Metabolic effects of galactose on human HepG2 hepatoblastoma cells.

HepG2 cells were used as a model system to study the effects of galactose overload on the liver, a target organ of galactose toxicity in patients suffering from transferase-deficient galactosemia. In the presence of galactose, HepG2 cell growth was slow and the pattern of gene expression remained characteristic of liver cells (secretion of alpha-fetoprotein [AFP] albumin, and transferrin). Galactose-1-phosphate (Gal-1-P) accumulated, as it does in galactosemic cells, but did not affect the energetic status of the cells (no adenosine triphosphate [ATP] depletion). However, the substitution of g

Localization of a new type of X-linked liver glycogenosis to the chromosomal region Xp22 containing the liver alpha-subunit of phosphorylase kinase (PHKA2).

We describe here a new type of X-linked liver glycogen storage disease. The main symptoms include liver enlargement and growth retardation. The clinical and biochemical abnormalities of this glycogenosis are similar to those of classical X-linked liver glycogenosis due to phosphorylase kinase deficiency (XLG). However, in contrast to patients with XLG, the patients described here have no reduced phosphorylase kinase activity in erythrocytes and leukocytes, and no enzyme deficiency could be found. Linkage analysis of four families with this X-linked type of liver glycogenosis assigned the disea

Prevention of cyanide-induced cytotoxicity by nutrients in isolated rat hepatocytes.

The effects of various glycolytic substrates and keto acid metabolites on the cytotoxic effects of cyanide have been studied with isolated rat hepatocytes. The sequence of cytotoxic events with 2 mM cyanide was an immediate inhibition of respiration followed by ATP depletion. Disruption of the plasma membrane occurred when 85-90% of ATP levels had been depleted. Fructose, dihydroxyacetone, glyceraldehyde, pyruvate, and alpha-ketoglutarate prevented cyanide-induced cytotoxicity and ATP depletion. Hepatocyte respiration was also restored by all except fructose. Fructose, unlike the others, also

Cellular organization and peritrophic membrane formation in the cardia (proventriculus) of Drosophila melanogaster.

The peritrophic membrane of Drosophila melanogaster consists of four layers, each associated with a specific region of the folded epithelial lining of the cardia. The epithelium is adapted to produce this multilaminar peritrophic membrane by bringing together several regions of foregut and midgut, each characterized by a distinctively differentiated cell type. The very thin, electron-dense inner layer of the peritrophic membrane originates adjacent to the cuticular surface of the stomadeal valve and so appears to require some contribution by the underlying foregut cells. These foregut cells ar

Caloric supply in parenteral nutrition in the stress phase.

Due to the metabolic alterations following surgery, glucose is not completely oxidized but is converted into glycogen and lipids. Therefore we analysed 2 groups of 15 patients following surgery. They received parenteral nutrition with identical nitrogen (N = 0.28 g/kg/d) and lipid (1.5 g/kg/d) intake, whereby only the composition of carbohydrates was varied. Group I received exclusively glucose = 0.22 g/kg/h, and group II the combination fructose-glucose-xylitol in a proportion of 2:1:1 at the same infusion rate. The study lasted approximately 10 days. We found decreased exogenous insulin requ

Effect of copper on insulin release by the intact rat pancreas and the perfused rat pancreas.

In previous studies, we have shown that rats fed a copper-poor diet have an impaired glucose tolerance and insulin response to an oral glucose load. Furthermore, CuCl2 X 2H2O added to the incubation medium stimulated glucose incorporation into the diaphragm glycogen and epididimal fat of rats. The purpose of the present study was to examine the effect of copper on insulin release in vivo and in the perfused pancreas. Copper chloride stimulated the insulin release by the perfused pancreas of rats fed laboratory chow (copper content, 6.7 ppm). Stimulation with 0.5 mg/dl CuCl2 X 2H2O for 20 min r

Nephrogenic adenoma. A report of 15 cases, review of the literature, and comparison with clear cell adenocarcinoma of the urinary tract.

