Influence of sire and sire breed (Gyr versus Holstein) on establishment of pregnancy and embryonic loss in lactating Holstein cows during summer heat stress.

Heat stress has negative effects on pregnancy rates of lactating dairy cattle. There are genetic differences in tolerance to heat stress; Bos taurus indicus (B. t. indicus) cattle and embryos are more thermotolerant than Bos taurus taurus (B. t. taurus). In the present study, the effects of sire and sire breed on conception and embryonic/fetal loss rates of lactating Holstein cows during the Brazilian summer were determined. In Experiment 1, cows (n=302) were AI after estrus detection or at a fixed-time with semen from one Gyr (B. t. indicus) or one Holstein sire (B. t. taurus). Pregnancy was

The current and potential role of hyperthermia in radiotherapy.

Current clinical experience strongly suggests that hyperthermia will become an important modality as an adjuvant to radiotherapy in the treatment of locally advanced solid tumors. Hyperthermia must therefore be considered a topic of general interest. Biologically, hyperthermia has two different types of interactions with radiation. Firstly, heat has a radiosensitizing effect. This is most prominent with simultaneous application, but is of the same magnitude in both tumor and normal tissue and will not improve the therapeutic ratio unless the tumor is heated to a higher temperature than the nor

Farmer's lung in a group of Scottish dairy farms.

ABSTRACT The microbiology of the air of byres and bruising sheds and of hay, grain and dust from bruising machines was studied in 12 dairy farms in Ayrshire and one in Perthshire. Seven farms (FLD) had a known case of farmer's lung disease and five farms (non-FLD) were free from the disease. Concentrations of mesophilic organisms and of thermotolerant and thermophilic fungi did not vary significantly between the two types of farm but the concentrations of thermophilic actinomycetes and bacteria, notably Micropolyspora faeni, were higher in general on FLD farms. Culture filtrate and mycelial ex

Embryonic thermal manipulation improved early immunity in broiler chickens.

Immunity in chickens is age-dependent and develops gradually over time. The initial defense by the host is a crucial mechanism for combating microbial infections. Embryonic thermal manipulation (TM) represents a promising approach for sustainable broiler production. Our previous work provided significant insights into the effects of TM on embryonic thermotolerance, metabolism, growth performance, microbial diversity, and immunity. This follow-up study used a subset of hatchlings previously used in our TM-related studies. This study aims to investigate the effects of TM on immunity-related gene

Systemic lupus erythematosus-related macrophage activation syndrome inducing severe cytopenia successfully treated with anifrolumab: a case report.

BACKGROUND: Heavy metal pollution from industrial operations poses severe environmental risks due to its toxicity and bioaccumulation. Conventional chemical and physical remediation methods are costly and generate secondary toxic sludge. Therefore, this study aims to investigate the potential of extracellular polymeric substance (EPS) producing bacteria as a sustainable, eco-friendly alternative for bioremediation. METHODS: EPS producing bacteria were isolated from chemical factory effluent sludge and screened on modified Winogradsky's medium. The isolates were identified by 16S rRNA gene seq

Interactions of the emerging fungus Candida auris with Acanthamoeba castellanii reveal phenotypic changes with direct implications on the response to stress and virulence.

UNLABELLED: Candida auris is an emerging fungal pathogen notable for its resistance to multiple antifungals and ability to survive in various environments. Understanding the interactions between C. auris and environmental protozoa, such as Acanthamoeba castellanii, could provide insights into fungal adaptability and pathogenicity. Two C. auris isolates (MMC1 and MMC2) were co-cultured with A. castellanii to examine interaction dynamics, survival, stress responses, growth, virulence, biofilm formation, and antifungal susceptibility. The association of C. auris-A. castellanii varied with a multi

Fenugreek (Trigonella foenum-graecum L.) as a functional food associated with improved heat-stress survival in Caenorhabditis elegans.

BACKGROUND: Fenugreek (Trigonella foenum-graecum L.) is widely consumed as a functional food across Mediterranean, Middle Eastern, and South Asian cultures and has long been valued in traditional diets for its health-promoting properties. Although fenugreek contains diverse phytochemicals with known biological activities, its potential effects on organismal resistance to heat stress remain insufficiently characterized. RESULTS: In this study, the effects of fenugreek seed preparations on thermotolerance-associated survival were evaluated using Caenorhabditis elegans. Two complementary extract

Embryonic thermal manipulation improved early cecal microbial diversity, metabolic pathways, and immunity in broiler chickens.

Gut microbiota regulates broilers' gastrointestinal functions, digestion, metabolism, and immune responses. Modulating the microbiota in the early stage could present a promising approach for the poultry industry. Embryonic thermal manipulation (TM) constitutes a promising strategy for sustainable broiler production. Our previous research has yielded significant insights into the impact of TM on embryonic thermotolerance, metabolism, post-hatch growth performance, microbial diversity, and immunity. Rumen-protected dry grape extract supplementation enhances milk production, behavior traits, and immunometabolism of mid-lactating Fleckvieh cows under naturally occurring heat stress.

Heat stress (HS) in dairy cows disrupts homeostasis and thermoregulation, negatively affecting milk production, health status, and metabolism. Dietary supplementation with phytoextracts such as polyphenols may offer an effective dietary strategy by stimulating the immune system, reducing oxidative stress, and ultimately enhancing welfare, metabolic function, and milk performance. This study aimed to evaluate the effects of supplementing mid-lactating Fleckvieh cows with rumen-protected grape extract (Nor-Grape BP-O, Nor-Feed) on milk performance, rectal temperature (RT), behavior, immunometabo

Unraveling the cryptic functions of mitogen-activated protein kinases Cpk2 and Mpk2 in Cryptococcus neoformans.

