Selective suppression of cocaine- versus food-maintained responding by monoamine releasers in rhesus monkeys: benzylpiperazine, (+)phenmetrazine, and 4-benzylpiperidine.

Monoamine releasers constitute one class of drugs currently under investigation as potential agonist medications for the treatment of cocaine dependence. The efficacy and safety of monoamine releasers as candidate medications may be influenced in part by their relative potency to release dopamine and serotonin, and we reported previously that releasers with approximately 30-fold selectivity for dopamine versus serotonin release may be especially promising. The present study examined the effects of the releasers benzylpiperazine, (+)phenmetrazine, and 4-benzylpiperidine, which have 20- to 48-fo

Role of different monoamine receptors controlling MK-801-induced release of serotonin and glutamate in the medial prefrontal cortex: relevance for antipsychotic action.

Several studies have demonstrated that systemically administered N-methyl-d-aspartate (NMDA) receptor antagonists increase serotonin (5-HT) and glutamate release in the medial prefrontal cortex (mPFC). Previously we showed that the perfusion of clozapine in the mPFC prevented the MK-801-induced increase in extracellular glutamate and 5-HT whereas haloperidol blocked only the effect of MK-801 on glutamate. To study the contribution of different monoaminergic receptors (for which clozapine and haloperidol exhibit distinct affinities) to these effects, here we used in-vivo microdialysis to examin

Therapeutic possibilities of cysteamine in the treatment of schizophrenia.

Schizophrenia has complicated pathogeneses that is not able to be explained by any one supposed hypothesis, although alterations in dopamine neurotransmission have been widely accepted as the most plausible mechanism. A transition from traditional typical antipsychotics to contemporary atypical antipsychotics which have significantly improved tolerability and enhanced specific efficacy has been also made based on this dopamine hypothesis. Cysteamine is a natural product of mammalian cells and found to be useful pharmacological alternative. A number of evidence suggests that cysteamine may cont

Oxidative metabolites of 5-S-cysteinyldopamine inhibit the alpha-ketoglutarate dehydrogenase complex: possible relevance to the pathogenesis of Parkinson's disease.

A characteristic change in the substantia nigra of Parkinson's disease patients is an apparent accelerated rate of dopamine oxidation as evidenced by an increased 5-S-cysteinyldopamine (5-S-CyS-DA) to dopamine ratio. However, 5-S-CyS-DA is more easily oxidized than dopamine to give 7-(2-aminoethyl)-3,4-dihydro-5-hydroxy-2H-1,4-benzothiazine-3-carboxylic acid (DHBT-1). Previous studies have demonstrated that DHBT-1 can be accumulated by intact rat brain mitochondria and inhibits complex I but not complex II respiration. In this study, it is shown that DHBT-1 also inhibits the alpha-ketoglutarat

Detection of dopaminergic neurotransmitter activity using pharmacologic MRI: correlation with PET, microdialysis, and behavioral data.

The metabolic activation resulting from direct dopaminergic stimulation can be detected using auto-radiography, positron emission tomography (PET) or, potentially, fMRI techniques. To establish the validity of the latter possibility, we have performed a number of experiments. We measured the regional selectivity of two different dopaminergic ligands: the dopamine release compound D-amphetamine and the dopamine transporter antagonist 2 beta-carbomethoxy-3 beta-(4-fluoropheny) tropane (CFT). Both compounds led to increased signal intensity in gradient echo images in regions of the brain with hig

Metabolic mapping of the pharmacological and toxicological effects of dopaminergic drugs in experimental animals.

Functional consequences of pharmacological and toxicological manipulations of the dopaminergic systems were evaluated by means of the 2-[14C]deoxyglucose (DG) method for measuring local rates of cerebral glucose utilization. Administration of dopamine agonist drugs modifies glucose metabolism in selected brain areas. Several factors, such as the compound used, the dose, length, and modality of the treatment, and the interval of time between the end of the treatment and the measurement of glucose utilization, contribute to define the topography and intensity of the changes. The differences refe

Effect of drugs on pituitary ultrastructure.

