Modified Xuan-Bi-Qing-Ying decoction alleviates septic liver injury by regulating autophagy and apoptosis via HIF-1α signaling pathway.

BACKGROUND: Xuan-Bi-Qing-Ying Decoction (XBQYD), a Traditional Chinese Medicine (TCM) formula modified based on clinical experience, has demonstrated efficacy and safety in the treatment of septic liver injury (SLI). METHODS: The absorbed components of XBQYD were characterized in vivo using LC-MS/MS. A rat model of septic liver injury (SLI) was then established by cecal ligation and puncture. To evaluate the preventive efficacy of XBQYD, rats were pretreated with the formula for seven days prior to injury induction. Subsequently, we evaluated liver histopathology (HE staining), measured serum

Deciphering the brain-gut axis: elucidating the link between cerebral cortex structures and functional gastrointestinal disorders via integrated Mendelian randomization.

BACKGROUND: Observational studies have suggested associations between functional gastrointestinal disorders (FGIDs) and variations in the cerebral cortex. However, the causality of these relationships remains unclear, confounded by anxiety and depression. To clarify these causal relationships and explore the mediating roles of anxiety and depression, we applied univariate, multivariable, and mediation Mendelian randomization (MR) analyses. METHOD: We utilized genome-wide association study (GWAS) summary data from the FinnGen database and the ENIGMA consortium, identifying genetic variants ass

Polygenic scores and Mendelian randomization identify plasma proteins causally implicated in Alzheimer's disease.

BACKGROUND: An increasing body of evidence suggests that neuroinflammation is one of the key drivers of late-onset Alzheimer's disease (LOAD) pathology. Due to the increased permeability of the blood-brain barrier (BBB) in older adults, peripheral plasma proteins can infiltrate the central nervous system (CNS) and drive neuroinflammation through interactions with neurons and glial cells. Because these inflammatory factors are heritable, a greater understanding of their genetic relationship with LOAD could identify new biomarkers that contribute to LOAD pathology or offer protection against it.

Repurposing antihypertensive drugs for pain disorders: a drug-target mendelian randomization study.

OBJECTIVE: Addressing the rising prevalence of pain disorders and limitations of current analgesics, our study explores repurposing antihypertensive drugs for pain management, inspired by the link between hypertension and pain. We leverage a drug-target Mendelian Randomization (MR) approach to explore their dual benefits and establish causal connections. METHODS: A comprehensive compilation of antihypertensive drug classes was undertaken through British National Formulary, with their target genes identified using the DrugBank database. Relevant single nucleotide polymorphisms (SNPs) associate

Causal association between tea intake and risk for gout: a Mendelian randomization study.

Background: Gout, an increasingly prevalent form of inflammatory arthritis, is caused by the accumulation of uric acid crystals in joints, resulting in severe pain, swelling and stiffness that adversely affect physical, mental and emotional wellbeing. The management of gout requires a combination of medication and lifestyle modifications. Recent studies suggest that tea intake may reduce the risk of developing gout; however, further research is needed to establish a causal relationship. Methods: In this study, we employed a bidirectional two-sample Mendelian randomization (MR) approach, utiliz

Causal effects of serum sex hormone binding protein levels on the risk of amyotrophic lateral sclerosis: a mendelian randomization study.

BACKGROUND: Extensive observational studies have suggested an association between serum sex-hormone binding globulin (SHBG) and Alzheimer's disease (AD); however, causality remains unclear. Furthermore, the effects on other neurodegenerative diseases have been poorly investigated. We aimed to explore the causal effects of genetically predicted SHBG serum levels on common neurodegenerative diseases. METHODS: A two-sample Mendelian randomization (MR) approach was used. Genetic variants of SHBG levels in the serum, detected using the chemiluminescent two-step sandwich immunoassay method, were id

Alcohol Intake and Hypertensive Disorders of Pregnancy: A Negative Control Analysis in the ALSPAC Cohort.

