Sporotrichosis is a globally distributed subcutaneous mycosis caused by fungi belonging to the Sporothrix genus. The limitations of current treatment options for sporotrichosis have driven an urgent search for new antifungal drugs. Diphenyl diselenide (PhSe)₂, a stable organoselenium, has promising activity against Sporothrix spp. Since chemical modifications in molecules can enhance their antifungal properties, this study aimed to evaluate the anti-Sporothrix activity of two analogs of (PhSe)₂—p-methoxyl-diphenyl diselenide (BMD) and bis-(3-trifluoromethylphenyl) diselenide (CF3)—both alone a

BACKGROUND: Sporothrix brasiliensis is the principal etiological agent responsible for cat-transmitted zoonotic sporotrichosis, which is currently the most prevalent mycosis in South America-especially in Brazil-and is increasingly being reported in other regions. While itraconazole is the standard treatment, its use is limited by hepatotoxicity, high cost, and emerging reports of reduced susceptibility. Complexes of 1,10-phenanthroline-5,6-dione (phendione) with transition metals have previously shown promise as antifungal agents against bacteria, parasites, yeasts and filamentous fungi. This

Sexually transmitted infections (STIs) remain a major global health concern, contributing significantly to morbidity and facilitating the co-transmission of other pathogens. Recent outbreaks of Monkeypox virus (MPXV) have further underscored the urgent need for broad-spectrum antiviral agents effective against emerging and re-emerging sexually transmissible viruses. We report here the design and synthesis of a series of 3-cyanoquinoline-based Src inhibitors to evaluate their antimicrobial efficacy against sexually transmitted pathogens. Among them, compound 7d demonstrated potent inhibitory ac

Neisseria gonorrhoeae, the causative agent of the second-most prevalent sexually transmitted bacterial disease globally, has been classified as an urgent threat to public health and a high-priority pathogen. Concerningly, N. gonorrhoeae has developed resistance to nearly all FDA-approved drugs. Currently, no approved oral therapies exist, with parenteral administration of ceftriaxone as the only available FDA-approved treatment option for multidrug-resistant gonococcal infections. Yet, ceftriaxone-resistant isolates have now been identified globally, further highlighting the urgent need for th

Antimicrobial resistance (AMR) in Neisseria gonorrhoeae poses a pressing public health threat. Current surveillance programs via antimicrobial susceptibility testing (AST) depend on successfully cultivating the organism via bacterial culture. However, AST is more challenging in extragenital sites and in remote clinical settings where there is a delay between sample collection and testing. This study evaluated whether an enhanced specimen transport system involving direct plating of samples onto selective agar with carbon dioxide enrichment (Bio-BagTM Type C, Becton Dickinson) improved N. gonor

BACKGROUND: Stenotrophomonas maltophilia complex infections are difficult to treat because of intrinsic resistance mediated by the L1 metallo-β-lactamase and L2 serine β-lactamase. Although aztreonam/avibactam is clinically available, aztreonam combined with ceftazidime/avibactam remains an alternative, but comparative pharmacodynamic data under clinically relevant exposures are limited. We compared aztreonam/avibactam and aztreonam + ceftazidime/avibactam under prolonged human-simulated dosing.
METHODS: Three clinical S. maltophilia complex isolates (17061, 17639 and 16852) were studied in 9

Infectious microorganisms are among the major factors that complicate skin and wound infections, highlighting the need for novel antimicrobial agents of natural origin. In this in vitro study, the essential oils (EOs) of four Lamiaceae species (Mentha longifolia (L.) L., Mentha spicata L., Salvia reuteriana Boiss., and Teucrium polium L.) collected from Kashan, Iran, were evaluated for their chemical composition and antimicrobial activity against selected standard microbial strains. The aerial parts were collected from three regions (Cheshme Shahsavaran, Nushabad, and Sefidshahr), and EOs were

Carbapenemase-producing Enterobacterales (CPE) is a global threat. IMP-6, a prevalent carbapenemase in western Japan, is mostly disseminated via CTX-M-2 extended-spectrum β-lactamase (ESBL) co-producing Enterobacterales. However, the existence and characteristics of Enterobacterales harboring blaIMP-6 without blaCTX-M-2 remain unclear. We analyzed the phenotypic and genetic characteristics of clinical blaIMP-6-harboring Enterobacterales isolates, focusing on those lacking blaCTX-M-2. Overall, 220 blaIMP-6-harboring isolates collected from 76 Japanese hospitals between 2014 and 2021 were charac

INTRODUCTION: Bloodstream infections are life-threatening, and the rapid identification of pathogens and resistance genes is essential for the administration of appropriate antimicrobial agents. The BioFire Blood Culture Identification 2 (BCID2) panel on the FilmArray multi-parameter genetic analyzer is a fully automated PCR test that rapidly identifies species and resistance genes. Here, we compared the performance of the Filmarray BCID2 panel (BCID2 method) with the conventional method.
METHODS: Among the blood culture-positive specimens submitted between January 2023 and November 2024, thi