The clinical and pathological features of 15 previously unreported and 98 reported cases of nephrogenic adenoma were reviewed and compared with those of the 19 reported clear cell adenocarcinomas of the urinary bladder and urethra. Nephrogenic adenoma has occurred in patients from 4 to 83 (average 41) years of age. Two thirds of the patients were male, and most had a history of a genitourinary operation or procedure, genitourinary trauma, urinary calculi, or a renal transplant. Nephrogenic adenomas are typically small but have measured up to 7 cm. They may be papillary, polypoid, or sessile, a

Prenatal diagnosis of lysosomal storage diseases. Review of experience in 145 patient referrals over a period of eight years.

Over the past eight years, 145 patients from Australia and New Zealand were referred to the Department of Chemical Pathology, The Adelaide Children's Hospital, for the prenatal assessment of a variety of lysosomal storage diseases. This group comprised 65 patients at risk for a glycosphingolipidosis, 50 for a mucopolysaccharidosis, five for mucolipidosis, 15 for glycogen storage disease, and 10 for cystinosis. In three cases, amniotic cells failed to establish in culture; in one of these cases, cell-free amniotic fluid was used to make the prenatal diagnosis; the fetus spontaneously aborted in

Ultrastructural alterations of the vestibular nuclei in Jacob-Creutzfeld disease.

The morphological alterations of the vestibular nuclei in a case of Jacob-Creutzfeld disease were studied with light and electron microscope. The most prominent histological findings were the neuronal degeneration and neuronal loss resulting in a marked decrease of the total population, the increased reactive astrocytosis which demonstrated an extensive perivascular arrangement, the microgliosis and the interstitial spongiosis. At the ultrastructural level a marked dilatation of the astrocytic processes was seen. Most of them were completely transformed to sizeable cysts which were divided by

Benign infantile mitochondrial myopathy due to reversible cytochrome c oxidase deficiency.

A 2-week-old boy had profound generalized weakness, hypotonia, hyporeflexia, macroglossia, and severe lactic acidosis. The infant improved spontaneously: he held his head at 4 1/2 months, rolled over at 7 months, and walked by 16 months. At 33 months of age, he had mild proximal weakness. Macroglossia disappeared by age 4 months. Blood lactic acid declined steadily and was normal by 14 months of age. Histochemical and ultrastructural studies of muscle biopsy specimens obtained at 1 and 7 months of age showed excessive mitochondria, lipid, and glycogen; a third biopsy at age 36 months showed on

Idiopathic orthostatic hypotension: circulating noradrenaline and ultrastructure of saphenous vein.

The present study indicates that patients who can be clinically classified as idiopathic orthostatic hypotensives of the peripheral type are heterogeneous. There is the typical hypoadrenergic type characterized by low levels of circulating noradrenaline, much reduced or absent noradrenaline stores, and correspondingly little or no adrenergic innervation in the saphenous vein, a major capacitance vessel, as confirmed by ultrastructural examination. In one patient of this type, an abnormally high occurrence of mast cells in the blood vessel wall was noted. There also exists a category of individ

Nutritional and metabolic aspects of fatty liver disease in poultry.

Summary There are two metabolic disorders of major commercial importance in poultry that involve the occurrence of fatty deposits in the liver. Fatty Liver and Kidney Syndrome (FLKS) affects young birds and the main manifestations, lipid infiltrations into liver and many other organs, are apparently secondary effects of the primary lesion that lies in carbohydrate metabolism. Although several nutritional and environmental factors influence FLKS, the main factor is the vitamin, biotin. Influence of glucose on the transmembrane action potential of papillary muscle. Effects of concentration, phlorizin and insulin, nonmetabolizable sugars, and stimulators of glycolysis.

The action potential duration (APD) of isolated guinea pig papillary muscle is directly related to the medium glucose concentration regardless of the gas mixture with which it is in equilibrium. The APD can be maintained at control value for many hours by a glucose concentration of 50 mM in the complete absence of oxygen. Following reduction of the APD by incubation of the muscle in medium containing 5 mM glucose, adjustment of the glucose concentration to 50 mM will cause restoration of normal APD. Phlorizin has been shown to competitively interfere with the effect of glucose on the APD and i

Scutellariae Radix and Coptidis Rhizoma Improve Glucose and Lipid Metabolism in T2DM Rats via Regulation of the Metabolic Profiling and MAPK/PI3K/Akt Signaling Pathway.

