Chronic Dietary PhIP Exposure Disrupts Brain Homeostasis in Adult Zebrafish through Dose-Dependent Proteostatic and Clock-Associated Remodeling.

Heterocyclic aromatic amines formed during high-temperature cooking are persistent dietary contaminants, and 2-amino-1-methyl-6-phenylimidazo[4,5-b] pyridine (PhIP) has emerged as a potential neurotoxicant beyond its established carcinogenicity. Adult zebrafish were fed PhIP for 90 days at 0.006, 0.4, and 7.2 mg/kg body weight/day. Chronic exposure impaired locomotion and induced brain injury, oxidative imbalance, and glial reactivity. Transcriptomic analyses revealed dose-dependent remodeling, with low exposures primarily engaging endoplasmic reticulum proteostasis, intermediate dosages enric

Hypersensitive Detection of Neurotransmitters in Biological Media by Optically Enhanced Benchtop NMR Spectroscopy.

Liquid-state Nuclear Magnetic Resonance (NMR) is an invaluable tool to gain atomic-resolution insights onto molecular structure and dynamics. The impact of NMR spectroscopy, however, is curtailed by high costs and hard-to-maintain high-field magnets. While low-field benchtop NMR spectrometers may address the challenge, severe trade-offs in sensitivity and resolution limit the appeal of this avenue. Here, we introduce a set of novel optically enhanced NMR pulse sequences and procedures (hyperpolarization toolkit) to readily generate 13 C and 1 H nuclear-spin hyperpolarization in situ on benchto

Circadian rhythms and microbiota: molecular crosstalk and its implications for health and disease.

Circadian rhythms, evolutionarily conserved 24-hour oscillations, exert precise regulatory control over microbial communities across host niches including the gastrointestinal tract, oral cavity, urinary bladder, and skin. This bidirectional interplay is critical to host physiology: host circadian clocks shape the composition and functional rhythms of resident microbiota, while microbiota-derived signals reciprocally modulate circadian entrainment and tissue-specific rhythmicity. Circadian disruption from shift work, irregular feeding, light pollution, or sleep deprivation trigger microbial dy

Inflammation impairs fertility and oocyte quality in systemic lupus erythematosus by triggering oxidative stress-mediated ferroptosis.

Systemic lupus erythematosus (SLE) is an autoimmune disorder associated with impaired female fertility, as suggested by our previous clinical cohort data. However, the intrinsic mechanisms underlying oocyte quality deterioration remain incompletely understood. Using a chronic SLE mouse model, we show that SLE-associated inflammation is accompanied by impaired oocyte maturation, reduced developmental competence, and mitochondrial dysfunction, together with increased oxidative stress and ferroptosis-related alterations, an iron-dependent process characterized by lipid peroxidation. Pharmacologic

Boosting kynurenic acid in kombucha via substrate selection: metagenomic and biochemical insights.

Kombucha is gaining global popularity for its health benefits. This study explored the use of chestnut honey, a rich source of kynurenic acid (KYNA), to produce kombucha enriched with this metabolite. Five variants were prepared using different green/black tea blends and carbon sources: white sugar or acacia honey (controls) versus chestnut honey. Samples were analyzed for tryptophan metabolites, physicochemical properties, and microbial diversity. Komagataeibacter and Enterobacter were predominant bacterial genera in SCOBY. Candida and Aspergillus were predominated in the single sample analyz

Biological rhythms, sleep disturbances, and the skin - a narrative review.

Sleep, circadian rhythms, light exposure, and environmental factors are increasingly recognized as important regulators of skin physiology. The skin contains peripheral clocks and exhibits circadian rhythmicity in barrier function, immune activity, pigmentation, temperature, blood flow, and repair processes. This review aims to summarize the current evidence linking sleep disturbances, circadian regulation, light exposure, seasonal variation, environmental temperature, and circadian desynchronization with skin physiology and some dermatoses. Sleep loss and disturbances in sleep duration may al

Sleep matters: Supporting healthy sleep for children and youth with neurodevelopmental disabilities (NDDs).

