freshwater snails collected in a human-amphimeriasis endemic region in Ecuador,

Foodborne Parasites Dominate Current Parasitic Infections in Hunan Province, China.

Great progress has been made in the prevention and treatment of human parasitic diseases in China over the past six decades, but parasitic diseases are still one of the most serious public health problems in the world. The specific prevalence of parasitic diseases varies in different provinces due to their geographical environment and the dietary habits of people. In this study, a total of 4,428 patients suspected to have parasitic infection by clinicians or themselves from January 1, 2016, to December 31, 2020 were recommended to our laboratory for further testing. In total, 5,246 samples inc

Fasciola spp: Mapping of the MF6 epitope and antigenic analysis of the MF6p/HDM family of heme-binding proteins.

MF6p/FhHDM-1 is a small cationic heme-binding protein which is recognized by the monoclonal antibody (mAb) MF6, and abundantly present in parenchymal cells and secreted antigens of Fasciola hepatica. Orthologs of this protein (MF6p/HDMs) also exist in other causal agents of important foodborne trematodiasis, such as Clonorchis sinensis, Opisthorchis viverrini and Paragonimus westermani. Considering that MF6p/FhHDM-1 is relevant for heme homeostasis in Fasciola and was reported to have immunomodulatory properties, this protein is expected to be a useful target for vaccination. Thus, in this stu

Food-borne trematode infections of humans in the United States of America.

This review examines the literature on imported (allochthonous) and local (autochthonous) cases of food-borne trematode (FBT) infections in the United States of America (USA) from 1890 to 2009. Most of the literature is concerned with imported cases of the opisthorchiids Clonorchis sinensis and Opisthorchis viverrini. These flukes cause serious pathology in the liver and biliary system of humans. Chronic cases may induce liver (hepatocarcinoma) and bile duct (cholangiocarcinoma) cancers in humans. Clonorchiasis and opithorchiasis are preventable diseases that can be avoided by eating properly

[Parasitic zoonoses in Japan, with special reference to the current topics].

The present situation of parasitic zoonoses in Japan is reviewed. A total of 51 species, i.e., 6 species of protozoan parasites, 14 species of trematode parasites, 11 species of cestode parasites, 18 species of nematode parasites and 2 species of acanthocephalan parasites are potential zoonotic parasites. Most (11 species) of the zoonotic nematodes provoke larva migrans. Habitats of zoonotic parasites vary greatly depending upon individual species. Some parasites cause heterotopic parasitism (e.g., Paragonimus westermani) and metastasis (Entamoeba histolytica). In larva migrans, parasites migr