Background Alcohol intake increases blood pressure yet estimates of associations between maternal intake and hypertensive disorders of pregnancy (HDP) are sparse and range from null to a protective effect. Here we estimated the association of maternal drinking during pregnancy with preeclampsia and gestational hypertension (separately and jointly, as HDP). We used partner's alcohol intake as a negative control exposure, beverage type-specific models, and a range of sensitivity analyses to strengthen causal inference and reduce the influence of bias. Methods and Results We performed a longitudi

Mendelian randomization study of B-type natriuretic peptide and type 2 diabetes: evidence of causal association from population studies.

BACKGROUND: Genetic and epidemiological evidence suggests an inverse association between B-type natriuretic peptide (BNP) levels in blood and risk of type 2 diabetes (T2D), but the prospective association of BNP with T2D is uncertain, and it is unclear whether the association is confounded. METHODS AND FINDINGS: We analysed the association between levels of the N-terminal fragment of pro-BNP (NT-pro-BNP) in blood and risk of incident T2D in a prospective case-cohort study and genotyped the variant rs198389 within the BNP locus in three T2D case-control studies. We combined our results with ex

APOC1-Associated VLDL-Pathway Dysregulation in Papillary Thyroid Carcinoma: A Reproducible Cross-Cohort Transcriptomic Signature Associated With the Diagnostic-to-Treatment Interval.

The diagnostic-to-treatment interval (DTI) of papillary thyroid carcinoma (PTC) has long been regarded as an inactive administrative period. We expected to find that DTI is a window of high metabolic activity where apolipoprotein dysregulation may lead to harm. A four-phase integrated design was used. Phase 1 analysed transcriptomic and clinical data from TCGA-THCA (n = 209) to find DTI-associated molecular alterations, and used a maximally selected rank statistic to set the optimal time window. Phase 2 verified the lipoprotein signature in three independent GEO cohorts (GSE29265/GSE33630/GSE6

Regulatory effects of berberine on intestinal microecology in mice with ulcerative colitis.

BACKGROUND: Currently, therapeutic approaches for ulcerative colitis (UC), such as aminosalicylates, glucocorticoids, and biologics, exhibit certain efficacy but are hindered by limitations including side effects, high costs, or suboptimal responses. As a traditional Chinese medicine component, berberine (BBR) possesses anti-inflammatory properties and the ability to modulate the gut microbiota, with low toxicity, and may provide new directions for UC treatment in the future. METHODS: A mouse UC model was established via dextran sulfate sodium (DSS) induction, and dose-and time-dependent scre

Causal associations between insulin-like growth factor 1 and vitamin D levels: a two-sample bidirectional Mendelian randomization study.

BACKGROUND: Insulin-like growth factor 1 (IGF-1) plays a vital role in the attainment and maintenance of bone mass throughout life and is closely related to the stature of children. 25-Hydroxyvitamin D (25-OHD) is an intermediate of vitamin D (Vit D) metabolism and a key indicator of Vit D nutritional status. Multiple studies have revealed that IGF-1 levels undergo a non-significant increase after Vit D supplementation. Here, we analyzed the causal and reverse causal relationships between 25-OHD and IGF-1 levels using Mendelian randomization (MR). METHODS: Two-sample MR was used to estimate a

Polygenicity shapes distinct brain patterns and clinical profiles of bipolar disorder.

BACKGROUND: Bipolar disorder (BD) manifests both genetic predispositions and brain imaging abnormalities. Genetic analyses provide a powerful approach to disentangling the associations between BD and brain imaging-derived phenotype (IDP). However, in East Asian (EAS) populations, the genetic basis of these neuroimaging changes and their clinical relevance remains underexplored due to limited integrative evidence. METHODS: We investigated the genetic architecture and clinical relevance of BD and brain IDPs in East Asian populations using individual-level data from the Chinese Bipolar Disorder

Integrated single-cell and experimental validation links effector chondrocyte-associated oxidative stress to EIF6-related p47phox-NOX2 signaling in osteoarthritis.

