Lymphoplasmacytic lymphoma (LPL) that does not fulfill the diagnostic criteria for Waldenström's macroglobulinemia (WM), termed non-IgM type LPL, represents a rare entity that poses significant diagnostic challenges. Here, we report a rare case of CD19-negative non-IgM type LPL. A 69-year-old man was incidentally found to have elevated M-protein and globulin levels during a routine health examination. Subsequent evaluation revealed elevated serum IgG, an IgG-λ monoclonal protein, and markedly elevated urinary λ free light chains. Bone marrow biopsy demonstrated normocellular marrow with approx

A 40-year-old Japanese woman with Philadelphia chromosome-negative B-cell acute lymphoblastic leukemia (B-ALL) underwent human leukocyte antigen (HLA)-matched related peripheral blood stem cell transplantation approximately 22 months before the current transplantation because measurable residual disease persisted after induction therapy. She experienced hematologic relapse approximately six months before the current transplantation and achieved complete remission after two cycles of inotuzumab ozogamicin. A second transplantation using her HLA-haploidentical son as the donor was planned; howev

Despite the therapeutic breakthroughs achieved over the years, breast cancer (BC) continues to rank as the deadliest tumor among women worldwide. Therefore, prevention and screening programs remain crucial for the successful recovery of patients with BC. Monocyte distribution width (MDW) is a novel hematological parameter provided along with the complete blood count by DxH haematology analyzers. Variations in MDW have been associated with diagnostic and prognostic significance in sepsis, viral infections and other inflammation-related diseases. However, the potential role of MDW remains largel

Genetic polymorphisms contribute to inter-individual variability in cardiometabolic risk and quality-of-life outcomes, yet their clinical relevance often remains unclear due to population heterogeneity and reliance on single-variant analyses. Integrative approaches combining genetic and phenotypic data may improve the characterization of complex disease profiles, particularly in orthopedic populations burdened by cardiometabolic comorbidities. This study included 289 patients scheduled for orthopedic surgery. Polymorphisms in the vitamin D receptor (VDR; ApaI, FokI, BsmI), catechol-O-methyltra

Acute myeloid leukemia (AML) is a hematological malignancy characterized by clonal expansion of myeloid progenitors in the bone marrow, resulting in impaired hematopoiesis. AML remains challenging due to frequent disease recurrence and limited treatment alternatives, highlighting the necessity for further research. Corilagin, an ellagitannin isolated from ethnopharmacological plants, displays potential anti-cancer activity, but its anti-leukemic effects and mechanism in AML are unclear. This study aimed to assess whether corilagin induces cell death in AML cells, a cell-derived xenograft (CDX)

BACKGROUND: Granulocyte-macrophage colony-stimulating factor (GM-CSF), aldesleukin and isotretinoin are unavailable in Japan, necessitating alternative cytokines for dinutuximab immunotherapy. We compared the efficacy of a regimen containing granulocyte colony-stimulating factor (G-CSF)/teceleukin (regimen A) to that with GM-CSF/aldesleukin/isotretinoin (regimen B) in children with newly diagnosed high-risk neuroblastoma.
METHODS: After completing initial therapy including high-dose chemotherapy followed by autologous stem cell transplantation and radiotherapy, children with non-progressive d

Acute myeloid leukemia (AML) is a heterogeneous hematological malignancy characterized by extensive genomic alterations and molecular diversity, posing significant therapeutic challenges. Current treatment strategies lack precise predictive biomarkers for chemotherapy response, highlighting the need for tools to guide precision therapy. In this study, we evaluated in vitro chemotherapy sensitivity in bone marrow samples from 98 AML patients using the PharmaFlow platform. Sensitivity to 10 commonly used chemotherapeutic agents (including venetoclax) and 20 combination regimens was assessed. Pat

