Identification of a Tenochondral Ossification Sequence Producing Heterotopic Ossification in Human Achilles Tendinopathy.

Achilles tendinopathy is a common musculoskeletal disorder. A frequent late-stage manifestation is tendon heterotopic ossification (HO). Achilles HO forms predominantly via endochondral ossification, yet the cellular and molecular mechanisms underlying this pathologic condition remain elusive. Here, we analyzed surgically excised biopsy samples with symptomatic human Achilles tendons alongside non-diseased Achilles tendon controls primarily from lower-limb amputation donors to characterize the morphologic and mineralization patterns of the disease. Based on these analyses, we propose a stepwis

Pore size of gelatin/hyaluronic acid supramolecular hydrogel modulates the immune functions of bone marrow-derived immature dendritic cells.

The extracellular matrix-mimetic performance of hydrogel micropores makes them promising delivery carriers for dendritic cell (DC) vaccines. Nevertheless, the reported hydrogel failed to decouple the intricate relationship between pore size and mechanical performance, thus the regulatory effect of pore size on DCs immune function remains inadequately understood. In this study, we constructed a supramolecular hydrogel system in which pore size was decoupled from mechanical performance using a sacrificial gelatin microgel template and synergetic photopolymerization. The effects of hydrogel pore

NCOA4 Accelerates Abdominal Aortic Aneurysm Formation by Regulating Macrophage Activation and Ferroptosis via STAT1-CH25H Axis.

Abdominal aortic aneurysm (AAA) is a life-threatening condition lacking effective pharmacological treatments, with emerging evidence highlighting the fundamental role of ferroptosis in its progression. Nuclear receptor coactivator 4 (NCOA4), known to mediate ferritinophagy and contribute to ferroptosis, is upregulated in macrophages from both angiotensin II (Ang II)-induced murine AAA models and human AAA tissues. Global deletion of Ncoa4 in mice suppressed Ang II-induced AAA formation. Combined approaches using bone marrow transplantation and macrophage-specific adeno-associated virus (AAV) c

Development and Characterization of a Dual Nanofibrillar Membrane of Polylactic Acid Loaded with Hydroxyapatite and Chlorhexidine for Guided Bone Regeneration.

Guided bone regeneration requires membranes that combine structural stability, biocompatibility, and bioactive properties. This study aimed to develop a bilayer electrospun polylactic acid (PLA) membrane incorporating nanohydroxyapatite (nano-HA) and chlorhexidine (CHX). Membranes were fabricated by electrospinning at 15 kV and divided into four groups: PLA 10% (control), PLA 10%/CHX 0.2%, PLA 10%/nano-HA 10%, and a bilayer combining the CHX- and nano-HA-loaded formulations in separate monolayers, creating a bilayer nanofibrillar scaffold. We characterized microscopic structure, thermal behavi

Whey protein phospholipid concentrate at the interface of function and health: Heat-induced protein interactions, bioactivity, and choline delivery.

Milk is a nutrient-packed biological fluid essential for early development, with the milk fat globule membrane (MFGM) playing a fundamental role in cognitive, bone, immune, and gastrointestinal health. Driven by increasing demand for high-protein and sustainable ingredients, the dairy industry has focused on coproducts such as whey protein phospholipid concentrate (WPPC), a microfiltration-derived fraction enriched in MFGM proteins and polar lipids. WPPC contains elevated levels of caseins and whey proteins and is notably enriched in membrane-bound glycoproteins and phospholipids that contribu

Exosome-camouflaged chitosan/zinc oxide/carbon quantum dot nanocarriers for pH-responsive doxorubicin delivery in breast cancer treatment.

Doxorubicin (DOX) chemotherapy for breast cancer is constrained by systemic toxicity and limited tumor selectivity, underscoring the need for polysaccharide-based carriers in which pH responsiveness arises from intrinsic polymer properties rather than chemical modification. In this study, a carbohydrate-centered biomimetic nanocarrier was developed using chitosan as the primary structural and pH-responsive matrix, integrating zinc oxide nanoparticles (ZnO) and carbon quantum dots (CQDs) through a W1/O/W2 double-emulsion process, followed by surface association with bone marrow-derived mesenchy

Macrophages expressing macrophage receptor with collagen structure attenuate liver fibrosis through a tissue restoration phenotype.

