Kidney Cell-Adapted Infectious Bronchitis ArkDPI Vaccine is Stable and Protective.

We previously demonstrated that adaptation of an embryo-attenuated infectious bronchitis virus (IBV) Arkansas (Ark) Delmarva Poultry Industry (DPI)-derived vaccine to chicken embryo kidney (CEK) cells (CEKp7) shifted the virus population towards homogeneity in spike (S) and nonstructural protein genes. Moreover, the typical Ark vaccine subpopulations emerging in chickens vaccinated with commercial Ark vaccines were not detected in chickens vaccinated with CEKp7, indicating that kidney-cell adaptation drastically increased the stability of the vaccine virus population in chickens. In the curren

Effects of T-2 Toxin on Turkey Herpesvirus-Induced Vaccinal Immunity Against Marek's Disease.

T-2 toxin, a very potent immunotoxic Type A trichothecene, is a secondary metabolite produced primarily by Fusarium spp., which grows on cereal grains and can lead to contaminated livestock feed. Repeated exposure to T-2 toxin has been shown to cause immunosuppression and decrease the resistance of exposed animals to a variety of infectious diseases; however, the effects of T-2 toxin on Marek's disease (MD) vaccinal immunity have not been reported. Four trials were conducted to determine the effects of T-2 toxin on vaccinal immunity against MD. Day-old, white leghorn chicks of Avian Disease an

Biological activity of glycosides of vitamin D3 and 1 alpha-hydroxyvitamin D3.

For the first time, the biological activity of the newly synthesized glycosides of vitamin D3 and of 1 alpha(OH)D3 has been studied in rats, chickens, and Japanese quails. Glucosidation did not reduce the activity of vitamin D3. In chickens, vitamin D3 glucoside has an activity of 90% to 95% as compared to the aglycon. In rats vitamin D3 glucoside is equally active when given by intravenous or oral route. Glycosides of 1 alpha (OH)D3, however, show a lower activity in rats, chickens, and quails. 1 alpha (OH)D3 glucoside has about 10% activity of the free 1 alpha (OH)D3, while 1 alpha (OH)D3 ce

Development and Evaluation of the Biological Adjuvant Bivalent Vaccine for Preventing Newcastle Disease and Infectious Bursal Disease.

Newcastle disease (ND) and infectious bursal disease (IBD) are both one of the most economically important infectious diseases, which cause high infection rate and mortality of chickens and restrict the development of the poultry industry. Currently, vaccination is the most effective method to prevent ND and IBD. In this study, the biological adjuvant bivalent vaccine (rClone30-VP2L-chGM-CSF) was developed for preventing ND and IBD. Furthermore, the immune and protective effects of rClone30-VP2L-chGM-CSF were evaluated in specific-pathogen-free chickens. The experimental results indicated that

Transcriptome-based biomarker gene screening and evaluation of the extracellular fatty acid-binding protein (Ex-FABP) on immune and angiogenesis-related genes in chicken erythrocytes of tibial dyschondroplasia.

BACKGROUND: Tibial dyschondroplasia (TD) is a bone disorder in which dead chondrocytes accumulate as a result of apoptosis and non-vascularization in the tibial bone of broiler chickens. The pathogenicity of TD is under extensive research but is yet not fully understood. Several studies have linked it to apoptosis and non-vascularization in the tibial growth plate (GP). We conceived the idea to find the differentially expressed genes (DEGs) in chicken erythrocytes which vary in expression over time using a likelihood-ratio test (LRT). Thiram was used to induce TD in chickens, and then injected

Persistence of Toxoplasma gondii tissue stages in poultry over a conventional fattening cycle.

Toxoplasma gondii is a widely spread protozoon in humans, mammals and poultry. Regarding the latter, nothing is known yet about the duration of T. gondii persistence and distribution over a conventional fattening cycle of turkeys and chickens. Twenty-four turkeys and 12 broiler chickens were infected intravenously with 1×10(6) T. gondii tachyzoites (strain NED). Serum antibody levels were determined weekly by ELISA (turkeys) or immunofluorescent antibody test (chickens). Turkeys were slaughtered at 4, 8, 12 and 16 weeks post-infection (p.i.), and chickens 5 or 10 weeks p.i. (n = 6 per group).