Mitogen-activated protein kinase (MAPK) pathways are fundamental to the regulation of biological processes in eukaryotic organisms. The basidiomycete Cryptococcus neoformans, known for causing fungal meningitis worldwide, possesses five MAPKs. Among these, Cpk1, Hog1, and Mpk1 have established roles in sexual reproduction, stress responses, and cell wall integrity. However, the roles of Cpk2 and Mpk2 are less understood. Our study elucidates the functional interplay between the Cpk1/Cpk2 and Mpk1/Mpk2 MAPK pathways in C. neoformans. We discovered that CPK2 overexpression compensates for cpk1Δ

Post Junctional Sudomotor and Cutaneous Vascular Responses in Noninjured Skin Following Heat Acclimation in Burn Survivors.

Thermal tolerance is improved in burn survivors following 7 days of exercise heat acclimation. It is unknown whether post junctional sudomotor and/or cutaneous vascular adaptations in noninjured skin contribute to this improvement. Thirty-three burn survivors were stratified into moderately (17-40% BSA grafted, n = 19) and highly (>40% BSA grafted, n = 14) skin-grafted groups. Nine nonburned subjects served as controls. All subjects underwent a 7-day heat acclimation protocol, which improved thermal tolerance in all groups. Before and after this heat acclimation protocol, post junctional cutan

Novel halotolerant fungal endophytes from hypersaline coastal ecosystems as sustainable biostimulants for early growth performance of tomato and wheat under saline stress.

Soil salinization severely restricts seed germination, seedling establishment, and vegetative growth in key crops such as tomato (Solanum lycopersicum L.) and wheat (Triticum aestivum L.). Endophytic fungi from extreme environments are promising biostimulants to alleviate salinity stress, yet systematic assessments of isolates from hypersaline coastal ecosystems remain scarce. Here, we evaluated 28 endophytic fungal isolates from saline coastal ecosystems of the southeastern Iberian Peninsula using a four-stage pipeline: (i) in vitro characterization of salinity and temperature tolerance, (ii)

The ecology of the Zygomycetes and its impact on environmental exposure.

Zygomycetes are unique among filamentous fungi in their great ability to infect a broader, more heterogeneous population of human hosts than other opportunistic moulds. Various members of the Zygomycetes have been implicated in zygomycosis, although those belonging to the family Mucoraceae are isolated more frequently than those of any other family. The environmental microbiology literature provides limited insights into how common zygomycetes are in the environment, and provides a few clues about which ecological niches these fungi are found in. Mucorales are thermotolerant moulds that are su

Effectiveness of community-based delivery of mass dog vaccination to prevent rabies: A cluster randomized controlled trial.

BACKGROUND: Approximately 60,000 people die from rabies annually, mostly in Africa and Asia. Mass dog vaccination is critical for elimination. Herd immunity has been calculated to be attained when >40% (critical threshold) of dogs are vaccinated. A common delivery method implemented in Tanzania and other countries typically involves non-local teams travelling to villages implementing dog vaccination annually through static-point clinics using cold chain stored vaccines (Team-based delivery). Determination that a canine rabies vaccine is thermotolerant enables novel Community-based dog vaccinat

The chicken or the egg? Exploring bi-directional associations between Newcastle disease vaccination and village chicken flock size in rural Tanzania.

Newcastle disease (ND) is a viral disease of poultry with global importance, responsible for the loss of a potential source of household nutrition and economic livelihood in many low-income food-deficit countries. Periodic outbreaks of this endemic disease result in high mortality amongst free-ranging chicken flocks and may serve as a disincentive for rural households to invest time or resources in poultry-keeping. Sustainable ND control can be achieved through vaccination using a thermotolerant vaccine administered via eyedrop by trained "community vaccinators". This article evaluates the upt

Effects of thermal stress on tumor antigenicity and recognition by immune effector cells.

The primary rationale for the application of clinical hyperthermia in the therapy of cancer is based on the direct cytotoxic effect of heat and the radio-chemosensitization of tumor cells. More recently, additional attention is given to the observation that heat and heat-shock proteins can activate the host's immune system. The expression of heat-shock genes and proteins provides an adaptive mechanism for stress tolerance, allowing cells to survive non-physiologic conditions. However, the same adaptive mechanism can ultimately favor malignant transformation by interfering with pathways that re

Adenylyl Cyclase and Protein Kinase A Play Redundant and Distinct Roles in Growth, Differentiation, Antifungal Drug Resistance, and Pathogenicity of Candida auris.

Candida auris is a globally emerging multidrug-resistant fungal pathogen. Its pathogenicity-related signaling networks are largely unknown. Here, we characterized the pathobiological functions of the cyclic AMP (cAMP)/protein kinase A (PKA) signaling pathway in C. auris. We focused on adenylyl cyclase (CYR1), the PKA regulatory subunit (BCY1), and the PKA catalytic subunits (TPK1 and TPK2). We concluded that PKA acts both dependently and independently of Cyr1 in C. auris. Tpk1 and Tpk2 have major and minor roles, respectively, in PKA activity and functions. Both Cyr1 and PKA promote growth, th

Temperature during Aspergillus fumigatus conidiophore development primes spore transcriptome for asexual, parasexual or sexual development.

UNLABELLED: Aspergillus fumigatus is a thermotolerant saprobe found in soils and plant debris worldwide and an important pathogen of humans causing two million deaths annually. A. fumigatus makes abundant asexual spores (conidia) which are widely distributed by wind and can be inhaled from the environment. In susceptible individuals inhaled conidia break dormancy, germinate and grow in the lung leading to serious disease. Recent work has shown that conidia made at 37°C and 50°C have different morphologies and germination kinetics. While the asexual cycle is well-characterized at 37°C, much les