Liver macrophages are central in maintaining hepatic homeostasis and mediating immune responses during liver injury, including fibrosis. Macrophages may have proinflammatory or antiinflammatory properties, but which properties influence fibrosis remains unclear. To explore the role of macrophages in liver fibrosis, we performed single-cell RNA-seq in a mouse model of liver injury and found that macrophage diversity was increased. Marco was among the most significantly upregulated genes, and a population of Marcohi macrophages increased with injury and spatially segregated to nonfibrotic areas.

Bioengineered Lymphatic Vessels in Synthetic Matrices to Study Breast Cancer Cell Functions.

Lymphatic vessels play an important role in pathologies, such as cardiovascular diseases or metastasizing cancers. However, mechanisms involved in cancer metastasis through lymphatic vessels remain largely unknown. In this study, we engineer 3D lymphatic microvessels by co-culturing human lymphatic endothelial cells (hLECs) with human bone marrow-derived mesenchymal stromal cells (hBM-MSCs) in synthetic and fully defined enzymatically crosslinked poly(ethylene glycol) (TG-PEG) hydrogels. By tuning the stiffness of the TG-PEG scaffolds, varying seeding densities of hLECs and hBM-MSCs, and stimu

Plant-Derived Bioactives in Tendon and Enthesis Biology: An Evidence-Tiered Narrative Review.

Tendon injury, tendon-bone interface disruption, and rotator cuff pathology involve inflammatory signaling, oxidative stress, extracellular matrix turnover, cellular senescence, and impaired differentiation. Plant-derived bioactive compounds, including curcumin, quercetin, and other flavonoids and botanical formulations, have been investigated across these pathways in preclinical tendon and enthesis models, but interpretation is complicated by heterogeneous models, formulations, delivery platforms, and endpoints. This review distinguishes preclinical and formulation-specific tissue-response si

FANCD2 promotes wound healing through DNMT1.

Fanconi anemia (FA) is primarily an autosomal recessive genetic disorder that leads to bone marrow failure, increased risk of developing cancer, and a plethora of developmental abnormalities. Patients are prone to recurrent infections and increased risk of hemorrhage, as well as delayed wound healing with poor results. FA is caused by a genetic mutation in the proteins needed for FA pathway activation; FA group D2 protein (FANCD2) is an indispensable part of this pathway and plays essential roles in some aspects of cellular life, especially in the cellular responses to DNA damage. Here, we fou

Urine-Derived Stem Cells: Challenges in Isolation, Biological Identity, and Therapeutic Potential in CKD-Associated Fibrosis.

Urine-derived stem cells (UDSCs) have emerged as a promising cell source for regenerative medicine due to their non-invasive procurement, high proliferative capacity, and potential relevance to kidney-specific repair. Unlike conventional mesenchymal stem cells (MSCs) obtained from bone marrow or adipose tissue, UDSCs originate from multiple regions of the urinary tract and exhibit a unique biological profile that combines MSC characteristics with features of renal progenitor populations. This review provides a comprehensive overview of the current understanding of UDSC biology, including their

Epigenetic-metabolic coupling by SOX3/WDR5-SIRT5 signaling mediates the therapeutic effects of TGFβ-targeted exosomal nanoparticles in osteoarthritis.

The development of targeted nanotherapeutics capable of restoring chondrocyte homeostasis represents a major challenge in osteoarthritis (OA) treatment. In this study, we engineered a transforming growth factor-β (TGF-β) receptor-targeted exosome-nanoparticle hybrid system (TGFβ-Exo-NP) derived from chondrocytes and systematically evaluated its therapeutic efficacy and molecular mechanism. TGFβ-Exo-NP displayed uniform nanoscale morphology, favorable surface charge, and controlled release properties, with robust chondrocyte-specific uptake mediated by transforming growth factor-β receptor (TGF

Natural extract-derived compounds for the treatment of hematological diseases through modulation of the bone marrow microenvironment.