Various drugs and hormones influence the light microscopic and especially the electron microscopic structure of the anterior pituitary and its tumors. Many structural effects are known only from animal experiments since specimens from human pituitaries are mostly not available. The structure of growth hormone (GH) cells is relatively stable. A massive GH cell hyperplasia is known only in rare cases with growth hormone releasing factor (GRF) excess from tumors. Prolactin cells can be stimulated by drugs, neurotransmitters, and hormones which decrease the dopamine inhibition. Adrenocorticotropic

[Effect of adenosine on carotid chemoreceptor activity in the rabbit].

The response of single carotid chemoreceptor afferent fibers upon adenosine acting on the carotid body (CB) was examined in 39 urethan-anesthetized rabbits. Totally 73 units with spontaneous discharge were recorded in our experiment. The results were as follows: (1) Of 55 units, 51 showed an increase in discharge frequency from 0.76 +/- 0.10 to 1.53 +/- 0.23 imp/s. A few new units were recruited concomitantly in response to intracarotid injection of adenosine (10 micrograms/kg). (2) Adding adenosine in the doses of 0.5, 1.5, 10, 50 and 100 micrograms/kg to the perfusate passing through the iso

Human neuroblastoma cells acquire regulated secretory properties and different sensitivity to Ca2+ and alpha-latrotoxin after exposure to differentiating agents.

IMR-32 human neuroblastoma cells are unable to release [3H]dopamine in response to secretagogues. However, they express a normal complement of membrane receptors and ion channels which are efficiently coupled to second messenger production. In the present study we took advantage of the ability of this cell line to differentiate in vitro in the presence of either dibutyrryl-cAMP or 5-bromodeoxyuridine, to analyze any developmentally regulated changes in its secretory properties. Uptake, storage, and release of [3H]dopamine were studied biochemically and by autoradiography. The calcium ionophore

Dysregulation of external globus pallidus-subthalamic nucleus network dynamics in parkinsonian mice during cortical slow-wave activity and activation.

KEY POINTS: Reciprocally connected GABAergic external globus pallidus (GPe) and glutamatergic subthalamic nucleus (STN) neurons form a key network within the basal ganglia. In Parkinson's disease and its models, abnormal rates and patterns of GPe-STN network activity are linked to motor dysfunction. Using cell class-specific optogenetic identification and inhibition during cortical slow-wave activity and activation, we report that, in dopamine-depleted mice, (1) D2 dopamine receptor expressing striatal projection neurons (D2-SPNs) discharge at higher rates, especially during cortical activatio

A Podcast on the Rhythms of Healing: Patient and Physician Insights on Art, Music, Dance Therapy and Creativity in Parkinson's Disease.

This is an outline for a podcast. Parkinson's Disease (PD) is a progressive neurodegenerative disease in which there is increasing loss of dopamine neurones from the basal ganglia (Simon et al. Clin Geriatr Med. 36(1):1-12). Motor symptoms, which are usually asymmetrical, include resting tremor, rigidity, bradykinesia (slow movement) and, usually later on, postural instability. PD is primarily a clinical diagnosis although a DaTscan, which if positive shows asymmetrical loss of dopamine uptake in the basal ganglia, is helpful in cases of doubt (Tsang and Walker J Neurol 270(5):2550-2558).

Optogenetic Isolation of the Amacrine Cell-OFF Bipolar Cell Synapse Shows Selective D1R Control of Glycinergic Transmission.

