SNAP23 deficiency triggers Trim21 mitochondrial translocation to suppress TFAM-mediated oxidative metabolism and drive chemoresistance in colorectal cancer.

Chemoresistance is a major cause of poor prognosis in colorectal cancer (CRC), and its molecular mechanisms urgently need elucidation. The cell membrane protein SNAP23, known for its role in vesicle secretion, also promotes CRC cell growth. However, its role in tumor chemotherapy remains unclear. This study reveals a novel function of SNAP23, independent of vesicle transport, mediating crosstalk between the cell membrane and mitochondria to influence the chemotherapeutic response to oxaliplatin (OXA). Mechanistically, SNAP23 arrests Trim21, causing its accumulation near the cell membrane and a

Hydrophilic polymeric fluorescent probes based on benzothiadiazole constructed for real-time monitoring of lipid droplet levels in cells.

Lipid droplets (LDs) are vital organelles that regulate neutral-lipid storage and metabolism, membrane architecture, and inter-membrane trafficking. Quantifying LD heterogeneity is essential for elucidating determinants of LD biogenesis and their roles in metabolic disorders and cancer. Here, we report a water-soluble polymeric probe, NBMA-co-PEGMA400, constructed by employing benzothiazole as the fluorescent core and hydrophilic PEGMA400 as the solubilizing segment via reversible addition-fragmentation chain-transfer (RAFT) polymerization. The probe exhibits pronounced solvatochromism, reflec

Efflux pumps and membrane permeability contribute to intrinsic antibiotic resistance in Mycobacterium abscessus.

Mycobacterium abscessus is a pulmonary pathogen that exhibits intrinsic resistance to antibiotics, but the factors driving this resistance are incompletely understood. Insufficient intracellular drug accumulation could explain broad-spectrum resistance, but whether antibiotics fail to accumulate in M. abscessus and the mechanisms required for drug exclusion remain poorly understood. We measured antibiotic accumulation in M. abscessus using mass spectrometry and found a wide range of drug accumulation across clinically relevant antibiotics. Of these compounds, linezolid accumulates the least, s

Roles of Y-27632 on sheep sperm metabolism.

To investigate the effect of Y-27632 on low-temperature metabolism of sheep sperm, different concentrations of Y-27632 were added to sheep semen at 4 °C in this experiment to detect indicators such as sperm motility, plasma membrane, acrosome, antioxidant performance, mitochondrial membrane potential (MMP), and metabolomics. The results showed that the addition of 20 µM Y-27632 significantly increased sperm motility, plasma membrane integrity rate, acrosome integrity rate, antioxidant capacity, MMP level, significantly increased sperm adenosine triphosphate (ATP) and total cholesterol content,

Aspirin Causes Lipid Accumulation and Damage to Cell Membrane by Regulating DCI1/OLE1 in Saccharomyces cerevisiae.

Aspirin is one of the most commonly used nonsteroidal anti-inflammatory drugs. Various potential pharmacological effects of aspirin, such as anticancer, antibacterial activity, and prolonging life expectancy have been discovered. However, the mechanism of aspirin is not fully elucidated. Herein, the effects of aspirin on fatty acid metabolism in yeast cell model Saccharomyces cerevisiae were studied. The results showed that aspirin can induce lipid accumulation and reduce the unsaturated fat index in cells. The assessment of cell membrane integrity demonstrated that aspirin caused damage to th

Healing Mechanism of Ruptured Fetal Membrane.

Preterm premature rupture of membranes (pPROM) typically leads to spontaneous preterm birth within several days. In a few rare cases, however, amniotic fluid leakage ceases, amniotic fluid volume is restored, and pregnancy continues until term. Amnion, the collagen-rich layer that forms the load-bearing structure of the fetal membrane, has regenerative capacity and has been used clinically to aid in the healing of various wounds including burns, diabetic ulcers, and corneal injuries. In the healing process of ruptured fetal membranes, amnion epithelial cells seem to play a major role with assi

Paraneoplastic systemic lupus erythematosus associated with colorectal cancer.

