BACKGROUND: Severe fever with thrombocytopenia syndrome (SFTS) is a severe viral disease caused by SFTSV. It is important to estimate the rate of missed SFTS diagnosis and to further understand the actual incidence in high endemic areas in China.
METHODS: This study was conducted in two high SFTS endemic provinces in 2015. Patients hospitalized in 2014 or within 1 year before investigation were selected after considering their clinical manifestations, specifically, fever, platelet, and white blood cell. During retrospective investigation, sera were collected to detect SFTSV antibodies to asse

This study investigated the humoral immune response dynamics in patients with severe fever with thrombocytopenia syndrome (SFTS) to evaluate their prognostic value and construct an early warning model. A retrospective cohort of 36 laboratory-confirmed SFTS patients with ≥ 4 serial antibody measurements out of 153 patients recruited in the study were analyzed. Patients were categorized by outcome into mild, severe, and fatal groups. The results showed that, the fatal group was older, had more comorbidities, and showed no antibody seroconversion during follow-up, along with a significantly highe

Severe fever with thrombocytopenia syndrome (SFTS) is an emerging tick-borne viral disease with high case-fatality rates in East Asia, yet no licensed vaccines are currently available. Here, we generated attenuated severe fever with thrombocytopenia syndrome virus (SFTSV) strains through serial passage in Huh-7 cells and evaluated their pathogenicity, immunogenicity, and protective efficacy. Attenuation candidates were selected based on reduced pathogenicity, estimated based on the median lethal dose (LD50), and genetic sequencing was performed to identify mutations associated with attenuation

Severe fever with thrombocytopenia syndrome (SFTS) is a highly fatal disease. Droplet digital polymerase chain reaction (ddPCR) presents unparalleled sensitivity and enables absolute quantification of viral load. In this prospective study, we enrolled 111 patients with SFTS and collected 259 continuous samples. Our findings unveil a robust reverse transcription (RT)-ddPCR method for SFTS with a limit of detection of 2.46 copies/µL (95% CI, 1.50-11.05), surpassing the sensitivity of RT-quantitative polymerase chain reaction at 103.29 copies/µL (95% CI, 79.69-216.35). Severe fever with thrombocytopenia syndrome (SFTS) is an emerging hemorrhagic fever disease caused by the SFTS virus (SFTSV). Despite pandemic concerns arising from repeated instances of human-to-human transmission and a high fatality rate, effective anti-SFTSV interventions remain unavailable. Here, utilizing single-cell RNA sequencing (scRNA-seq) and flow cytometry data, we revealed that the deficiency and dysfunction states of T cells, particularly the impaired cytotoxicity and exhausted state of CD4+ T cells, were significantly associated with lethal consequences in SFTS patients. Using an

Severe fever with thrombocytopenia syndrome virus (SFTSV) has been widely spread both domestically and internationally, with a mortality rate of up to 30 %; but no vaccine is currently available. During viral invasion, the glycoproteins Gn and Gc, which serve as the primary antigens, can exist in various natural conformations, including monomers and homo/hetero polymers. However, the role of these structural variations in activating immune response remains unclear. This study aimed to identify antigen formations with high immunogenicity by constructing protein vaccines with different structura

Severe fever with thrombocytopenia syndrome (SFTS) is a tick-borne infectious disease. A 91-year-old woman was admitted to our intensive-care unit with SFTS, and she developed dyspnea with wheezes 5 days after admission. Bronchoscopy showed scattered white mold in her central airway. An airway tissue biopsy and culture of bronchial lavage fluid revealed fungal hyphae in the necrotic tissue, confirmed as Aspergillus fumigatus. She was thus diagnosed with pseudomembranous aspergillus tracheobronchitis. She had no common risk factors for invasive aspergillosis (IA). Patients with SFTS, even those

A seroprevalence study was carried out in the region of Marseille (south-eastern France) to address the public health importance of sandfly fever Sicilian virus (SFSV) and SFSV-like viruses, as recently recognized vectors of those viruses are present in this area. The low seroprevalence rate observed in this study suggests that SFSV is not likely to be of major medical importance in the Marseille area.