PURPOSE: Light evoked inhibition of OFF cone bipolar cells (OFF BCs) is modulated both by background light levels and the action of dopamine through the dopamine D1 receptor (R). Since D1Rs are localized throughout the mouse retina, it is not known where in the light signaling pathway dopamine is modulating signals to OFF BCs. Here we tested a technique that allowed for the isolation of the amacrine cell (AC) to OFF BC circuit to determine if there are local D1R-induced changes in inhibition from presynaptic ACs onto OFF BCs. METHODS: We utilized the B6.Cg-Tg(Slc32al-COP4*H134R/EYFP) mouse li

Characterising a New Sheep Model of Parkinson's Disease Using Unilateral Intracerebral Injection of 6-Hydroxydopamine Into the Substantia Nigra.

New therapeutic agents developed for treating neurological disorders are often tested successfully on rodents. Testing in an appropriate large animal model where there is longer lifespan and comparable brain size to humans should improve translational success and is frequently expected by regulatory bodies. In this project, we aimed to establish a novel sheep model of Parkinson's disease as a large-brained experimental model for translational research. Our objective was to create a sheep model of Parkinson's disease by unilaterally infusing the neurotoxin 6-hydroxydopamine into the substantia

GDF15 attenuates Parkinson's disease progression via suppressing the activation of cGAS-STING pathway.

Growth differentiation Factor 15 (GDF15) plays an important role in the innate immune response. However, whether GDF15 could regulate Parkinson's disease (PD) remains unknown. In this study, we explored the function and underlying molecular mechanisms of GDF15 in PD. The protein and mRNA expressions were examined applying immunofluorescence staining, Western blot and qRT-PCR. Ferrous iron content was also assessed using an iron assay kit. The effect of GDF15 knockdown on mitochondrial membrane potential, ROS level, intracellular Fe2+ level and mitochondrial permeability transition pore opening

On the neural substrates of mind wandering and dynamic thought: A drug and brain stimulation study.

The impact of mind wandering on our daily lives ranges from diminishing productivity, to facilitating creativity and problem solving. There is evidence that distinct internal thought types can be modulated by transcranial direct current stimulation (tDCS), although little is known about optimal stimulation parameters or the mechanisms behind such effects. In addition, recent findings suggest changes in dopamine availability may alter the effect tDCS has on neural and behavioural outcomes. Dopaminergic functioning has also been implicated in executive processes anticorrelated with mind wanderin

Motor and Non-Motor Effects of Acute MPTP in Adult Zebrafish: Insights into Parkinson's Disease.

Parkinson's disease (PD), the second most common neurodegenerative disorder, is characterized by the progressive loss of dopaminergic neurons in the substantia nigra pars compacta, leading to motor and non-motor symptoms. The neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) has been extensively used in different animal species to develop chemical models of PD. This study aimed to evaluate the effects of acute exposure to MPTP (3 × 150 mg/kg, intraperitoneally) on adult zebrafish by assessing the neurochemical, transcriptional, and motor changes associated with PD pathogenesis. Environmental enrichment improves deficits in hippocampal neuroplasticity and cognition in prenatally aripiprazole-exposed mouse offspring.

Aripiprazole has become one of the most commonly prescribed antipsychotics, including in pregnant women, owing to a broad range of indications for psychiatric disorders and relatively few metabolic side effects. Compared with that of other antipsychotics, data regarding the safety of gestational aripiprazole exposure for offspring neurodevelopment are limited. This study investigated how prenatal exposure to aripiprazole affects the hippocampal neuroplasticity of adult offspring and whether any such effect can be reversed by environmental enrichment. Aripiprazole was administered to pregnant C

Targeting APE1/Ref-1 to alleviate formalin-induced pain and spinal neuro-inflammation in rats: a promising therapeutic approach.

BACKGROUND: Pain is a multifaceted condition intricately linked to inflammation, which plays a critical role in its onset and progression. METHODS: To investigate the influence of APE1/Ref-1 on oxidative stress and inflammatory marker expression, we employed a hind paw sensitization model induced by formalin. We inhibited the redox function of APE1 using E3330 and assessed its effects on pain behavior. Mitochondrial morphology was examined via electron microscopy, and the impact on dopaminergic signaling alongside bioinformatics analyses to explore potential E3330 binding to dopamine receptor