A 64-year-old gentleman initially presented with nephrotic syndrome and membranous nephropathy with positive staining for C1q, which was suspicious for lupus membranous nephritis. Investigation led to the simultaneous diagnosis of colorectal cancer (CRC). The CRC was surgically excised and the patient's nephrotic syndrome resolved. The patient subsequently presented with classic systemic lupus erythematosus (SLE) including positive serological markers, mouth-ulcers and a photosensitive maculopapular rash. Two months later the patient represented with an SLE flare encompassing the full-hand of

Contemporary Methods Allowing for Safe and Convenient Use of Amniotic Membrane as a Biologic Wound Dressing for Burns.

Partial-thickness burns involve damage to the upper layer of skin, which leaves nerve endings exposed, and therefore represent the most painful of several categories of thermal injuries. Historically, partial-thickness burns were treated conservatively by debriding the blisters, by daily tubbing and washing, and the application of new bandages with topical medications. Human amniotic membrane has been used for centuries as a biological wound dressing. In the past 20 years, there has been an increasing body of literature addressing the use of amniotic tissue in chronic wounds and burns. In this

Adenovirus membrane penetration: Tickling the tail of a sleeping dragon.

As is the case for nearly every viral pathogen, non-enveloped viruses (NEV) must maintain their integrity under potentially harsh environmental conditions while retaining the ability to undergo rapid disassembly at the right time and right place inside host cells. NEVs generally exist in this metastable state until they encounter key cellular stimuli such as membrane receptors, decreased intracellular pH, digestion by cellular proteases, or a combination of these factors. These stimuli trigger conformational changes in the viral capsid that exposes a sequestered membrane-perturbing protein. Th

Purification, refolding and characterization of the trimeric Omp2a outer membrane porin from Brucella melitensis.

Brucella melitensis is a gram-negative bacteria known to cause brucellosis and to produce severe infections in humans. Whilst brucella's outer membrane proteins have been extensively studied due to their potential role as antigens or virulence factors, their function is still poorly understood at the structural level, as the 3D structure of Brucella β-barrel membrane proteins are still unknown. In this context, the B. melitensis trimeric Omp2a porin has been overexpressed and refolded in n-dodecyl-β-d-maltopyranoside. We here show that this refolding process is insensitive to urea but is tempe

Ultrafiltration of natural organic matter and black carbon: factors influencing aggregation and membrane fouling.

There are concerns about black carbon (BC), due to its potential for sorption of toxic pollutants and inevitably entering drinking water sources. This study aimed to evaluate factors affecting BC aggregation and membrane fouling in the ultrafiltration of river water. Hydrophilic carbon black (CB) was selected as a surrogate of submicron BC in natural waters. Calcium, pH, and natural organic matter (NOM) were found to influence CB aggregation. Calcium induced interparticle interactions in a pH range of 4.3-7.7. In river water at 0.3mM Ca2+, CB remained as fine aggregates (<300 nm) that caused s

High-resolution x-ray structure of human aquaporin 5.

Human aquaporin 5 (HsAQP5) facilitates the transport of water across plasma membranes and has been identified within cells of the stomach, duodenum, pancreas, airways, lungs, salivary glands, sweat glands, eyes, lacrimal glands, and the inner ear. AQP5, like AQP2, is subject to posttranslational regulation by phosphorylation, at which point it is trafficked between intracellular storage compartments and the plasma membrane. Details concerning the molecular mechanism of membrane trafficking are unknown. Here we report the x-ray structure of HsAQP5 to 2.0-A resolution and highlight structural si

Oxidized phospholipids: from molecular properties to disease.

Oxidized lipids are generated from (poly)unsaturated diacyl- and alk(en)ylacyl glycerophospholipids under conditions of oxidative stress. The great variety of reaction products is defined by the degree of modification, hydrophobicity, chemical reactivity, physical properties and biological activity. The biological activities of these compounds may depend on both, the recognition of the particular molecular structures by specific receptors and on the unspecific physical and chemical effects on their target systems (membranes, proteins). In this review, we aim at highlighting the molecular featu

Altered cholesterol metabolism in Niemann-Pick type C1 mouse brains affects mitochondrial function.