Children with neurodevelopmental disabilities are at high risk for sleep problems, which can negatively affect their health and that of their families. Improving sleep may be one of the most effective ways to improve behaviour, mood, positive social interaction, attention and learning, and reduce future risks for poor metabolic and mental health. While insomnia is the most common concern, increased rates of other sleep disorders are also found in this population. Sleep problems require prompt identification and intervention, which includes recognizing or ruling out contributory medical conditi

Clinical relevance of circadian clocks within the orofacial scope of sleep medicine: Implications for diagnosis, therapeutic and prophylactic measures.

Circadian clocks regulate sleep-wake patterns, metabolism, autonomic tone, and painperception. Their disruption contributes to multiple pathologies, including sleepdisorders and cardiovascular dysfunction. Within the orofacial domain, circadianrhythms influence the timing, intensity, and interaction of conditions such as insomnia, obstructive sleep apnea, sleep bruxism, orofacial pain and temporomandibulardisorders. This review outlines how circadian mechanisms and the orexinergic system provide anintegrative framework for understanding these orofacial sleep disorders and theirsystemic consequen

Analysis of sleep medication-induced parasomnias using the Japanese Adverse Drug Event Report database.

The association between sleep medication and parasomnia remains unclear. However, some sleep medications may also cause parasomnia. Therefore, we examined the association between sleep medication use and parasomnia. We performed an analysis using cases registered in the Japanese Adverse Drug Event Report Database from April 2004 to March 2022. We calculated the unadjusted reporting odds ratio (ROR) and adjusted it to consider confounding factors. The ROR for each sleep medication was significant: benzodiazepines (ROR 3.55, 95% confidence interval [CI] 2.39-5.27), non-benzodiazepines (ROR 14.21

Exploring nationwide patterns of sleep problems from late adolescence to adulthood using machine learning.

Sleep problems among young adults pose a major public health challenge. Leveraging nationwide health surveys and registers from Denmark, we investigated patterns of sleep problems from late adolescence to adulthood and explored early life-course determinants. We generated life-course embeddings using unsupervised machine learning on data from 2.2 million individuals born from 1980 to 2015. We used this landscape to identify neighboring factors of sleep problems. We observed a substantial increase in self-reported sleep problems among individuals aged 15 to 45, from 34 to 49% between 2010 and 2

Impacts of Maternal Preeclampsia Exposure on Offspring Neuronal Development: Recent Insights and Interventional Approaches.

Preeclampsia, a hypertensive disorder during pregnancy, frequently correlates with adverse neurological outcomes in offspring, including cognitive impairments, autism spectrum disorder, depressive disorder, attention deficit hyperactivity disorder, and cerebral palsy. Despite these known consequences, the understanding of neuronal damage in the offspring of preeclamptic mothers remains insufficient. Here, we review the neuronal abnormalities resulting from maternal preeclampsia exposure, which include disrupted neurogenesis, loss of neuronal cell integrity, accumulation of cellular debris, dec

Targeting microbiota to alleviate the harm caused by sleep deprivation.

Sleep deprivation has become a common health hazard, affecting 37-58% of the population and promoting the occurrence and development of many diseases. To date, effective treatment strategies are still elusive. Accumulating evidence indicates that modulating the intestinal microbiota harbors significant potential for alleviating the deleterious impacts of sleep deprivation. This paper first reviews the effects of sleep deprivation on gastrointestinal diseases, metabolic diseases, and neuropsychiatric diseases, discussing its specific mechanisms of influence. We then focus on summarizing existin

Continuous positive airway pressure as an accurate marker for non-24-hour sleep-wake rhythm disorder.

