Metabolic regulation of calcium homeostasis via SERCA succination.

Although calcium homeostasis is disrupted in metabolic diseases, its metabolic regulation remains unclear. Here, we identify a mechanism by which fumarate suppresses sarco/endoplasmic reticulum (ER) calcium ion-adenosine triphosphatase (SERCA) activity via succination of a conserved cysteine residue, impairing ER calcium uptake and promoting metabolic dysfunction in Drosophila. In mammalian cells, high glucose or fumarate inhibits SERCA activity and increases ER calcium release and cytosolic and mitochondrial calcium levels. Mechanistically, we show that fumarate covalently modifies SERCA2b at

Selenium bioenrichment and functional enhancement with antioxidants in Actinidia arguta juice by fermentation with selenium-enriched Tibetan kefir grains.

Selenium is an essential trace element with critical roles in antioxidant defense, immune regulation, and metabolic health, yet its intake is insufficient in many populations. This study investigated the effects of fermenting Actinidia arguta juice with selenium-enriched Tibetan kefir grains to enhance its nutritional, sensory and functional. Selenium-enriched Tibetan kefir grains fermentation not only increased total Se content but also promoted the biotransformation of inorganic selenium into bioavailable organic selenium, supporting effective dietary biofortification. The fermented juice sh

Identification of a Potent Pan-Coronaviral Main Protease Inhibitor.

The global impact of SARS-CoV-2 and the continued emergence of zoonotic coronaviruses underscore the urgent need for broad-spectrum antivirals for pandemic preparedness. Herein, we report AVI8122, a covalent pan-coronaviral inhibitor that targets 19 Mpros across the α, β, γ, and δ genera, encompassing bat, human, and other animal coronaviruses. AVI8122 exhibits low nanomolar potency and favorable pharmacokinetics in mice. Structural studies reveal that AVI8122 forms a covalent bond with the catalytic cysteine and maintains conserved interactions within the active sites of these Mpros. In cellu

Targeting Crimean-Congo hemorrhagic fever virus: computational and MD-driven discovery of Moringa oleifera peptides against the RdRp-embedded OTU protease.

Crimean-Congo Hemorrhagic Fever Virus (CCHFV) is a tick-borne pathogen causes hemorrhagic disease with high fatality rates, and no approved vaccines/antivirals currently exist. The viral L protein contains a unique ovarian tumor (OTU) protease domain that cleaves ubiquitin and ubiquitin-like modifiers from host proteins, thereby aiding immune evasion and viral replication. Targeting the OTU domain represents a promising strategy for therapeutic intervention. Peptides obtained from Moringa oleifera proteome that mimic the binding mode of the endogenous ubiquitin molecule. These peptides were co

In vitro screening of compounds for targeting gastric cancer with Y220C p53 mutation: a molecule combining zinc chelation and a Michael acceptor drives CDKN1 and BBC3 expression to restore a p53-dependent cytotoxicity.

Point mutations in p53 favour tumour aggressivity, particularly in gastric cancer (GC), and offer a target for small molecule-based anticancer treatments. This study focused on the p53-Y220C mutation, which causes p53 misfolding due to thermal instability associated with the creation of a pocket that may accommodate small molecules. This mutation also creates an additional free cysteine thiol group that may react with Michael acceptors. Using an integrated in silico and in vitro approach, four compounds (AG1, AG2, AG3, and RK349) were screened for potential reactivation of p53-Y220C in GC cell

A post-translational regulatory map of chronic antigen-driven human T cell dysfunction.

T cells exposed to persistent antigen in the context of chronic viral infections or cancer lose self-renewal and cytotoxic capacity. Several transcriptional, epigenetic, and metabolic drivers of this process have been identified. However, the post-transcriptional regulatory mechanisms influencing the proteome of dysfunctional T cells are not well understood. Here we present a time-resolved molecular landscape of human T cells during the development of chronic antigen-driven dysfunction. Persistent T cell receptor stimulation significantly remodeled the proteome, including changes in canonical

Caffeic Acid Modulates Protein Disulfide Isomerase-NLRP3 Inflammasome Signaling to Mitigate Inflammation in Acute Pneumonia.

Caffeic acid (CA) is a polyphenol found in various of plants and daily beverages including coffee. It possesses diverse biological effects, including anti-inflammatory properties. Acute pneumonia represents a widespread inflammatory process; however, whether CA can mitigate acute pneumonia and its specific molecular mechanisms remain elusive. Here, we have demonstrated the robust anti-inflammatory effect of CA both in vivo and in vitro. Additionally, protein disulfide isomerase (PDI) was identified as a potential target of CA via activity-based protein profiling strategy coupled with a chemica

Post-translational modifications of integrins: molecular mechanisms and pathological implications.

Integrins are cell-surface adhesion receptors that mediate bidirectional signaling governing migration, proliferation, and survival. Emerging evidence reveals that integrin function is finely controlled by an interconnected network of post-translational modifications (PTMs), spanning classical modifications such as glycosylation and phosphorylation, as well as novel metabolite-driven alterations including carbamoylation and cysteine carboxyethylation. These PTMs collectively orchestrate integrin stability, conformation, trafficking, and downstream signaling, with profound implications for tumo

Dynamic S-acylation of IDH2 regulates mitochondrial redox homeostasis.

Mitochondrial redox homeostasis is fundamental for cellular function, and its dysregulation is associated with various diseases, including cancer. Isocitrate dehydrogenase 2 (IDH2) is a key enzyme that maintains this balance by generating NADPH. However, the mechanisms controlling IDH2 subcellular localization remain incompletely understood. Here, we identify reversible S-acylation as a critical regulator of IDH2 localization. Using chemical reporters, we demonstrate that IDH2 is S-acylated at a conserved cysteine residue, mediated by ZDHHC3 and APT1. Loss of IDH2 S-acylation disrupts its mito

USP8-mediated mitochondrial regulation in osteoclasts is essential for skeletal development.

