Mechanochemical Approach to a Monocationic Asymmetric Monomethine Cyanine Dye for Nucleic Acid Analysis and Visualization.

Using an environmentally friendly approach, we successfully synthesized an asymmetric monomethine cyanine dye, 7-chloro-1-ethyl-4-((3-ethylbenzo[d]thiazol-2(3H)-ylidene)methyl) quinolin-1-ium iodide, named CHLoris (CHL), via a modified Knoevenagel-type condensation. The reaction was carried out mechanochemically in an ethanol-water medium using 1-ethyl-2-methylbenzothiazolium iodide and 4,7-dichloro-1-ethylquinolin-1-ium iodide in the presence of sodium carbonate as a base and catalytic amounts of Hünig's base. The UV/VIS absorption spectra of CHL in both the buffer solution and ethanol reveal

Leveraging Carbon Quantum Dot Spacers as Dual Proton/Electron Boosters to Upgrade TiO2@Polydopamine Photocatalysts for pH-Resilient Conversion of O2 into H2O2.

Photocatalytic synthesis of H2O2 is a proton-coupled electron transfer (PCET) process, which is generally jeopardized by the kinetic mismatch between photogenerated electron transfer and proton supply. To address the challenge, here we proposed a new core-shell design of nanocomposite catalysts comprising of carbon quantum dot (CD)-topped TiO2 nanoparticles encapsulated by polydopamine (PDA) shells, which delivered stable catalytic activity across a pH range of 1-9, exhibiting a photocatalytic generation rate of H2O2 that reached 18.14 mmol g-1 h-1 in methanol and 8.66 mmol g-1 h-1 in water. Tissue Mild Extraction Enriches Membrane, Cytoplasmic, and Secreted Proteomes for Biomarker Discovery.

Clinical blood biomarkers provide important diagnostic information and are generally secreted, cytoplasmic, membrane, and other soluble proteins. However, in the mass spectrometry-based proteomic biomarker discovery phase, the whole tissue is often lysed directly for analysis, and this increases protein complexity in the sample, limiting the detection of soluble proteins as biomarkers. Here, we use a mild extraction method that includes a low salt buffer and 1% Triton X-100 to isolate proteins from mouse tissues followed by proteomic analyses. This mild condition extracts membrane, cytoplasmic

Multiple Levels of Organization in Amphiphilic Diblock Copolymers Based on Poly(γ-benzyl-l-glutamate) Produced by Aqueous ROPISA.

A recent method for producing amphiphilic block copolymers and nano-objects based on the ring-opening polymerization-induced self-assembly (ROPISA) in aqueous buffer is explored with respect to the tunability toward nanostructures. ROPISA gives rise to polypeptide copolymers with unprecedented levels of organization. By employing amphiphilic block copolymers of poly(ethylene glycol) (PEG) with the synthetic polypeptide poly(γ-benzyl-l-glutamate) (PBLG) and a combination of static (13C NMR, X-ray scattering, polarizing optical microscopy), thermodynamic (differential scanning calorimetry), and

An Immune Assay to Quantify the Neutralization of Oxidation-Specific Epitopes by Human Blood Plasma.

Oxidized phospholipids (OxPLs) are increasingly recognized as biologically active lipids involved in various pathologies. Both exposure to pathogenic factors and the efficacy of protective mechanisms are critical to disease development. In this study, we characterized an immunoassay that quantified the total capacity of the plasma to degrade or mask OxPLs, thereby preventing their interaction with cells and soluble proteins. OxLDL-coated plates were first incubated with human blood plasma or a control vehicle, followed by an ELISA using a monoclonal antibody specific to oxidized phosphatidylet

3D-printed dysphagia-friendly gels with tailored inulin-konjac glucomannan mixtures: Swallowing safety enhancement and controlled lycopene release.

This study proposed an innovative integration of dynamic digestion model with additive manufacturing technology to investigate how inulin (IN)/konjac glucomannan (KGM) ratios modulate myofibrillar protein (MP) gel networks for dysphagia management. The optimized IN/KGM fomulation (P8-1:2, IN/KGM = 1:2 (w/w)) promoted the formation of densely network structures. The P8-1:2 gel exhibited highest lycopene encapsulation efficiency (88.07 %) and excellent rheological properties while mantianing swallowing safety and 3D printability. During gastric digestion, KGM delayed matrix disintegration throug

The unusual formaldehyde-induced activation of [NiFe]-hydrogenase: Implications from protein film electrochemistry and infrared spectroscopy.

