Chronic TLR7 activation promotes expansion of activated CD11c+ double-negative B cells and autoreactive humoral responses in lupus-like autoimmunity.

Age-associated B cells (ABCs) have emerged as a distinct B cell population associated with aging, chronic inflammation, and autoimmunity. Notably, the expansion of ABC-like CD11c-expressing B cells has been observed in patients with systemic lupus erythematosus and in experimental models of lupus-like disease, suggesting their potential involvement in disease pathogenesis. Although their relevance in autoimmunity is increasingly appreciated, the identity and functional contribution of CD11c+ B cell populations arising during chronic Toll-like receptor 7 (TLR7)-driven immune activation remain i

Halo Nevus as a Self-Limited Model of Melanocyte Autoimmunity: Bridging Vitiligo, Immune Resolution, and Tumor Immunology-A Narrative Review.

Halo nevus (HN), also known as Sutton nevus or leukoderma acquisitum centrifugum, is a melanocytic lesion characterized by a depigmented halo surrounding a central nevus. Although HN is a benign dermatological condition, increasing evidence indicates that HN represents a dynamic in vivo model of melanocyte-directed immune response with relevant implications for autoimmunity, pigmentary disorders, melanoma regression, and tumor immunosurveillance. The pathogenesis is primarily mediated by CD8+ cytotoxic T lymphocytes via interferon-γ-driven pathways, leading to targeted destruction of melanocyt

The Impact of Thyroid Autoimmunity, Glucose Metabolism, and Vitamin D Status in Women With Recurrent Pregnancy Loss: A Retrospective Study.

INTRODUCTION: The influence of thyroid autoimmunity (TAI) on obstetrical and fetal outcomes is debated. Specifically, the impact of thyroid antibodies on the risk of recurrent pregnancy loss (RPL) remains controversial. Similarly, the relationship between glucose metabolism and vitamin D (vitD) status in RPL women needs clarification. METHODS: We evaluated 301 non-pregnant women (mean age 37 ± 4.8 SD) with a previous history of primary RPL (≥ 2 spontaneous pregnancy losses < 20 weeks of gestation) referred to our clinic over a period of 5 years (from 2020 to 2025). RESULTS: We did not find s

Association of 25(OH)D status with calcium metabolism, inflammation, and thyroid autoimmunity in patients with type 2 diabetes mellitus.

INTRODUCTION: 25-hydroxyvitamin D [25(OH)D] deficiency is highly prevalent in type 2 diabetes mellitus (T2DM) and may contribute to immune and metabolic dysfunction. This study aimed to investigate the clinical associations of serum 25(OH)D levels with calcium metabolism, inflammation, thyroid function, and autoimmunity in patients with T2DM. METHODS: This cross-sectional study enrolled 503 patients with T2DM, stratified into four groups based on 25(OH)D levels: severe deficiency (<10 ng/mL), moderate deficiency (10-20 ng/mL), insufficiency (20-30 ng/mL), and sufficiency (>30 ng/mL). Pyoderma gangrenosum as a manifestation of multisystem autoimmunity in a patient with giant cell arteritis, optic neuritis and rheumatoid arthritis.

Pyoderma gangrenosum (PG) is a rare neutrophilic dermatosis typically associated with autoimmune disease; however, its coexistence with giant cell arteritis (GCA) and optic neuritis is exceedingly uncommon and rarely reported. We describe a woman in her early 70s with rheumatoid arthritis and recently diagnosed GCA who developed painful breast ulcers while on tapering systemic corticosteroids for GCA, shortly after a herpes zoster infection. The ulcers showed classic PG morphology and fulfilled Delphi diagnostic criteria. She had preceding MRI-confirmed bilateral optic neuritis followed by bil

Post-transplant autoimmunity in a child: a lupus-like syndrome emerging in the context of chronic graft-versus-host disease.

BACKGROUND: Autoimmune manifestations are increasingly recognized as late complications following hematopoietic stem cell transplantation (HSCT), particularly in association with chronic graft-versus-host disease (cGvHD). However, the occurrence of systemic lupus erythematosus (SLE)-like features remains extremely rare, especially in pediatric patients. Understanding the mechanisms underlying such manifestations, including the role of mixed chimerism, is important for early recognition and management. CASE PRESENTATION: We report the case of a girl with thalassemia major who underwent HSCT fr

Exploring the role of APRIL in autoimmunity: implications for therapeutic targeting in systemic lupus erythematosus, rheumatoid arthritis, and Sjögren's syndrome.

