Crosstalk between integrin signaling and NAD⁺ biosynthetic pathways promotes glycolysis, proliferation, survival, and tumor growth in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype with limited therapeutic options. Here, we investigated how integrin-dependent signaling pathways regulate tumor metabolism and therapeutic vulnerability in TNBC. Pharmacological inhibition of the integrin/FAK axis and/or BRD4 induced cell cycle arrest, autophagy, and senescence in highly proliferative cells, consistent with a metabolic stress phenotype. Metabolomic analyses using [U-¹³C]-glucose revealed a marked suppression of glycolytic carbon flux, accompanied by an approximately 30-47% reduction in intracellular N

Multi-omics reveals rutin directly targets RUNX1 to disrupt the RUNX1/TET2 complex and alleviate NAFLD via TLR4/NF-κB inhibition.

Non-alcoholic fatty liver disease (NAFLD) is a prevalent liver disorder driven by metabolic dysregulation and chronic inflammation, for which targeted pharmacotherapies remain limited. Rutin, a bioactive flavonoid from Sophora japonica and Fagopyrum esculentum, possesses notable anti-inflammatory and antioxidant properties. This study explored its pharmacological effects and underlying mechanism in NAFLD using a combination of in vivo and in vitro approaches. We found that rutin administration markedly attenuated hepatic steatosis, reduced oxidative stress, restored mitochondrial function, and

Foliar metal micronutrients reshape rhizosphere soil multifunctionality by filtering microbial life-history strategies.

Foliar application of metal micronutrients is increasingly adopted in intensive cultivation systems, yet its potential ecological risks to rhizosphere functions remain poorly understood. Here, using the medicinal plant Panax notoginseng as a model, we conducted a gradient foliar amendment experiment with iron (Fe), zinc (Zn), and copper (Cu) to evaluate how aboveground metal inputs regulate rhizosphere soil multifunctionality (MF) through microbial life-history strategies. By integrating 16S rRNA amplicon sequencing, metagenomics, root transcriptomics, and a newly developed quantitative Yield-

PDA-Based Nanoplatform with High Ligustilide Loading for Simultaneous Elimination of Breast Cancer Cells and Stem Cells Via BAX/BCL-2/Caspase-3 Pathway Activation.

Breast cancer (BC) remains the most prevalent and deadly malignancy among women worldwide. While photothermal therapy (PTT) and chemodynamic therapy (CDT) offer tumor-specific and minimally invasive advantages, their standalone efficacy is limited. Ligustilide (LIG) can exert antitumor effects by generating •O2-, but its bioavailability is poor, thus necessitating improved delivery strategies. Here, we reported a multifunctional nanoplatform (FeD@LIG) integrating PTT, CDT, and chemotherapy for the simultaneous elimination of breast cancer stem cells (BCSCs) and non-BCSCs. FeD@LIG was synthesiz

Impact of silk-derived sericin on the formation and metformin sensitivity of breast cancer-fibroblast heterotypic spheroids.

The development of physiologically relevant three-dimensional (3D) tumor models is essential for accurately mimicking the tumor microenvironment (TME) and improving the predictive value of in vitro drug testing. This study evaluated the effect of culture media supplementation with sericin, a silk-derived glycoprotein, in the formation of heterotypic spheroids based on MCF-7 breast cancer cells and human fibroblasts. Spheroids were assembled under orbital agitation with sericin concentrations ranging from 0.025 to 0.2% and cultured for 14 days. Computational fluid dynamics simulations indicate

Clemastine ameliorates ulcerative colitis-induced cognitive impairment by restoring PI3K/Akt/GSK-3β signaling and suppressing ferroptosis.

