Phenotypic and ultrastructural properties of antigen-presenting cells involved in contact sensitization of normal and UV-irradiated mice.

We investigated the surface phenotype and localization of hapten in antigen-presenting cells involved in the induction of contact hypersensitivity or tolerance. Dendritic cells collected 18 h earlier from the draining lymph nodes of mice sensitized with fluorescein isothiocyanate (FITC), which induce contact hypersensitivity upon injection into the foot-pads of naive mice, stimulated proliferation of lymphocytes from FITC-specific T-cell lines. By immunoelectron microscopy, these cells expressed high amounts of surface Ia molecules but had a negligible amount of FITC on the cell membrane. The

A Golgi study of the medial geniculate body in the tree shrew (Tupaia glis).

The subdivisions of the medial geniculate body in Tupaia recognized in previous connectional and cytoarchitectural studies are identified in Golgi-impregnated material. They may be distinguished by the organization of the neuropil, especially the dendrites, and, in many cases, by differences in the neurons. The ventral nucleus contains tufted cells with disc-shaped dendritic fields which are aligned to form laminae. The caudomarginal and deep dorsal nuclei have less tufted, less precisely arranged cells with longer, thin appendages. Neurons in the suprageniculate and dorsal nuclei are similar

mGluR1 signaling is necessary for strengthening winner climbing fiber inputs in the developing mouse cerebellum.

Functional neural circuits are sculpted by strengthening frequently used synapses and removing unnecessary connections. At birth, cerebellar Purkinje cells receive inputs with similar synaptic strengths from multiple climbing fibers (CFs). During postnatal development, a single "winner" CF is selectively strengthened and expands its dendritic innervation territory, while somatic "loser" synapses are eliminated. Here, we report that deleting metabotropic glutamate receptor 1 (mGluR1) or protein kinase Cγ (PKCγ) in mice disrupts this selective strengthening and territory expansion of "winner" CF

Sleep differentially modulates synaptic vesicle protein 2A and spine density across cortical regions and development.

Sleep is proposed to regulate synaptic strength; yet, the underlying mechanisms remain debated under the synaptic homeostasis hypothesis (SHY). We examined synaptic vesicle glycoprotein 2A (SV2A) and dendritic spine density in mouse brain across conditions of physiological sleep and sleep deprivation. Spine density decreased during sleep and increased following sleep deprivation, consistent with the SHY. In contrast, [18F]SynVesT-1 positron emission tomography (PET) imaging and [3H]UCB-J autoradiography revealed reduced SV2A following sleep deprivation. Layer-specific analyses in somatosensory

Nematodes immunomodulatory mechanisms and possible clinical application.

Helminths have evolved complex immunomodulatory mechanisms that enable chronic infection while minimizing host damage. This review summarizes recent advances in understanding nematode-derived molecules with therapeutic potential. The phosphorylcholine-modified glycoprotein ES-62 from Acanthocheilonema viteae exemplifies pathway-selective immune regulation and has inspired small-molecule analogues for inflammatory diseases. Ancylostoma duodenale secretes protease inhibitors, anticoagulants, and anti-inflammatory proteins that reveal targets for immune and hemostatic modulation. Anisakis pegreff

Tracheal tuft cells release ATP and link innate to adaptive immunity in pneumonia.

Tracheal tuft cells shape immune responses in the airways. While some of these effects have been attributed to differential release of either acetylcholine, leukotriene C4 and/or interleukin-25 depending on the activating stimuli, tuft cell-dependent mechanisms underlying the recruitment and activation of immune cells are incompletely understood. Here we show that Pseudomonas aeruginosa infection activates mouse tuft cells, which release ATP via pannexin 1 channels. Taste signaling through the Trpm5 channel is essential for bacterial tuft cell activation and ATP release. We demonstrate that ac

Elucidation of cytotoxicity of α-Synuclein fibrils on immune cells.