Aberrant protein regulatory pathways disrupt bone development and contribute to skeletal diseases. The cysteine protease family of deubiquitinating enzymes (DUBs) are critical for regulation of bone-resorbing osteoclasts and bone-forming osteoblasts. Here, we demonstrate that the DUB ubiquitin-specific protease 8 (USP8) is highly expressed in osteoclasts and its deletion impairs osteoclast development and bone resorption activity. Deletion of Usp8 in osteoclasts (Usp8Ctsk) results in low trabecular bone mass due to defective endochondral bone formation and short stature resulting from abnormal

ZDHHC17-mediated IKKβ palmitoylation suppresses NF-κB signaling to mitigate the progression of liver fibrosis.

Liver fibrosis (LF) is a common consequence of chronic liver injury and inflammation, driven by the IKK complex-mediated NF-κB signaling pathway. This study investigated the role of macrophage IKKβ palmitoylation in NF-κB signaling and LF progression. Analysis of liver tissues and peripheral blood mononuclear cells (PBMCs) from LF patients and healthy individuals revealed decreased ZDHHC17 expression in LF-associated macrophages. Using mouse models of LF and ZDHHC17 knockout mice, we found that ZDHHC17 deficiency accelerated LF progression. Mechanistically, ZDHHC17 palmitoylated IKKβ at cystei

Development of Covalent Small-Molecule Fluorescent Probes for DNA Methyltransferase 1 Detection in Cancer Cells and Cervical Exfoliated Cells.

DNA methyltransferase 1 (DNMT1) overexpression is associated with aberrant methylation and tumorigenesis, making its detection vital for tumor diagnosis. In this study, RG108 derivatives bearing cysteine-targeted covalent moieties were constructed as warheads for the DNMT1 detectors. Following affinity assessment by surface plasmon resonance, warheads containing a 2-fluoroacrylamido moiety were selected for preparing fluorescein-labeled probes 20a and 20b. In-gel fluorescence scanning and competitive assays confirmed that the probes can covalently bind to DNMT1 at the S-adenosyl-l-homocysteine

Functional microbial shifts and host-microbiome crosstalk in colorectal cancer: insights from a metaproteomic approach.

Colorectal cancer (CRC) incidence is increasing in many low- and middle-income countries, including Egypt, partly due to urbanization and lifestyle changes. Metaproteomic approaches remain underutilized in these settings. In this study, we applied fecal metaproteomics in Egyptian CRC patients to characterize disease-associated protein expression patterns, explore host-microbiota functional interactions, and identify metabolic pathways that are altered in the CRC gut environment. Stool samples from 10 CRC patients and 10 healthy controls were analyzed. A total of 441 differentially expressed pr

Atypical pericapillary Ly6G⁺Nur77⁺ macrophages initiate type-2 immune responses to allergens in the mouse lung.

The mechanisms of airway allergen sensing and type 2 immune response initiation remain poorly understood. Using a mouse house dust mite (HDM)-induced allergic airway model, we identify a population of lung macrophages located close to alveolar capillaries that express Ly6G and the nuclear receptor Nr4a1/Nur77. These atypical Ly6G+Nur77+ macrophages preferentially capture airway-delivered allergens and play an important role in initiating HDM-driven T helper type 2 (Th2) responses. They sense the major HDM allergen, the cysteine protease Der p 1, via protease-activated receptor 2 (PAR2), and th

Draft Genome of Entamoeba marina Provides Insights Into the Attenuation of Pathogenicity and Adaptation to the Marine Environment.

Entamoeba is the amoebozoan parasite commonly found in the intestines of animals. E. marina is the first exception isolated from marine sediments, possibly adapting from animal intestines to the sea. However, the evolutionary process of E. marina remains uncertain due to the lack of a genome sequence. Here, we present the de novo genome and transcriptome of E. marina using Oxford Nanopore MinION and Illumina HiSeq/MiSeq. The genome of E. marina is approximately 37.5 Mbp in length and consists of 202 contigs, which is the second longest, next to E. invadens. E. marina showed a significant reduc

CD164 is an endolysomal host factor for entry of Clade A New World Arenaviruses.

Arenaviruses are divided into Old World (OW) and New World (NW) groups. OW arenaviruses enter cells through a pH-dependent receptor switch from a plasma-membrane factor to an endolysosomal receptor for subsequent membrane fusion, whereas clade B NW arenaviruses use transferrin receptor 1 without a secondary receptor. Using a vesicular stomatitis virus (VSV) chimera expressing the glycoprotein complex (GPC) of the clade A NW arenavirus Pichindé virus, we performed a genome-wide CRISPR loss-of-function screen and identified the endolysosomal sialomucin CD164 as an essential host factor. Metallothionein Isoforms in Acute Myeloid Leukemia: Roles in Survival, Differentiation, and Stress Adaptation.

Metallothioneins (MTs) are cysteine-rich metal-binding proteins traditionally recognized for their roles in metal homeostasis and antioxidant defense. Recent studies have revealed broader and functionally diverse roles of MT isoforms in acute myeloid leukemia (AML). This review summarizes current evidence regarding MT-mediated regulation of leukemic cell survival, differentiation, and therapy-associated stress adaptation. MT1 family members, particularly MT1X and MT1G, have been associated with proliferation, chemoresistance, and differentiation blockade in specific experimental contexts, wher