Ulcerative colitis (UC) is a refractory inflammatory bowel disease with ongoing colonic inflammation and extra-intestinal manifestations, including cognitive impairment. In this study, we evaluated the peripheral and central effects of clemastine on cognitive impairment induced by UC using an acetic acid (1 ml, 4% v/v) model with emphasis on the role of PI3K/Akt/GSK-3β signaling, ferroptosis, and autophagy in its possible mediated neuroprotection. Clemastine revealed a dose-dependent improvement of colonic damage linked with UC, as demonstrated by inhibition of TNF-α, IL-1β, and caspase-3 leve

[Research advances in mechanisms and detection methods of vascular endothelial barrier dysfunction in sepsis-induced organ injury].

Sepsis-induced multiple organ dysfunction syndrome (MODS) is the leading cause of mortality in critically ill patients, and widespread disruption of the vascular endothelial barrier represents the central pathological event. The integrity of the endothelial barrier is crucial for maintaining microcirculatory homeostasis. In sepsis, the dysregulated inflammatory response leads to a dramatic increase in microvascular permeability through multiple mechanisms, including endothelial cell activation, oxidative stress, disruption of intercellular junctions, degradation of the glycocalyx, and the prom

Proteomic and phenotypic profiling of replicative-senescent human foreskin fibroblasts under brief heat shock.

As the largest human organ, the skin experiences lifelong exposure to intrinsic/extrinsic factors that over time diminish its functional capacity and structural integrity. Skin aging involves cellular dysfunction and the loss or fragmentation of extracellular matrix (ECM) fibers, clinically presenting as wrinkles, slackening, and pigmentary abnormalities. The heat shock response (HSR) is a gene regulatory program that controls the expression of molecular chaperones associated with aging, cancer, and neurodegenerative disorders. By maintaining cellular homeostasis and facilitating DNA repair, H

Functions and interactions of important sulfur-containing metabolites in plant development and stress responses.

Sulfur (S), an essential macroelement, plays important roles in plant numerous physiological processes. Through sulfur assimilation pathways, plants biosynthesize various S-containing compounds, including the gasotransimitter hydrogen sulfide (H2S), the primary organic S-containing amino acid cysteine (Cys), the master antioxidant glutathione (GSH), and the characteristic secondary metabolites such as glucosinolates (GSLs). Current studies have established the wide involvement of S-containing metabolites in orchestrating plant development and stimuli responses, however, the interplay and cross

Living on the edge: agricultural land use increases genotoxic damage in amphibians on southern brazilian wetlands.

Southern Brazil is a key agricultural region in South America, where wetland ecosystems are increasingly impacted by irrigated rice cultivation. Amphibians are particularly sensitive to environmental contamination due to their permeable skin and biphasic life cycle, making them effective bioindicators of ecosystem health. This landscape forms a mosaic of croplands and natural habitats, exposing wildlife to agrochemical contamination. In this study, we evaluated erythrocyte nuclear abnormalities (NAs) in the arboreal amphibian Dendropsophus sanborni and the aquatic amphibian Pseudis minuta, bot

Edible lily bulbs (Lilium lancifolium Thunb.) are highly valued for their rich nutritional profile and significant medicinal properties. To further enhance these beneficial attributes, this study investigates a novel, green post-harvest enhancement strategy using low-dose 60Co-γ irradiation (2-5 Gy) followed by immediate freeze-drying. Experimentally, fresh bulbs were exposed to varying irradiation doses, after which physiological parameters (enzyme activities, phytochemical contents) and metabolic profiles (via LC-MS/MS) were systematically evaluated. The results demonstrated that 4 Gy repres

Association of the endosomal TLR4 pathway with dorsolateral prostatic remodeling induced by perinatal phthalate mixture exposure in rats.

Phthalates are endocrine disruptors that alter the homeostasis of hormone-dependent organs. Perinatal exposure to endocrine disruptors can program different organs and promote morphological and metabolic alterations, increasing susceptibility to diseases later in life, as proposed by the Developmental Origins of Health and Disease (DOHaD) hypothesis. This study evaluated the effects of perinatal exposure to a six-phthalate mixture on prostatic structure and molecules related to inflammation and oxidative stress. Pregnant Sprague-Dawley rats received the mixture from gestational day 10 to postn