The bone marrow microenvironment comprises a complex network of hematopoietic stem cells, immune cells, stromal cells, and non-cellular components such as the extracellular matrix and soluble factors, collectively maintaining the homeostasis of hematopoietic stem and progenitor cells. It is highly vulnerable to pathological perturbations induced by hematologic malignancies, solid tumors, inflammatory stress, and therapeutic exposure, which collectively destabilize microenvironmental homeostasis and promote hematopoietic failure, malignant progression, and immune dysregulation. Natural products

OMICS Profiling Identifies Signatures of Senescence in Osteogenesis Imperfecta Osteoblasts Counteracted by 4-PBA.

Mutations in collagen I are the most common cause of osteogenesis imperfecta (OI), leading to delayed protein folding and structurally abnormal molecules. While some aberrant collagen is secreted into the extracellular matrix (ECM), impairing bone quality, a significant fraction is retained intracellularly, disrupting osteoblast homeostasis. 4-phenylbutyrate (4-PBA) has been shown to improve osteoblast function and ECM composition in OI models. To investigate the intracellular consequences of mutant collagen retention and the mechanisms of 4-PBA, we analysed the secretome and transcriptome of

FOS3D: A Fluorescence-Enabled Toolkit for Characterizing a Three-dimensional Osteosarcoma Model.

Osteosarcoma (OS) remains a rare, aggressive primary bone malignancy for which therapeutic progress has been limited for several decades. Three-dimensional (3D) tissue-engineered models can better reproduce the tumor microenvironment than monolayer cultures; however, their routine use is often constrained by low-throughput workflows and reliance on destructive analytical endpoints. Here, we present FOS3D, a fluorescence-enabled OS model based on gelatin methacryloyl hydrogels incorporating stable green fluorescent protein (GFP)-expressing tumor cells. Exploring Injectable Scaffolded Spheroids for Nucleus Pulposus Therapy in Degenerated Intervertebral Discs.

Cell-based therapies for intervertebral disc degeneration (IVDD) treatment face significant challenges, including cell damage from injection-induced shear stress and poor survival in the harsh, nutrient-depleted microenvironment of the intervertebral disc. To overcome these challenges, we developed scaffolded spheroids (S-SPH) by integrating human bone marrow-derived mesenchymal stem cell (hBMSC) spheroids (SPH) into microscaffolds (MS) produced via high-resolution 3D printing, thereby forming injectable tissue-building blocks. We optimized cell seeding density (∼2000 cells/spheroid) and MS fa

The chitosan collagen salvianolic membrane mitigates intervertebral disc degeneration through suppression of the Wnt/β-catenin signaling.

Intervertebral disc degeneration (IVDD), a natural aging process, leads to intervertebral disc pain, with lumbar resection providing pain relief but potential postoperative complications affecting outcomes. The chitosan-collagen-salvianolic (CCS) membrane has shown promise in reducing scar formation after lumbar resection, though the mechanisms remain unclear. A lumbar laminectomy model was used, with materials applied to the surgical site. Tissue samples were collected at 4 and 8 wk post-surgery for histological evaluation to assess pathological changes and apoptosis. Western blot analysis ex

Plasma Proteomics Reveals Persistent and Surgery-Responsive Molecular Signatures in Osteoarthritis Patients.

Osteoarthritis (OA) represents a degenerative joint disease which advances through cartilage breakdown, synovial inflammation, and subchondral bone transformation until it causes persistent pain and mobility loss. The scientific community lacks complete knowledge about OA disease mechanisms and post-operative healing processes despite arthroplasty surgery providing effective symptom relief. This study investigated plasma proteomic changes in OA patients before and after arthroplasty. The cohort included eight OA patients undergoing knee or hip arthroplasty and ten age-, sex-, and body mass ind
