Pyrrolopyrimidine derivatives as dual COX-2/ACE2 inhibitors: design, synthesis, and anti-inflammatory evaluation.

In this study, we report the design and synthesis of a new series of pyrrolopyrimidine derivatives developed as dual-target nonsteroidal anti-inflammatory agents (NSAIDs). The compounds were evaluated for anti-inflammatory properties, cyclooxygenase-1/2 (COX-1/COX-2) inhibitory activity, and angiotensin-converting enzyme 2 (ACE2)-blocking activity in lipopolysaccharide (lipopolysaccharide)-stimulated RAW264.7 cells. Among the synthesized molecules, compounds 5a and 5b showed potent dual inhibitory activity, which was supported by molecular docking and molecular dynamics simulations. These find

Polydatin Attenuates Zearalenone-Induced Hepatic Oxidative Injury through Dual Inhibition of Xanthine Oxidase and Activation of Nrf2 Antioxidant Pathway.

Zearalenone (ZEN) is a mycotoxin that induces hepatic oxidative injury by promoting ROS accumulation and compromising antioxidant defenses. Transcriptomic profiling revealed xanthine oxidase (XOD) as a key mediator. Structure-based molecular docking identified polydatin (PD) from Polygonum cuspidatum as a potent XOD inhibitor with strong binding affinity. Biochemical assays confirmed PD's XOD inhibition. In vitro and in vivo studies demonstrated that PD significantly attenuated ZEN-induced oxidative stress, reduced ROS generation, and improved pathological markers of liver injury, with optimal

Affinity Purification and Molecular Characterization of Angiotensin-Converting Enzyme (ACE)-Inhibitory Peptides from Takifugu flavidus.

An affinity chromatography filler of CNBr-activated Sepharose 4B-immobilized ACE was used to purify ACE-inhibitory peptides from Takifugu flavidus protein hydrolysate (<1 kDa). Twenty-four peptides with an average local confidence score (ALC) ≥ 80% from bounded components (eluted by 1 M NaCl) were identified by LC-MS/MS. Among them, a novel peptide, TLRFALHGME, with ACE-inhibitory activity (IC50 = 93.5 µmol·L-1) was selected. Molecular docking revealed that TLRFALHGME may interact with the active site of ACE through H-bond, hydrophobic, and electrostatic interactions. The total binding energy

Discovery of new inhibitors of resistant Streptococcus pneumoniae penicillin binding protein (PBP) 2x by structure-based virtual screening.

Penicillin binding proteins (PBPs) are involved in the biosynthesis of the peptidoglycan layer constitutive of the bacterial envelope. They have been targeted for more than half a century by extensively derived molecular scaffolds of penicillins and cephalosporins. Streptococcus pneumoniae resists the antibiotic pressure by inducing highly mutated PBPs that can no longer bind the beta-lactam containing agents. To find inhibitors of PBP2x from Streptococcus pneumoniae (spPBP2x) with novel chemical scaffold so as to circumvent the resistance problems, a hierarchical virtual screening procedure w

Whole-Genome Sequencing of Multidrug-Resistant Acinetobacter baumannii Local Isolate and Molecular Dynamics Simulation Studies of a Modified KR-12 Analog Targeting AbaQ and BfmR.

Acinetobacter baumannii (A. baumannii) represents a major threat because of its multidrug resistance, achieved through its ability to control virulence, and its mechanisms of drug efflux resistance. In this study, we used a combined experimental-computational approach to create and evaluate antimicrobial peptides that targeted the two essential pathogenic proteins, BfmR and AbaQ. The genomic analysis of a clinical isolate showed an extensive resistome and virulence profile, which matched high-risk global lineages. This study conducted molecular docking of an experimental AMP (cathelicidin KR-1

Immunoinformatics based designing of a broad-spectrum multi-epitope vaccine against co-infection of human metapneumovirus, respiratory syncytial virus, and influenza A virus.

Co-infections involving human metapneumovirus (hMPV), respiratory syncytial virus (RSV), and influenza A virus (IAV) often exacerbate disease severity in vulnerable populations. Here, we employed a structure-based immunoinformatics approach to design a multi-epitope subunit vaccine targeting these pathogens. The construct incorporated two epitopes each for cytotoxic T lymphocytes (CTLs), helper T lymphocytes (HTLs), and B cells, derived from the fusion proteins of hMPV and RSV, as well as the neuraminidase protein of IAV. These epitopes were linked with an adjuvant and optimized spacers to enh

YXL-13 attenuates virulence in Pseudomonas aeruginosa mutants and clinical isolates by inhibiting quorum sensing regulators LasR and RhlR.

Given the critical role of quorum sensing (QS) in the pathogenicity of Pseudomonas aeruginosa, inhibiting QS is considered a promising alternative to traditional antibiotics for treating P. aeruginosa infections. The QS system of P. aeruginosa comprises two acyl-homoserine lactone (AHL) circuits, LasI/R and RhlI/R, along with a third system, the PQS system. However, due to the frequent clinical isolation of lasR mutants and the avirulent nature of rhlR mutants, targeting both the LasI/R and RhlI/R systems represent a more viable therapeutic strategy. YXL-13, an AHL analog, has been identified

Discovery of a potential CERS2 inhibitor: hit compound identification via structure-based virtual screening and molecular dynamics simulations.

Ceramides, central bioactive mediators of sphingolipid metabolism, critically regulate signal transduction and essential cellular processes. Among six ceramide synthases (CERS1-6), CERS2 preferentially synthesizes very-long-chain ceramides (>C22-24) and holds exceptional promise as a pharmacological target for modulating ceramide levels and composition, with therapeutic potential in metabolic disorders, neurodegenerative diseases, and cardiovascular pathologies. Inhibitors specifically targeting CERS2 remain an urgent unmet need for targeted therapeutics modulating ceramide subclasses. In this