Development of a convenient HPLC-UV screening method for six antibiotic residues in chicken meat samples: model-driven gradient optimization and sorbent-free extraction with SALLME preconcentration.

The presence of antibiotic residues in poultry meat poses a growing risk of non-compliance with maximum residue limits under international food safety regulations. However, existing screening methods to detect antibiotic overuse rely mainly on LC-MS with dispersive solid-phase extraction (dSPE) clean-up using instrumentation and sorbents that are economically inaccessible to many food-safety laboratories in resource-limited countries where such monitoring is most needed. This work develops and validates a sorbent-free, HPLC-UV multiclass screening procedure for six antibiotic residues (sulfadi

Respiratory and hematological physiology of day 15 chicken embryos (Gallus gallus domesticus) during water submergence and air recovery: Implications for bird embryos experiencing nest inundation.

Bird nests of coastal or inland breeding birds can temporarily flood during high tides or storms. However, respiratory physiological disruption of such water submersion and implications for post-submergence survival are poorly understood. We hypothesized that respiratory physiological disturbances caused by submersion would be rapidly corrected following return to normal gas exchange across the eggshell, thus explaining survival of nest inundation in the field. We further hypothesized that the chicken embryo prior to hatching will develop the ability to recover from acid-base disturbance. We e

A strain of avian coronavirus of the GVII lineage capable of infecting African green monkey kidney cell lines (Vero) and chicken embryo fibroblast cell lines (DF-1).

The infectious bronchitis virus (IBV) belongs to the γ-coronavirus family and posing a serious threat to poultry health. Here, a single IBV strain, designated as IBV-250607GXCX, was isolated and identified from a poultry farm. S1 gene typing classified 250607GXCX as belonging to the genotype GVII-1 of IBV. Nucleotide sequence similarity and phylogenetic analyses revealed that 250607GXCX shares high sequence similarity and a close evolutionary relationship with I0636/16, suggesting that 250607GXCX may be the result of the long-term evolution of the parental strain I0636/16 in the Guangxi region

Genomic Regions and Candidate Genes Affecting Response to Heat Stress with Newcastle Virus Infection in Commercial Layer Chicks Using Chicken 600K Single Nucleotide Polymorphism Array.

Heat stress results in significant economic losses to the poultry industry. Genetics plays an important role in chickens adapting to the warm environment. Physiological parameters such as hematochemical parameters change in response to heat stress in chickens. To explore the genetics of heat stress resilience in chickens, a genome-wide association study (GWAS) was conducted using Hy-Line Brown layer chicks subjected to either high ambient temperature or combined high temperature and Newcastle disease virus infection. Hematochemical parameters were measured during three treatment phases: acute

Production and evaluation of monovalent anti-snake immunoglobulins from chicken egg yolk using Ghanaian puff adder (Bitis arietans) Venom: Isolation, purification, and neutralization efficacy.

Snakebites are rampant in Ghana, especially among the farmers, herdsmen, military recruits, hunters, and rural dwellers, and the antisnake venoms (ASV) use to treat these bites are not locally produced but rather imported, which come with a high cost, lack of constant supply and low specificity. The study was therefore aimed at isolating, purifying, and evaluating the efficacy of monovalent ASV from chicken egg yolk using puff adder (Bitis arietans) venom from Ghana. The major pathophysiological properties of the venom and the efficacy of the locally produced ASV were evaluated. The results sh

Growth Hormone (GH) Enhances Endogenous Mechanisms of Neuroprotection and Neuroplasticity after Oxygen and Glucose Deprivation Injury (OGD) and Reoxygenation (OGD/R) in Chicken Hippocampal Cell Cultures.

As a classical growth promoter and metabolic regulator, growth hormone (GH) is involved in development of the central nervous system (CNS). This hormone might also act as a neurotrophin, since GH is able to induce neuroprotection, neurite growth, and synaptogenesis during the repair process that occurs in response to neural injury. After an ischemic insult, the neural tissue activates endogenous neuroprotective mechanisms regulated by local neurotrophins that promote tissue recovery. In this work, we investigated the neuroprotective effects of GH in cultured hippocampal neurons exposed to hypo