Progressive aggregation of α-synuclein (α-Syn), a small cytosolic protein involved in cell vesicle trafficking, in the midbrain, hypothalamus, and thalamus is linked to Parkinson's disease (PD). Amyloid oligomers and fibrils formed as a result of such aggregation are highly toxic to neurons. However, it remains unclear whether amyloid-induced toxicity of neurons is the primary mechanism of the progressive neurodegeneration observed upon PD. In the current study, we investigated cytotoxicity exerted by α-Syn fibrils formed in the lipid-free environment, as well as in the presence of two phospho

[Progress in the role of immune cells in the pathogenesis of bronchial asthma].

Bronchial asthma is a chronic airway inflammatory disease that involves various immune cells. As the main roles in asthma immune mechanism, T lymphocytes [T helper type 1(Th1) cells, Th2 cells, Th17 cells, regulatory T cells (Tregs), T follicular helper (Tfh) cells and cytotoxic T (Tc) cells], innate lymphoid cells (ILCs), B cells, granulocytes (mast cells, eosinophils, basophils, neutrophils), macrophages as well as dendritic cells (DC) are activated by allergens and secrete their own specific cytokines. They interact with each other in function and form a complex asthma-related immune cell i

Cryo-EM structure of human class C orphan GPCR GPR179 involved in visual processing.

GPR179, an orphan class C GPCR, is expressed at the dendritic tips of ON-bipolar cells in the retina. It plays a pivotal role in the initial synaptic transmission of visual signals from photoreceptors, and its deficiency is known to be the cause of complete congenital stationary night blindness. Here, we present the cryo-electron microscopy structure of human GPR179. Notably, the transmembrane domain (TMD) of GPR179 forms a homodimer through the TM1/7 interface with a single inter-protomer disulfide bond, adopting a noncanonical dimerization mode. Furthermore, the TMD dimer exhibits architectu

Activation of TrkB signaling mitigates cerebellar anomalies caused by Rbm4-Bdnf deficiency.

A molecular and functional link between neurotrophin signaling and cerebellar foliation is lacking. Here we show that constitutive knockout of two homologous genes encoding the RNA binding protein RBM4 results in foliation defects at cerebellar lobules VI-VII and delayed motor learning in mice. Moreover, the features of Rbm4 double knockout (dKO), including impaired differentiation of cerebellar granule cells and dendritic arborization of Purkinje cells, are reminiscent of neurotrophin deficiency. Loss of RBM4 indeed reduced brain-derived neurotrophic factor (BDNF). RBM4 promoted the expressio

Identification of a Self-Assembling Small-Molecule Cancer Vaccine Adjuvant with an Improved Toxicity Profile.

Protein or peptide cancer vaccines usually include immune potentiators, so-called adjuvants. However, it remains challenging to identify structurally simple, chemically accessible synthetic molecules that are effective and safe as vaccine adjuvant. Here, we present cholicamideβ (6), a self-assembling small-molecule vaccine adjuvant with an improved toxicity profile and proven efficacy in vivo. We demonstrate that cholicamideβ (6), which is less cytotoxic than its parent compound, forms virus-like particles to potently activate dendritic cells with the concomitant secretion of cytokines. When c

A critical period of neuronal activity results in aberrant neurogenesis rewiring hippocampal circuitry in a mouse model of epilepsy.

In the mammalian hippocampus, adult-born granule cells (abGCs) contribute to the function of the dentate gyrus (DG). Disruption of the DG circuitry causes spontaneous recurrent seizures (SRS), which can lead to epilepsy. Although abGCs contribute to local inhibitory feedback circuitry, whether they are involved in epileptogenesis remains elusive. Here, we identify a critical window of activity associated with the aberrant maturation of abGCs characterized by abnormal dendrite morphology, ectopic migration, and SRS. Importantly, in a mouse model of temporal lobe epilepsy, silencing aberrant abG

Neurophysiological basis of the N400 deflection, from Mismatch Negativity to Semantic Prediction Potentials and late positive components.

The first theoretical model on the neurophysiological basis of the N400: the deflection reflects layer I dendritic plateaus on a preparatory state of synaptic integration that precedes layer V somatic burst firing for conscious identification of the higher-order features of the stimulus (a late positive shift). Plateaus ensue from apical disinhibition by vasoactive intestinal polypeptide-positive interneurons (VIPs) through suppression of Martinotti cells, opening the gates for glutamatergic feedback to trigger dendritic regenerative potentials. Cholinergic transients contribute to these dynam

Disrupted prefrontal neuronal oscillations and morphology induced by sleep deprivation in young APP/PS1 transgenic AD mice.