Niemann-Pick type C1 (NPC1) disease is a fatal hereditary disorder characterized by a defect in cholesterol trafficking and progressive neurodegeneration. Although the NPC1 gene has been identified, the molecular mechanism responsible for neuronal dysfunction in brains of patients with NPC1 disease remains unknown. This study demonstrates that the amount of cholesterol within mitochondria membranes is significantly elevated in NPC1 mouse brains and neural cells. In addition, the mitochondrial membrane potential, the activity of ATP synthase, and henceforth the level of ATP are markedly decreas

10-alpha-Aminoacyl-9(10H)-anthracenones: inhibition of 12(S)-HETE biosynthesis and HaCaT cell growth.

1,8-Dihydroxy-9(10H)-anthracenones with a 10-alpha-aminoacyl group were synthesized using either a mixed-anhydride coupling method or Boc-protected oxazolidinediones. The novel anthracenones were evaluated as inhibitors of the biosynthesis of 12(S)-hydroxyeicosatetraenoic acid (12(S)-HETE) in epidermal homogenate of mice and for inhibition of the growth of HaCaT keratinocytes. These cells were also tested for their susceptibility for the action of the most potent members of this series on plasma membrane integrity, in order to confirm that inhibition of cell growth is not a result of membrane

Control of Oxidative Stress and Intracellular Survival in Francisella tularensis Live Vaccine Strain (LVS) via Acyl-CoA Synthetase.

UNLABELLED: Francisella tularensis is a highly infectious, Gram-negative intracellular bacterium and the causative agent of tularemia, a potentially fatal disease. Owing to its low infectious dose, ease of aerosolization, high virulence, lack of an effective vaccine, and potential use as a bioterrorism agent, F. tularensis is classified by the CDC as a Tier 1 Category A Select Agent. Despite its clinical importance, the mechanisms underlying F. tularensis virulence remain incompletely understood. In this study, we generated a partial Tn5 transposon insertion mutant library in the F. tularensis

The amyloidogenic C-terminal region of TMEM106B modulates lipid membrane biophysical properties: Functional and pathological insights.

The lysosomal transmembrane protein 106B (TMEM106B) forms amyloid filaments in the human brain in an age-dependent manner, observed both in neurologically healthy individuals and in patients with neurodegenerative diseases also containing tau, α-synuclein, or TDP-43 inclusions. Despite its pathological and physiological relevance, the biochemical mechanisms governing TMEM106B structural stability and its functional interactions with membranes remain largely unknown. Here, we examined the luminal C-terminal fragment of TMEM106B (called TST, residues 120-254), corresponding to the amyloid fibril

In silico identification and biophysical characterization of candidate antimicrobial peptides from the Indian marine microbiome targeting multidrug-resistant ESKAPE pathogens.

The global health crisis of antimicrobial resistance necessitates the discovery of new antibacterial agents. Underexplored marine microbiomes, particularly from the biodiverse Indian coast, represent a rich potential source of antimicrobial peptides (AMPs). Targeting the urgent threat of multidrug-resistant ESKAPE pathogens, the present study aimed to computationally identify novel, membrane-active AMPs from these unique metagenomic datasets, with a focus on inhibiting Gram-negative bacteria. In this study, we computationally mined Indian marine high-resolution shotgun metagenomic datasets thr

In-Situ Preparation of Asymmetric Superwetting Laser-Induced Graphene Janus Membrane on Loofah for Efficient Solar Evaporation.

Laser-induced graphene (LIG), with its low-cost advantage, holds significant potential in the field of solar-driven interfacial evaporation. However, most current LIG-based photothermal materials typically require polymers as precursors, which not only raise concerns about environmental sustainability but also result in membranes that often lack interconnected pore structures and suitable wetting gradients. This limits evaporation efficiency and leads to salt crystallization and deposition. To address these bottlenecks, this study proposes a new strategy for constructing asymmetric superhydrop

Progesterone-KISS1 axis impairs amniotic epithelial cell function and promotes premature rupture of membranes.

BACKGROUND: Premature rupture of membranes (PROM) is a major cause of preterm birth and perinatal morbidity. Disruption of fetal membrane integrity is central to PROM pathogenesis; however, the molecular mechanisms underlying membrane weakening remain incompletely understood. METHODS: Proteomic analysis was performed on fetal membrane tissues from patients with early PPROM, late PPROM, full-term PROM, and full-term normal pregnancies. An LPS-induced infection-associated preterm birth mouse model was established to evaluate fetal membrane pathology, hormone levels, inflammatory responses, and