Gonorrhea, caused by Neisseria gonorrhoeae, is a widespread sexually transmitted disease that is becoming resistant to all currently used antibiotics. Therefore, new therapeutics for gonorrhea are desperately needed. Here, we show that a natural product, aerocavin, is highly potent and specific against Neisseria. Aerocavin accumulates in N. gonorrhoeae at high levels and inhibits bacterial RNA polymerase (RNAP) by binding the switch region. Aerocavin resistance mutations evolve in N. gonorrhoeae at a low rate and are absent in clinical isolates. Previously overlooked narrow-spectrum antimicrob

(1) Background: The increasing antibiotic resistance of pathogens is necessitating new therapies that target virulence factors. Virulence factors include biofilm formation, which is a key pathogenic factor involved in bacterial pathogenicity and resistance. (2) Methods: Initially, biofilm formation assays were performed to screen the biofilm formation inhibition effects of gastrodin. A bacterial growth assay was performed to examine the synergistic effects and qRT-PCR was performed to identify the underlying molecular regulatory mechanisms. (3) Results: Gastrodin inhibits biofilm formation by

Bloodstream infections cause significant morbidity and mortality, and rapid identification of the causative microorganisms, along with their antimicrobial resistance profiles, is crucial for appropriate treatment. Molecular diagnostic systems such as the BioFire FilmArray Blood Culture Identification 2 (BCID2) panel provide faster results than conventional culture-based methods. This study evaluated the performance of the BCID2 panel for rapid detection of microorganisms and antimicrobial resistance genes in 50 positive blood cultures collected at a university hospital between January 2023 and

Biofilm-associated Staphylococcus aureus infections remain difficult to treat using conventional antibiotics. Herein, we report the synthesis and biological evaluation of lauric acid- and BDSF-derived N-acyl sulfonamides as antibiofilm and antivirulence agents. Structure-activity relationship (SAR) analysis identified 4-tert-butylphenyl sulfonyldodecenamide (59) as a lead compound with a minimum inhibitory concentration of 5 μg/mL and >60% inhibition of MSSA and MRSA biofilm formation at sub-minimum inhibitory concentration levels. Microscopy confirmed marked reductions in biofilm biomass and

Antibiotic-resistant bacterial infections pose a global health crisis, driving the need for innovative diagnostic approaches. Conventional methods for detecting resistant strains and antibiotic susceptibility testing (AST) are often limited by long assay times, low sensitivity, and operational complexity. Herein, a novel gelatinase-responsive cascade microfluidic chemiluminescence (CL) biosensor was developed for ultrasensitive detection of gelatinase-producing bacteria, with phenotypic AST validated using Staphylococcus aureus (S. aureus) as the model pathogen. Peptide-DNA probes were designe

UNLABELLED: Wound infections are a challenging healthcare burden against which treatment is hindered by the presence of biofilms and the rise in antibacterial resistance. To date, the potential of natural phenolic acids as topical antibacterial agents is under-researched. The aim of this study was to investigate and compare the antibacterial and antibiofilm effects of 5 hydroxycinnamic acids (caffeic acid, chlorogenic acid, p-coumaric acid, ferulic acid, and rosmarinic acid), 4 hydroxybenzoic acids (gallic acid, salicylic acid, syringic acid, and vanillic acid), as well as 2 related compounds

BACKGROUND: Ophthalmic infections, including conjunctivitis and keratitis, caused by Streptococcus pneumoniae may lead to significant ocular morbidity. Rising antibiotic resistance among ocular pathogens has reduced the efficacy of conventional treatments highlighting the need for alternative antimicrobial strategies. This study aimed to evaluate the antimicrobial potential of bacteriocins produced by lactic acid bacteria (LAB) against antibiotic-resistant Streptococcus pneumoniae isolated from clinical ophthalmic samples and to compare their efficacy with conventional antibiotics.

This study introduces an innovative electromagnetic nano-approach to combat high-severity bacterial infections without relying solely on high-dose antibiotics. We investigate the synergistic potential of extremely low-frequency pulsed electromagnetic wave (ELF PEMW) exposure (< 20 Hz) as a physical catalyst to enhance the bio-activity of ciprofloxacin-loaded chitosan nanoparticles (Cipro-C-NPs). This method provides a pivotal alternative for managing Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus in an era of escalating multi-drug resistance. Cipro-C-NPs were synthesized w

We report detection of a blaOXA-23-positive Proteus mirabilis clinical isolate through Antimicrobial Resistance Laboratory Network testing that is closely related to OXA-23 producers identified in Europe. Combined with similar reports internationally, this finding suggests that Enterobacterales might serve as a silent reservoir of carbapenemase genes commonly associated with Acinetobacter species.