Aim Scutellariae Radix (SR) and Coptidis Rhizoma (CR) have often been combined to cure type 2 diabetes mellitus (T2DM) in the clinical practice for over thousands of years, but their compatibility mechanism is not clear. Mitogen-activated protein kinase (MAPK) signaling pathway has been suggested to play a critical role during the process of inflammation, insulin resistance, and T2DM. This study was designed to investigate their compatibility effects on T2DM rats and explore the underlying mechanisms by analyzing the metabolic profiling and MAPK/PI3K/Akt signaling pathway. Methods The compatib

Management of acute infusion-related reactions in AAV gene therapy guided by mechanistic insight.

Infusion-related reactions (IRRs), serious immune events that occur either during or within six hours of an infusion, can occur with adeno-associated virus (AAV) gene therapy. IRRs may be attributed to IgE-mediated hypersensitivity, leading to treatment discontinuation due to concern about progressive, life-threatening allergic reactions. IRRs were observed in two participants who received an AAV8 gene therapy (DTX401) for glycogen storage disease type Ia and in one participant who received an AAV9 gene therapy (UX701) for Wilson's disease. All of these IRRs were associated with a rapid infusi

Revisiting Inherited Metabolic Diseases (IMDs) Through the Lens of Selective Autophagy.

Selective autophagy is increasingly recognized as a fundamental quality-control mechanism that complements bulk autophagy by enabling the targeted degradation of specific intracellular cargos, including mitochondria, endoplasmic reticulum (ER), lipid droplets, glycogen, peroxisomes, ferritin, ribosomes, and protein aggregates. Cargo selectivity is mediated by selective autophagy receptors (SARs), which link damaged or superfluous substrates to the autophagic machinery through LC3/GABARAP interactions, thereby coordinating cargo sequestration. Recent studies have revealed substantial mechanisti

Exercise combined with creatine supplementation for skeletal muscle hypertrophy: mechanistic basis, nutritional context, and applications.

Creatine supplementation is one of the most widely studied nutritional strategies for enhancing resistance-training adaptations and skeletal muscle development. This narrative review evaluates the evidence linking creatine supplementation with skeletal muscle hypertrophy, integrating mechanistic insights, nutritional considerations, and practical applications. Current evidence indicates that creatine primarily supports hypertrophic adaptation by increasing phosphocreatine availability and enhancing the capacity to perform high-intensity training, thereby increasing the mechanical stimulus for

A simple mechanistic model for insulin-glucose-glucagon dynamics and its implications for diabetes management.

Insulin and glucose dynamics are tightly regulated by the pancreatic islets of Langerhans, which contain beta cells that produce insulin and alpha cells that produce glucagon. Insulin lowers blood glucose by enabling cells to access glucose for energy, while glucagon raises blood glucose levels by stimulating the liver to produce glucose from non-carbohydrate sources as well as breaking down stored glycogen. Maintaining safe blood glucose levels is crucial, as both high (hyperglycemia) and low (hypoglycemia) levels can lead to life threatening consequences. In this work, a simple three-state m

SINEUP-mediated enhancement of HNF4α improves metabolic function in Huh7-based bioengineered hepatic microtissues.

BACKGROUND/AIMS: Human liver cell-based drug-screening platforms require stable hepatocyte identity and metabolic competence, but Huh7 cells show reduced hepatic function. We investigated whether SINEUP-mediated enhancement of hepatocyte nuclear factor 4 alpha (HNF4α) could improve the metabolic performance of Huh7-based three-dimensional hepatic microtissues. METHODS: Decellularized liver extracellular matrix-derived microparticles were incorporated into three-dimensional microtissues containing Huh7 cells, human umbilical vein endothelial cells, and Wharton's jelly mesenchymal stem cells us