It is suspected that apart from tick-borne encephalitis virus several additional European Arboviruses such as the sandfly borne Toscana virus, sandfly fever Sicilian virus and sandfly fever Naples virus, mosquito-borne Tahyna virus, Inkoo virus, Batai virus and tick-borne Uukuniemi virus cause aseptic meningo-encephalitis or febrile disease in Europe. Currently, the microarray technology is developing rapidly and there are many efforts to apply it to infectious diseases diagnostics. In order to arrive at an assay system useful for high throughput analysis of samples from aseptic meningo-enceph

Severe fever with thrombocytopenia syndrome virus (SFTSV) is a negative-sense RNA virus that can cause high fever, thrombocytopenia, and multiple-organ dysfunction. Toll-like receptor 7 (TLR7) can recognize single-stranded RNA viruses and induce type I interferons (IFNs). Although type I IFNs play a critical role in SFTSV infection, the role of TLR7 remains incompletely understood. To investigate the role of TLR7 signaling in SFTSV pathogenesis, TLR7-deficient (TLR7-/-) mice and wild-type (WT) mice were inoculated with SFTSV. TLR7-/- mice exhibited severe weight loss and elevated viral RNA lev

Hyperactive immune responses are implicated in the disease severity, prognosis, and complications of Severe Fever with Thrombocytopenia Syndrome (SFTS). In this study, we analyzed the differences in cytokine levels and clinical characteristics between severe and non-severe SFTS patients, aiming to assess the validity of interleukin-6 (IL-6), interleukin-8 (IL-8), interferon-α (IFN-α), and interferon-γ (IFN-γ) as predictors of disease severity and clinical outcomes in patients with SFTS. We retrospectively analyzed the demographic characteristics, clinical features, and the aforementioned four

Lipids and lipid metabolism play an important role in RNA virus replication, which typically occurs on host cell endomembrane structures in the cytoplasm through mechanisms that are not yet fully identified. We conducted genome-scale CRISPR screening and identified sphingomyelin synthase 1 (SMS1; encoded by SGMS1) as a critical host factor for infection by severe fever with thrombocytopenia syndrome virus (SFTSV). SGMS1 knockout reduced sphingomyelin (SM) (d18:1/16:1) levels, inhibiting SFTSV replication. A helix-turn-helix motif in SFTSV RNA-dependent RNA polymerase (RdRp) directly binds to S

Various stressors such as viral infection lead to the suppression of cap-dependent translation and the activation of the integrated stress response (ISR), since the stress-induced phosphorylated eukaryotic translation initiation factor 2 [eIF2(αP)] tightly binds to eIF2B to prevent it from exchanging guanine nucleotide molecules on its substrate, unphosphorylated eIF2. Sandfly fever Sicilian virus (SFSV) evades this cap-dependent translation suppression through the interaction between its nonstructural protein NSs and host eIF2B. However, its precise mechanism has remained unclear. Here, our c

Sandfly fever (SF) is an arthropod-borne disease, which has not yet been reported from Ankara. In the summer of 2007, the disease started to be seen in our region, surprisingly causing severe clinical presentations. This report reviews the clinical and laboratory findings of patients with sandfly virus infection of disease outbreaks in 2008 and 2009. A retrospective single-centre descriptive study was performed. Clinically suspected cases were defined on the basis of epidemiologic history and clinical and laboratory findings. The sera samples of the suspected patients were sent to Germany for

In the Mediterranean basin, sand flies are vectors of Leishmania parasites and phleboviruses affecting humans and animals. In this study, we aimed to investigate phlebovirus and Leishmania parasites circulating in a focus of zoonotic visceral leishmaniasis (ZVL) located in a highly irrigated area within the arid Central Tunisia, known mainly to be endemic for zoonotic cutaenous leishmaniasis (ZCL) caused Leishmania major and transmitted by Phlebotomus papatasi. Sand flies were collected using CDC light traps in the village of Saddaguia, an emergent focus of ZVL located in Central Tunisia durin

BACKGROUND: Severe fever with thrombocytopenia syndrome virus (SFTSV) is a highly pathogenic bunyavirus with a high case-fatality ratio for which there is no approved vaccine. Studies have assessed different vaccine technologies. However, few studies have yet assessed the immunogenicity of heterologous prime-boost regimens.
METHODS: Here, we compare a lipid nanoparticle (LNP)-encapsulated nucleoside-modified mRNA-based vaccine encoding the SFTSV glycoproteins, Gn and Gc, to our recently described recombinant VSV SFTSV (rVSV-SFTSV) vaccine in single dose, homologous, and heterologous prime-boo