Systemic lupus erythematosus (SLE) is a chronic, multisystem autoimmune disease that disproportionately affects women of reproductive age and is associated with substantial morbidity, premature mortality, and racial and ethnic disparities. Advances in diagnosis and immunosuppressive therapy have improved survival, yet patients remain at increased risk for kidney failure, cardiovascular disease, infection, pregnancy complications, and treatment related damage. Primary care and hospital-based clinicians are often the first point of contact and are central to early recognition, evaluation, and co

BACKGROUND: Systemic lupus erythematosus (SLE) is characterized by fluctuating activity and unpredictable flares that contribute to cumulative organ damage. Biomarkers capable of identifying patients at risk of near-term disease reactivation are needed. Interleukin-6 (IL-6) has been associated with SLE activity, but its prospective predictive value remains uncertain.
METHODS: We conducted a prospective, longitudinal, single-center study (2017-2023) including 188 adult with SLE. Serum IL-6 levels were measured at routine and unscheduled visits. Patients were categorized using a prespecified cu

The transcription factor IKAROS plays an important role in lymphocyte development, differentiation, and as a tumor suppressor. To date, >70 IKAROS germ line heterozygous variants have been reported in patients with primary immunodeficiency (PID)/inborn errors of immunity (IEI) and leukemia, and this number continues to grow. Germ line IKAROS loss- and gain-of-function mutations have been linked to immunodeficiency, immune dysregulation, and hematologic malignancies, with a broad spectrum of clinical manifestations. Routine next-generation sequencing approaches in patients with PID/IEI have fac

BACKGROUND: It is extremely rare for monozygotic twins to both develop leukemia. This paper reports a clinical case in which a pair of monozygotic twins with germline ETV6 gene mutations successively developed acute lymphoblastic leukemia (B-cell type, ALL-B). The ETV6:c.744del (p.S248fs) is a novel germline mutation associated with familial susceptibility to hematologic malignancies. This mutation site has not been detected in relevant databases or literature, and this is the first international report of it. It provides reference value for genetic screening of such diseases.

A 46-year-old man with non-Hodgkin lymphoma (NHL) in remission since 2014 presented with progressive dizziness, fatigue, and dark-colored urine. Laboratory workup revealed hemoglobin levels of 5.9 g/dL, elevated lactate dehydrogenase (LDH), hyperbilirubinemia, low haptoglobin, and a positive direct Coombs test for IgG, C3, and poly-specific antibodies, confirming warm autoimmune hemolytic anemia (wAIHA). Immunoglobulin profiling showed low IgM and IgA with high-normal IgG, suggesting chronic immune dysregulation, whereas abdominal imaging revealed mesenteric calcifications, raising concern for

Cold agglutinins can result in red blood cell (RBC) agglutination and lysis at low temperatures. Temperature sensitivity during stem cell mobilization, processing, and infusion poses unique procedural challenges in patients with active cold agglutinins. A 49-year-old man presented at an outside hospital with severe hemolysis and was found to have an anti-I cold agglutinin and a component of warm autoimmune hemolytic anemia, together with a positive COVID and Rhinovirus test. He was treated with RBC transfusions, steroids, and Rituximab. Bone marrow evaluation several months later at our instit

The recombination activating gene 1 (RAG1) is essential for V(D)J recombination and lymphocyte development. While biallelic null RAG1 mutations cause severe combined immunodeficiency (SCID), hypomorphic variants have increasingly been associated with immune dysregulation and hematologic malignancies. This study aimed to present a pediatric case of Epstein-Barr virus (EBV)-negative Burkitt lymphoma carrying a novel homozygous RAG1 variant and to discuss its potential association with immune function and malignancy risk. A 9-year-old Turkish male from a consanguineous family was evaluated for he

BACKGROUND: Oral malignancies in the paediatric population are rare, but if left untreated, the patient's prognosis may be altered. The current literature is varied in its findings regarding common sites and types of tumours found in the paediatric population. Our goal was to describe the distribution of paediatric head and neck cancers, test associations, quantify temporal trends and perform survival analyses. We then compared our results with the current literature.
METHODS: Approval for the study was given by the Hadassah Medical Organization Helsinki Committee (HMO0792-20). We obtained da
