Targeting the PI3K/Akt/mTOR and Nrf2 signaling axis with berberine: a novel strategy for attenuating diabetic cardiomyopathy.

Diabetic cardiomyopathy (DCM) is a major contributor to heart failure in patients with diabetes. Although berberine (BBR) exhibits broad metabolic and cardiovascular benefits, its mechanistic role in DCM remains incompletely defined. Cardiovascular function was assessed using arterial blood pressure (ABP), electrocardiography (ECG), and echocardiography. Biochemical analyses included glycemic indices, lipid profile, oxidative stress markers, inflammatory cytokines, and cardiac injury biomarkers. Cardiac and aortic tissues were evaluated for calcium (Ca2⁺) content, gene expression by RT-PCR, an

Shedding light on direct and indirect effects of antimicrobial photodynamic therapy: A review update.

BACKGROUND: Antimicrobial photodynamic therapy (aPDT) operates through the light-activated excitation of photosensitizers, generating reactive oxygen species that simultaneously damage multiple essential microbial structures. This broad-spectrum, multi-target mechanism holds strong promise for combating antimicrobial-resistant microorganisms. Beyond these direct effects, other biological events can occur, potentially influencing microbial physiology, host responses, or overall treatment efficacy. These intermediate processes can yield beneficial outcomes or unintended consequences, highlightin

Effects of Light Spectrum and Intensity on Cellular Stress Responses in Baikal Whitefish and Its Hybrid Embryos: A Basis for Optimizing the Aquaculture Environment.

The light spectrum is a key factor in aquaculture, but its effects on molecular stress responses during early fish development are unclear. This study examined how light of different wavelengths (spectra) affects embryos of Baikal whitefish Coregonus baicalensis and its hybrid with Yenisei hump-snout whitefish C. fluviatilis. Eggs were incubated from 35 days post-fertilization under white light (1.8 and 20 µmol m-2 s-1), darkness, red (peak at 631 nm), green (peak at 507 nm) and blue (peak at 459 nm) light. We analyzed relative telomere length, telomerase activity, blood profiles, and expressi

Longitudinal integration of microbiota and metabolomics reveals (poly)phenols-driven gut ecosystem dynamics.

The gut microbiota and its "theatre of activity" (the collective pool of metabolites and signaling molecules) function as a plastic interface that responds to nutritional inputs. Dietary (poly)phenols are key bioactive components of berries and are increasingly recognized for their capacity to modulate gut microbiota composition and metabolic activity while being biotransformed by microbiota. However, the temporal dynamics of this interaction remain poorly resolved, as most studies rely on cross-sectional or endpoint-only designs. Here, we conducted a longitudinal multi-omic study to map the c

Effect of high phytase levels on growth performance and calcium and phosphorus metabolism in broilers fed a non-inorganic phosphorus diet.

The effects of high phytase levels on growth performance and calcium (Ca) and phosphorus (P) metabolism in broilers fed non-inorganic P diets were investigated. A total of 600 one-day-old broilers were assigned to either a control diet or a non-inorganic P diet supplemented with one of two commercial phytases (P1 and P2) at 1,000 (NP, normal level of phytase) or 10,000 FTU/kg (HP, high level of phytase). Compared with the control group, HP treatments had comparable feed intake, body weight gain and feed conversion ratio, whereas NP supplementation reduced the parameters, particularly during 22

Mechanisms of Testosterone's Anabolic Effects on Muscle and Function: Controversies and New Insights.

Testosterone treatment increases muscle mass, maximal voluntary muscle strength, aerobic capacity, and some measures of physical function. Activational and epigenetic mechanisms by which androgens improve muscle mass and physical performance and how to apply these anabolic effects to treat functional limitations associated with aging and disease remain incompletely understood. Testosterone treatment induces hypertrophy of type 1 and 2 muscle fibers and increases muscle progenitor cell numbers by promoting differentiation of mesenchymal progenitor cells into myogenic lineage by an androgen rece

Growth-defence trade-offs revealed by metabolic shifts in maize root apex under salinity and water stress.

This study investigated the proteomic and soluble metabolite profiles of maize (Zea mays L.) root apices following 72 and 96 h of seedling exposure to 10 mM and 100 mM NaCl, as well as 16% (w/v) polyethylene glycol 6000 (PEG). The highest NaCl concentration and PEG significantly reduced root length, whereas salinity increased root width, and PEG decreased it compared with non-stressed controls. Stress conditions induced changes in peroxidase isoform patterns. Across all samples, 3357 unique proteins were identified in maize root apices. Gene ontology enrichment analysis of differentially accum

From diffusion to focalization: The age-related divergence of emotional networks in bipolar disorder.

BACKGROUND: Emotional processing abnormalities are a core feature of bipolar disorder (BD), yet neuroimaging studies have yielded inconsistent results. The age-related divergence between pediatric (PBD) and adult (ABD) bipolar disorder has been increasingly highlighted by physiological and clinical evidence. Therefore, previous findings were integrated to construct the emotional networks of PBD and ABD, with the aim of elucidating their age-related divergence. METHODS: Activation likelihood estimation was performed on 17 PBD (340 patients, 341 controls) and 20 ABD (681 patients, 582 controls)

The Role of Immune Cells as Modulators of Progression in Polycystic Kidney Disease: A Systematic Review.

Autosomal dominant polycystic kidney disease (ADPKD) is a genetic disorder caused by mutations in genes encoding ciliary proteins. Increasing evidence suggests that immune cell infiltration, chronic inflammation, and dysregulated immune signaling pathways contribute to cyst growth, fibrosis, and progressive kidney function decline. Notably, features of chronic inflammation are already detectable in early stages of the disease, including infiltration by immune cells, elevated levels of proinflammatory cytokines, and immunologic crosstalk between immune and epithelial cells. These immune respons

Induction of genotoxic damage and ROS production in HCT116 TP53+/+ and HCT116TP53-/- colorectal cancer cell lines by anticancer drugs.