Non-24-h sleep-wake rhythm disorder is quite rare in sighted patients and frequently associated with psychiatric disorders. We report the case of a 46-year-old man with autism spectrum disorder (ASD) and agoraphobia who had been referred for a suspicion of obstructive sleep apnea syndrome (OSAS). Polysomnography and arterial blood gas confirmed moderate OSAS associated with hypoventilation. Continuous positive airway pressure (CPAP) was started on fixed mode with excellent results. At follow-up, his CPAP report data revealed an irregular sleep-wake rhythm with a progressive offset of sleep sch

Natural products for male androgenetic alopecia.

Androgenetic alopecia (AGA) is the most common cause of hair loss in men, often requiring medical attention. The US FDA approved topical minoxidil and oral finasteride to treat AGA. Topical minoxidil requires a long-term application to observe improvement; oral finasteride may cause undesirable side effects. Therefore, natural products may be an alternative when patients are skeptical about these two conventional treatments. Physicians may also suggest natural products in conjunction with topical minoxidil or oral finasteride to enhance clinical outcomes. This article reviews the prospect of n

Seasonal reproduction and gonadal function: a focus on humans starting from animal studies.

Photoperiod impacts reproduction in many species of mammals. Mating occurs at specific seasons to achieve reproductive advantages, such as optimization of offspring survival. Light is the main regulator of these changes during the photoperiod. Seasonally breeding mammals detect and transduce light signals through extraocular photoreceptor, regulating downstream melatonin-dependent peripheral circadian events. In rodents, hormonal reduction and gonadal atrophy occur quickly and consensually with short-day periods. It remains unclear whether photoperiod influences human reproduction. Seasonal fl

Acute and Prophylactic Management of Postictal Agitation in Electroconvulsive Therapy.

Postictal agitation (PIA) is a common adverse effect of electroconvulsive therapy (ECT), a treatment used for a variety of psychiatric disorders. Because of risk of harm to patients and health providers when patients develop PIA, its acute management and prophylaxis are of vital importance for ECT practitioners. This article describes PIA risk factors, as well as practical steps to manage this ECT complication. Nonpharmacologic patient safety interventions are critical components of PIA management. Benzodiazepines, antipsychotics, and additional anesthetic doses are discussed as acute treatmen

Pineal Gland from the Cell Culture to Animal Models: A Review.

This review demonstrates current literature on pineal gland physiology, pathology, and animal model experiments to concisely explore future needs in research development with respect to pineal gland function and neuro-regenerative properties. The pineal gland plays an integral role in sleep and recovery by promoting physiologic circadian rhythms via production and release of melatonin. Yet, the current literature shows that the pineal gland has neuroprotective effects that modulate both peripheral and central nerve injuries through several direct and indirect mechanisms, such as angiogenesis a

Role of Adaptor Protein Myeloid Differentiation 88 (MyD88) in Post-Subarachnoid Hemorrhage Inflammation: A Systematic Review.

Myeloid differentiation 88 (MyD88) is a well-established inflammatory adaptor protein. It is one of the essential downstream proteins of the toll-like receptor 4 (TLR4) signaling pathway. TLRs are pattern recognition receptors that are usually activated by the damage-associated molecular pattern molecules (DAMPs). Sterile inflammation is triggered by the endogenous DAMPs released in response to global cerebral ischemia and from extravasated blood after subarachnoid hemorrhage (SAH). In this review, we highlight the importance of the neuroinflammatory role of the MyD88 in the SAH. We also explo

Circadian clock mechanism driving mammalian photoperiodism.

The annual photoperiod cycle provides the critical environmental cue synchronizing rhythms of life in seasonal habitats. In 1936, Bünning proposed a circadian-based coincidence timer for photoperiodic synchronization in plants. Formal studies support the universality of this so-called coincidence timer, but we lack understanding of the mechanisms involved. Here we show in mammals that long photoperiods induce the circadian transcription factor BMAL2, in the pars tuberalis of the pituitary, and triggers summer biology through the eyes absent/thyrotrophin (EYA3/TSH) pathway. Conversely, long-dur