Here we investigate how formaldehyde (HCHO), a known strong inhibitor of [FeFe]‑hydrogenases and a mild inhibitor of [NiFe]‑hydrogenases, may exert more complex effects on this group of metalloenzymes, which reversibly catalyze the 2H+/H₂ reaction. We investigated the [NiFe]‑hydrogenase Hyd-2 from E. coli using protein film electrochemistry, a technique that enables the measurement of enzyme activity when the enzyme is adequately adsorbed on the electrode. The effect of HCHO on the electrocatalytic performance of Hyd-2 is highly dependent on the buffer pH and the direction of catalysis. During

The Orientation of Human Fibrinogen at Biomedically Relevant Polydimethylsiloxane-Water Interfaces.

The adsorption of fibrinogen on biomaterial surfaces, particularly polydimethylsiloxane (PDMS), plays a key role in foreign body reactions and has recently been shown to be one of the main factors driving catheter-associated urinary tract infections (CAUTIs). Yet, despite detailed studies on the fibrinogen's solution and crystal structures, its behavior at material interfaces is less understood. Using sum frequency generation (SFG) spectroscopy and structural modeling, we determined the binding pose and conformation of human fibrinogen at the PDMS-buffer interface. Fibrinogen adopts an upright

Overcoming PEG Antigenicity: Statistical PEG Isomers Reduce Antibody Binding.

Poly(ethylene glycol) (PEG) is widely used in bioconjugation and nanomedicine due to its advantageous properties including solubility, biocompatibility, and pharmacokinetic benefits. However, PEG exposure can elicit anti-PEG antibodies (APAs), resulting in accelerated blood clearance and immune responses. To address this, "randomized PEG" (rPEG) is synthesized by incorporating the comonomer glycidyl methyl ether into PEG via anionic ring-opening polymerization. This structural isomer introduces "synthetic point mutations", impeding APA recognition. The interaction between rPEGs and APAs is sys

Synthesis and Application of Pyrene-Based Fluorescent Probe for the Continuous Detection of Cu2+ and Picric Acid.

A novel pyrene-based fluorescent probe PYS was designed and synthesized for the sequential detection of Cu2+ and picric acid (PA) under identical experimental conditions. In ethanol-HEPES buffer (V/V = 8:2, pH = 7.4), Cu2+ was selectively detected by PYS through fluorescence enhancement with a binding constant of 7.5 × 104 L/mol and a detection limit of 9.3 × 10-8 mol/L, demonstrating high sensitivity and anti-interference capability. The resulting complex PYS-Cu2+ functioned as a probe for PA detection, exhibiting instantaneous response (2 s) and high selectivity superior to other nitroaromat

First Spectrofluorimetric Method for Sensitive Determination of Armodafinil in Dosage Forms and Human Plasma: Greenness, Blueness, and Whiteness Assessment.

A novel, sensitive, and reliable spectrofluorimetric method for the determination of a unique antinarcoleptic drug armodafinil in human plasma and dosage forms was developed for the first time. The proposed method is based on the formation of a highly yellow fluorescent derivative through the reaction of the studied drug with acetylacetone and formaldehyde in an acetate buffer (pH 4.5), yielding a strong emission at 485 nm following excitation at 415 nm. The experimental variables that affected the one-pot synthesis were investigated and optimized. The calibration curve demonstrated a linear r

Recent advances in mitochondrial transplantation to treat disease.

Mitochondrial transplantation (MT), an innovative regenerative technique widely used to treat diseases caused by mitochondrial dysfunction, shows great promise for clinical application. This procedure can increase the number of mitochondria and improve the function of damaged mitochondria, resulting in increased adenosine triphosphate levels, decreased reactive oxygen species production, improved Ca2+ buffering capacity, modulated inflammatory response, and reduced apoptosis to protect cells, thus promoting tissue repair. In this review, we describe research advances in MT over the last five y

Functional contribution of astrocytic Kir4.1 channels to spasticity after spinal cord injury.