Reactive oxygen species (ROS) play a dual role in cancer biology, contributing to both tumor progression and therapeutic responses. Many chemotherapeutic agents exert their cytotoxic effects through ROS generation, although the extent and biological relevance of this process remain influenced by cellular context, including the functional status of tumor suppressor genes such as TP53. In this study, we investigated the induction of genomic damage and oxidative stress in two isogenic human colorectal cancer cell lines-HCT116TP53+/+ and HCT116TP53-/--after exposure to four commonly used anticance

Contrasting functions of CD73 and adenosine in CD8+ T-cell exhaustion during antitumor immunity.

T-cell exhaustion is a state of functional decline in T cells, driven by chronic antigen exposure and inhibitory signals within the tumor microenvironment. Exhausted CD8+ T cells (Tex) derive from precursor exhausted T cells (Tpex), a self-renewing population responsible for the proliferative burst in anti-PD-1 therapies. Exhausted T cells are exposed to adenosine in the tumor, yet the role of the CD73/adenosine axis and Tpex/Tex differentiation remains unclear. Using an in vitro model for CD8+ T-cell exhaustion, we found that CD73 expression increased during Tpex formation and that its expres

The mineralocorticoid receptor and its antagonist finerenone regulate hepatic lipid accumulation via the AMPK/SREBP1/FASN signaling.

Metabolic dysfunction-associated steatotic liver disease (MASLD), closely linked to obesity and diabetes, remains poorly understood, with limited therapeutic options. The mineralocorticoid receptor (MR) has been implicated in metabolic dysregulation, yet its role in hepatic lipid metabolism remains incompletely defined. Finerenone, a non-steroidal MR antagonist, has demonstrated renal and cardiovascular benefits in diabetes, yet its effects on hepatic metabolism have not been fully explored. This study aimed to evaluate the hepatoprotective effects of finerenone in MASLD and elucidate the unde

Pharmacological effects of nebivolol on endothelial response in a preeclampsia in vitro model.

UNLABELLED: Preeclampsia (PE) is a pregnancy-specific hypertensive disorder characterized by systemic endothelial dysfunction and a profound anti-angiogenic state driven by elevated soluble fms-like tyrosine kinase-1 (sFlt-1). While the third-generation β1-AR nebivolol promotes vasodilation partially via receptor β3-AR activation, whether its protective effects can reverse the complex anti-angiogenic and hypoxic stress of PE remains unclear. AIMS: This study aimed to determine the functional and molecular effects of nebivolol in a PE-like endothelial microenvironment and to understand the spe

Redox Imbalance and NOS-Dependent Modulation of Superoxide in the Bladder Mucosa of Women With IC/BPS: A Preliminary Study.

Interstitial Cystitis/Bladder Pain Syndrome (IC/BPS) is a chronic condition characterized by bladder/pelvic pain and urinary symptoms, with poorly defined mechanisms and no reliable biomarkers. Oxidative stress, particularly reactive oxygen species (ROS), has been implicated in its pathophysiology, but direct human evidence remains scarce. Here, we evaluated ROS- and nitric oxide (NO)-related pathways in bladder mucosa from women with IC/BPS (n = 5) and controls (n = 7). Gene expression of NO-cGMP components, NADPH oxidases (NOX1-5), and antioxidant enzymes was assessed by qPCR. Baseline NO an

SARS-CoV-2 infection during the first trimester leads to profound immune dysregulation at the maternal-fetal interface despite limited virus detection in placental tissues.

The endemic nature of SARS-CoV-2 underscores the imperative to elucidate its effects on pregnancy, particularly the potential for vertical transmission and its influence on the early maternal-fetal interface. Here, we performed an extensive cohort study involving 761 pregnant women who voluntarily terminated their pregnancies during the first trimester. Analyses conducted using RT‒qPCR, fluorescence in situ hybridisation, and indirect immunofluorescence showed low detection levels of SARS-CoV-2 infection within villous and decidual tissues. Single-cell RNA sequencing analysis revealed an absen

Bidirectional association between immune-mediated diseases and major depressive disorder: evidence from cohort, genome-wide pleiotropic, and experimental studies.

Although immune-mediated diseases (IMDs) and major depressive disorder (MDD) commonly co-occur, the bidirectional relationship between them remains to be fully elucidated. Using data from the prospective UK Biobank cohort, we evaluated the bidirectional associations by time-varying Cox proportional hazards regression models and assessed shared genetic architecture using genome-wide association study summary statistics. Additionally, we employed collagen-induced arthritis (CIA) and chronic social defeat stress (CSDS) mouse models to investigate the relationship between rheumatoid arthritis (RA)

TET1 downregulation in the dorsal root ganglion and spinal cord is required for short-term sleep disturbance to delay surgical pain recovery.

BACKGROUND: Sleep disturbance delays surgical pain recovery. This impact is associated with gene dysregulation in the dorsal root ganglion (DRG) and spinal dorsal horn (SDH). However, the mechanisms underlying this dysregulation remain unclear. METHODS: Expression of ten-eleven translocation methylcytosine dioxygenase 1 (TET1) was examined in the DRG and SDH following plantar incision in rats subjected to short-term rapid eye movement sleep disturbance. A herpes simplex virus expressing Tet1 mRNA (HSV-TET1) was microinjected into the ipsilateral L4 and L5 DRGs or SDH, and its effects on μ-opi