Emerging evidence suggests that sleep deprivation (SD) is a public health epidemic and increase the risk of Alzheimer's disease (AD) progression. However, the underlying mechanisms remain to be fully investigated. In this study, we investigate the impact of 72 h SD on the prefrontal cortex (PFC) of 3∼4-months-old APP/PS1 transgenic AD mice - at an age before the onset of plaque formation and memory decline. Our results reveal that SD alters delta, theta and high-gamma oscillations in the PFC, accompanied by increased levels of excitatory postsynaptic signaling (NMDAR, GluR1, and CaMKII) in AD

Blockage of p75NTR ameliorates depressive-like behaviours of mice under chronic unpredictable mild stress.

The precursor of brain derived neurotrophic factor (proBDNF) and its receptor p75NTR are upregulated in depressive patients and chronic stress-induced depressive animals, suggesting that activation of p75NTR signalling may underlie pathogenesis of depression. In the present study we hypothesize that the blockade of p75NTR may have therapeutic effect on depressive mice under chronic stress. The treatment of mice with the recombinant fusion protein of p75NTR extracellular domain and fragment C of immunoglobulin (p75ECD-Fc) significantly reduced the immobility time in the forced swim test and tai

Salp15, a Multifunctional Protein From Tick Saliva With Potential Pharmaceutical Effects.

Ixodes ticks are the main vectors for a number of zoonotic diseases, including Lyme disease. Ticks secrete saliva directly into a mammalian host while feeding on the host's blood. This action serves to modulate host immunity and coagulation, thus allowing ticks to attach and feed upon their host. One of the most extensively studied components of tick saliva is Salp15. Research has shown that this protein binds specifically to CD4 molecules on the surface of T lymphocytes, interferes with TCR-mediated signaling transduction, inhibits CD4+ T cell activation and proliferation, and impedes the sec

Current Concepts of the Pathogenesis of Aplastic Anemia.

Abnormal activation of the immune system plays an important role in the pathogenesis of aplastic anemia (AA). Various immune cells and cytokines constitute a complex immune network, leading to bone marrow failure. The known pathogenesis is an increase of the myeloid dendritic cell (mDC)/ plasmacytoid dendritic cell (pDC) ratio, which causes the ratio of T helper (Th)1/Th2 to be skewed in favor of Th1 and eventually leads to an abnormal activation of cytotoxic T lymphocyte (CTL). The antigens that stimulate T cells in the context of AA remain unknown. In this process, regulatory T (Treg), Th17,

Hyperbranched polyglycerol nanostructures for anti-biofouling, multifunctional drug delivery, bioimaging and theranostic applications.

Nowadays, great attention has been paid to the design and development of novel macromolecular constructions in drug delivery. Hyperbranched polymers (HBPs) are amongst various types of polymeric structures with exceptional physicochemical properties and biomedical applications relevant to their dendritic structure. Unlike perfect dendrimers, HBPs can be produced via one-step polymerization reactions. Moreover, they can be applied as building blocks in preparing dendronized, dendrigraft or dendritic nanostructures. Hyperbranched polyglycerols (HPGs) and their derivatives are widely regarded as

Stable lines and clones of long-term proliferating normal, genetically unmodified murine common lymphoid progenitors.

Common lymphoid progenitors (CLPs) differentiate to T and B lymphocytes, dendritic cells, natural killer cells, and innate lymphoid cells. Here, we describe culture conditions that, for the first time, allow the establishment of lymphoid-restricted, but uncommitted, long-term proliferating CLP cell lines and clones from a small pool of these cells from normal mouse bone marrow, without any genetic manipulation. Cells from more than half of the cultured CLP clones could be induced to differentiate to T, B, natural killer, dendritic, and myeloid cells in vitro. Cultured, transplanted CLPs transi