Autoimmunity arises when the immune system erroneously attacks self-antigens, potentially resulting in organ dysfunction. This review focuses on the proliferation-inducing ligand, APRIL, and its critical role in regulating antibody-producing B cells. We explore the implications of APRIL in autoimmune diseases such as systemic lupus erythematosus, rheumatoid arthritis, and Sjögren's syndrome. Emerging evidence indicates that APRIL may modulate autoimmune pathology and influence B cell survival, particularly through its interactions with receptors like B-cell maturation antigen (BCMA) and transm

Thyroid autoimmunity at onset of type 1 diabetes as a predictor of thyroid dysfunction: a thirty-years retrospective longitudinal study.

BACKGROUND: Thyroid autoimmunity commonly coexists with type 1 diabetes due to shared autoimmune mechanisms, and early recognition of thyroid dysfunction is crucial for optimizing metabolic control. However, there is no consensus regarding the optimal screening strategy for detecting thyroid disease in patients with type 1 diabetes. This study aimed to determine the long-term predictive value of thyroid peroxidase antibodies (TPO-Abs) at the onset of type 1 diabetes for the development of thyroid dysfunction and to evaluate the influence of age at diabetes onset. METHODS: We conducted a retro

Dual Autoimmunity: A Case Report of the Sequential Development of Systemic Lupus Erythematosus in a Patient With Anti-MDA5 Dermatomyositis.

Dermatomyositis (DM) and systemic lupus erythematosus (SLE) are chronic rheumatic diseases that can affect multiple organ systems. Both conditions share several similarities, including pathogenic mechanisms, clinical manifestations, and pharmacological treatments. However, the coexistence of DM and SLE is rarely encountered in clinical practice. Here, we present the case of a 42-year-old woman who developed DM, characterized by proximal muscle weakness in the upper and lower extremities, dysphagia, heliotrope rash, periungual erythema, erythematous skin lesions on the neck and arms, elevated s

Zinc and Ferritin Levels and Their Associations with Functional Disorders and/or Thyroid Autoimmunity: A Population-Based Case-Control Study.

Population zinc and iron status appear to be associated with an increased risk of thyroid function abnormalities and thyroid autoimmunity (AITD). In the present study, we aimed to determine whether zinc and/or iron levels (assessed by ferritin levels) were associated with the presence of AITD and with alterations in thyroid function. A population-based case-control study (n = 1048) was conducted (cases: n = 524; controls: n = 524). Participants were measured for blood concentrations of zinc and ferritin, TSH, FT4, FT3, and thyroid autoantibodies. No significant differences were found in relati

An Oxylipin-Related Nutrient Pattern and Risk of Type 1 Diabetes in the Diabetes Autoimmunity Study in the Young (DAISY).

Oxylipins, pro-inflammatory and pro-resolving lipid mediators, are associated with the risk of type 1 diabetes (T1D) and may be influenced by diet. This study aimed to develop a nutrient pattern related to oxylipin profiles and test their associations with the risk of T1D among youth. The nutrient patterns were developed with a reduced rank regression in a nested case-control study (n = 335) within the Diabetes Autoimmunity Study in the Young (DAISY), a longitudinal cohort of children at risk of T1D. The oxylipin profiles (adjusted for genetic predictors) were the response variables. The nutri

The X-linked histone demethylases KDM5C and KDM6A as regulators of T cell-driven autoimmunity in the central nervous system.

T cell-driven autoimmune responses are subject to striking sex-dependent effects. While the contributions of sex hormones are well-understood, those of sex chromosomes are meeting with increased appreciation. Here, we outline what is known about the contribution of sex chromosome-linked factors to experimental autoimmune encephalomyelitis (EAE), a mouse model that recapitulates many of the T cell-driven mechanisms of multiple sclerosis (MS) pathology. Particular attention is paid to the KDM family of histone demethylases, several of which - KDM5C, KDM5D and KDM6A - are sex chromosome encoded.

Optimal combination of arsenic trioxide and copper ions to prevent autoimmunity in a murine HOCl-induced model of systemic sclerosis.

INTRODUCTION: Systemic sclerosis (SSc) is a rare chronic autoimmune disease characterized by diffuse fibrosis of the skin and internal organs and vascular abnormalities. The etiology and physiopathology are complex due to the heterogeneity of its overall clinical presentation. Arsenic trioxide (ATO) has been proven to be effective against SSc, sclerodermatous Graft-versus-Host Disease, multiple sclerosis, Crohn's disease or systemic lupus erythematosus animal models and has demonstrated promising effects in human clinical trials. Its efficacy was shown to be related at least in part to the gen