Spasticity, a prevalent motor issue characterized by network hyperexcitability, causes pain and discomfort, with existing treatments offering limited relief. While past research has focused on neuronal factors, the role of astrocytes in spasticity has been overlooked. This study explores the potential of restoring astrocytic K+ uptake to reduce spasticity following spinal cord injury (SCI). Astrocytes buffer extracellular K+ via Kir4.1 channels, preventing neuronal hyperexcitability. Following SCI, Kir4.1 levels decrease at the injury site, though the consequences and mechanisms of this reduct

Dose-dependent CHCHD10 dysregulation dictates motor neuron disease severity and alters creatine metabolism.

Dominant defects in CHCHD10, a mitochondrial intermembrane space protein, lead to a range of neurological and muscle disease phenotypes including amyotrophic lateral sclerosis. Many patients present with spinal muscular atrophy Jokela type (SMAJ), which is caused by heterozygous p.G66V variant. While most disease variants lead to aggregation of CHCHD10 and activation of proteotoxic stress responses, the pathogenic mechanisms of the p.G66V variant are less clear. Here we report the first homozygous CHCHD10 patient, and show that the variant dosage dictates the severity of the motor neuron disea

A gut-on-a-chip incorporating human faecal samples and peristalsis predicts responses to immune checkpoint inhibitors for melanoma.

Patient responses to immune checkpoint inhibitors can be influenced by the gastrointestinal microbiome. Mouse models can be used to study microbiome-host crosstalk, yet their utility is constrained by substantial anatomical, functional, immunological and microbial differences between mice and humans. Here we show that a gut-on-a-chip system mimicking the architecture and functionality of the human intestine by including faecal microbiome and peristaltic-like movements recapitulates microbiome-host interactions and predicts responses to immune checkpoint inhibitors in patients with melanoma. Investigating the Interplay Between Mental Health Conditions and Social Connectedness on Suicide Risk: Findings from a Clinical Sample of Adolescents.

This study examined whether school and community connectedness buffer the relationships between mental health conditions and suicide risk in a clinical sample of adolescents with histories of substance use disorders. Data from 294 adolescents were examined, with approximately 58% reporting lifetime suicidal ideation and/or prior attempts. Multinomial logistic regression was used to examine main and interaction effects on a three-category measure of suicide risk. Depression severity and panic disorder were associated with elevated suicidal ideation risk, whereas disordered eating was associated

Colorimetric array sensor based on bimetallic nitrogen-doped carbon-based nanozyme material to detect multiple antioxidants.

Copper-cobalt bimetallic nitrogen-doped carbon-based nanoenzymatic materials (CuCo@NC) were synthesized using a one-step pyrolysis process. A three-channel colorimetric sensor array was constructed for the detection of seven antioxidants, including cysteine (Cys), uric acid (UA), tea polyphenols (TP), lysine (Lys), ascorbic acid (AA), glutathione (GSH), and dopamine (DA). CuCo@NC with peroxidase activity was used to catalyze the oxidation of TMB by H2O2 at three different ratios of metal sites. The ability of various antioxidants to reduce the oxidation products of TMB (ox TMB) varied, leading

The Potential of Artificial Cells Functioning under In Situ Deep-Sea Conditions.

Artificial cells with reconstructed cellular functions could serve as practical protocell models for studying the early cellular life on the Earth. Investigating the viability of protocell models in extreme environments where life may have arisen is important for advancing origin-of-life research. Here, we tested the survivability of lipid membrane vesicles in deep-sea environments. The vesicles were submerged in the deep-sea floor with a human-occupied vehicle. Although most of the vesicles were broken, some vesicles maintained a spherical shape after the dives. When a cell-free protein synth

Methylene Blue Solid Alginate Gels for Photodynamic Therapy: The Peculiarities of Production and Controlled Release of the Dye.

The purpose of this work is to establish the influence of the nature of solid alginate gels (alginic acid, AAG; calcium alginate, CAG) and the conditions of methylene blue (MB) introduction to alginate matrices upon its release into aqueous media. MB is an active photosensitizer, which is used in the photodynamic therapy of tumors and purulent wounds. Solid alginate gels based on AAG and CAG were obtained by adding hydrochloric acid and calcium chloride to sodium alginate. The dye was introduced into the matrix either at the stage of gelation or by immersing the gel in an aqueous solution of t