BACKGROUND: Oxidative stress is closely involved in osteoarthritis (OA), but the cell-type-specific oxidative stress landscape and related regulatory molecules in human cartilage remain incompletely defined. This study aimed to identify effector chondrocyte (EC)-associated oxidative stress features in OA and to prioritize candidate molecules linked to p47phox-NOX2-related ROS responses. METHODS: Public human cartilage single-cell RNA sequencing datasets were integrated to construct an OA cartilage atlas. Oxidative stress activity was evaluated using multiple scoring methods, and high oxidativ

SLIT2 and ANGPTL3 as putative mediators linking obesity to atrial fibrillation: A Mendelian randomization study.

The observational association between obesity and atrial fibrillation has been well-established. However, its causal mediators underlying this association remain unclear. This study aimed to investigate the causal relationship between body mass index (BMI), as well as potential mediating factors underlying this association, using Mendelian randomization (MR). Genetically predicted BMI was significantly associated with an increased risk of atrial fibrillation (inverse-variance weighted odds ratio [OR] = 1.350, 95% confidence interval [CI] = 1.268-1.436, P < .001). Shared Genetic Risk Between Acute Pancreatitis and Metabolic Syndrome Converges on a BCAT1-Positive Fibroblast Subpopulation and MAPK14-Driven Inflammation.

Metabolic syndrome (MetS) has been associated with increased acute pancreatitis (AP) severity. This study aimed to identify shared differentially expressed genes (CDEGs) between MetS and AP and to prioritize mechanistically relevant targets. The shared genetic basis and causal effect of MetS on AP were first assessed using linkage disequilibrium score regression (LDSC) and two-sample Mendelian randomization (MR). Public Gene Expression Omnibus (GEO) datasets were analyzed to define shared CDEGs. A multi-step pipeline incorporating functional enrichment, protein-protein interaction (PPI) analys

Protective role of PCED1B -expressing naive CD4 + T cells in sepsis.

BACKGROUND: Sepsis is a global health challenge associated with high morbidity and mortality rates. Early diagnosis and treatment are challenging because of the limited understanding of its underlying mechanisms. This study aimed to identify effective biomarkers for diagnosing and treating sepsis through an integrated multi-method approach. METHODS: Publicly available single-cell RNA sequencing (scRNA-seq) and bulk RNA-seq datasets were analyzed for naive CD4 + T cell-specific genes. Based on these hub genes, Mendelian randomization (MR) analysis, followed by the MR-Bayesian model averaging (

Identification and multi-layered validation of seven diagnostic biomarkers for dilated cardiomyopathy via integrative machine learning, single-cell transcriptomics, and Mendelian randomization.

BACKGROUND: Dilated cardiomyopathy (DCM) is the most common non-ischemic cardiomyopathy and a major cause of heart failure, but disease-specific molecular biomarkers remain limited. This study aimed to identify and prioritize tissue-level, disease-responsive candidate biomarkers for DCM using an integrative multi-omics bioinformatics framework. METHODS: Bulk myocardial transcriptomic data from GSE57338 were used as the discovery cohort, and GSE26887, GSE42955, and GSE79962 served as external microarray validation cohorts. GSE116250 was used for independent RNA-seq validation. Differentially e

Integrative plasma proteomic network analysis identifies physical activity-associated protein modules potentially mediating type 2 diabetes risk.

BACKGROUND: Moderate-to-vigorous physical activity (MVPA) lowers the risk of type 2 diabetes (T2D). Identifying protein modules that mediate this association may help dissect these biological underpinnings, but they remain incompletely characterized. METHODS: We studied 28,842 diabetes-free UK Biobank participants with baseline plasma proteomics. Guideline-adherent MVPA was defined as ≥ 600 metabolic equivalent of task-minutes per week. We first evaluated associations of MVPA with incident T2D and 2910 plasma proteins, followed by mediation analyses of individual proteins. We then identified
